Abstract
The invention provides a fermentation type small molecule peptide mixed solution for regulating blood sugar, a preparation method and application thereof, and relates to the technical field of small molecule peptide mixed solutions. The invention utilizes the common fermentation of tuckahoe bacterial liquid, compound probiotics, mulberry leaf, sealwort, dendrobium, schisandra, astragalus, ginseng, yam and medlar, the zymolysis of the zymotic fluid is carried out by cellulase, pectinase and papain, the fermentation fluid is ultrafiltered to obtain the fermentation type small molecular peptide mixed solution which is rich in a plurality of bioactive factors such as polypeptide, oligosaccharide, saponin, flavone and the like, and the various bioactive factors have synergistic effect. According to the invention, by selecting and proportioning various components in the fermentation liquor, the small molecular peptide mixed liquor capable of regulating intestinal flora, improving immunity and blood circulation is finally obtained, and the healthy level of blood sugar can be maintained on the whole. Therefore, the fermented small molecule peptide mixed solution can be used for preparing food or medicine for maintaining healthy blood sugar level.

(3) and intercepting components with the molecular weight of less than 2000Da in the enzymolysis liquid to obtain the fermentation type small molecular peptide mixed liquid.
Preferably, the mass ratio of the poria cocos to the ginseng, the Chinese yam, the sealwort, the medlar, the dendrobium and the mulberry leaf in the poria cocos bacterial liquid in the step (1) is (150-200): (3-10): (10-20): (10-15): (15-20): (10-20): (10-15).

The mass ratio of the poria cocos to the ginseng, the Chinese yam, the sealwort, the wolfberry fruit, the dendrobium and the mulberry leaf in the poria cocos bacterial liquid is preferably (150-200): (3-10): (10-20): (10-15): (15-20): (10-20): (10-15), more preferably (150-190): (3-5): (10-15): (10-14): (10-16): (15-18): (10-15): (10-13).
The composite probiotics are preferably purchased from Shandong Jungle Biotech limited; and the adding mass of the bacterium powder of the composite probiotics is 1 percent of the mass of the fermentation liquor.
The preparation method of the poria cocos bacterial liquid preferably comprises the following steps of crushing poria cocos, mixing with water according to the ratio of 1: 10 by mass ratio. According to the invention, the poria cocos bacterial liquid is used for fermenting various medicinal and edible homologous substances, the materials are simple to obtain, the effect is obvious, the poria cocos bacterial liquid is easy to prepare, and the fermentation is easy to succeed; and the tuckahoe can promote diuresis, eliminate dampness, invigorate spleen and calm heart, and has certain regulation effect on spleen deficiency, anorexia, uneasiness, palpitation and insomnia.
In the invention, the mulberry leaves are suitable for treating the effects of reducing blood sugar and blood fat, resisting inflammation and the like; rhizoma Polygonati has effects of invigorating qi, nourishing yin, invigorating spleen, moistening lung, and invigorating kidney; dendrobe, sweet and light in nature and taste, slightly salty and cold in property, enters stomach, kidney and lung channel, benefits stomach, promotes fluid production, nourishes yin and clears heat, is used for yin deficiency and fluid deficiency, dry mouth, polydipsia, anorexia, retching, asthenic fever after illness and dim and unclear eyes; the ginseng can stimulate the secretion function of an endocrine system, enhance the function of a pituitary adrenal cortex system, has the function of reducing blood sugar, can reduce urine sugar of a light diabetic patient and improve the general symptoms of a moderate diabetic patient by taking the ginseng, so that the symptoms of craving, hyperhidrosis, weakness and the like disappear or are relieved, and the dosage of insulin can be reduced by taking the ginseng; the Chinese yam is sweet, warm, mild and nontoxic, is mainly used for treating impairment of middle warmer, tonifying deficiency and win, removing cold and heat evil, tonifying middle warmer, benefiting strength, being taken for a long time, improving hearing and eyesight, reducing weight, not hunger, prolonging life, tonifying five consumptive diseases and seven injuries, removing cold wind, calming mind, calming soul, tonifying heart-qi deficiency, and keeping heart-hole for many stories; the medlar is sweet in taste and neutral in nature, enters liver and kidney channels, has the effects of tonifying qi and strengthening essence, nourishing liver and kidney, resisting aging, relieving diabetes and warming the body, and the medlar extract can obviously and durably reduce the blood sugar of rats and increase the sugar tolerance and has lower toxicity.
The sources of the traditional Chinese medicines are not particularly limited, and the traditional Chinese medicines can be prepared by using conventional commercially available medicines in the field.

The pH value of the first enzymolysis is preferably 4.5-6.5, the temperature is preferably 45-55 ℃, and the time is preferably 0.4-2.2 h; the pH value of the second enzymolysis is preferably 3.0-6.0, the temperature is preferably 15-55 ℃, and the time is preferably 0.4-2.2 h; the pH value of the third enzymolysis is preferably 6-7, the temperature is preferably 55-65 ℃, and the time is preferably 0.4-2.2 h. In the enzymolysis, the addition amounts of the cellulase, the pectinase and the papain are preferably 0.1-0.3%. In the invention, the cellulase and the pectinase carry out wall-breaking enzymolysis on the raw materials, and the papain hydrolyzes macromolecular protein into small peptides.
When the enzymolysis liquid is obtained, the invention intercepts the components with the molecular weight less than 2000Da in the enzymolysis liquid to obtain the fermentation type small molecular peptide mixed liquid. The retention according to the invention preferably comprises ultrafiltration with an ultrafiltration membrane. The dosage form of the food or pharmaceutical product of the present invention preferably includes a solid agent or a liquid agent, and more preferably includes an oral liquid.
The following examples are provided to describe the fermented small molecule peptide mixture for regulating blood sugar and the preparation method and application thereof in detail, but they should not be construed as limiting the scope of the present invention.
Example 1
Mashing 30g of poria cocos, and mixing with 300ml of water to obtain 300ml of bacterial liquid;
mixing 6g of ginseng, 20g of Chinese yam, 20g of rhizoma polygonati, 20g of schisandra chinensis, 30g of wolfberry fruit, 20g of dendrobium, 20g of mulberry leaf and 300ml of poria cocos liquid, adding 4.54g of composite probiotic bacterium powder (Shandong Junle Biotech Co., Ltd.), fermenting for 24h at room temperature of 27 ℃ and humidity of below 70 wt.% to obtain fermentation liquid, performing enzymolysis on the fermentation liquid for 1.2h by using cellulase at pH 5 and temperature of 50 ℃ to obtain first enzymolysis liquid, performing enzymolysis on the first enzymolysis liquid for 1.2h by using pectinase at pH 4 and temperature of 50 ℃ to obtain second enzymolysis liquid, and performing enzymolysis on the fermentation liquid for 1.2h by using papain at pH 6.5 and temperature of 60 ℃ to obtain third enzymolysis liquid; wherein the dosage of the cellulose is 0.2 percent of the mass of the fermentation liquor; the dosage of the pectinase and the papain is 0.1 percent of the mass of the fermentation liquor.
And (3) performing ultrafiltration on the obtained enzymolysis liquid by using a filter membrane with the molecular weight cutoff of 2000Da through an ultrafiltration membrane to obtain 50ml of fermentation type micromolecular peptide mixed liquid.
30 SPF-grade Kunming male mice (Liaoning Biotechnology Limited) are selected, and the body mass is (25 +/-5) g. After the mice were purchased, the mice were divided into a blank group, a model group and a dosing group. The model group mice are molded by adopting a mode of injecting Streptozotocin (STZ) solution into the abdominal cavity, the concentration is 5mg/ml, the injection amount is 35mg/kg, the injection is continuously carried out for 3 days, the mice are fed with tap water and are fed with high-fat high-sugar feed, and when the blood sugar of the mice rises to more than 11.1mmol/L, the molding is successful.
The mice in the blank group are injected with sodium citrate buffer solution with the same amount per day in the abdominal cavity, and the mice drink tap water and conventional feed. And (3) respectively adopting a tail vein blood sampling method before and after modeling, and measuring blood sugar by using a glucometer. After fasting for 8h without water deprivation, the blood glucose concentration of the mice was measured. After the molding was successfully performed, 80ml/kg of the drug was administered by gavage in the afternoon administration group every day, and after 8 weeks of continuous gavage of the same dose of physiological saline in the blank group and the model group, the blood glucose values were measured, and the results are shown in table 1.

Example 2
The preparation protocol was the same as in example 1, except that the formulation was different from that of example 1: 330ml of poria cocos bacterial liquid (33g of poria cocos and 330ml of water), 8g of ginseng, 26g of Chinese yam, 28g of rhizoma polygonati, 28g of schisandra chinensis, 34g of wolfberry fruit, 28g of dendrobium nobile, 24g of mulberry leaf and 5.26g of composite probiotic bacteria powder (Shandong bacteria-le biotechnology limited).
In this example, 30 mice similar to those in example 1 were used, and the mice were divided into a blank group, a model group and an administration group, and the molding method was the same as that in example 1.
After successful molding, the blank group and the model group were subjected to gavage administration of 80ml/kg of normal saline, and after 8 weeks, the blood glucose values were measured, and the results are shown in table 2.

As is clear from the experimental data shown in Table 2, the blood glucose level of the type 2 diabetic mice successfully molded can be reduced to some extent by using the fermented small molecule peptide mixture of the present invention
Example 3
The preparation protocol was the same as in example 1, except that the formulation was different from that of example 1: 380ml of tuckahoe bacterial liquid (38g of tuckahoe and 380ml of water) are fermented with 10g of ginseng, 30g of yam, 26g of rhizoma polygonati, 32g of schisandra, 36g of medlar, 30g of dendrobium, 26g of mulberry leaf and 5.93g of composite probiotic bacterial powder (Shandong Junle Biotech Co., Ltd.).
The same molding experiment was performed using the same number of mice as in example 2.
After successful molding, the blank group and the model group were subjected to gavage administration of 80ml/kg of normal saline, and after 8 weeks, the blood glucose values were measured, and the results are shown in table 3.

As can be seen from the experimental data shown in table 3, the blood glucose level of the type 2 diabetic mice successfully molded can be significantly reduced by using the fermented small molecule peptide mixture of the present invention.
Example 4
Selecting two groups of people of 35-55 years old, diabetic patients and 80 people according to the ages, wherein each group comprises 40 people, 35-45 years old is one group, and 46-55 years old is one group. 50ml of the small molecule peptide mixed solution prepared in the third example is taken twice a day in the morning and at night, and the blood sugar value change of 4 courses, 8 courses and 10 courses before taking is recorded, as shown in table 4.

The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
Claims (10)
1. A preparation method of a fermentation type small molecule peptide mixed solution for regulating blood sugar is characterized by comprising the following steps: (1) mixing Poria bacteria liquid and composite probiotic with Ginseng radix, rhizoma Dioscoreae, rhizoma Polygonati, fructus Lycii, herba Dendrobii and folium Mori, and fermenting together to obtain fermentation liquid; the fermentation temperature is 26-28 ℃, the humidity is 62-70%, and the time is 16-24 h; the strains of the composite probiotics comprise: bifidobacterium adolescentis, Bifidobacterium lactis, Bifidobacterium longum, Lactobacillus acidophilus, Lactobacillus casei, and Lactobacillus rhamnosus;
(2) sequentially carrying out enzymolysis on the fermentation liquor by using cellulase, pectinase and papain to obtain enzymolysis liquid;
(3) and intercepting components with the molecular weight of less than 2000Da in the enzymolysis liquid to obtain the fermentation type small molecular peptide mixed liquid.
2. The preparation method according to claim 1, wherein the mass ratio of the poria cocos to the ginseng, the yam, the polygonatum rhizome, the wolfberry fruit, the dendrobium and the mulberry leaf in the poria cocos bacterial liquid in the step (1) is (150-200): (3-10): (10-20): (10-15): (15-20): (10-20): (10-15).
3. The method according to claim 1 or 2, wherein the mass ratio of Poria cocos to water in the Poria cocos solution of step (1) is 1: 10.
4. the preparation method according to claim 1, wherein the enzymolysis in step (2) comprises performing a first enzymolysis on the fermentation broth with cellulase to obtain a first enzymolysis solution, performing a second enzymolysis on the first enzymolysis solution with pectinase to obtain a second enzymolysis solution, and performing a third enzymolysis on the second enzymolysis solution with papain to obtain the enzymolysis solution;
the pH value of the first enzymolysis is 4.5-6.5, the temperature is 45-55 ℃, and the time is 0.4-2.2 h;
the pH value of the second enzymolysis is 3.0-6.0, the temperature is 15-55 ℃, and the time is 0.4-2.2 h;
the pH value of the third enzymolysis is 6-7, the temperature is 55-65 ℃, and the time is 0.4-2.2 h.
5. The preparation method according to claim 1 or 4, wherein the addition amount of the cellulase, the pectinase and the papain in the enzymolysis in the step (2) is 0.1-0.3%.
6. The method of claim 1, wherein the retaining of step (3) comprises ultrafiltration using an ultrafiltration membrane.
7. A fermented small molecule peptide mixture prepared by the method according to any one of claims 1 to 6.
8. Use of the small molecule peptide mixture of claim 7 for the preparation of a food or pharmaceutical product for maintaining healthy blood glucose levels.
9. A food or medicine for maintaining healthy blood sugar level, wherein the effective component of the food or medicine comprises the small molecule peptide mixture of claim 7.
10. The food or pharmaceutical product of claim 9, wherein the food or pharmaceutical product is in a dosage form comprising a solid or liquid dosage form.
