技术领域
本发明涉及一种利用水溶化乳香胶(mastic gum)以及山胡椒(Lindera glauca)提取物的生发促进或脱发抑制用组合物。
背景技术
毛发是在毛囊(hair follicles)中形成,成年人的毛囊大约有五百万个,分布于人体皮肤中除手掌以及脚掌之外的整个身体上,一百万个存在于头部,而其中的十万个存在于头皮(Bulletin of Food Technology 2009,22(4):651-663)。头皮毛发占人体毛发的大约20%,不仅可以起到如汗液蒸发以及体温调节等毛发的生理功能,还可以通过对头部的缓冲作用起到保护头部免受冲击伤害的功能,而且因为现代人注重外表的审美倾向,对于个人展现出自己的个性以及形象具有非常重要的社会功能(Clin Dermatol 2001,19(2):161-166)。
虽然直至目前为之并没有完全查明导致脱发的准确原因,但是已经提出的观点包括与毛发周期调节相关的毛乳头细胞(dermal papilla cells)的增殖抑制或功能下降、因为雄性激素作用而导致的毛发周期的非正常化、因为头皮血流量的下降而导致的毛发周期的非正常变化、抗癌剂等药物作用、精神压力、物理性刺激以及环境污染等(J InvestDermatol,1999,113:873-877;Mol Cell Endocrinol,2002 198:89-95;J.Dermatol.Sci.2011,62:154-159)。
毛乳头细胞是在毛球(Hair bulb)内发生特化的成纤维细胞(Dermalfibroblast)的一种,可以与毛囊内的多种类型的上皮细胞进行相互作用,而且已经得知可以对毛囊的形成、毛发的再生以及毛发的生长起到重要的作用,因此毛乳头细胞的生长以及增殖、细胞凋亡(apoptosis)是开发用于维持毛发的生长期并对脱发进行抑制的药物方面的一个重要靶标(Physiol.Rev.81:449-494;J.Dermatol.Sci.2011,62:154-159;FASEBJ.2008,22:1725-1736)。
细胞的增殖与由G0/G1、S以及G2/M期构成的细胞周期(cell cycle)的进程有着密切的关联,而且细胞周期蛋白E/细胞周期蛋白依赖性激酶2复合体(cyclin E/CDK2complex)的活性化、细胞周期蛋白D1(cyclin D1)的增加、成视网膜母细胞瘤蛋白(pRB)的磷酸化的增加以及细胞周期蛋白依赖性激酶抑制因子(p27KIP1)的减少等会促进如上所述的细胞周期的进程(Mar.Drugs 2013,11:1783-1799)。
尤其是在如上所述的细胞周期蛋白中,已经得知细胞周期蛋白D1(cyclin D1)作为在细胞周期中从G0/G进展到S期(S phase)时增加的蛋白,是细胞外因子/β-连环蛋白(Wnt/β-catenin)信号传递的靶标基因(target gene)之一(Johnson et al.,1999;Sherr1996;Prall et al.,1997),而且细胞外因子/β-连环蛋白(Wnt/β-catenin)信号传递在毛发生长以及毛发再生方面也可以起到重要的作用。目前认为细胞外因子/β-连环蛋白(Wnt/β-catenin)信号传递可以通过糖原合酶激酶3β(GSK3β,glycogen synthase kinase-3β)、蛋白激酶A(PKA,protein kinase A)以及蛋白激酶B(Akt)等的作用进行调节(Dev.Biol.2005,185:82-91;Mol.Cell.Biol.2005,25:9063-9072;J.Immunol.2001,166:4713-4720)。
目前,已经获得美国食品药品监督管理局(Food and Drug Administration,FDA)批准的用于促进毛发生长的药物包括米诺地尔(minoxidil)以及非那雄胺(finasteride)两种。米诺地尔最初的开发目的是作为用于治疗高血压的血管扩张剂,但是因为有报告指出其具有多毛症的副作用,因此转而开发成生发剂,虽然直至目前为止并没有明确查明所述米诺地尔在生发效果方面的作用机制,但是普遍认为是通过血管扩张的营养供应增加、腺嘌呤核苷三磷酸敏感性钾通道(KATP通道,ATP-sensitive K+-channel)开放(opening)效果、细胞外因子/β-连环蛋白(Wnt/β-catenin)信号传递的活性化以及毛乳头细胞的细胞凋亡(apoptosis)抑制效果等诱导毛发生长(J Dermatol Sci.2004,34:91-98)。此外,由默克(Merck)公司开发的非那雄胺是一种可以对作用于雄性激素代谢的酶即5α-还原酶5α-reductase)的活性进行抑制的物质,最初是开发目的是作为前列腺肥大增生治疗剂,但是因为有报告指出其具有促进毛发生长的副作用,因此后来被批准为生发剂类药物(MolCell Endocrinol,1999,198:89-95)。5α-还原酶是用于将雄性激素睾酮(testosterone)转换成二氢睾酮(dihydrotestosterone,DHT)的酶,但是通过利用非那雄胺对5α-还原酶的活性进行抑制,可以对二氢睾酮(DHT)的生成进行抑制并借此减少将二氢睾酮(DHT)作为激动剂(agonist)结合的雄性激素受体(androgen receptor),从最终防止脱发症状。
因为脱发治疗需要长期治疗,因此减少副作用显得至关重要。因此,人们正在努力从对人体的副作用相对较少的天然物质提取出全新的脱发治疗材料。
发明内容
本发明的目的在于提供一种利用水溶化乳香胶以及山胡椒提取物的生发促进或脱发抑制用组合物。
本发明的其他目的或具体目的将在后续内容中进行公开。
本发明如下述实施例以及试验例所证实,确认了水溶化乳香胶以及山胡椒提取物的混合物可以促进毛乳头细胞的增殖并对5α-还原酶的表达进行抑制,从而完成了本发明。
在考虑到如上所述的试验结果的情况下,本发明的生发促进或脱发抑制用组合物,其特征在于:作为有效成分包括(i)水溶化乳香胶或(ii)所述水溶化乳香胶与山胡椒提取物的混合物。
在本说明书中,“水溶化乳香胶”是指对乳香胶进行微粒化(平均粒径为1~999μm范围)至纳米粒子化(平均粒径为1~999nm)之后悬浮到作为溶剂的水中的悬浮液(suspension)(即,在作为溶剂的水中维持无凝聚·沉淀的分散状态)。如上所述的水溶化乳香胶可以通过在对乳香胶进行粉碎并执行微粒化至纳米离子化之后向其加水搅拌混合的方式获得,尤其是通过在对乳香胶进行粉碎并执行微粒化之后向其中加水混合,进而通过对所述混合物(或混合液)进行粉碎并对所述混合物中的乳香胶的粒子进行纳米离子化的方式获得为宜。此时,为了对所分散的粒子进行稳定化而防止其发生凝聚·沉淀,可以在将微粒化至纳米离子化的乳香胶与水混合前后以及粉碎前后添加适量的悬浮剂(suspending agents)。可以使用本行业所公知的任意悬浮剂,例如,可以使用如阿拉伯树胶、黄芪胶(tragacanth gum)、琼脂、卡拉亚胶(karaya gum)、槐树豆胶(locust beangum)、瓜尔豆胶(guar gum)、黄原胶(xanthan gum)、印度树胶(ghatti gum)以及果胶(pectin)等水溶性高分子;如环糊精(cyclodextrin)、甘油或其脂肪酸酯衍生物;如甲基纤维素或羟甲基纤维素等改性纤维素(modified cellulose);以及各种表面活性剂等。所述悬浮剂可以单独使用或将两种以上混合使用,而且可以在适合于进行悬浮的适量范围即相对于乳香胶100重量份的20至200重量份的范围内使用。与水溶化乳香胶的制造相关的具体信息,可以参阅下述的实施例或参阅在发明名称为“通过纳米离子化提升其分散粒以及水溶化力的乳香胶水溶液的制造方法”的韩国注册专利第10-1865712号中所公开的方法等。其中,乳香胶是指从乳香黄莲木(mastic tree,Pistacia lentiscus)获取到的树脂(reisn)。
此外,在本说明书中,“山胡椒提取物”是指将提取对象即山胡椒叶、茎、地上部分、根茎、根、地下部分、全株或所述的混合物利用如水、碳数量为1至4的低级醇(如甲醇、乙醇以及丁醇等)、二氯甲烷、乙烯、病痛、己烷、乙醚、氯仿、乙酸乙酯、乙酸丁酯、N,N-二甲基甲酰胺(DMF)、二甲基亚砜(DMSO)、1,3-丁二醇、丙二醇或所述的混合溶剂进行浸提而获得的提取物,以及利用如二氧化碳以及戊烷等超临界提取溶剂获得的提取物或对所述提取物进行分馏而获得的分馏物,而作为提取方法,可以在考虑到活性物质的极性、提取程度以及保存程度等的情况下适用如冷浸、对流、加热、超临界放射以及超临界提取等任意方法。经过分馏的提取物,是包括在将提取物悬浮到特定溶剂之后通过与极性不同的溶剂进行混合、静置而获得的分馏物,以及在将所述粗提取物吸附到填充有硅胶等的色谱柱之后将疏水性溶剂、亲水性溶剂或所述的混合溶剂作为流动相获得的分馏物的含义。此外,在所述提取物的含义中,还包括通过如冻结干燥、真空干燥、热风干燥以及喷雾干燥等方式去除提取溶剂的经过浓缩的液态提取物或固态提取物。较佳地,是指作为提取溶剂使用水、碳数量为1至4的醇或所述的混合溶剂获得提取物,更较佳地,是指作为提取溶剂使用水与碳数量为1至4的醇的混合溶剂,尤其是使用乙醇与水的混合溶剂获得的提取物。
此外,在本说明书中,“有效成分”是指可以单独呈现出所需要的活性,或者可以与自身没有活性的载体一起呈现出活性的成分。
此外,在本说明书中,所述“脱发”应该理解为是无论其直接、间接诱因,包括在本行业中分类为脱发的所有症状的含义。具体来讲,是包括血液循环不良、因为生成过量的二氢睾酮等而导致的皮脂分泌过剩、因为过氧化物或细菌等而导致的头皮功能下降、老化、遗传因素、压力以及所述的复合作用等因素所导致的所有脱发症状的含义。
在本发明的组合物中,水溶化乳香胶与山胡椒提取物的混合物可以是以任意的比例混合而成,而在考虑到下述的实施例以及试验例的情况下,所述混合物可以是相对于水溶化乳香胶100重量份包含山胡椒提取物0.1至10重量份范围的混合物。
本发明的组合物只要可以呈现出生发促进或脱发抑制活性,可以根据其用途、剂型以及混合目的等包含任意量(有效量)的所述有效成分,通常来讲,在以组合物的整体重量为基准的情况下,有效量可以是在0.001重量%至20.0重量%范围内决定。其中,“有效量”是指在医疗专家等的建议下,在给药期间内利用本发明的组合物向其适用对象即哺乳动物、较佳地向人进行给药时,可以呈现出如生发促进等所需要的功能、药理学效果的包含于本发明的组合物中的有效成分的量。如上所述的有效量,可以在相关从业人员的一般的能力范围内通过试验决定。
本发明的组合物为了提升、强化生发促进或脱发抑制效果或为了通过附加如血压调节活性等类似活性而提升其服用或摄入的便利性,除了有效成分之外,还可以追加包含在相关行业中已经对其安全性进行验证且已经知晓具有相应活性的任意的化合物或天然提取物。在如上所述的化合物或提取物中,可以包含各国药典(在韩国为“大韩民国药典”)、各国健康功能食品公典(在韩国为食药处公告的“健康功能食品标准及规范”)等法定文件中公开的化合物或提取物、根据用于对医药品的制造以及销售进行约束的各国法律(在韩国为“药事法”)获得许可的化合物或提取物以及根据用于对健康功能食品的制造以及销售进行约束的各国法律(在韩国为“健康功能食品相关法律”)获得独立的功能性认证的化合物或提取物。
例如,如上所述的化合物或提取物,可以包含与韩国化妆品相关的在食药处公告的“功能性化妆品标准及其试验方法”中刊登的如右泛醇、生物素、L-薄荷醇、吡啶硫酮锌等具有脱发症状缓解功能的成分;以及如含有源自于L-谷氨酸的γ-氨基丁酸(GABA)粉末、鲣节寡肽、纳豆菌培养粉末、鼠目太(鼠目豆)肽复合物、鲑鱼肽、橄榄叶提取物、沙丁鱼肽、酪蛋白水解物、辅酶Q10、葡萄籽酶分解提取粉末、海苔寡肽等具有血压调节功能的成分。
作为本发明的组合物的具体形态,可以是食品组合物。
本发明的食品组合物可以制造成任意形态,例如,可以制造成如茶水、果汁、碳酸饮料以及离子饮料等饮料类;如牛奶以及酸奶等加工乳制品类;如口香糖、饼干、韩式点心、面包以及面条等食品类;以及如片剂、胶囊剂、丸剂、颗粒剂、粉磨剂、片剂、糊剂、糖浆剂、凝胶剂、果冻剂以及棒状剂等健康功能食品制剂类。
此外,本发明的食品组合物在法律、功能上的分类方面,可以分类成符合制造、流通时的实施法规的任意产品类型。例如,可以根据韩国“健康功能食品相关法律”分类成健康功能食品,或者根据韩国“食品卫生法”的食品公典(食药处公告的“食品标准及规范”)上的各个食品类型分类成如饼干类、豆类、茶类、饮料类以及特殊用途食品类等。
本发明的食品组合物中,除了所述有效成分之外还可以包含食品添加剂。食品添加剂通常可以理解为是在对食品进行制造、加工或保存时添加到食品中进行混合或浸润的物质,因为会与食品一起每天且长时间摄入,因此必须确保其安全性。在用于对食品的制造、流通进行约束的各国法律(在韩国为“食品卫生法”)的食品添加剂公典中,在成分方面或功能方面限定性地规定了可以保证安全性的食品添加剂。在韩国食品添加剂公典(食药处公告的“食品添加剂标准及规范”)中,将食品添加剂在成分方面的分类规定为化学合成品、天然添加剂以及混合制剂类,而如上所述的食品添加剂在功能方面分类成如甜味剂、调味剂、保鲜剂、乳化剂、酸味剂以及增稠剂等。
甜味剂的作用在于为食品赋予适当的甜味,在适用本发明的食品组合物中可以使用天然或合成的甜味剂。较佳地,使用天然甜味剂为宜,而作为天然甜味剂,可以使用如玉米糖浆固态物、蜂蜜、蔗糖、果糖、乳糖、麦芽糖等甜味剂。
调味剂的作用在于改善味道或者香气,可以使用天然或合成的调味剂。较佳地,使用天然调味剂为宜。当使用天然调味剂时,除了改善风味之外还可以达成营养强化的目的。作为天然调味剂,可以使用从如苹果、柠檬、柑橘、葡萄、草莓、桃子等提取的调味剂或从如绿茶叶、玉竹、竹叶、桂皮、菊花叶、茉莉花等提取的调味剂。此外,还可以使用从如人参(红参)、竹笋、芦荟汁、银杏等提取的调味剂。天然调味剂可以是液状的浓缩液或固态状的提取物。在一部分情况下,可以使用合成调味剂,而作为合成调味剂,可以使用如酯、醇、醛或萜烯等。
作为保鲜剂,可以使用如山梨酸钙、山梨酸钠、山梨酸钾、苯甲酸钙、苯甲酸钠、苯甲酸钾以及乙二胺四乙酸(EDTA)等,而作为乳化剂,可以使用如阿拉伯胶、羧甲基纤维素、黄原胶以及果胶等,作为酸味剂,可以使用如枸橼酸、苹果酸、富马酸、己二酸、磷酸、葡萄糖酸、酒石酸、抗坏血酸、乙酸以及磷酸等。酸味剂除了改善味道的目的之外,还可以以抑制微生物增殖的目的添加到食品组合物中直至达到适当的酸度。作为增稠剂,可以使用如悬浮剂、沉降剂、凝胶成型剂、膨化剂等。
适用本发明的食品组合物除了如上所述的食品添加剂之外.还可以以补充、强化功能性以及营养性的目的包含本行业公知且可以保障其作为食品添加剂的稳定性的生理活性物质或矿物质类。
作为如上所述的生理活性物质,可以使用如包含于绿茶等的儿茶素、如维生素B1,维生素C,维生素E,维生素B12等维生素类、生育酚、二苯酰硫胺素等,而作为矿物质类,可以使用如枸橼酸钙等钙制剂、如硬脂酸镁等镁制剂、如枸橼酸铁等铁制剂、氯化铬、碘化钾、硒、锗、钒、锌等。
在适用本发明的食品组合物中,可以根据产品类型添加可达成其添加目的的适量的所述食品添加剂。
关于可以包含于适用本发明的食品组合物中的其他食品添加剂,请参考各个国家的食品公典或食品添加剂公典。
作为本发明的组合物的另一具体形态,可以是药剂学组合物。
适用本发明的药剂学组合物中除其有效成分之外还可以包含药剂学所容许的载体,并利用本行业公知的一般方法,根据给药路径制造成口服剂型或非口服剂型。其中,给药路径可以是包括局部路径、经口路径、静脉内路径、肌肉内路径以及通过粘膜组织的直接吸收在内的任意的适当路径,也可以组合使用两种以上的路径。
在不同的给药路径或剂型中,药学所容许的载体属于本行业所周知的事项,具体请参阅包括“大韩民国药典”在内的各个国家的药典。
当适用本发明的药剂学组合物被制造成口服剂型时,可以与适当的载体一起利用本行业公知的方法制造成如粉末、颗粒、片剂、丸剂、糖衣片剂、胶囊剂、液态剂、凝胶剂、糖浆剂、悬浊液、薄片等剂型。此时,作为适当的载体的实例,可以包括如乳糖,葡萄糖,蔗糖,右旋糖,山梨糖醇,甘露糖醇,木糖醇等糖类、如玉米淀粉,土豆淀粉,小麦淀粉等淀粉类、如纤维素,甲基纤维素,乙基纤维素,羧甲基纤维素钠,羟丙基甲基纤维素等纤维素类、聚乙烯吡咯烷酮、水、对羟基苯甲酸甲酯、对羟基苯甲酸丙酯、硬脂酸镁、矿物油、麦芽、明胶、滑石粉、多元醇、植物油、乙醇、丙三醇等。当进行制剂化时,可以根据需要包含适当的结合剂、润滑剂、崩解剂、着色剂、稀释剂等。作为适当的结合剂,可以使用如淀粉、硅酸镁铝、淀粉糊、明胶、甲基纤维素、羧甲基纤维素钠、聚乙烯吡咯烷酮、葡萄糖、玉米甜味剂、海藻酸钠、聚乙二醇、蜡等,作为润滑剂,可以使用如油酸钠、硬脂酸钠、硬脂酸镁、苯甲酸钠、乙酸钠、氯化钠、二氧化硅、滑石、硬脂酸或其镁盐和钙盐、聚乙二醇等,而作为崩解剂,可以使用如淀粉、甲基纤维素、琼脂(agar)、膨润土、黄原胶、淀粉、海藻酸或其钠盐等。此外,作为稀释剂,可以使用如乳糖、右旋糖、蔗糖、甘露糖醇、山梨糖醇、纤维素、甘氨酸等。
当适用本发明的药剂学组合物被制造成非口服剂型时,可以与适当的载体一起利用本行业公知的方法以如注射剂、经皮给药剂、鼻腔吸收剂以及栓剂等形态进行制剂化。作为以注射剂的形态进行制剂化时的适当的载体,可以使用如水性等渗溶液或悬浊液,具体来讲可以使用如包含三乙醇胺的磷酸盐缓冲液(PBS,phosphate buffered saline)或注射用灭菌水、5%右旋糖等等渗溶液。当以经皮给药剂的形态进行制剂化时,可以以软膏剂、霜剂、乳剂、凝胶剂、外用液剂、粉剂、搽剂、气雾剂等形态进行制剂化。当以鼻腔吸入剂的形态进行制剂化时,可以利用如二氯氟甲烷、三氯氟甲烷、二氯四氟乙烷、二氧化碳等适当的推进剂以气雾喷雾剂的形态进行制剂化,而当以栓剂的形态进行制剂化时,作为其载体可以使用如威泰索尓(Witepsol)、吐温(Tween)61、聚乙二醇类、可可脂、月桂酸甘油酯、聚氧乙烯脱水山梨糖醇脂肪酸酯类、聚氧乙烯硬脂酸酯类、脱水山梨糖醇脂肪酸酯类等。
药剂学组合物的具体的制剂化方法属于本行业所公知的事项,可以参考如文献【雷明顿药物科学(Remington's Pharmaceutical Sciences)(1995年第19版)】等。上述文献将被视为本说明书的一部分。
适用本发明的药剂学组合物的较佳给药量,根据患者的状态、体重、性别、年龄、患者的严重程度以及给药路径,可以是每天0.001mg/kg~10g/kg范围,较佳地可以是每天0.001mg/kg~1g/kg范围。给药可以以每天一次或多次的方式执行。无论从哪一个方面,如上所述的给药量均不应该被解释为是对本发明的范围作出的限定。
作为本发明的组合物的又一具体形态,可以是化妆品组合物。
当适用本发明的组合物被定义为是化妆品组合物时,上述化妆品组合物同样可以在其用途上、法律上的分类中采用任意产品类型,具体来讲可以是具有脱发缓解、生发促进、皮肤问题改善以及特应性皮炎改善等用途的功能性化妆品、非功能性一般化妆品等。在产品形态方面也可以采用任意的产品形态,具体来讲可以采用如溶液、悬浊液、乳浊液、糊剂、凝胶剂、霜剂、乳剂、粉末、香皂、洗发水、含表面活性剂的洁面剂、油剂、粉末基材、乳浊液基材、蜡基材、喷雾剂等产品形态。在具体的产品形态方面,可以是如柔肤化妆水、营养化妆水、营养霜、按摩霜、精华液、眼霜、洁面霜、洁面泡沫、洁面水、面膜、喷雾剂或粉末剂型等。
适用本发明的化妆品组合物中除其有效成分之外还可以包含在化妆品组合物中普遍使用的成分,如稳定剂、增溶剂、表面活性剂、维生素、色素以及香料等一般的辅助剂以及载体。
当本发明的剂型为糊剂、霜剂或凝胶剂时,作为载体成分,可以使用如动物油、植物油、蜡、石蜡、淀粉、黄芪胶、纤维素衍生物、聚乙二醇、硅树脂、膨润土、二氧化硅、滑石粉或氧化锌等。
当本发明的剂型为粉末或喷雾剂时,作为载体成分,可以使用乳糖、滑石粉、二氧化硅、氢氧化铝、硅酸钙或聚酰胺粉末,尤其是当剂型为喷雾剂时,还可以追加包含如氯氟烃、丙烷/丁烷或二甲醚等推进剂。
当本发明的剂型为溶液或乳浊液时,作为载体成分,可以使用如溶剂、增溶剂或乳浊化剂,具体来讲可以使用如水、乙醇、异丙醇、碳酸乙酯、乙酸乙酯、苯甲醇、苯甲酸苄酯、丙二醇、1,3-丁基乙二醇油、甘油脂族酯、聚乙二醇、脱水山梨糖醇的脂肪酸酯等。
当本发明的剂型为悬浊液时,作为载体成分,可以使用如水,乙醇或聚乙二醇等液状的稀释剂、如乙氧基化异硬脂醇,聚氧乙烯山梨糖醇酯,聚氧乙烯脱水山梨糖醇酯等悬浊剂、微晶纤维素、偏氢氧化铝(Aluminium metahydroxide)、膨润土、琼脂等。
当本发明的剂型为含表面活性剂的洁面剂时,作为载体成分,可以使用如脂肪醇硫酸盐、脂肪醇醚硫酸盐、磺基琥珀酸单酯、羟乙基磺酸盐、咪唑啉衍生物、甲基牛磺酸酯、肌氨酸盐、脂肪酸酰胺醚硫酸盐、烷基酰胺甜菜碱、脂肪醇、脂肪酸甘油酯、脂肪酸二乙醇酰胺、植物油、羊毛脂衍生物或乙氧基化甘油脂肪酸酯等。
本发明的化妆品组合物,除了包含呈现出生发促进或性等的所述有效成分之外,可以按照相关行业通常所采用的化妆品组合物的制造方法进行自造。
如上所述,本发明可以提供一种利用水溶化乳香胶以及山胡椒提取物的生发促进或脱发抑制用组合物。本发明的生发促进或脱发抑制用组合物可以被产品化成如功能性食品、药品以及功能性化妆品等。

具体实施方式
接下来,将结合实施例以及试验例对本发明进行说明。但是,本发明的范围并不因为下述的实施例以及试验例而受到限定。
<实施例>水溶化乳香胶与山胡椒提取物混合物的生发促进或脱发抑制功效
1.试料的准备
1.1水溶化乳香胶的准备
通过对乳香胶粉末进行肉眼检查,执行了颜色确认以及异物检查。接下来,在投入经过筛选的乳香胶粉末20重量%、纯净水75.7重量%、环糊精4重量%以及添加剂(印度树胶)0.3重量%之后,利用均质混合器(KNS Company株式会社)在30~50℃下以2,000~3,000rpm进行30~60分钟的分散,从而使其粒子大小达到大约20~50μm。利用球磨机(DNTek株式会社)将所述乳香胶的分散液在20~40℃下执行3~6小时的分散,从而使其最终分散粒子大小达到平均600~1,000nm。接下来,利用40~60μm的过滤器进行过滤,从而获得悬浮液状态的乳香胶水溶液。在接下来的试验中,直接使用了所述乳香胶。
1.2山胡椒提取物的准备
将山胡椒(提取部位:叶片,细枝)利用4~8倍重量的70%酒精在70~85℃5下执行4~8小时的第一次提取,接下来将过滤的滤液在60℃以下执行减圧浓缩之后冻结干燥,从而获得粉末状的提取物。
2.生发促进或脱发抑制功效试验
2.1毛斧头细胞的增殖功效
利用包含1%胎牛血清(FBS,Fetal Bovine Serum)的杜氏改良伊格尔氏培养基(DMEM)将毛乳头细胞(Dermal papilla cell,PromoCell)以1×103/孔的浓度接种到96孔板中,并在37℃的培养器中进行24小时的培养。接下来,在按照不同的试料浓度进行处理并培养24~96小时之后,利用D-Plus CCK cell viability assay kit(DONGIN Biotech)对450nm下的吸光度进行观察,从而对不同时间的增殖率变化进行了评估。与未处理组(1%胎牛血清(FBS))相比的水溶化乳香胶相关结果如图1所示,而水溶化乳香胶与山胡椒提取物的混合物相关结果如图2所示。将米诺地尔(minoxidil,Mnx,100μM)和/或非那雄胺(finasteride,1μM)作为阳性对照组使用。
参阅图1以及图2,水溶化乳香胶以及水溶化乳香胶与山胡椒提取物的混合物均呈现出了依赖于浓度的毛乳头细胞增殖促进效果。
2.2脱发相关因子即5α-还原酶的表达程度测定
利用包含1%胎牛血清(FBS,Fetal Bovine Serum)的杜氏改良伊格尔氏培养基(DMEM)将毛乳头细胞以1×103/孔的浓度接种到96孔板中,并在37℃的培养器中进行24小时的培养,接下来,在按照不同的试料浓度进行处理并培养48小时以及72小时之后,对培养液进行回收并利用ELISA assay(Human 5AR ELISA kit,Cusabio)对影响脱发的5α-还原酶(5α-reductase,5AR)的表达量进行了评估。与未处理组(1%胎牛血清(FBS))相比的水溶化乳香胶相关结果如图3所示,而水溶化乳香胶与山胡椒提取物的混合物相关结果如图4所示。此时,同样将米诺地尔(minoxidil,Mnx,100μM)和/或非那雄胺(finasteride,1μM)作为阳性对照组使用。
参阅图3以及图4,水溶化乳香胶以及水溶化乳香胶与山胡椒提取物的混合物均依赖于浓度对5α-还原酶的表达进行了抑制。
2.3统计分析
按照不同的时间,对未处理组、米诺地尔、非那雄胺以及乳香胶、山胡椒提取物、混合物处理组的毛乳头细胞增殖功效以及5α-还原酶表达变化率进行了比较。为了对统计学意义进行验证而执行了独立t-试验(Independent t-test),并将其换算成p-值(p-value)进行了图示。如上所述的所有统计处理是利用Excel程序进行了分析。

1.一种生发促进或脱发抑制用组合物,其特征在于:
作为有效成分包括(i)水溶化乳香胶或(ii)所述水溶化乳香胶与山胡椒提取物的混合物。
2.根据权利要求1所述的生发促进或脱发抑制用组合物,其特征在于:所述山胡椒提取物为水、乙醇或所述的混合溶剂的提取物。
3.根据权利要求1或权利要求2所述的生发促进或脱发抑制用组合物,其特征在于:
所述组合物为食品组合物。
4.根据权利要求1或权利要求2所述的生发促进或脱发抑制用组合物,其特征在于:
所述组合物为药剂学组合物。
5.根据权利要求1或权利要求2所述的生发促进或脱发抑制用组合物,其特征在于:
所述组合物为化妆品组合物。
