Description

Traditional Chinese medicine composition for resolving depression and soothing nerves as well as pharmaceutical preparation and application thereof
[ field of technology ]
The invention relates to the technical field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition for resolving depression and soothing nerves, a pharmaceutical preparation and application thereof.
[ background Art ]
Sleep disorders are often caused by long-term thought contradiction or mental overload, mental work, improper working and escape combined with long-term treatment, weakness after illness, and the like. Generally, four general categories are defined: 1. difficulty in initiating and maintaining sleep (insomnia); 2. excessive daytime sleepiness (somnolence); 3.24 hours sleep-wake cycle disorder (sleep-wake rhythm disorder); 4. abnormal activities and behaviors during sleep (sleep disorder, night terrors, nightmares).
Investigation has shown that adults have a proportion of sleep disorders up to 35% and the elderly over 60 years old have a proportion of sleep disorders of 27% -50% in some cities in children 2-6 years old. Furthermore, pregnant women may have a rate of up to 75% of sleep disorders at the end of gestation. Some stroke, parkinson's disease, diabetes and psychotic patients also experience varying degrees of sleep disorder, with 55.5% of the patients experiencing varying degrees of social dysfunction.
Sleep disorders are among the most common among them as insomnia. Insomnia (insomnia) is a subjective experience that affects daytime social functions by failing to meet normal physiological needs in terms of sleep quality or quantity due to sleep and/or sleep maintenance difficulties, and is the most common sleep disturbance disorder. Today, the acceleration of social rhythm and the aggravation of competition, insomnia is a very common phenomenon. The prevalence rate of insomnia is very high, the prevalence rate of countries such as Europe and America is about 20% -30%, and the prevalence rate of countries such as Europe and America is about 10% -20%. This difference in prevalence is related to the different individual demands for quality of life and subjective experiences, and there is no relevant epidemiological data in our country. Insomnia can cause inattention, hypomnesis, impaired judgment and daily work, and severe cases with anxiety, compulsions and depression.
Especially for naval vessel officers and soldiers, due to factors such as small personal space, large working pressure, moist climate, noisy environment and the like, the sleeping state is easy to influence, and sleeping disorder is easy to occur, so that various negative moods such as anxiety, depression and the like can be induced. Feng Chenxi [1] And the anxiety and depression emotion in the long voyage of sea crews mainly in young and strong years are obviously higher than those before voyage, and the sleep quality also can influence the survival quality of officers and soldiers, so that the quality of training, duty and other tasks is not facilitated. Recent research evidence suggests that insomnia is an important risk factor for post-traumatic stress disorder, depression, and anxiety disorder after military combat mission is performed by soldiers [2] During the sea going training of ships, the sleeping quality of officers and soldiers is improved, and normal training is not disturbed. However, the conventional hypnotizing mode by means of medicines is extremely easy to produce side effects, tolerance and dependence can be produced after long-term taking of medicines, withdrawal symptoms can be produced after stopping taking medicines, and therefore the long-term taking of medicines is not suitable.
In order to overcome the above-mentioned drawbacks, the positive idea meditation is a hot spot for researching non-drug treatment of sleep disorder in recent years, and many researches have proved that the positive idea is toHas good curative effect on chronic insomnia. Black etc [3] In a randomized controlled clinical trial, it was found that: the reduction in the pittsburgh sleep quality index was more pronounced in subjects taking part in meditation training than in the control group receiving sleep hygiene education. Domestic Wang Xiaoming, etc [4] Studies have found that positive-sense meditation levels and sleep quality of subjects show a significant positive correlation, and meditation training can effectively improve sleep quality by reducing anxiety levels of subjects. However, the effect of positive training is affected by experience and technology of therapists, foreign excessive syndrome documents can mention training qualification of positive trainers when reporting positive therapeutic effects, and most of domestic documents at present do not mention qualification and training experience of trainers, which may be a defect of positive meditation in practical application, so that clinical popularization is difficult.
Western medicine treatment mainly comprises insomnia, and common hypnotics are tranquilizer, such as short-acting alprazolam, medium-acting tranquilizer and sulbactam, and long-acting chlordiazepoxide; western medicine is improved but not fully controlled for most patients. Has limited curative effect on part of patients with difficult treatment, is easy to relapse after stopping the medicine, and has obvious side effect of western medicines.

The results prove that the traditional Chinese medicine composition can effectively improve insomnia and has the effects of calming, helping sleep and soothing the nerves.
Such traditional Chinese medicine compositions for resolving depression and soothing nerves have many but more or less disadvantages, such as excessive medicine taste, difficult material acquisition, high price, insignificant medicine effect and the like. Therefore, a traditional Chinese medicine composition with obvious depression relieving and nerve soothing effects, moderate medicine taste and convenient preparation is needed.

2.2 grouping
The experimental group of sachets was randomly divided into 20 persons, the positive thought meditation group 20 persons and the blank group 19 persons. The selected officers and soldiers are men, and the comparison difference has no statistical significance (P is more than 0.05) in the aspects of age, academic, marital, post, personnel quality and the like. There was no statistical difference between the PSQI scores and the SAS scores before treatment for the three officials and soldiers, and the scores were comparable.
2.3 Experimental methods
The experimental group uses self-made traditional Chinese medicine sachet (grassleaf sweelflag rhizome, platycladi seed, chinese angelica, fineleaf schizonepeta herb and lavender) for resolving depression and soothing nerves. The sachet is placed at the bed head, the sachet is opened before sleeping at night, the nose and mouth are placed at a distance of about ten centimeters from the sachet mouth, the sachet is deeply smelled for 3 minutes, the sachet is replaced for officers and soldiers every 2 weeks, and the treatment is carried out for 8 weeks.
The positive idea meditation training mainly refers to the eight-week course structure of positive idea cognitive therapy (V.West, williams Mark, tim Stokes John. Internal cognitive therapy of depression, beijing company of world book publishing company, 2008), and is intensively trained 1 time per week under guidance audio guidance, each time for 2 hours, including training and experience sharing, and the rest of time is trained by self according to post-class operation.
The blank control group keeps the same work and rest as the sachet experimental group and the positive idea meditation group, and works. The differences in PSQI and SAS scores before and after treatment were statistically compared in three groups.
3. Observation index
PSQI scale scores, total score > 8 indicating sleep disorder, higher score indicating more severe sleep disorder. Efficacy evaluation criteria: and evaluating according to the PSQI score reduction rate of the Pittsburgh sleep quality index scale.
(1) And (3) healing: the PSQI score reduction rate is above 75%;
(2) The effect is shown: the PSQI score reduction rate is between 50% and 74%;
(3) The method is effective: the PSQI score reduction rate is 25-49%;
(4) Invalidation: the PSQI score reduction rate is within 25%.
SAS scale score <50 score no anxiety, > 50 score mild anxiety, > 60 score moderate anxiety, and > 70 score severe anxiety.
4. Statistical method
Statistical analysis of the data was performed using SPSS21.0 software, and the metrology data was expressed as mean.+ -. Standard deviation (x.+ -. S). The difference between the three groups of PSQI scores and the SAS score is tested by adopting single factor analysis of variance and SNK, and all tests take P <0.05 as a test standard.
5. Analysis of results
Results of scoring conditions of the PSQI and SAS scales before and after the experiment of the three experimental groups are shown in Table 1:
table 1 scoring Condition (score, X.+ -. S) of PSQI and SAS scales before and after three official experiments

Electric shock cage, ventilation cage, one-way contact type alternating current voltage regulator (Zhejiang power supply equipment limited company).
2 method
2.1 preparation of Insomnia animal model
The model is built by adopting a multi-condition fear method, rats are placed into electric shock cages, 5 rats are placed into each cage, and insomnia rats are built by adopting a method of associating negative stimulation (such as plantar electric shock) with neutral prompt (such as sound and illumination) or specific environment (experimental room) except normal feeding: firstly, placing a rat in an electric shock cage, adding used cat litter, exposing for 90min, and performing fear memory training; noise interference is then carried out, the intensity is generally 85-90 dB, and most of the used sounds are random combinations of noise with different properties, intensity and frequency, and can comprise traffic noise, alarm, music, artificial synthetic noise and the like. Then, 15V voltage is selected, and the current is supplied and the electric shock quantity is intermittent by 1:5, so that the rats are deprived of sleep time for 2.5 hours at random every day for 14 days. The arousal time of the rat is obviously increased, the sleep quality is reduced and the sleep rhythm is disturbed, namely the modeling is considered successful, otherwise the modeling is considered to be failed.
2.2 test protocol
The 50 rats were randomly divided into five groups, 10 in each of the inventive group I, inventive group II, control group, western medicine group and model group. By observing the weight change of rats, the group of the invention does not fall off; the weight of the second group of the invention does not reach the standard, and 1 case of the second group falls off; one body weight of the control group does not reach the standard, and the other body weight exceeds the standard, and 2 cases of falling off; the western medicine group body weight exceeds standard, and 1 case of western medicine is dropped; the model group is free from falling off.
The treatment (opening the sachet before sleeping, the rats sniff for 30min each day, replacing the sachet for every 1 week, and treating for 4 weeks) of the invention group and the control group respectively, the western medicine group and the model group were subjected to gastric lavage treatment. Daily observation and recording body surface characteristics of each group of rats; after the completion, the rat is sacrificed by head breaking, the skull is removed, the brain tissue is completely exposed, the medulla oblongata is rapidly sheared at the occipital bone macropore, the brain tissue is taken out, the brain and cerebellum are removed, and the weight is carried out. The absorbance of 5-HT, 5-HIAA, NA and DA of each sample of the model group is converted to mug/g by referring to simultaneous extraction and fluorescence spectrophotometry of 5-hydroxytryptamine, 5-oxindole acetic acid, norepinephrine and dopamine in brain tissue.
2.3 statistical methods
The counting data and the grade data are compared by Fisher accurate inspection; metering dataTo show, the comparison between groups uses one-factor analysis of variance (variance non-uniformityThe latter were tested using the Tam hanesT test). Statistical analysis, P, using GraphPadprism 8 software<0.05 is statistically significant.
2.4 results
2.4.1 body surface characteristics of rats before and after treatment
The rats in the group, the control group, the model group and the western medicine group have poor mental state, slow response, withered hair, obviously reduced drinking water, more stool and sometimes no formation before treatment. After four weeks of treatment, compared with a model group, rats in the group disclosed by the invention have good mental state, smooth and moist hair, active response, normal stool and normal drinking water. The western medicine group rats have smooth and glossy hair, are sensitive to irritation, are not easy to be irritated, and have normal defecation times. The control rats had slightly weaker mental states, sometimes irregular stool shape, and increased drinking water.
2.4.2 control of index content of rats 5-HT, 5-HIAA, NA, DA in groups (μg/g)
As can be seen from the comparison of the two compositions, after continuous administration to insomnia rats: the content of 5-HT in the first, second, control and western medicine groups is increased, but the first and second groups are obviously increased and are obviously different from the model group (P is less than 0.05); the 5-HIAA content of the first group and the second group of the invention is obviously increased compared with that of the model group, and the difference is obvious (P is less than 0.05); the first group and the second group of the invention can obviously reduce the NA content in brain tissues of insomnia rats, and are superior to a control group and a western medicine group (P is less than 0.05). The results of the dopamine content show that the DA content can be reduced by the first group and the second group of the invention, which is superior to the control group and the western medicine group (P is less than 0.05). See Table 5
Table 5 controls of index levels (. Mu.g/g) for rats of each group 5-HT, 5-HIAA, NA, DA
Note that: comparison with model group 1) (P<0.05 A) is provided; comparison with the control group 2) (P<0.05 A) is provided; comparison with Western medicine group 3) (P<0.05)。
The foregoing is merely a preferred embodiment of the present invention, and it should be noted that modifications and additions may be made to those skilled in the art without departing from the method of the present invention, which modifications and additions are also to be considered as within the scope of the present invention.
