涉及一种用于激活和增幅NK细胞的经基因改造的细胞株以及其用途,更具体地,涉及一种使用所述细胞株增殖NK细胞的方法。
背景技术
NK细胞(自然杀伤细胞;natural killer cell)是一种对先天免疫很重要的细胞毒性淋巴细胞。NK细胞对病毒感染的细胞或癌细胞有反应,这些反应取决于NK细胞的多个激活受体(activating receptor)和抑制受体(inhibitory receptor)与靶细胞中各自的配体(ligand)反应而发出的信号(signal)。通常,当激活信号强于抑制信号时,NK细胞可以攻击靶细胞,但当抑制信号更强时,NK细胞不作出攻击。因此,正常细胞不会受到NK细胞的攻击,因为NK细胞的抑制受体配体(MHC;major histocompatibility complex)存在并发出抑制信号。
癌细胞中的一部分在细胞表面的MHC发生异常而可能会减少。在这种情况下,NK细胞没有抑制信号,因此NK细胞攻击靶细胞。NK细胞分泌穿孔素(perforin)刺破感染细胞或癌细胞的细胞膜,并具有释放颗粒酶(granzyme)来杀死这些细胞的细胞毒性。在具有B细胞淋巴瘤和许多其他癌症患者的情况下,发现NK细胞的数量或抗癌活性存在缺陷,并已知NK细胞功能异常与这些癌症的发生密切相关。
因此,利用这些NK细胞的抗癌能力对癌症患者进行治疗的方法正在兴起,而开发一种用于确保大量的高性能NK细胞的增幅方法很重要。此外,已知具有NK细胞功能下降或异常的人与癌症和各种疾病有关,因此需要开发一种测量NK细胞的活性的方法。
现时被报道的用于增殖NK细胞的方法是使用各种高浓度的昂贵细胞因子或同时使用外周血单核细胞或癌细胞株的方法。但是,这需要很高的成本,并且增幅率不是很高。此外,在许多研究中,除了NK细胞之外,也伴随着诸如T细胞等的其他淋巴细胞的增幅,因此也显示出在基于NK细胞的免疫细胞治疗中不适合的方面。因此,开发一种用于仅选择性增幅NK细胞的有效方法是一项重要问题,虽然已经对此进行了研究(韩国注册专利10-1525199),但仍不完善。
发明内容
技术问题
一方面提供一种细胞株或饲养细胞(feeder cell),其用于培养NK细胞,其经基因改造以表达膜结合白细胞介素-18(membrane bound interleukin-18:mbIL-18)、膜结合白细胞介素-21(membrane bound interleukin-21:mbIL-21)和/或OX40L。
另一方面提供一种增殖NK细胞的方法,包括收获含有混合的NK细胞群(mixedimmune cells)的血液样品;以及使所述NK细胞群的至少一部分与用于激活NK细胞的经基因改造的细胞接触,其中,所述经基因改造的细胞经基因改造以表达膜结合白细胞介素-18(membrane bound interleukin-18:mbIL-18)、膜结合白细胞介素-21(membrane boundinterleukin-21:mbIL-21)和/或OX40L。

所述细胞株可用作仅选择性增幅NK细胞的饲养细胞。一般而言,作为用于NK细胞增殖的具有代表性的饲养细胞的K562细胞株,其仅用于NK细胞的增殖,因此K562细胞株本身不应增殖。因此,在与NK细胞一起培养之前,通过进行对所述K562细胞株照射强放射线(例如,50Gy至100Gy)的预处理,使所述K562细胞株本身完全不增殖,而仅有助于NK细胞的增殖。一方面,为了NK细胞的有效增殖,大多数细胞因子如IL-2和IL-15必须持续露出于NK细胞。因此,当进行基因改造在基于癌细胞株的饲养细胞中以表达细胞因子如IL-2和/或IL-15时,经放射线照射等预处理的癌细胞株在培养过程中不能长期存活,存在NK细胞的选择性增殖效率降低的问题。在一实施例中,当NK细胞在K562-OX40L存在下短期露出于IL-18或IL-21时,细胞增幅效果直到第21天也没有出现,但当同时露出于IL-18和IL-21时,确认细胞增幅效果造在28天后出现。因此,在IL-18和IL-21的情况下,尽管在培养开始日只有一次露出和短期露出,但对NK细胞的增殖有效果。换言之,在根据一方面的经基因改造以表达mbIL-18和mbIL-21的饲养细胞株中,尽管IL-18和IL-21的短期表达,NK细胞增殖的效率并未降低,而仅NK细胞可以被选择性增幅。

根据一具体实施例的用于培养NK细胞的组合物,其包括经基因改造以表达mbIL-18、mbIL-21和OX40L的细胞,且可以诱导NK细胞(例如,自然杀伤细胞)的增幅和/或激活,因此可有效用于NK细胞的培养、分离或增殖等。
另一方面提供一种增殖NK细胞的方法,包括收获含有混合的NK细胞(mixedimmune cells)群的血液样品;以及使所述混合的NK细胞群的至少一部分与用于激活NK细胞的经基因改造的细胞接触,其中,所述经基因改造的细胞经基因改造以表达膜结合白细胞介素-18(membrane bound interleukin-18:mbIL-18)、膜结合白细胞介素-21(membranebound interleukin-21:mbIL-21)和/或OX40L。
在一具体实施例中,使接触的步骤包括共培养与所述经基因改造的细胞混合的NK细胞群以刺激、激活或增幅(expanding)所述NK细胞的亚群(subpopulation)。

在下文中,公开优选的实施例以帮助理解本发明。然而,提供以下实施例只是为了更容易理解本发明,本发明的内容不受以下实施例的限制。
[实施例]
实施例1.制备经基因改造的饲养细胞(feeder cell)
制备饲养细胞,其表达膜结合白细胞介素-18(membrane bound interleukin-18:mbIL-18)、膜结合白细胞介素-21(membrane bound interleukin-21:mbIL-21)和OX40L。具体地,将人源mbIL18基因(SEQ ID NO:1)和mbIL21基因(SEQ ID NO:2)克隆到慢病毒载体pCDH-CMV-RFP中,从而制备重组慢病毒生产用载体。然后,通过使用lipofectamin3000(Invitrogen)和包装载体将所制备的重组基因(pCDH-CMV-RFP-mbIL-1821)转染到293FT细胞。一段时间后换上新鲜培养基,培养细胞48小时,然后回收含有病毒的培养基。将所回收的培养基以500×g离心10分钟,并使用0.45μm的过滤器仅分离含有病毒的纯培养基,从而生产表达mbIL18-mbIL21的慢病毒。然后,将1ml的表达mbIL18-mbIL21的慢病毒溶解在9ml的包括以下比较例1的K562-OX40L细胞的培养基中,并加入聚凝胺(polybrene)(8μg/ml),从而进行细胞培养48小时。接着,为了仅选择受感染的细胞,使用高速自动细胞分选仪仅选择表达绿色和红色荧光的细胞,并通过荧光激活细胞分类(FACS;fluorescence-activatedcell sorting)仪分析有否表达mbIL18-mbIL21,从而生产表达mbIL18-mbIL21的K562细胞株(在下文称为“K562-OX40L-mbIL18-mbIL21”)。

制备表达OX40L的饲养细胞。具体地,将人源OX40L基因(SEQ ID NO:3)克隆到作为慢病毒载体的pLVX-IRES-ZsGreen中,从而制备用于重组慢病毒生产用载体。然后,为了生产病毒,将所述重组基因(OX40L-pLVX-IRES-ZsGreen)通过使用lipofectamin3000(Invitrogen)和包装载体转染到293FT细胞中。一段时间后换上新鲜培养基,培养细胞48小时,然后回收含有病毒的培养基。将所回收的培养基以500×g离心10分钟,并使用0.45μm的过滤器仅分离含有病毒的纯培养基,从而生产表达OX40L的慢病毒。之后,将1ml的表达OX40L的慢病毒溶解在9ml的包括K562细胞的培养基(RPMI 1640)中,并加入聚凝胺(polybrene)(8μg/ml),从而进行细胞培养48小时。接着,为了仅选择受感染的细胞,使用高速自动细胞分选仪仅选择表达绿色荧光的细胞,并通过FACS分析有否表达OX40L,从而生产表达OX40L的K562细胞株(在下文称为“K562-OX40L”)。
[试验例]
试验例1.经基因改造的饲养细胞的细胞特性
为了确认根据一方面的经基因改造的饲养细胞的mbIL-18和mbIL-21的表达特性,确认mRNA和表面蛋白的表达水平。
具体地,通过使用RNeasy迷你试剂盒(Mini Kit)(Qiagen,芬洛,荷兰)分离在所述实施例1中制备的K562-OX40L-mbIL18-mbIL21细胞株和在比较例1中制备的K562-OX40L细胞株的总RNA,并且使用IMPLEN超微量分光光度计(Nanophotometer)P330(IMPLEN,慕尼黑,德国)进行量化。之后,使用QuantiTect反转录试剂盒(Reverse Transcription Kit)(Qiagen)将分离的RNA转换为cDNA,然后使用QuantiTect SYBR绿PCR试剂盒(Qiagen)和Rotor-Gene Q(Qiagen)在标准20ml的反应体积中进行聚合酶链式反应(PCR;PolymeraseChain Reaction)。在此,使用下表1中的引物,并且所有试验重复实施3次。

图1a是确认根据一方面的饲养细胞中的mbIL-18和mbIL-21的表达特性的图表。
图1b是根据一方面的饲养细胞与mIL-18和mIL-21的组合图像(左-)以及通过荧光强度量化mIL-18和mIL-21的量的图表(右)。
结果,如图1a-图1b所示,在实施例1的饲养细胞中检测到mbIL-18和mbIL-21mRNA,但在比较例1的饲养细胞中检测不到mbIL-18和mbIL-21mRNA。另外,如图1b所示,可以确认出,与比较例1的饲养细胞相比,实施例1的饲养细胞表面持续表达更大量的IL-18和IL-21。
试验例2.通过添加K562-OX40L细胞、IL18和IL21确认NK细胞激活
为了预测根据一方面的经基因改造的饲养细胞的NK细胞激活效能,在预备试验中确认在根据K562-OX40L细胞中添加细胞因子的NK细胞激活程度。
具体地,在提供有5%的CO2的培养基中,将在所述比较例1中制备的K562-OX40L细胞株培养于T-25烧瓶中,所述烧瓶为37℃且装有25ml的完全RPMI 1640培养基(含有胎牛血清(FBS;Fetal Bovine Serum)、青霉素和链霉素)。此后,在400×g条件下离心3分钟,并将细胞沉淀物重悬于5ml的完全RPMI 1640培养基中以收获饲养细胞(feeder cell)。之后,为了防止所述饲养细胞过度生长,使用Gammacell 3000Elan辐射器以100Gy的伽马射线照射所述饲养细胞并用于后续试验。
为了分离外周血单核细胞(PBMCs;peripheral blood mononuclear cells),将正常供体的全血和PBS以1:2比率(10ml的全血:20ml的PBS)稀释,并覆盖在15ml的人淋巴细胞分离液(Lymphoprep)上。接着,在室温下以1200×g的条件下无刹车离心25分钟(加速1,减速0),并从棕黄层(buffy coat layer)收获细胞,且在400×g下离心7分钟的同时,用PBS洗涤3次。将3×106个的所分离的外周血单核细胞和0.5×106个经100Gy-照射的K562-OX40L接种到包括1ml的NK细胞培养基的24孔板上,然后加入含有20U/ml的IL-2的1ml的NK细胞培养基,使培养基总体积为2ml/孔且IL-2的终浓度为10U/ml,然后轻轻吸取(pipetting)且混合后,在37℃下且5%的CO2培养基中进行培养。培养第7天,向培养基中加入100U/ml的IL-2和5ng/ml的IL-15,并在每2天至3天更换一次新培养基的同时,进行培养14天。此后,在培养的第0天加入5ng/ml的IL-21和/或25ng/ml、50ng/ml、100ng/ml的IL-18,并进一步培养14天。
NK细胞的增幅通过使用异硫氰基荧光素(FITC;Fluorescein isothiocyanate)-偶联的小鼠抗人CD3和PE-Cy5-偶联的小鼠抗人CD56单克隆抗体进行确认,并且当用K562细胞和IL-2/IL-15处理时,以NK细胞的纯度作为基线(baseline)确认每个扩增倍数(Foldexpansion)。
图2a是显示当NK细胞在K562-OX40L细胞存在下短时间露出于单独的IL-21或IL-18和IL-21时NK细胞的扩增倍数(Fold expansion)的图表。
图2b是显示当NK细胞在K562-OX40L细胞存在下短时间露出于单独的IL-21或IL-18和IL-21时NK细胞的纯度的图表。
结果,如图2a所示,NK细胞短期露出于IL-18和IL-21的影响直到第21天还未被确定。然而,NK细胞的增幅被确认在第28天后显着增加。另外,如图2b所示,可以确认出,相比于仅单独处理IL-21,在第28天,短时间露出于Il-18和IL-21两者的NK细胞的NK细胞纯度更高。
图2c是显示当NK细胞在K562-OX40L细胞存在下长时间露出于单独的IL-21或IL-18和IL-21时NK细胞的扩增倍数的图表。
结果,如图2c所示,可以确认出,与分别处理IL-18或IL-21时相比,当用IL-18和IL-21一起处理K562-OX40L时,NK细胞的活性增加了约2倍至4倍。
这些结果表明,表达OX40L的细胞不仅显着增加NK细胞的活性,而且在短时间内通过额外的IL-18和IL-21处理协同增加NK细胞的活性。换言之,表达OX40L的饲养细胞可以通过用IL-18和IL-21处理对NK细胞的活性表现出协同效果。
试验例3.通过K562-OX40L-mbIL18-mbIL21细胞的NK细胞激活和增幅
基于所述试验例2的结果,确认了在所述实施例1中制备的K562-OX40L-mbIL18-mbIL21细胞对NK细胞活性的影响。
具体地,在提供有5%CO2的培养基中,将所述实施例1的K562-OX40L-mbIL18-mbIL21细胞株在T-25烧瓶中进行培养,所述烧瓶为37℃且装有25ml的完全RPMI1640培养基。此后,在400×g条件下离心3分钟,并将细胞沉淀物重悬于5ml的完全RPMI 1640培养基中以收获饲养细胞(feeder cell)。之后,为了防止所述饲养细胞过度生长,使用Gammacell3000Elan辐射器以100Gy的伽马射线照射所述饲养细胞并用于后续试验。以与所述试验例2相同的方法,将3×106个分离的外周血单核细胞和0.5×106个经100Gy照射的K562-OX40L-mbIL18-mbIL21细胞接种到包括1ml的NK细胞培养基的24孔板上,然后加入含有20U/ml的IL-2的1ml的NK细胞培养基,使培养基总体积为2ml/孔且IL-2终浓度为10U/ml,然后然后轻轻吸取且混合后,在37℃下且5%的CO2培养基中进行培养。培养第7天,向培养基中加入100U/ml的IL-2和5ng/ml的IL-15,并在每2天至3天更换一次新培养基的同时,进行培养14天。作为对照组,使用K562细胞。通过以与所述试验例2中相同的方法确认NK细胞的纯度和扩增倍数来确认NK细胞的激活程度。
图3是比较使用根据一方面的饲养细胞的NK细胞的增幅(左)和纯度(右)的图表。
结果,如图3所示,可以确认出,与对照组相比,实施例1的饲养细胞显着增加NK细胞的扩增倍数。特别地,在对照组的情况下,NK细胞在培养后增加至第35天后都没有显示出任何进一步的增加。然而,在实施例1的情况下,在培养后到第42天显示出显着增加,之后显示出减少的模式,但可以确认能够持续增幅。此外,可以确认出,使用实施例1的饲养细胞培养的NK细胞的纯度显着高于使用对照组培养的NK细胞的纯度。
即,根据一方面的饲养细胞不仅显着增加NK细胞的活性,而且允许NK细胞的长期培养,使得NK细胞可以大量增殖并用作细胞治疗剂。
试验例4.通过K562-OX40L-mbIL18-mbIL2细胞增幅的NK细胞的表型特性
确认了通过K562-OX40L-mbIL18-mbIL2细胞增幅的NK细胞的表型特性。具体地,在用FACS缓冲液(含有1%FBS的PBS)洗涤在所述试验例2中增幅的来自正常供体(HD)的2×105个NK细胞后,用APC-Cyanine7-偶联的小鼠抗人CD3和PE-Cyanine7-偶联的抗人CD56膜抗体处理15分钟。然后,收获细胞,并用分别不同的荧光-偶联的抗人CD16、CD69、NKG2D、NKp30、NKp44、NKp46、CD94、CD158a和CD158b膜抗体进一步染色30分钟。之后,用FACS缓冲液洗涤所述细胞,然后使用FACS Calibur获取数据,并用Kaluza分析。
图4a和图4b是比较通过使用根据一方面的饲养细胞从正常供体(HD)增幅的NK细胞中表达的标志物的图表。
结果,如图4a-图4b所示,通过表达OX40L、mbIL18和mbIL21的饲养细胞增幅的NK细胞不仅表现出高NK纯度(>90%)),而且与培养的初始阶段相比,在第14天显示出NK细胞激活标志物的表达增加。
即,可以看出通过根据一方面的饲养细胞增幅的NK细胞具有NK细胞的完整功能特性。
试验例5.确认通过K562-OX40L-mbIL18-mbIL2细胞增幅的NK细胞的激活程度
随着激活受体的表达增加,增幅的NK细胞的激活也一定要增加,因此,评估了NK细胞的细胞因子释放和通过细胞毒性的免疫调节活性,所述NK细胞通过使用实施例1的饲养细胞增幅并来源于正常供体(HD)。
5-1.细胞内IFN-γ测量
为了评估免疫调节活性,通过测量细胞内IFN-γ来评估细胞的细胞毒性。具体而言,在布雷非德菌素A(brefeldin A)(BD生物科学;BD Biosciences)和莫能菌素(BD生物科学)的存在下,将使用实施例1的饲养细胞增幅且来自正常供体(HD)的2×105个NK细胞在37℃和5%CO2的条件下在96孔圆底板中培养5小时。然后,收获细胞,用FACS洗涤,然后用抗人CD3和CD56膜抗体染色20分钟。在洗涤、固定和穿透后,将NK细胞在冰上进一步用PE偶联的抗人IFN-γ抗体染色30分钟。此后,洗涤所述细胞并通过使用FACS Calibur流式细胞仪进行分析。
图5a是确认通过使用根据一方面的饲养细胞从正常供体(HD)增幅的NK细胞中表达的细胞因子的图表。
结果,如图5a所示,可以确认出,来自正常供体的NK细胞中的IFN-γ的表达水平在NK细胞增幅的同时显着增加。
5-2.CD107a脱粒和细胞毒性的确认
将2×105个的在所述5-1中增幅的来自正常供体(HD)的NK细胞、2×105个靶细胞(K562、U266、RPMI8226)以及PE偶联的抗人CD107a在96孔圆底板(96-well U-bottomplate)培养。1小时后,加入莫能菌素和布雷非德菌素A(BD生物科学),然后进一步培养4小时。然后,通过用抗人CD3和CD56抗体染色来获得NK细胞。
图5b显示通过使用根据一方面的饲养细胞从正常供体(HD)增幅的NK细胞的CD107a脱粒分析结果的图表。
结果,如图5b所示,可以确认出,与培养初期(第0天)相比,14天后,在与来自正常供体的NK细胞一起培养的所有细胞(K562、U266、RPMI8226)中,CD107a的表达显着增加。
即,这是指从正常供体的外周血单核细胞中成功地生成了活性增加的NK细胞。因此,根据一具体实施例的表达OX40L、mbIL18和mbIL21的饲养细胞对于生成具有增加的活性的NK细胞是有效的。
试验例6.确认通过K562-OX40L-mbIL18-mbIL2细胞增幅的NK细胞的细胞毒性
为了确认由根据一方面的饲养细胞增幅的NK细胞作为抗体治疗剂的效能,确认细胞毒性。
具体而言,通过基于CFSE的分析测量第14天的NK细胞对靶细胞的细胞毒性4小时。具体而言,靶细胞在FACS缓冲液中用0.5μm的羧基荧光素琥珀酰亚胺酯(CFSE;carboxyfluorescein succinimidyl ester)在37℃下染色10分钟,然后用完全培养基洗涤两次。然后,将5×104个靶细胞三重置于96孔圆底板中,并分别以0.5:1、1:1和2:1的效能细胞对目标细胞(E:T;effector-to-target)的比率将来自正常供体(HD)的NK细胞和来自具有多发性骨髓瘤(MM;Multiple myeloma)患者的的NK细胞混合。将板以1,500rpm离心3分钟,然后在37℃下在5%CO2的培养基中培养4小时。此后,然后将混合的细胞转移到FACS管中,并将1μl的1mg/mL的碘化丙啶(PI;propidium iodide)(SigmaAldrich,圣路易斯,密苏里州,美国)添加到每个管中。然后通过使用FACS Calibur收获细胞并使用Kaluza软件进行分析。在减去自发死亡的靶细胞的百分比后,计算死亡的靶细胞之百分比(CFSF阳性和PI阳性)。
图6a是确认通过使用根据一方面的饲养细胞从正常供体(HD)增幅的NK细胞的细胞毒性的图表。
图6b是确认通过使用根据一方面的饲养细胞从正常供体(HD)增幅的NK细胞的ADCC细胞毒性的图表。
结果,如图6a所示,可以确认出,使用实施例1的饲养细胞增幅的NK细胞表现出与使用K562饲养细胞增幅的NK细胞相同的细胞毒性。并且,如图6b所示,可以确认出,使用实施例1的饲养细胞增幅的NK细胞在结合于利妥昔单抗的Raji细胞表现出与使用K562饲养细胞增幅的NK细胞相似的抗体依赖的细胞介导的细胞毒性作用(ADCC;antibody-dependentcell-medicated cytotoxicity)活性。
换言之,通过根据一方面的饲养细胞增幅的NK细胞表现出细胞毒性,因此可以用作抗体治疗剂。

Claims (12)
1.一种饲养细胞,其用于培养NK细胞,且经基因改造以表达膜结合白细胞介素-18、膜结合白细胞介素-21以及OX40L。
2.根据权利要求1所述的饲养细胞,其中,所述饲养细胞选自由K562、RPMI8866、EBV_LCL、721.221、HFWT以及NK-92细胞组成的组中。
3.根据权利要求1所述的饲养细胞,其包括膜结合白细胞介素-21和编码膜结合白细胞介素-21的核酸。
4.一种用于培养NK细胞的组合物,其包括根据权利要求1至3中任一项所述的饲养细胞。
5.一种增殖NK细胞的方法,包括:
收获含有NK细胞群的血液样品;以及
使混合的NK细胞群的至少一部分与用于激活NK细胞的经基因改造的细胞接触,其中,所述经基因改造的细胞经基因改造以表达膜结合白细胞介素-18、膜结合白细胞介素-21和OX40L。
6.根据权利要求5所述的方法,其中,所述接触的步骤包括共培养与所述经基因改造的细胞混合的NK细胞群以增幅NK细胞亚群。
7.根据权利要求6所述的方法,其中,所述共培养在细胞因子存在下进行。
8.根据权利要求7所述的方法,其中,所述细胞因子为选自由IL1、IL2、IL3、IL4、IL5、IL6、IL7、IL8即CXCL8、IL9、IL10、IL11、IL12、IL13、IL14、IL15、IL16、IL17、IL18、IL19、IL20、IL21、IL22、IL23、IL24、IL25、IL26、IL27、IL28、IL29、IL30、IL31、IL32、IL33、IL35、和IL36组成的组的至少一种。
9.根据权利要求8所述的方法,其中,所述细胞因子为IL-18和IL-21。
10.根据权利要求6所述的方法,其中,所述共培养进行2天至30天。
11.根据权利要求5所述的方法,其中,所述血液样品是全血样品。
12.根据权利要求5所述的方法,其中,所述经基因改造的细胞经50Gy至300Gy的放射线处理。
