Description
Technical Field
The invention relates to the field of skin care products and medical dressings, in particular to a plant extract with wound repair effect, a composition, a preparation method and application thereof.
Background
The skin is the first barrier between the body and the nature, and can prevent the invasion of environmental pathogenic microorganisms and the loss of body fluid.
With the development of modern society, people can choose leisure activities such as going to the seaside or walking in holidays, and before outdoor activities, people begin to perform physical sun protection and chemical sun protection in order to prevent sunburn and sunburn. However, no matter how the sun is protected, ultraviolet rays can generate certain stimulation to the skin, so that the moisture loss of the skin is serious, the skin is lack of water, and the skin barrier is damaged; it also causes a series of inflammatory mediators to be released, and the skin becomes reddish, burned, and fever, and if not repaired in time, the skin may also become photoaged in the past. Meanwhile, as people pursue beauty more and more with the development of society, many people have tried various aesthetic treatments such as facial laser, photon skin rejuvenation, micro-acupuncture, tartaric acid skin activation, etc. in order to have admirable skin, not only the use of daily skin care products has been satisfied. Proper medical and cosmetic treatment can solve a plurality of skin problems and make our skin brand new, but medical and cosmetic operations can also bring different degrees of damage to the skin epidermis, so that skin symptoms such as erythema, edema, pigmentation and the like are easy to appear. Therefore, the method becomes an important research hotspot for the repair after solarization and the repair after medical and cosmetic operations.
The skin is in the golden period of recovery after the sun and after the medical and artistic works within 72 hours. compared with a negative control group, the IL-6 secretion level of the positive control group is remarkably reduced (P is less than 0.01), which indicates that the positive control group is effective in the experiment;
the compositions obtained in examples 1 to 3 significantly reduced the IL-6 content, had anti-inflammatory effects, and anti-inflammatory effects, as compared to the negative control group: composition 2> composition 3> composition 1.
As can be seen from the results of the tests of 4 and 5, the compositions obtained in examples 1 to 3 have excellent anti-inflammatory activity, and can reduce the skin stress and rapidly relieve the symptoms of swelling caused by vasodilatation due to allergic reaction.
From the test results of 1-5, the overall effect is as follows: composition 2> composition 1> composition 3. Example 2 the amount of Bidens pilosa was reduced based on example 1, so the anti-inflammatory effect, cell proliferation effect, and cell migration were better than those of composition 1, indicating that increasing the amount of Bidens pilosa did not increase the anti-inflammatory and cell proliferation and migration effects, but rather decreased the effects; example 3 the amount of phoenix-tail fern was reduced based on example 2, the expression of FLG and LOR genes, cell proliferation and cell migration effects were all reduced, and the anti-inflammatory effects were almost identical. Therefore, the composition obtained in example 2 is most effective in repairing skin barrier from the viewpoint of the effect of repair ability and economy.
Test example 3
Lactic acid stinging test was performed in alternative example 2, and the results are shown in FIGS. 10-13
1. Tivi sensitivity test:
the blank group has sensitivity (TiVi) fluctuating around the basic value at different test time points, and the difference has no statistical significance (P > 0.05), which indicates that the skin barrier function of the testee keeps stable and consistent in the test process, so the influence of the skin state of the testee on the experimental result is negligible.
After the negative control group is successfully modeled, the skin sensitivity is slowly reduced along with the time, and the difference has statistical significance compared with the negative control group after modeling (P < 0.05); and the difference significance of the skin sensitivity (TiVi) of the negative control group at the time point of 90min after the use is lower than that of the blank group (P < 0.05), which indicates that the skin (the negative control group) has certain repair capacity.
The composition obtained in example 2 showed a significant decrease in the mean sensitivity (TiVi) (p < 0.05) at the time points of 60min and 90min after application, which is better than that of the negative control group, indicating that the composition can reduce skin sensitivity.
The Tivi bar graph (shown in FIG. 10) in combination with the Tivi700 test image (shown in FIG. 11) further illustrates that the composition obtained in example 2 promotes skin soothing and reduces skin sensitivity at the time points of 40min, 60min and 90min after the lactic acid sting test.
2. The skin erythema (E) test, the results are shown in FIG. 12.
The blank group has skin erythema (E) fluctuating around the basic value at different test time points, and the difference has no statistical significance (P > 0.05), which indicates that the skin barrier function of the subjects keeps stable and consistent in the test process, so the influence of the skin state of the subjects on the experimental result is negligible.
After the negative control group is successfully modeled, the skin sensitivity is slowly reduced along with the time, and the difference has statistical significance compared with the negative control group after modeling (P < 0.05); the skin erythema (E) fluctuated around the basic value, and the difference had no statistical significance (P > 0.05), which indicates that the skin reddening condition remained stable and consistent in the test process.
The skin erythema (E) value of the composition obtained in example 2 is obviously reduced (p is less than 0.005) at the time points of 40min, 60min and 90min after the use, and the composition is superior to a negative control group, which shows that the composition has better capability of improving the skin erythema state;
3. the results of the skin redness (a) test are shown in fig. 13.
The blank group had a fluctuation of redness (a @) around the basal value at different test time points, and the difference was not statistically significant (P > 0.05), indicating that the skin barrier function of the subjects remained stable and consistent during the test, and therefore the influence of the skin condition of the subjects on the test results was negligible.
After the negative control group is successfully modeled, the skin sensitivity is slowly reduced along with the time, and the difference has statistical significance compared with the negative control group after modeling (P < 0.05); the redness (a) fluctuated around the basal value, and the difference was not statistically significant (P > 0.05), indicating that the redness of the skin remained stable and consistent during the test.
The redness (a) value of the composition is obviously reduced (p is less than 0.05) at the time points of 40min, 60min and 90min after the composition is used, and the redness (a) value is better than that of a negative control group, so that the composition has better redness removing capability.
The lactic acid sting test is combined with the test results of 1, 2 and 3, so that the composition has better efficacy of removing red and soothing allergy.
Combining the results of the above tests, it is demonstrated that the composition of example 2 can repair skin barrier, relieve skin inflammation, reduce redness caused by inflammatory reaction of skin, and can be used in the field of after-sun repair skin care products.
It will be understood that the invention is not limited to the examples described above, but that modifications and variations will occur to those skilled in the art in light of the above teachings, and that all such modifications and variations are considered to be within the scope of the invention as defined by the appended claims.
