背景技术
黑色素(Melanin)主要由人体皮肤表皮基底层的黑色素细胞(Melanocyte)产生,它能减小紫外线对皮肤的伤害。黑色素的含量及分布决定了皮肤的颜色。但是,如果因过度暴露于紫外线、空气污染、压力等外界环境而在皮肤中生成过度,则会发生色素沉着现象,从而成为皮肤变黑或暗斑、雀斑等的原因。紫外线通过促进作为黑色素生成细胞的黑素细胞的活性、促进黑色素生物合成刺激激素的分泌、促进黑色素的氧化或促进酪氨酸酶的活性等,成为诱导黑色素生成过度的主要因素。
皮肤干燥或者油脂分泌过于旺盛都是常见的皮肤问题;皮肤保湿能力的好坏,能够反应出皮肤屏障功能的正常与否。皮肤保湿能力降低导致皮肤屏障功能下降,皮肤屏障功能下降又进一步降低皮肤保湿能力,造成恶性循环。当肌肤缺水时,细胞代谢和信息传递等受到影响,甚至出现障碍,皮肤的生理结构发生变化,如干燥,粗糙,失去光泽,弹性降低,皱纹增多或加深,色素增多,最糟糕的是缺水引起皮肤屏障功能障碍,不但增加了皮肤病的易感性,而且可诱发和加重一些皮肤病,例如皮炎,湿疹,银屑病及痤疮。而皮肤油脂分泌过于旺盛,同样也会照成不良后果。

步骤(2)中,原料组、水和纤维素酶的质量比为100:2-4:0.1-0.3。
步骤(2)中,酶解温度为40-55℃,酶解时间为1.5-3h。
步骤(3)和步骤(4)中,夹带剂为75-85(V/V%)的乙醇水溶液。
步骤(3)中,夹带剂的添加量为原料组质量的1.2-1.8倍;浸泡时间为6-10h。
步骤(4)中,夹带剂流速为2.0-3.0ml/min;CO2流体压力为160-220bar;CO2流量为3.5-4.5L/min;萃取温度为30-45℃;萃取时间为3-4h。
本发明的第二个目的,在于提供一种美白淡斑组合物,以质量百分比计算包括以下成分:
特别地,所述乳化剂为PPG-2-鲸蜡硬脂醇聚醚-9、鲸蜡硬脂醇聚醚-21、油醇聚醚-10中的至少一种。
特别地,所述植物油脂为霍巴巴油、白池花籽油中的至少一种。
本发明的第三个目的,在于提供前述美白淡斑组合物的制备方法,包括以下步骤:
(1)制备植物组合物;
(2)将甘油、植物油脂、植物组合物混合均匀,得油相;
(3)将水、海藻糖、乳化剂丙二醇混合均匀,得水相;
(4)油相、水相混合并均质。
所述步骤(2)和步骤(3)中,混合速率为400-800rpm,时间为10-20min。
所述步骤(4)中,均质速率为1200-1500rpm,时间为4-6min。
与现有技术相比,本发明的有益效果为:
(1)本发明特别选取的三种植物原料,其中马黛茶(巴拉圭草)含有丰富的咖啡因、多酚类物质和多种维生素和微量元素;本发明研究发现,三种植物原料的提取物具有抗菌消炎、美白的效果;此外还能与其他已经证明其美白效果的成分协同,进一步增强其美白效果。
(2)本发明所用的植物原料来源广泛,不属于《国家重点保护野生植物名录》内的物种;且不含过敏源,适用范围广。
(3)本发明的植物组合物通过多种活性成分协同,达到温和美白的效果。
(4)本发明的美白淡斑组合物的制备方法除了美白效果外,还具有平衡油脂分泌,保湿的效果。

具体实施方式
为了更好地理解本发明,下面结合具体实施例对本发明作进一步的描述,其中实施例中使用的术语是为了描述特定的具体实施方案,不构成对本发明保护范围的限制。
本发明中:
巴拉圭草为冬青科巴拉圭冬青Ilex paraguariensis.的干燥叶子,又名马黛茶。
杜松子为柏科杜松JuniperusrigidaS.etZ.或欧洲刺柏uniperus communis L.的成熟果实。
广藿香为唇形科植物广藿香Pogostemon cablin(Blanco)Benth.的干燥地上部分。
实施例一植物提取物的制备
植物提取物的制备,包括以下步骤:
(1)按下表1称取对应的植物原料,然后混合,研磨至≥12目,得原料组;
(2)原料组加入水和纤维素酶进行酶解;
(3)酶解后加入夹带剂浸泡;
(4)加入夹带剂CO2超临界萃取,收集萃取液脱溶剂处理,得巴拉圭草提取物、杜松子提取物和广藿香提取物混合的植物提取物。
步骤(2)、(3)、(4)的工艺参数如下表1所示。

对实施例一制备的植物提取物进行安全性测试,具体为:
将制备的植物提取物溶解于DMSO(Dimethyl sulfoxide,二甲基亚砜)后,处理在皮肤细胞(HaCaT)上,以确认是否影响细胞生物活性。将浓度为2×105细胞/mL的皮肤细胞分别以100μL分株到96孔细胞培养板上,并进行24小时的培养后,在其上用10μg/mL至100μg/mL浓度的花津浦芽孢杆菌菌株的提取物进行处理,再进行24小时的培养。
对于每个序号,重复实验3次。利用MTT分析法来比较细胞的生物活性,基于将未处理组的生物活性视为100%,表示提取物处理组的生物活性。
其结果显示对皮肤细胞的生长没有影响,从而对皮肤细胞具有安全性,如图1所示。
对实施例一制备的植物提取物进行美白效果测试,具体为:
具体地,将制备的植物提取物处理在黑色素生成细胞(B16黑色素瘤细胞)上,以评估美白效果。
将浓度为4×104细胞/孔的黑色素生成细胞分株到24孔细胞培养板上,然后进行24小时的培养,以用于实验。
为了诱导黑色素的生成,用1μM的α-MSH(α-Melanocyte-stimulating hormone,α-黑素细胞刺激素)进行处理,同时用不同序号的植物提取物进行处理。重复实验3次。再进行72小时的培养后,测量405nm处的吸光度基于将诱导黑色素生成的组的吸光度视为100%,用%来表示植物提取物处理组的诱导黑色素生成的程度;结果如图2所示。
实施例二植物组合物的制备
在实施例一的基础上,添加下表2中所示的成分,得到对应的植物组合物。

将表2中的植物组合物用DMSO稀释成10%的体积浓度;以实施例一相同的美白测试方法进行测试,结果如下图3所示。
实施例三美白淡斑组合物的制备
美白淡斑组合物的制备的制备,包括以下步骤:
(1)按下表2称取对应的原料,备用;
(2)将甘油、植物油脂、植物组合物混合均匀,得油相;
(3)将水、海藻糖、乳化剂丙二醇混合均匀,得水相;
(4)油相、水相混合并均质。
步骤(1)(2)、(3)、(4)的工艺参数如下表3所示。

对实施例三的成品进行美白测试。
实验原理
采用Lab色度系统测量皮肤颜色的变化。测试指标为皮肤黑色素含量MI值、皮肤白度/亮度L值和ITA°。
△MI=Tn-T0;式中Tn为受试区域随时间变化数值;T0为受试区域起始值。
△L*=Tn-T0;式中Tn为受试区域随时间变化数值;T0为受试区域起始值。
△ITA°=Tn-T0;式中Tn为受试区域随时间变化数值;T0为受试区域起始值。
实验仪器
多功能皮肤测试系统(MPA580,德国CK);
皮肤色差测试探头及(CL400,德国CK);
皮肤红黑色素测试仪及测试探头(Mexameter MX18,德国CK)。
实验方法
招募24-50岁志愿者130人,签订知情同意书;随机分为7组,每组20名作为试用者,其中6组为实验组,第7组为对照组,对照组的样品为实施例二中,序号7产品不添加植物组合物,用水补齐100%的不含美白成分的基础护肤品。
试用样品前对其面部皮肤进行黑色素和色差数据采集分析测试,数据作为皮肤本底值。试用者早晚各使用一次样品,连续使用4周。分别于第0周、第4周、第6周时进行数据采集。评测受试者使用化妆品前后皮肤白度(亮度)和皮肤黑色素含量的变化。结果如下表4、表5、表6所示。

表4、5、结果显示,本方具有明显的美白效果。

Claims (10)
2.根据权利要求1所述的植物组合物,其特征在于,所述巴拉圭草提取物、杜松子提取物、广藿香提取物的制备方法为:
(1)将巴拉圭草、杜松子、广藿香混合后研磨,得原料组;
(2)原料组加入水和纤维素酶进行酶解;
(3)酶解后加入夹带剂浸泡;
(4)加入夹带剂CO2超临界萃取,收集萃取液脱溶剂处理,得巴拉圭草提取物、杜松子提取物和广藿香提取物的混合物;
步骤(2)中,原料组、水和纤维素酶的质量比为100:2-4:0.1-0.3;
步骤(3)和步骤(4)中,夹带剂为75-85(V/V%)的乙醇水溶液;
步骤(3)中,夹带剂的添加量为原料组质量的1.2-1.8倍;浸泡时间为6-10h。
3.根据权利要求2所述的植物组合物,其特征在于,步骤(2)中,酶解温度为40-55℃,酶解时间为1.5-3h。
4.根据权利要求2所述的植物组合物,其特征在于,步骤(4)中,夹带剂流速为2.0-3.0ml/min;CO2流体压力为160-220bar;CO2流量为3.5-4.5L/min;萃取温度为30-45℃;萃取时间为3-4h。
6.根据权利要求5所述的美白淡斑组合物,其特征在于,所述乳化剂为PPG-2-鲸蜡硬脂醇聚醚-9、鲸蜡硬脂醇聚醚-21、油醇聚醚-10中的至少一种。
7.根据权利要求5所述的美白淡斑组合物,其特征在于,所述植物油脂为霍巴巴油、白池花籽油中的至少一种。
8.权利要求5-7任一所述的美白淡斑组合物的制备方法,其特征在于,包括以下步骤:
(1)制备植物组合物;
(2)将甘油、植物油脂、植物组合物混合均匀,得油相;
(3)将水、海藻糖、乳化剂丙二醇混合均匀,得水相;
(4)油相、水相混合并均质。
9.根据权利要求8所述的美白淡斑组合物的制备方法,其特征在于,所述步骤(2)和步骤(3)中,混合速率为400-800rpm,时间为10-20min。
10.根据权利要求8所述的美白淡斑组合物的制备方法,其特征在于,所述步骤(4)中,均质速率为1200-1500rpm,时间为4-6min。
