3.1 materials and instruments
The formula is as follows: 90470 antidepressant formulation and 90472 antidepressant formulation.
Data acquisition was performed using a digital electroencephalograph (model ND-97, high tech medical instruments, shanghai).
3.2 test sites: brain wave testing room of aromatic plant research and development center of Shanghai university of transportation
3.3 brief introduction to digital electroencephalograph
The brain electricity is an electrophysiological signal generated by the electrical activity of the brain cortex nerve cell cluster, generally ranging from a few microvolts to hundreds of microvolts. The brain waves are composed of various rhythmic waves and are divided into delta waves, theta waves, alpha waves and beta waves according to different frequencies, and the frequencies of the delta waves, the theta waves, the alpha waves and the beta waves are 0.5-3.5 Hz respectively; 4.0-7.5 Hz; 8.0-13.5 Hz; 14.0 to 30.0 Hz. Alpha and beta waves belong to fast waves in brain waves; alpha waves are the normal waveform when an adult is awake and quiet closing the eyes, and beta waves occur when a person senses an external stimulus, is alert, or is subjected to a particular psychological stress. The delta and theta waves are slow waves in brain waves, and their components are increased during the sleep period of a normal adult. The occurrence and amplitude of each wave vary greatly among individuals, and are affected by factors such as age and sex.
3.4 electroencephalogram data acquisition
3.4.1 recruitment of volunteers
All volunteers in this study were recruited by voluntary roll-in, with 47 people co-recruited, from age 17 to 34 years, for the most students at school. Wherein 90470 groups had 25 people; 90472 the population is 22.
3.4.2 volunteer electroencephalogram data acquisition
The brain wave experiment is carried out in the electroencephalograph room of the aromatic plant research and development center of Shanghai university of transportation. The conventional indoor environment arrangement has the advantages that the surrounding environment is relatively quiet, and no obvious bad smell influence exists. The tested volunteers receive the brain wave test in a normal sitting posture mode and are connected with the electroencephalograph in a 16-lead human body potential mode. After the brain wave signals are led out by the scalp electrodes, the fast Fourier transform is carried out by the preamplifier and the central processing unit, the wave amplitude-time relation of the brain waves is converted into a power-frequency relation, a brain wave curve is synchronously displayed on a screen in real time, and brain wave absolute power values of delta, theta, alpha and beta in each brain area position of four frequency domains are marked on a skull model chart.
Each volunteer was first controlled by recording 3 minutes of brain electrical activity as a blank without sniffing any essential oils or other significant odors. Then, sniffing for 3min without adding blank solid fragrance of essential oil and recording EEG signals. The third sniffing treatment was performed by giving solid sniffing to volunteers 90470 or 90472 for 3min and recording brain electrical signals. Each sniffing is carried out in a closed room with the same space size, the interval of each sniffing is 10min, and each volunteer carries out the second same test at the same time after one week. The brain wave data is collected to avoid the interference of various external factors such as the order of the orders, and all volunteers are required to keep a relaxed state, and the bodies are not easy to blink and shake. These representative signals are then superimposed. The analysis result is that the percentage of delta, theta, alpha and beta 4 frequency domains in the total power of 1-30Hz is calculated, and the results of the two tests are averaged.

2. Olfaction preference survey of different kinds of anti-depression fragrance
The purpose of the test is as follows: the olfactory preference of the odorant for the four numbered antidepressant fragrances (90470, 90471, 90472, 90473) was investigated, i.e. whether the test person liked or disliked a fragrance of a certain number.
The test method comprises the following steps: the testee smells the incense in a closed room for 30 min. And finally, finishing the aroma evaluation table. Including preference, feeling after smelling fragrance, etc. Each numbered incense corresponds to about 90 persons, and the total number of persons to be investigated is 348 persons.

and (4) counting results:
Figure 325466DEST_PATH_IMAGE002
the results of the test showed 90470 to be the most favoured, next to 90472 and 90473 and finally 90471. The feeling of calmness, comfort and warmth is the most common after smelling the fragrance. Meanwhile, because the unfavorable rate of 90472 is far lower than that of 90473, two formulas are finally selected for the next experiment.
3. Evaluation of efficacy of different species of antidepressive incense
1) Animal model test:
the purpose of the test is as follows: animal experiments are carried out to investigate the efficacy of different species of the anti-depression fragrance.
The test method comprises the following steps: the experimental animals are Kunming mice and 60 male animals. The tail suspension experiment and forced swimming experiment are carried out. After suspending the tail or putting into water, the mice give up struggling, and show disappointed state in a discontinuous immobile or floating state.
The basic idea of the test is as follows: grouping and administration: randomly dividing the mice into 6 groups, each group comprises 10 mice, and the mice are respectively 2 blank control groups; 2 non-perfuming coil smelling groups; 2 preferably perfumed coil sniffing groups. Feeding for 3d, and performing the experiment after smelling for 15min on 4 d.
Smelling the fragrance: igniting the incense coil outdoors until the burning is stable, putting the incense coil into an animal incense smelling cage, stably expanding the incense for 15min, and putting the animal incense for 15 min. Animals were free to feed and drink during the experiment.
Time for which mice remained immobile in both depression models
Figure 304924DEST_PATH_IMAGE003
After smelling 90470 and 90472, the mice had shorter immobility time in both tail suspension experiment and forced swimming experiment. In the tail suspension experiment, compared with the blank fragrance, both fragrances are greatly reduced (P is less than 0.01) compared with the blank fragrance. In forced swimming experiments, the time for the mice to remain immobile after smelling the two smells also significantly decreased, but did not reach a significant level.
2) Effect of different kinds of antidepressive incense on human brain waves
There are multiple sites in the brain that participate in the process of producing emotions. Scientists have demonstrated that there are indeed sensation-related systems in the brain and that they end up as limbic systems.
The Papez theory of loops holds that: the peripheral lobe, including the hippocampus, cingulate gyrus, papillary body and hypothalamus, constitutes a loop that is associated with the emotional sensory experience. The circuit formed by hippocampus, fornix, papillary body, papillary thalamic fascicle, cingulate gyrus, frontal lobe of cerebral cortex and hippocampus is used as the emotional thought pathway. The frontal, parietal, occipital and temporal 4 lobes all partially extend to the medial aspect of the cerebral hemisphere. Above the diencephalon there is a corpus callosum connecting the left and right hemispheres. The groove above the corpus callosum is called the corpus callosum groove, which bypasses behind the corpus callosum and moves forward to the hippocampal groove. The cingulum is arranged above the callosum groove, and the cingulum is arranged between the other callosum grooves.
The electroencephalogram experiment records electroencephalograms before and after the human smells the antidepressant aroma, and analyzes the change of the electroencephalogram. Namely, the mental state change of the person is analyzed according to the brain wave change condition of each frequency domain range, and the efficacy of the incense is evaluated.
The number of tested persons: the first formula is 25 persons, and the second formula is 22 persons. Volunteers smell fragrance twice, and data were averaged over the two tests.
1. The experimental results are as follows:
Figure DEST_PATH_IMAGE005
2. and (3) analyzing an experimental result:
the first formula and the second formula have antidepressant effects, and are characterized in that the power percentages of low-frequency waves delta and theta in the electroencephalogram waves are obviously reduced after smelling fragrance, the alpha waves are obviously increased, and the high-frequency waves beta waves are also reduced.
Comparing the two formulas, wherein the rising of alpha wave of the formula I is more reflected as alpha 1, namely low-frequency alpha wave; the beta wave reduction is significant. And the rising of alpha 2 in the rising of the formula two alpha waves is more remarkable than that of alpha 1, namely, the rising of high-frequency alpha waves is dominant. The beta wave is not reduced or is not obvious before and after smelling the fragrance, and particularly the beta wave is not changed after the fragrance is smelled by the beta wave 1.
This shows that both formula one and formula two have good antidepressant effect, and formula one and formula two have different emphasis on the use of antidepressant effect.
The formula I has the advantages that the low-frequency alpha wave is increased predominantly, and the beta wave is reduced remarkably, so that the formula can better relieve the tension mood and the anxiety feeling of depressed people when relieving the depression symptoms of the depressed people.
The elevation of high-frequency alpha waves is dominant in the formula II, and beta waves have no obvious change, so that the depressed mood of people with obvious depressed mood and depressed mood is relieved after smelling fragrance, and meanwhile, the people do not feel too much sinking.
Formula one and formula two have different emphasis on the use of antidepressant efficacy.
The present invention has been described in detail, and it should be understood that the detailed description and specific examples, while indicating the preferred embodiment of the invention, are intended for purposes of illustration only and are not intended to limit the scope of the invention.
