已知一定数量的辣木种子提取物更特别地用于化妆品领域。在皮肤病学领域,从文件FR2776519中得知,来自辣木种子的蛋白质提取物以其对浑浊水的澄清作用而闻名,对皮肤和粘膜具有润肤、生理调节、保湿、重组、修复和抗污染的作用。在所述文件中,活性成分是具有包括在6500和8800Da之间的分子量的蛋白质,它们是通过从辣木饼的水性提取获得的。
在制药领域,已知一种来自KR20140143655的组合物含有来自辣木叶的水溶性提取物作为活性成分,以治疗或预防癌症。对辣木叶提取物的酶法水解可以用于分离具有分子量包括在6000至8000Da之间的蛋白质。
文件CN 107012190涉及使用辣木种子以在使用微波辐射进行酶法水解后提取油之后从种子蛋白质中获得多肽。获得的产品用于口服并且具有抗癌活性。事实上,这种粉末状的产品被描述为具有减少化疗副作用以及增加肝癌患者食欲或控制白细胞指数和体温的作用。
最后,从文件IN2009CH02906,包含硫代葡萄糖苷与阳离子蛋白组合的辣木饼的水提物可用作抗糖尿病的产品。
所有上述的引用文件都涉及物种辣木的用途;它们中都没有描述来自物种阿拉伯辣木的提取物在药物或皮肤病学领域中的用途。
此外,ABU TARBOUSH等人的文件XP055753092,2005是已知的,其描述了经过10小时的时间段通过酶法水解产生的从去壳阿拉伯辣木种子中提取水解产物。该提取的目的是获得具有高吸油和吸水能力的产品。
使用(1/1)二氯甲烷/甲醇混合物从埃及种子生产的阿拉伯辣木油的提取物在三种人癌细胞系MCF-7(乳腺癌)、Hep-G2(肝细胞癌)和HCT-116(结肠癌)上表现出活性,IC50值为2.92、9.40和9.48μg/mn(ABD el BAKY et al.,2013,Characterization of EgyptianMoringa peregrina seed oil and its bioactivities,International Journal ofManagement Sciences and Business Research,2(7):p.98-108)。作者还通过DPPH(2,2-二苯基1-苦基肼(DPPH(2,2-diphenyl 1-picrylhydrazyl)))、ABTS(阴离子清除能力和还原能力)和抗增殖测定证明了高抗氧化活性。由于获得提取物的方法与本发明的方法不同,因此获得的分子具有非常不同的极性。
ABOU-HASHEM等人(ABOU-HASHEM,M.M.M.et al.,2019,Induction of sub-G0 arrestand apoptosis by seed extract of Moringa peregrina(Forssk.)Fiori in cervicaland prostate cancer cell lines,Journal of Integrative Medicine17:p.410-422)也证明了宫颈(HELA)和前列腺(PC-3)癌细胞系上亚G0期阻滞和凋亡的诱导活性。该方案涉及埃及阿拉伯辣木种子缩小成粉末并用95%乙醇提取,然后溶解在水溶液中,从中研究三种分级的提取物(使用石油醚(PE)、CHCl3和EtOAc的部分)以及水醇提取残留物。该活性在总氯仿级分得到证实,并且归因于饱和和不饱和脂肪酸以及多酚,而没有对机制的具体研究。由于获得提取物的方法与本发明的方法不同,因此获得的分子具有非常不同的极性。
鉴于上述情况,本发明提出要克服的问题是开发基于辣木属和辣木科的物种阿拉伯辣木的提取物的新产品,该产品可用于药学或皮肤病学并且易于使用。
因此,申请人出人意料地开发了一种从阿拉伯辣木种子饼中获得的特定蛋白质水解产物,用于其作为药物的用途,并且特别是用于治疗纤维化疾病(作为弗林(furin)转化酶的抑制剂),治疗炎症、癌症、细菌或病毒类型的感染性疾病以及治疗遗传漂变和与皮肤色素沉着相关的病理。
物种阿拉伯辣木生长在非常干旱的气候中。因此,它耐旱和在极端条件下繁殖的能力使其获得了独特的特征,申请人已经能够通过对阿拉伯辣木种子饼实施特定的提取方法来识别这些特征。根据本发明的蛋白质水解产物已显示具有作为药物的性质,并且已经证明特别是,它具有非常高的弗林转化酶的抑制活性。
弗林转化酶(以下简称弗林)是在不同细胞类型中表达的具有794个氨基酸的1型跨膜蛋白。弗林是一种蛋白质,已被显示参与大量生物过程(BRAUN E.et al.,2019,Furin-mediated protein processing in infectious diseases and cancer,Clinical&Translational Immunology https://doi.org/10.1002/cti2.1073)。它特别地参与伤口的愈合以及其中纤维化是主要组织修复机制或过度纤维化导致病理破坏和组织功能障碍的疾患的治疗(在这方面参见WO 2004/09113文件,其涉及使用转化酶以减少损伤治愈过程中的愈合和治疗纤维化状态中减少纤维化)中纤维化的调节。成人的伤口愈合是一个复杂的修复过程。伤口可能涉及组织或内脏诸如肺、肾脏、心脏、肠、肌腱或肝脏的伤害、损伤或创伤。在组织中的伤口愈合过程通常由皮肤血管损伤引发的止血反应的起始。在此过程中,在愈合过程中形成的结缔组织通常本质上是纤维状的,并且通常作为结缔组织疤痕形成(称为纤维化的过程)。因此,纤维化可以包括肺纤维化、肾纤维化、肝纤维化、皮肤纤维化、眼部纤维化、心脏纤维化和其它各种纤维化状态。本发明的蛋白质水解产物还可以用于治疗由弗林介导的其它病理状态,包括但不限于:高血压、癌症、感染性疾病(细菌的和病毒的)和遗传疾患(例如囊性纤维化(CF))和神经系统变性疾患(在这方面参见WO 2019/215341,其涉及新型弗林转化酶抑制剂或化合物以及含有它们的药物组合物,并且其引用了许多证明抑制剂药物活性的出版物;这些引用特此通过引用并入本说明书)。
通过法兰西共和国政府与沙特阿拉伯王国于2018年4月10日达成的政府间协议,申请人法国AlUla发展局(AFALULA)[French Agency for AlUla]和皇家AlUla委员会(RCU)[Royal Commission for AlUla]发起了一个联合项目,特别地为了发展负责任的农业和当地经济、特别地通过当地生产源自乡土植物的天然产品,以及保护沙特阿拉伯王国AlUla地区的生物多样性和法律。沙特阿拉伯王国自2020年10月8日起成为《名古屋议定书》(NagoyaProtocol)的成员。在起草本专利时,关于《名古屋议定书》将纳入当地法律相关方面的实施法规在考虑之中。因此,在此阶段,沙特阿拉伯王国对本专利申请和《名古屋议定书》没有特定要求。因此,在提交该专利申请之日,没有关于获得遗传资源的合规证书的要求。
发明内容
在第一方面,本发明提供从阿拉伯辣木种子饼获得蛋白质水解产物的方法,包括以下步骤,其中:
a)从成熟的阿拉伯辣木果实中收集未去壳的成熟种子并且进行干燥,以获得小于8%的内部含水率,
b)以使得将油与种子的剩余部分分离的方式,以使得获得包括按重量计小于6%的残油的饼的方式,压榨干燥的种子,
c)碾磨步骤b)中获得的所述饼,
d)将步骤c)中获得的碾磨饼分散在水相中,
e)在pH大于13和包括在16℃至25℃之间的温度下,对步骤d)中获得的水性分散体进行化学蛋白水解约2小时的时间,
f)中和所述蛋白水解以稳定获得的蛋白质水解产物,
g)通过固/液分离回收所述蛋白质水解产物,

在第二方面,本发明涉及来自种子饼的蛋白质水解产物,该种子饼已从成熟的阿拉伯辣木果实中收获并且未去壳,包括分子量包括在1500Da和5000Da之间的氨基酸衍生物、氨基酸、肽和糖肽的主要部分P1,分子量包括在10000和17000Da之间的大约20%的部分P2,分子量大约为23000Da大约20%的部分P3,其中,它是通过在pH大于13的条件下在包括在16℃和25℃之间的温度下化学蛋白水解约2小时的时间获得的,并且其中它是液体并且具有大于1,优选约1.1的密度。
由于其特性,根据本发明的阿拉伯辣木蛋白质水解产物从未在辣木属和辣木科中公开。将证明的是,物种阿拉伯辣木的提取物具有特定的肽谱,其不同于从该属的其他物种,尤其是申请人已经能够展示的物种辣木。

-“主要是氨基酸衍生物、氨基酸、肽和糖肽”是指量超过20%、更优选超过30%和可能高达约40%(重量/重量)干物质的氨基酸衍生物、氨基酸、肽和糖肽,更优选按重量计50%的干物质。
-“有效量”是指获得所需结果,即保证所需的治疗活性所必需的活性分子的数量。
-“蛋白水解”是指通过化学水解将蛋白质分割成肽、寡肽及其碱基片段(氨基酸)以及其残基。
-“局部给药”是指直接施加于其作用点的药物对疾患的确切部位发挥其药理作用。局部给药的目的是限制有效成分从其给药点的扩散,使得将不良影响降至最低。皮肤、鼻腔和呼吸给药、眼部给药、耳部给药、阴道给药和口腔给药是采用的主要局部给药方式。

收集未去壳的种子,即在果实成熟时保留壳,并且优选在壳体开始分裂时保留壳。
将种子干燥以获得小于8%且优选约6%的内部含水率;干燥优选在避光的通风架上进行,优选在露天阴凉处进行。
接下来,用冷压即时研磨干燥的种子,这可用于将油与压榨种子的其余部分(即饼)机械分离。
然后用任何类型的机械磨机诸如锤磨机、连枷磨机、刀磨机、压碎/切碎磨机、球磨机或冲击式碾磨机(beater mill),而且还用任何类型的冷冻磨机(cryomill)对饼进行机械碾磨。
根据步骤d)形成水相的分散和根据步骤e)的蛋白水解在连续搅拌下有利地进行,从而使固体在液体中分散和均质化,因此改善了交换的总表面积,并且因此改善了蛋白水解。
获得具有的密度大于1并且优选为约1.1的液体蛋白质水解产物,其包括在10%和15%之间、优选约12.5%的干物质内容物,所述干物质内容物包括在1%和6%之间的含氮化合物,尤其是比例为0.5%至1.5%、优选约0.8%的挥发性腈衍生物,以及20mg/升的多酚(0.002%)。
根据本发明的方法的优选实施方式,步骤f)的蛋白水解温度大约为22℃。
根据本发明的方法的优选实施方式,步骤g)的固/液分离通过各种方法诸如离心、脱水或过滤进行。
在获得液体阿拉伯辣木蛋白质水解产物的方法的一种实施方式中,蛋白质水解产物通过蒸馏、微滤、超滤和/或纳滤纯化,以便将其浓缩成为目的化合物,该化合物在也被提取的有机物质之上,特别是在也被提取的其它衍生物之上。这些纯化步骤可用于将目的化合物库浓缩在已引用的其他提取的化合物之上。
依据根据本发明的方法的另一种优选实施方式,纳滤以使得分离蛋白质水解产物的3条带或部分的方式进行,包括分子量小于10000Da的带P1,分子量在包括在10000和17000Da之间的带P2和分子量大约为23000Da的带P3。
通过纳滤分离三条带也是有利的,以便通过用于获得蛋白质水解产物的方法获得:

根据本发明的一种实施方式,从阿拉伯辣木种子饼获得的液体蛋白质水解产物例如以获得将水分蒸发掉的阿拉伯辣木种子的固体水解产物的方式通过雾化、冻干或沸腾的方式干燥。干燥可以在有机载体诸如麦芽糊精、环糊精或菊糖存在下,或在无机载体诸如层状硅酸盐、硅酸镁或碳酸盐及其盐存在下进行。
本发明还涉及通过根据本发明的提取方法获得的来自阿拉伯辣木种子饼的蛋白质水解产物。

在本发明的组合物中,根据本发明的蛋白质水解产物以按重量计相对于组合物的总重量的0.0001%至40%的量使用。
在优选的实施方式中,根据本发明的蛋白质水解产物以相对于组合物的总重量按重量计0.001%至10%,更优选相对于组合物的总重量按重量计0.01%至5%的量使用。
最后,在第五方面,本发明涉及药物组合物或皮肤病学组合物,其用作治疗以下疾病的药物:

实施例
实施例1:从阿拉伯辣木饼开始制备根据本发明的植物蛋白质水解产物
阿拉伯辣木(Forssk.)的种子将Fiori干燥以获得小于8%且优选约6%的内部含水率,然后用无头螺杆机械压榨机以将油从种子的剩余部分中分离出来的方式压榨,以便一方面获得初榨油,另一方面获得饼。然后将饼以预切挤出物的形式分离成1至2cm的块,在其上进行提取。
所用的起始原料如下。

a)制备一种浓缩的1摩尔的强碱诸如氢氧化钠、氢氧化钾或氢氧化钙的溶液,特别地或具有1摩尔浓度的强碱,但优选氢氧化钠的水性混合物;这种强碱性溶液具有在13和14之间的pH,

上述干提取物是通过基于蒸发之前和之后液体提取物中存在的质量的重量法获得的。
实施例2:根据本发明的阿拉伯辣木蛋白质水解产物作为弗林转化酶(称为弗林) 抑制剂的作用
本研究的目的是评估根据实施例1获得的来自阿拉伯辣木种子饼的蛋白质水解产物的抑制活性。
方案:该研究是使用重组人弗林进行的,其催化特定荧光参比底物的裂解。
使用100nM癸酰基-Arg-Val-Lys-Arg-CMK作为用于弗林活性的参比抑制剂。
将弗林在不存在(对照)或存在参比产品或增加的测试化合物浓度的情况下,在环境温度下预温育10分钟:
“阿拉伯辣木蛋白水解物”;0.02;0.2和2%(v/v)。
在预温育步骤结束时,加入弗林底物,并且将实验条件再次在远离光照的环境温度下温育5分钟。所有实验一式三份进行。
化合物的制备:
将测试的阿拉伯辣木蛋白质水解产物直接溶解在测定缓冲液中然后稀释以获得上述测试浓度。
评价方案
在加入底物后,通过读取485nm/535nm处的荧光对荧光弗林底物的裂解监测5分钟。
统计学
结果以RFU(相对荧光单位)+/-S.D.(标准偏差)表示。
通过t检验评价“对照”和“参比产品”组之间观察到的差异的统计显著性(p<0.001)。
通过单因素ANOVA,然后Holm-Sidak检验评价“对照”或“参比产品”和“测试化合物”组之间观察到的差异的统计显著性(p<0.05)。
在100nM下测试的弗林的参比抑制剂,称为癸酰基-Arg-Val-Lys-Arg-CMK,显著抑制了97.9%的弗林活性(p<0.001)。
这个结果是预期的并且验证了研究。
下面给出了与获得的碱活性相比的弗林酶活性的结果。

结论:在2%的浓度下,根据本发明的阿拉伯辣木蛋白质水解产物能够抑制98.8%的弗林转化酶的活性。在超过0.2%的浓度下,根据本发明的阿拉伯辣木蛋白质水解产物能够抑制达26.8%的弗林转化酶的活性。
实施例3:根据本发明的阿拉伯辣木蛋白质水解产物在抑制组蛋白脱乙酰酶 (HDAC)和sirtuin I酶中的作用
本研究的目的是证明根据本发明的阿拉伯辣木蛋白质水解产物对HDAC和sirtuinI酶的抑制活性,这些酶通过调节染色质的缩合/脱缩参与控制遗传漂变,其提供或阻断对DNA携带的基因的获取。将HDAC和sirtuin I的缓冲溶液与底物在37℃下反应超过20分钟,并将其转化以形成化合物,在37℃下温育10分钟后,该化合物在显色剂存在下显色。因此,可以通过测量在405nm处的吸光度来评价sirtuin的最大脱乙酰活性。将根据本发明的阿拉伯辣木蛋白质水解产物或参比产品“曲古抑菌素A(STA)抑制剂1μM”同时作为酶底物与sirtuin溶液在37℃下接触20分钟,并通过添加显色剂对通过酶转化的底物染色。然后通过测量在405nm处的吸光度来评价在存在活性成分下HDAC和sirtuin I的脱乙酰活性。在不存在活性成分下,即仅在存在HDAC和sirtuin I酶的底物下,将此活性的调节表达为HDAC和sirtuin I的最大活性的抑制百分比或激活百分比。
方案:在不存在(对照)或存在参比产品或渐增浓度的待测试产品下,将sirtuin酶溶液在其底物中温育20min。将本发明的阿拉伯辣木蛋白质水解产物在以下浓度下进行测试:2%;1%;0.1%(V/V)。在温育期结束时,通过使用显影剂溶液(在37℃下10min)染色来显示并通过测量在405nm处的反应介质的吸光度来评价具有和不具有测试产品或参比产品的sirtuin酶的活性。对于每个测试浓度,通过使用以下公式计算测试产品对组蛋白脱乙酰酶和sirtuin I酶的脱乙酰活性的调节。

如果结果为阴性,则将百分比表示为酶促反应的抑制;如果结果为阳性,则将百分比表示为酶促反应的激活。以下给出了组蛋白脱乙酰酶(HDAC)的抑制结果。

结论:在2%时,根据本发明的阿拉伯辣木蛋白质水解产物表现出对HDAC的显著抑制;这种抑制揭示了促进皮肤细胞自我保护免受遗传漂变的能力。因此,该提取物似乎可用于对抗皮肤表面上最常见的遗传漂移之一,即纤维化,其通过肉的“肿块”(纤维化隆凸)的出现来表现。提取物可以有利地干扰皮肤表面的纤维化现象。
实施例4:根据本发明的阿拉伯辣木蛋白质水解产物对抑制内皮素1(ET-1)作用的 效果
内皮素是一种源自内皮细胞的激素肽,能够通过其受体作用于各种细胞和组织。例如,已知内皮素会导致平滑肌血管细胞和其他细胞中钙的细胞内浓度增加(OHBA T.等人,WO 2012/081370)。
近年来有报道称,内皮素1型(ET-1)是通过直接和间接作用来收缩平滑血管和非血管肌肉细胞的生物活性因子。据认为,内皮素作用的增加为外周部位中、肾脏中和大脑中的血管提供了持续的血管收缩,并且可能是多种疾病诸如高血压、心肌梗死、脑血管意外、急性肾功能不全,雷诺综合征(Raynaud sydrome)、动脉粥样硬化、哮喘和前列腺癌的起源(MIYAGAWA K.&Emoto N.et al.,2014,Current state of endothelin receptorantagonism in hypertension and pulmonary hypertension,Therapeutic Advances inCardiovascular Diseases,vol.8(5)202-216;SCHINZARI F.et al.,2018,IncreasedEndothelin-1-Mediated Vasoconstrictor Tone in Human Obesity:Effects of GutHormones,Physiological Research 67suppl.1:S69-S81)。来自具有相似结构的内皮素家族的三种类型的肽存在于动物中,包括人类(KADONO,S.et al.,2001,The Role of theEpidermal Endothelin Cascade in the Hyperpigmentation Mechanism of LentigoSenilis,Journal of Investigative Dermatology 116 4,p.571-577;ANONYMOUS,2006,American Society for Biochemistry and Molecular Biology,New CosmeticsHandbook,p.527-529)。所有这些肽都具有血管收缩作用和血管抑制作用。

其目的是在暴露于根据本发明的阿拉伯辣木蛋白质水解产物24h后测定人微血管内皮细胞中的1型内皮素。
方案:人微血管内皮细胞由PELOBiotech公司提供,并根据供应商的生产程序在96孔板中培养。这意味着允许提取物在不同浓度在80%汇合下作用于内皮细胞24小时,然后借助ELISA PicoKine(EDN1)试剂盒对细胞上清液中的内皮素1进行量化。进行了先前的生存力测试,以对在测定内皮素1时将使用的无毒剂量进行限定。将培养基中未处理的细胞形成阴性对照。生存力测试中的阳性对照为0.5%SDS。所有条件均在培养基中准备并且随后将细胞在36.5℃/5%CO2下温育24小时。
a)将测试溶液施用至内皮细胞:
将测试产品与在96孔板中的处于亚汇合的内皮细胞接触。对每种浓度的3个孔进行测试。将板在36.5℃/5%CO2下温育24小时±1小时。
b)生存力测试:
与产品一起温育后,用MTT方法评价细胞的细胞生存力。温育24小时后,将上清液回收并且储存在-20℃下用于测定。然后用200μL的PBS冲洗孔一次。向每个孔中添加50μL的0.5mg/ml MTT溶液:在36.5℃/5%CO2下温育3小时。向每个孔中添加100μL的异丙醇。均质化后,在550nm处记录吸光度。对于每种条件,细胞的光密度平均值与阴性对照的光密度平均值的比率确定生存力比率。
c)内皮素1测定:
测定是在ELISA试剂盒的帮助下进行的。以下给出了对内皮素作用的抑制的结果。

结论:在处理结束时进行的生存力测试没有示出对测试的浓度的任何毒性作用。
内皮素1测定在无毒浓度的细胞上清液中进行。借助于ELISA试剂盒测定每种条件下的内皮素1的量。
对于基础水平对照细胞,该值为大约134.94pg/mL。对于用不同浓度的水解产物处理的细胞,基础水平降低47.09pg/mL(用2%的根据本发明的提取物),并且降低17.58pg/mL(用1%的根据本发明的提取物)。这证明了1%的根据本发明的提取物的非常显著的抑制作用,对1型内皮素的产生具有大约13%的抑制作用,并且2%的根据本发明的提取物具有高达34.90%的抑制作用。
内皮素及其特异的剂量依赖性抑制的结果表明,根据本发明的蛋白质水解产物由于其显著降低内皮素的能力而具有抗血管生成和抗纤维化作用。
下面描述了用于评估实施例12中描述的P1、P2、P3和复合物P1+P2的补充研究,用于内皮素的生产:正常人内皮细胞的细胞测试模型。
如实施例12所述,从根据本发明的水解产物中分离出3条带或蛋白质部分,其特征为它们的质量P1(分子量小于10000Da)、P2(分子量包括在10000和17000Da之间)和最后P3(分子量约为23000Da)。这些部分在下文中也称为“提取物”。

结论:称为“提取物P1”和“提取物P2”的化合物对1型内皮素的调节没有显著作用,只有具有根据本发明的提取物的0.3%的“提取物P3”显著降低了单层培养物中正常人内皮细胞释放到培养基中的内皮素-1,其抑制得分为52.3%。当P1和P2组合时,未观察到协同效应。
提取物P3特别地表现出抗癌作用(BAGNATO A.et al.,2011,Role of theendothelin axis and its antagonists in the treatment of cancer,BritishJournal of Pharmacology,163:220-233)。
实施例5:根据本发明的阿拉伯辣木蛋白质水解产物对干细胞保护的作用
成人组织,包括皮肤表皮、胃肠上皮和造血系统,具有非常高的细胞更新水平。维持组织稳态的生理过程归因于在器官更新中维持恒定数量的细胞。胚胎干细胞(ESC)对于维持和再生皮肤组织至关重要。
表皮是从胚胎表面的外胚层发育而来的。它从神经胚形成之后覆盖胚胎的单层非特异性祖细胞开始,并且发育成基底表皮层。基底表皮层富含ESC(表皮干细胞)。事实上,这一层的细胞产生所有的表皮结构,包括分层表皮(也称为毛囊间表皮)和表皮附件,诸如毛囊、皮脂腺和汗腺。下层真皮主要来源于外胚层下的中胚层。中胚层是间充质干细胞的主要来源,间充质干细胞产生生成胶原蛋白的成纤维细胞、下层脂肪细胞和皮肤的免疫细胞。

方案:从62岁的供体获得人角质形成细胞。为了进行实验,将角质形成细胞以单层培养,直到它们达到80%的汇合。
然后按照Goodell,M.等人(1996,Hoescht 33342HSC staining and stem cellpurification protocol,Journal of Experimental Medicine 183,p.1797-806)描述的方法将细胞培养物富集在表皮干细胞中。
参比产品:本研究使用1μM槲皮素作为参比产品。槲皮素购自于Sigma Aldrich。

端粒酶是核糖核蛋白,它催化将端粒重复添加到端粒末端。端粒是长段的重复序列,其覆盖染色体末端并且已知稳定染色体。在人类中,端粒通常为7至10kb长,并包括序列-TTAGGG-的多个重复。

方案:从44岁的供体获得人成纤维细胞。为了进行实验,使用第2代和第5代细胞。成纤维细胞在没有(对照)或存在阿拉伯辣木蛋白质水解产物的测试浓度增加的情况下连续培养3代:0.01%;0.1%和0.5%(V/V)。

在温育结束时,将细胞胰酶消化。通过专用的DNA提取试剂盒从细胞中提取DNA。通过nanodrop对DNA进行定量。
通过定量PCR(q-PCR)测量端粒长度。对于每个样品,使用SCR(单拷贝参比)基因作为参比基因,通过相对定量测量端粒长度的变化。对于每个样品,使用识别和扩增端粒序列的一组端粒引物进行q-PCR,并且使用识别和扩增人17号染色体上100bp区域的一组SCR引物进行第二q-PCR并充当用于数据归一化的参比。
结果以对应于相对于第2代细胞端粒的长度的相对单位表示(平均值±S.D.)。借助Student检验评估第2代和第5代与“对照”的显著水平(*:p<0.05)。通过单因素方差分析(单因素ANOVA)、随后的Holm-Sidak检验独立评价每种产品的“对照”和“测试化合物”之间的显著性水平(*:p<0.05)。

实施例8:根据本发明的阿拉伯辣木蛋白质水解产物对调节DKK1和DKK3测定的作
用

(ACE2)的作用-抑制剂作用的无细胞研究。

实施例10:通过防止“刺突SARS-CoV-2假型冠状病毒”的渗透来评估水解提取物在
抑制纤维素中弗林的抗感染作用:在“SARS-CoV-2假型慢病毒”存在的情况下对人HEK细胞
的纤维素模型。
本评价使用了两种主要组分。一方面,组成型表达全长人ACE2的稳定的重组克隆HEK293细胞(Genbank#NM_021804.3),具有经流式细胞术证实的ACE2的表面表达。另一方面,刺突SARS-CoV-2假型慢病毒是用刺突SARS-CoV-2(Genbank登录号QHD43416.1)作为包膜糖蛋白而不是传统使用的VSV-G产生的。这些假病毒粒子还含有由CMV启动子引导的萤火虫荧光素酶基因;因此,可以通过荧光素酶报告体的活性以实用的方式测量由峰值介导的细胞进入。刺突SARS-CoV-2假型慢病毒可用于筛选生物安全2级设施中的应用。

实施例12中鉴定的带P1、P2、P3和复合物P1+P2对弗林转化酶的评价:无细胞测试模型。
对鉴定的带进行的研究并未证明用P1、P2、P3和P1+P2测试的所有浓度下对弗林转化酶有任何抑制作用。

实施例11:从阿拉伯辣木种子中获得的不同制剂对弗林转化酶的抑制的比较试验

方案:在使用之前将乙醇(96%)中的阿拉伯辣木提取物和阿拉伯辣木蛋白质水解产物避光储存在+4℃。阿拉伯辣木油在环境温度下储存在黑暗区域。
参比产品:使用100nM癸酰基-Arg-Val-Lys-Arg-CMK作为用于弗林活性的参比抑制剂。
温育方案:将弗林在不存在(对照)或存在参比产品或渐浓度的测试化合物的情况下,在环境温度下预温育10分钟:
-0.3%、1%和3%(V/V)的阿拉伯辣木油。
-0.02%、0.2%和2%(V/V)的在乙醇(96%)中的阿拉伯辣木提取物。
-0.02%、0.2%和2%(v/v)的根据本发明的阿拉伯辣木蛋白质水解产物。在预温育步骤结束时,加入弗林底物,并且将实验条件再次在避光的环境温度下温育5分钟。所有实验一式三份进行。
化合物的制备:将96%乙醇中的阿拉伯辣木提取物和阿拉伯辣木蛋白质水解产物直接溶解在测定缓冲液中,然后稀释以达到测试浓度,如上所述。将阿拉伯辣木油溶解在测定缓冲液中的0.05%吐温20溶液中至3%。然后将溶液稀释以获得上述浓度。

实施例12:阿拉伯辣木蛋白质水解产物-蛋白质带的分离电泳凝胶色谱
在含有十二烷基硫酸钠的聚丙烯酰胺上进行凝胶电泳,称为SDS-PAGE电泳。它是一种技术,包括在电场的影响下通过SDS使负电荷饱和,使变性蛋白质在聚丙烯酰胺凝胶中迁移,从而使它们能够分离。这是一种变性技术,通过使用带负电荷的离子洗涤剂(SDS)解离非共价蛋白质复合物。这种洗涤剂通过疏水键无选择性地与两种氨基酸结合。因此,该技术可用于分析蛋白质并且根据其分子量的进行分离。

1.确定对细菌菌株鼠伤寒沙门菌(Salmonella typhimurium)(TA 100)的致突变 活性–反向细菌突变测试
测试分3个主要阶段进行:
-进行初步实验,以评价待测试元素的细胞毒性,并为随后实验选择剂量范围,
-第一遗传毒性实验(测试1),具有和不具有代谢活化,在初步研究规定的剂量范围内,在最小凝胶上直接掺入测试系统和测试(或对照),

结果的表达和解释
许多准则可以用于确定结果是否为阳性,尤其是与测试项的剂量相关的回复体的数量增加,或在一种或多种浓度下,在具有或不具有代谢活化下,回复体数量的可再现增加。
-如果在验证步骤的最后,在具有和/或不具有代谢活化的5种菌株中的一种或多种上以可再现的方式获得剂量-作用关系,则将测试元素视为是致突变的。当回复体的数量对于菌株TA98、TA100和TA102至少等于自发回复率的两倍(R≥2)以及对于菌株TA1535和TA1537至少等于自发回复率的三倍(R≥3)时,才将给定浓度视为致突变性。
-如果在试验1和试验2结束时,在有和没有代谢活化下,并且前提是致突变作用已被证实与测试浓度的毒性有关,回复体的数量仍然小于已测试的菌株TA98、TA100和TA102的所有浓度的自发回复率的两倍(R<2),并且小于菌株TA1535和TA1537的自发回复率的三倍(R<3),认为测试元素是非致突变的。
初步研究未证明测试元素有任何细胞毒性;因此,该浓度范围用于遗传毒性试验1。
由于测试1获得的结果,决定对测试2使用相同的稀释范围。回复体的分析示出:
-没有观察到细胞毒性作用,
-在具有和不具有代谢活化下,没有测试提取物的浓度示出比率R对于TA98、TA100和TA102大于或等于自发回复率的至少两倍,或对于TA1535和TA1537,大于或等于自发回复率的三倍,
-无论测试体系或测试条件如何,均未观察到剂量反应。
鉴于在本研究期间获得的结果,根据实施例1的蛋白质水解产物可被认为既不具有致突变活性也不具有促致突变活性。
2.体外3T3 NRU光毒性测试
测试的原理是在存在和不存在非细胞毒性剂量的UVA下,根据实施例1的蛋白质水解产物对培养物中细胞的细胞毒性的比较。在具有或不具有UVA辐照下,在用参比元素和根据本发明的阿拉伯辣木蛋白质水解产物处理后24h,通过活体染料:中性红,辅助确定细胞生存力来评价细胞毒性。使用的细胞是Balb/c 3T3克隆31系(ATCC–CCL163)的小鼠胚胎成纤维细胞。阳性对照为氯丙嗪溶液(CAS号:69-09-0)。阴性对照是测试和参比提取物的稀释液(缓冲盐+/-1%溶剂)。在存在或不存在UVA下,在8个浓度下,在至少四个培养孔/研究的浓度中测试阿拉伯辣木蛋白质水解产物。将成纤维细胞进行胰酶消化并且在两个96孔培养板中接种在完全培养基中的100μL的2×105个细胞/mL(即2x106个细胞/孔)的细胞悬浮液。
将温育的板在37℃,/5%CO2下在烘箱中温育24小时。在温育结束时,检查细胞垫(cell mat)的半汇合。在将它们沉积在细胞上之前制备稀释液。测量最高浓度的pH;它包括在6.5和7.8之间。去除培养基;每个孔都已用在环境温度下保持的150μL的PBS仔细冲洗,然后用100μL的每种提取物稀释液或参比稀释液进行处理。将培养板在37℃和5%CO2下在暗处温育1h±5分钟。辐照是在BIO SUN太阳辐照系统(Vilber Lourmat RMX3W)的帮助下进行的。BIO SUN是借助可编程微处理器控制UV辐照的系统。该系统持续监测UV光的发射。当递送的能量等于程序编入的能量时,辐照自动停止。使用校准的光谱辐射计在250至700纳米的波长范围内测量测试装置的光谱辐照度。
将2块板中的一块在环境温度下用其盖(cover)辐照,并且另一块板被保护免受UVA,并在辐照期间保持在环境温度。辐照后,抽取处理介质并冲洗细胞。接下来,小心添加100μL的完全培养基,并将板在37℃和5%CO2下温育18至22h。第二天,通过使用相差显微镜观察来评价细胞生存力(生长、形态、单层完整性)。去除培养基,并且在用100μL的染色溶液处理之前,每个孔已经被冲洗并保持在环境温度。在相同条件下,将板放回培养箱3h。去除染色溶液并冲洗细胞,然后去除冲洗溶液,并向每个孔中添加150μL的解吸(desorption)溶液。摇动板,直到晶体完全溶解。在450nm处测量吸光度值。
测试验证:
在暴露于增加剂量的照射后通过评估细胞的生存力,在所有大约10代中监测细胞对UVA的敏感性。以测试中使用的密度培养细胞。第二天以2.5至9J/cm2的剂量对它们辐照,并且一天后借助于NRU测试确定细胞生存力。如果细胞在5J/cm2下的UVA辐照后生存力是或大于保持在黑暗中的参比的生存力的80%;则细胞符合品质准则;在最高剂量9J/cm2的UVA下,生存力必须至少为保持在黑暗中的参比的生存力的50%。
结果:
阴性对照具有的吸光度为0.4或更高。阳性对照氯丙嗪在存在UVA下具有的CI50值为在0.1和2μg/ml之间,以及在不存在UVA下为在7和90μg/ml之间。这些结果使测试得以验证。不能估计在存在或不存在UVA下,提供50%细胞死亡的根据实施例1的阿拉伯辣木蛋白质水解产物的浓度。死亡率从未达到50%。不能估计在存在或不存在UVA下,产生50%细胞生存力的阿拉伯辣木蛋白质水解产物的浓度。生存力始终大于50%。
结论:在所采用的实验条件下,根据本发明的阿拉伯辣木蛋白质水解产物可以被认为是无光毒的。
3.根据对于SIRC细胞系的中性红的浸出方法,通过体外细胞毒性研究评估眼部刺 激性可能性
这项体外研究基于通过借助中性红浸出技术确定细胞单层上导致50%细胞死亡(IC50)的浓度来评价阿拉伯辣木蛋白质水解产物的细胞毒性。使用的细胞是SIRC,兔角膜成纤维细胞(ATCC–CCL60),其不含支原体。
将阿拉伯辣木蛋白质水解产物用生理血清稀释至25%和50%。将成纤维细胞进行胰酶消化并且在两个24孔培养板中接种在完全培养基中的1mL的量的2×105个细胞/mL的细胞悬浮液。将温育的板在37℃和5%CO2下在烘箱中温育。在温育结束时,检查细胞垫的汇合。在完全培养基中以0.5mg/mL的浓度制备染色溶液。去除培养基;在每个孔中沉积1mL的染色溶液。将板放回在37℃和5%CO2/下的培养箱中3h±15分钟。在该接触时间后,去除染色溶液,并用每孔1mL的完全培养基替换。在与提取物或参比接触之前,将板在环境温度下保持至少30分钟,以稳定体系。将每个孔用2mL的PBS冲洗,保持在环境温度下,并且然后将500μL的阿拉伯辣木蛋白质水解产物或参比的每种稀释液沉积与细胞垫接触。接触时间为60秒(阳性对照为30秒)。逐孔进行处理,在沉积阿拉伯辣木蛋白质水解产物或参比的那一刻开始计时。在整个处理期间手动摇动板。55秒(或阳性对照为25秒)后,抽取稀释液。在精确的60或30秒时,进行5次依次冲洗(5x2mL PBS,保持在环境温度下)。每次冲洗后抽取上清液,并且最后一次冲洗后,在等待显示阶段时,保持孔中没有介质。完成培养板的处理之后,在每个孔中沉积1mL的染色溶液。将板摇动约15分钟,直到获得均匀的染色。将每个培养孔获得的溶液取出并分入96孔板的2个孔中,即150μL/孔。
结果:
提供50%细胞死亡的阿拉伯辣木蛋白质水解产物的浓度评价为>50%。阿拉伯辣木蛋白质水解产物产物在50%下的细胞死亡百分比被评价为17%。
结论:在所采用的实验条件下,根据本发明的阿拉伯辣木蛋白质水解产物可以被归类为:细胞毒性可忽略不计。
4.在皮肤病学控制下,在包扎疗法下单次施用48小时后评价根据本发明的阿拉伯 辣木蛋白质水解产物的皮肤相容性
此研究的目的是通过在手臂的前外表面进行48小时的表皮(epicutaneous)测试来评价阿拉伯辣木蛋白质水解产物的皮肤相容性程度;并且通常评价阿拉伯辣木蛋白质水解产物保持皮肤良好状态的能力。将来自18至65岁,不具有干性皮肤也不具有敏感皮肤并且处理区域没有任何皮肤病学病变的10名健康的女性或男性志愿者将纳入研究中。在去除敷料后初始施用30至40分钟后,48小时评价了阿拉伯辣木蛋白质水解产物的皮肤相容性,该阿拉伯辣木蛋白质水解产物以具有5%的根据实施例1的阿拉伯辣木蛋白质水解产物和95%丙二醇/山梨糖醇混合物的洗剂形式制备。根据以下等级对皮肤反应(红斑和水肿)给予0至3的得评分:

1)眼睛和皮肤刺激测试为阴性
2)光毒性测试为阴性
3)致突变性测试为阴性。
已经证明根据本发明的阿拉伯辣木蛋白质水解产物的安全性,并且它是理想的用于大规模皮肤病学使用,而不受目标群体的限制。

19.根据权利要求15至18中任一项所述的组合物,用于其用作治疗纤维化疾病和治疗炎症的药物的用途,其特征在于,其包括作为活性剂的有效量的根据权利要求10或11,优选根据权利要求10所述的蛋白质水解产物。
20.根据权利要求15至18中任一项所述的组合物,用于其用作治疗癌症的药物的用途,其特征在于,其包括作为活性剂的有效量的根据权利要求12所述的蛋白质水解产物。
21.根据权利要求15至18中任一项所述的组合物,用于其用作治疗细菌或病毒类型的感染性疾病,并且特别是为了抑制刺突COV2蛋白的药物的用途,其特征在于,其包括作为活性剂的有效量的根据权利要求6或7所述的蛋白质水解产物。
22.根据权利要求15至18中任一项所述的组合物,用于其用作治疗遗传漂变和治疗与皮肤色素沉着相关的病理的药物的用途,其特征在于,它包括作为活性剂的有效量的根据权利要求13所述的蛋白质水解产物。
