Description

Traditional Chinese medicine peptide oral liquid for relieving fatigue and preparation method thereof
Technical Field
The invention relates to the technical field of traditional Chinese medicine preparation, in particular to a traditional Chinese medicine peptide oral liquid for relieving fatigue and a preparation method thereof.
Background
Fatigue, a physiological phenomenon, means that the physiological process of the body cannot last for its function at a certain level and/or cannot maintain a predetermined exercise intensity. With the acceleration of modern life rhythm and the increase of working pressure, the bodies of many people are in sub-health state, namely, the bodies are free from organic lesions, but functional disorders, such as fatigue, poor spirit, low immunity, easy to cause common diseases, natural aging acceleration and other symptoms, are in the early stages of cardiovascular and cerebrovascular diseases or other chronic diseases, are generated. If the sub-health state is not improved in time, the problems of abnormal regulation of the nervous system and the endocrine system of the organism and the like can be caused, and finally, organic lesions are generated.
Along with the continuous improvement of living standard, people's health consciousness and health preserving concept are stronger and stronger, and the disease prevention is changed from the past disease treatment to the disease prevention so as to achieve the purpose of obtaining healthy body. According to data statistics, more than half of people in China are in the state of chronic fatigue syndrome, and about 70% of people in urban population are occupied. Therefore, it is necessary to provide a medicine or health food effective in relieving fatigue. However, since fatigue itself is not and does not directly cause a serious disease in the relevant population, the necessity or meaning of treatment by a drug is not great from the viewpoint of pharmaceutical economics, and there are some adverse effects of the drug itself more or less. Therefore, in recent years, a health food for relieving fatigue has been attracting attention of researchers. Although there are many health foods for relieving fatigue on the market, these health foods generally have the defects of insignificant effect or single effect. Therefore, it is necessary to develop a safe and effective health food for relieving fatigue.

Each test group had 10 mice, and the example 1 group had three dose groups (1 low dose group, 1 medium dose group, 1 high dose group).
Test sample:
the products prepared in the embodiment 1 are designed into a low dose group (0.42 mL/Kg), a medium dose group (0.83 mL/Kg) and a high dose group (2.50 mL/Kg), the doses of the products are 5 times, 10 times and 30 times of the recommended amount of a human body respectively, and the products are subjected to a gastric lavage method once a day; comparative example 1 uses the product of comparative example 1 of the present invention, comparative example 2 uses the product of comparative example 2 of the present invention, and the dosages of the comparative example 1 and the comparative example 2 are: low dose (0.42 mL/Kg), medium dose (0.83 mL/Kg) and high dose (2.50 mL/Kg); the negative control group was given an equal volume of distilled water and was continuously gavaged for 30 days.
Test method and results:
1. weight bearing swimming test
After the last administration of the test object for 30min, the mice were put in a swimming box to swim with a water depth of about 35cm and a water temperature (25 soil 1) DEG C, and the roots of the rat tails were loaded with 5% of the weight of the lead skin. The time from the start of the swimming to death of the mice was recorded as the swimming time (min) of the mice. And the body weight of the mice was measured before the start of the test and after the end of the test.

The statistical results of the combination of example 1, comparative example 1 and comparative example 2 show that the sample of example 1 has an effect of significantly prolonging the load swimming time of mice compared with other samples.
2. Serum urea nitrogen and lactic acid content determination
After the last administration for 30min, the mice swim for 90min in water with the temperature of 30 ℃ without load, draw out eyeballs for blood sampling after resting for 60min, place the eyeballs in a refrigerator with the temperature of 4 ℃ for 3h, centrifuge for 15min at 2000r/min after blood coagulation, and take serum to measure urea nitrogen.

Serum urea levels of mice in the medium and high dose groups were very significantly different (P < 0.01) compared to the negative control group. The statistical results of the combination of example 1, comparative example 1 and comparative example 2 show that the samples of the examples can delay the occurrence of fatigue or accelerate the rapid elimination of fatigue.
3. Determination of liver glycogen
After the last administration for 30min, the mice are sacrificed, the livers are rinsed by normal saline and then sucked by filter paper, tissue homogenate is taken, centrifugation is carried out for 15min at 3000r/min, supernatant is taken, and the content of liver glycogen is measured.
