Abstract
本发明公开了一种利用植物弹状病毒载体对植物染色体DNA进行定点修饰的方法及病毒载体组合物。本发明提供了对植物基因组目的DNA进行定点编辑的方法,该方法无需引入选择性标记基因,包括如下步骤:利用植物弹状病毒载体系统侵染植物并表达序列特异性的核酸酶,所述序列特异性核酸酶靶向所述被侵染植物基因组靶位点并切割靶位点,植物的DNA修复机制完成对所述靶位点的定向编辑。本发明利用植物弹状病毒表达载体递送序列特异性核酸酶,不仅提高了对靶位点的编辑效率,而且可获得非转基因的可稳定遗传的编辑植株。
