Description
Technical Field
The invention relates to a medicine pillow and a preparation method thereof, in particular to a heart-clearing sleep-aiding and intelligence-improving medicine pillow and a preparation method thereof.
Background
The health status of a person can be divided into three states, healthy, sub-healthy and disease. According to a global investigation by the World Health Organization (WHO), people in a healthy state account for 5% of the general population, people in a diseased state account for 20% of the general population, and less than 25% of the total population, more than 75% of the people are in a transitional state between healthy and diseased, also referred to as a "sub-healthy state". Sub-health is reversible, and if it is in sub-health state for a long period, it can develop into sub-clinical state and further into disease state.
Insomnia is the most common symptom of sub-health status, which is a functional disorder caused by imbalance of brain function balance, and is a status that is often difficult to fall asleep, easy to wake after sleeping, or wake early, caused by disorder of sleep rhythm caused by reasons related to modern life style. Questionnaire analysis, wherein most people are insomniac with symptoms such as dysphoria, dreaminess, drowsiness, lazy after waking, memory decline and the like, and the biological clock is disturbed due to irregular long-term work and rest, and the circadian rhythm is regulated and controlled to be unbalanced; the study and research pressure is great, and the thinking is that liver qi cannot reach smoothly, and the gallbladder fire disturbs the heart and the fire disturbs the mind.
At present, the modern medicine has the treatment principle of 'taking on demand' and 'intermittent small dosage' of hypnotic sedative. However, the study shows that the sedative hypnotic drug used for a long time is easy to generate various adverse reactions such as dependence, tolerance, withdrawal symptoms, rebound effect and the like, thereby becoming a cause for inducing insomnia. The modern medicine pillow has the advantages of multiple functions, complex using steps, single medicine flavor, low universality and the like, and needs to be further improved.

TABLE 1 solid phase microextraction extraction of sleep-aiding Chinese herbal Compound volatile Components
By virtue of the volatile oil characteristics of the alpha-asarone and the beta-asarone, the alpha-asarone and the beta-asarone can quickly penetrate through the blood brain barrier from blood to enter the brain to exert pharmacological activity, and the beta-asarone can be widely distributed in cortex, cerebellum, brain stem and hippocampus. Research shows that they have potential therapeutic advantages for nervous system diseases, such as insomnia, mild cognitive impairment and the like. Glutamate, the major neurotransmitter responsible for excitotoxicity, has a key effect in both nervous system diseases, insomnia and cognitive dysfunction, and alpha-asarone can regulate Ca 2+ Concentration, for glutamate receptor (AMPA, NMDA), transporter GLT-1, and controlled release of glutamate synaptorin protein (SYNI) to maintain glutamate concentration, alpha-asarone can also act directly on insomnia direct related protein CYP2D6. The alpha-asarone and the beta-asarone can regulate the excitatory neurotransmitter and the inhibitory neurotransmitter in the brain and maintain the balance level of the two. At the same time, the two can also play a role in inhibiting the expression of inflammatory factors, and the beta-asarone can reduce Abeta 25-35 The induced neuroblastoma inflammatory factors TNF-alpha, IL-1 beta and IL-6 levels reduce the inflammatory response. Alpha-asarone subtractionLight inflammation is the reduction of production of the inflammatory factors TNF- α, IL-1β by inhibiting NF- κB signaling pathways. The cinnamon volatile oil has anxiolytic and antidepressant effects on mice, and trans-cinnamic aldehyde is a main volatile component in cinnamon, and can participate in the antidepressant process through a 5-hydroxytryptamine energy receptor and an adrenergic receptor. Meanwhile, in vitro experimental researches show that the cinnamaldehyde can obviously inhibit the expression of proinflammatory cytokines IL-1 beta and TNF-alpha and the generation of NO, and can be used for treating neurodegenerative diseases closely related to neuroinflammation. The tunicarene is a main volatile component in the schisandra volatile oil, can strengthen the excitation and inhibition process of human cerebral cortex, and improve the sleep of people. Experimental study shows that the cinnamon can remarkably promote the synthesis of 5-hydroxytryptamine, epinephrine, dopamine and cerebral nerve neurotrophic factors, and increase the secretion of monoamine neurotransmitters and the sensitivity of the monoamine neurotransmitters, so that the HPA axis dysfunction is improved. And cinnamaldehyde can alleviate symptoms of mice with systemic inflammatory response syndrome, and reduce the levels of NO, TNF-alpha and peritoneal IL-10. The volatile components of cortex Cinnamomi can play neuroprotective role in Hippocampus and multiple pathological links by interaction with central monoaminergic nerve, inhibiting inflammatory reaction, etc.
Test experiment 2: ulaise subthreshold dose synergistic sleep experiments
Mice were successfully model-built and randomly screened for PCPA insomnia (blank, model, low, medium, high, n=10), and were dosed 9.1 times as much as adults according to 380g of drug used in example 3 of 60kg of daily drug pillow according to the body surface area method calculated from the human dose and experimental animal dose conversion of "pharmacological experimental methodology", so that the mice were dosed 57.63g/kg. 2 times (115.27 g/kg) of the equivalent dose of the drug pillow formula of the mice and the human is set as the medium dose of the experiment, 4 times of the equivalent dose is set as the high dose (230.53 g/kg), and the experimental intervention treatment is started on the next day. The control group does not need any treatment, and the crushed and packed medicine cores are paved above the squirrel cage in the high-medium-low dose group, and the using time of the medicine cores is 20:00-8:00 in the next day. Mice were tested for various indicators after 30d of continuous dosing.

Compared with the blank control, the drug pillow high, medium and low dose groups can obviously shorten the sleep latency period (P < 0.05) of the aconite sugar of the mice and improve the sleep rate of the mice.
Test experiment 3: influence of the drug pillow Compound on the 5-HT, TNF-alpha content in brain tissue of insomnia mice
As shown in fig. 1, the 5-HT content in the brain tissue of mice in the model group was significantly reduced compared to the normal group; the drug pillow example 3 showed a significant increase in 5-HT levels in the high, medium and low dose groups compared to the model group.
As shown in fig. 2, TNF- α content was significantly elevated in the serum of the model group compared to the normal group. Drug pillow example 3 the TNF-alpha content in the serum of the high, medium and low dose groups was significantly reduced (P < 0.05).
The experimental research results show that the invention can shorten the sleep latency of the insomnia mice induced by the uliose, improve the sleep rate of the insomnia mice, increase the 5-HT level in the brain tissues of the mice and reduce the TNF-alpha level in serum.
Test experiment 4: 50 subjects with sleep quality degradation problems were selected, aged 18-55 years, and divided into 4 test groups and 1 control group by random number method. Giving sleep knowledge to the control group, and teaching and only filling a blank control drug pillow filled with filling cotton; the test group underwent sleep knowledge education while the drug pillow treatments in examples 1 to 4 were administered respectively. Patients were assessed for sleep quality using PSQI prior to and 4 weeks after group entry.
