Abstract

背景技术
向圣锦等采用中药眼部敷贴联合穴位按摩治疗方案,通过比较治疗前以 及治疗后2周、4周的UCVA、屈光度、视疲劳症状积分、眼调节功能的变化, 发现中医药疗法更有利于改善视疲劳患者的调节滞后及调节灵活度,而在低 度近视,还有利于改善正相对调节(中华眼视光学与视觉科学杂志.2015), 但该研究只选择了青少年患者,疗程也较短,操作也不够便捷,具有一定局 限性;李仕明等通过对一种中药眼贴(含有野菊花、丹参、珍珠粉和薄荷脑 等成分)贴敷后,视疲劳受试者在近距离(33cm)时的调节反应显著增大, 有助于减轻视频终端视疲劳(中华眼科医学杂志,2011)。然而该研究仅观察 了使用1副眼贴后短时间内的效果,至于多次、连续使用一段时间后是否会 使得调节功能显著改善,从而有益于控制近视进展,尚有待进一步研究。此 外还有一些中药眼贴产品宣称具有缓解视疲劳效果,但缺乏有说服力的试验 数据,并且效果也单一,不能满足人们在缓解视疲劳同时改善眼部干涩和眼 部皮肤健康的需求。
基于上诉原因和前期的研究,从缓解视疲劳、改善眼部皮肤健康出 发,希望能找到一种有效中药配方及制备方法。
发明内容
本发明的目的是设计开发了一种中药凝胶眼贴,通过多种中药材的组合 配比,达到缓解眼部疲劳、清肝明目、改善眼部皮肤血液循环的作用。
本发明还设计开发了一种中药凝胶眼贴的制备方法,通过多种中药材的 配比混合,同时结合预处理技术,实现了缓解眼部疲劳、改善眼部肌肤血液 循环的作用。
本发明提供的技术方案为:
一种中药凝胶眼贴,所述中药凝胶眼贴的组分及各组分的质量份数包括:
野菊花5-15份、透骨草5-10份、决明子5-10份、珍珠1-10份、丹参1-10 份、石菖蒲1-10份、人参1-5份、红景天1-5份、黄连1-5份、番红花0.1-0.5 份、冰片0.05-0.5份、薄荷脑0.05-0.5份。
优选的是,所述中药凝胶眼贴的组分及各组分的质量份数为:
野菊花5份、透骨草5份、决明子5份、珍珠1份、丹参1份、石菖蒲 2份、人参1份、红景天1份、黄连1份、番红花0.1份、冰片0.05份、薄 荷脑0.05份。
优选的是,所述中药凝胶眼贴的组分及各组分的质量份数为:
野菊花15份、透骨草10份、决明子10份、珍珠10份、丹参10份、 石菖蒲10份、人参5份、红景天5份、黄连5份、番红花0.5份、冰片0.5 份、薄荷脑0.5份。
一种中药凝胶眼贴的制备方法,包括如下步骤:
步骤一、按照质量份数,将野菊花5-15份、透骨草5-10份、决明子5-10 份、珍珠1-10份、丹参1-10份、石菖蒲1-10份、人参1-5份、红景天1-5 份、黄连1-5份和番红花0.1-0.5份分别进行预处理后获得野菊花提取物、透 骨草提取物、决明子提取物、珍珠提取物、丹参提取物、石菖蒲提取物、人 参提取物、红景天提取物、黄连提取物和番红花提取物;
步骤二、将所述野菊花提取物、透骨草提取物、决明子提取物、珍珠提 取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提取物、黄连提取 物和番红花提取物混合均匀,45℃真空干燥至水分含量小于5%,冷却至室温 后粉碎获得提取物干粉;
步骤三、按照质量份数,取冰片0.05-0.5份和薄荷脑0.05-0.5份用无水乙 醇溶解后,喷加到所述提取物干粉上,混合均匀直至挥散乙醇,获得药物组 合物,将所述药物组合物填充至凝胶眼贴中获得中药凝胶眼贴。
优选的是,所述野菊花的预处理为:
将所述野菊花以75%乙醇作溶剂,按照料液比1:20g/ml回流提取2小 时,提取温度为60℃,回收乙醇提取液,过滤后减压蒸馏至干获得野菊花提 取物;
所述透骨草的预处理为:
将所述透骨草置于烧杯中,加入65%乙醇后,置于700W微波炉中提取 4分钟,再将提取液以转速为3500rpm离心8分钟,取上清液置于45℃干燥 箱中,浓缩至膏状获得透骨草提取物。
优选的是,所述决明子的预处理为:
将决明子粉碎后以70%乙醇提取2次,第1次1.5h,第2次1.5h,合并 提取液,过滤,滤液回收乙醇,水浴蒸干得干膏,以45℃真空低温干燥至 恒定质量获得决明子提取物;
所述珍珠的预处理为:
将珍珠研磨成粉后置于锥形瓶中,加入纯净水,煮沸13分钟后降至室温, 加入中性蛋白酶,将锥形瓶至于摇床,调节温度为43℃,转速为110rpm,酶 解190分钟后,再次加入中性蛋白酶,继续酶解200分钟,酶解结束后,将 提取液煮沸340分钟,冷却后在转速为3800rpm下离心15分钟,取上清液后 继续离心15分钟,再次取上清液,真空浓缩40℃下浓缩至干获得珍珠提取 物。
优选的是,所述丹参的预处理为:
将丹参切段后至于锥形瓶中,加入55%乙醇,超声提取20分钟,过滤, 滤液置水浴锅蒸干获得丹参提取物;
所述石菖蒲的预处理为:
将石菖蒲研磨成粉末后置于锥形瓶中,加丙酮溶解后封口,放入30℃, 150r/min的恒温振荡器上混合振荡10h,振荡后配置料液比为1:18,将其 放入35℃,超声功率为700W的超声器中,工作1.5h后取出,对得到的提取 液抽滤浓缩后获得石菖蒲提取物。
优选的是,所述番红花的预处理为:
将番红花置于55℃烘箱烘干,粉碎后,置于锥形瓶中,加入95%乙醇, 室温下磁力搅拌24h后抽滤,滤渣重复提取3次,将所得滤液合并,置于旋 转蒸发仪中浓缩至深棕色膏状物获得番红花提取物;
所述黄连的预处理为:
将黄连研磨成粉末,按料液比1:8加入75%乙醇,于50℃下超声提取 30分钟,提取3次,过滤,合并提取液,蒸干获得黄连提取物。
优选的是,所述人参的预处理为:
将人参切段后至于锥形瓶中,加入60%乙醇,超声提取30分钟,过滤, 滤液置于水浴锅中蒸干获得人参提取物;
所述红景天的预处理为:
将干燥红景天用粉碎机粉碎后过40目筛,加入8倍量的水煎煮1.5h,抽 滤后收集滤液,将分离后得到的滤渣加入6倍量的水再次进行煎煮1.5h,抽 滤后合并2次所得滤液并旋转蒸发,浓缩到原来体积的1/3获得红景天提取 物。
本发明所述的有益效果:
(1)、本发明设计开发的一种中药凝胶眼贴,采用的多种中药材之间能 够进行搭配产生协调效果,具有缓解眼部疲劳、改善眼部肌肤的作用,此组 合物原料易获得。
(2)、本发明设计开发的一种中药凝胶眼贴,由多种中药材构成的组合 物达到的效果是目前已有的化学药品所不具备的,同时本发明具有天然香料 的化学成分及抗菌化学成分,所以本组合物不用添加任何防腐剂及香精。
(3)、本发明设计开发的一种中药凝胶眼贴的制备方法,通过多种中药 材的配比混合,同时结合各种药材的预处理技术,具有组合物的药理活性的 同时体现了中药组合物的协同作用,使得制备的中药眼贴实现了缓解眼部疲 劳、改善眼部肌肤血液循环的作用。
具体实施方式
下面结合对本发明做进一步的详细说明,以令本领域技术人员参照说明 书文字能够据以实施。
本发明提供的一种中药凝胶眼贴,其组分及各组分的质量份数包括:
野菊花5-15份、透骨草5-10份、决明子5-10份、珍珠1-10份、丹参1-10 份、石菖蒲1-10份、人参1-5份、红景天1-5份、黄连1-5份、番红花0.1-0.5 份、冰片0.05-0.5份、薄荷脑0.05-0.5份。
本发明还提供一种中药凝胶眼贴的制备方法,具体包括:
步骤一、按照质量份数,将野菊花5-15份、透骨草5-10份、决明子5-10 份、珍珠1-10份、丹参1-10份、石菖蒲1-10份、人参1-5份、红景天1-5 份、黄连1-5份和番红花0.1-0.5份分别进行预处理后获得野菊花提取物、透 骨草提取物、决明子提取物、珍珠提取物、丹参提取物、石菖蒲提取物、人 参提取物、红景天提取物、黄连提取物和番红花提取物;
其中,所述预处理包括:
(1)取野菊花,以75%乙醇作溶剂,料液比为1:20g/ml,回流提取2 小时,温度为60℃,回收乙醇提取液,滤过,然后减压蒸馏至干获得野菊花 提取物。
(2)取透骨草置于烧杯中,加入65%乙醇,放在700W微波炉中提取4 分钟,再将提取液3500rpm离心8分钟,取上清液置于45℃干燥箱中,浓缩 至膏状获得透骨草提取物。
(3)取决明子粉碎后以70%乙醇提取2次,第1次1.5h,第2次1.5h, 合并提取液,过滤,滤液回收乙醇,水浴蒸干得干膏,真空低温(t=45℃) 干燥至恒定质量获得决明子提取物。
(4)取珍珠研磨成粗粉,置于锥形瓶中,加入纯净水,煮沸13分钟后 降至室温,加入中性蛋白酶,将锥形瓶至于摇床,调节温度为43℃,转速为 110rpm,酶解190分钟后,再次加入中性蛋白酶,继续酶解200分钟,酶解 结束后,将提取液煮沸340分钟,冷却后在3800rpm下离心15分钟,取上清 后继续离心15分钟,再次取上清,真空浓缩40℃下浓缩至干获得珍珠粉提 取物,其中,通过酶解得到活性成分。
(5)取丹参,切成小段,置于锥形瓶中,加入55%乙醇,超声提取20 分钟,过滤,滤液置水浴锅蒸干获得丹参提取物。
(6)取石菖蒲,研磨成粉末,置于锥形瓶中,加丙酮溶解后封口,放入30℃,150r/min的恒温振荡器上混合振荡10h,振荡后配置料液比为1:18, 将其放入35℃,超声功率为700W的超声器中,工作1.5h后取出,对得到提 取液抽滤,浓缩获得石菖蒲提取物。
(7)取番红花,置于55℃烘箱烘干,粉碎。置于锥形瓶中,加入95% 乙醇,室温下磁力搅拌24h后抽滤,滤渣重复提取3次,将所得滤液合并, 于旋转蒸发仪(45℃、0.1MPa)浓缩至深棕色膏状物获得番红花提取物。
(8)取黄连,研磨成粉末,按料液比1:8加入75%乙醇,于50℃下超 声提取30分钟,各提取3次,趁热过滤,合并提取液,蒸干获得黄连提取 物。
(9)取人参,切成1cm左右小段,至于锥形瓶中,加入60%乙醇,超 声提取30分钟,过滤,滤液置水浴锅蒸干获得人参提取物。
(10)取干燥红景天,用粉碎机粉碎后过40目筛,加入8倍量的水煎煮 1.5h,抽滤后收集滤液。将分离后得到的滤渣加入6倍量的水再次进行煎煮 1.5h,抽滤后合并2次所得滤液并旋转蒸发,浓缩到原来体积的1/3获得红景 天提取物。
步骤二、将步骤一中的所有提取物混合均匀,45℃真空干燥至水分含量 小于5%,冷却至室温后粉碎获得提取物干粉;
步骤三、按照质量份数,取冰片0.05-0.5份和薄荷脑0.05-0.5份用无水乙 醇充分溶解后,喷加到所述提取物干粉上,混合均匀直至挥散乙醇,获得药 物组合物,将所述药物组合物填充至凝胶眼贴中获得中药凝胶眼贴。
实施例1
步骤一、按照质量份数,将野菊花5份、透骨草5份、决明子5份、珍 珠1份、丹参1份、石菖蒲2份、人参1份、红景天1份、黄连1份和番 红花0.1份分别进行预处理后获得野菊花提取物、透骨草提取物、决明子提 取物、珍珠提取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提取 物、黄连提取物和番红花提取物;
步骤二、将所述野菊花提取物、透骨草提取物、决明子提取物、珍珠提 取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提取物、黄连提取 物和番红花提取物混合均匀,45℃真空干燥至水分含量小于5%,冷却至室温 后粉碎获得提取物干粉;
步骤三、按照质量份数,取冰片0.05份和薄荷脑0.05份用无水乙醇充分 溶解后,喷加到所述提取物干粉上,混合均匀并充分挥散乙醇后,获得药物 组合物,将所述药物组合物填充至凝胶眼贴中获得中药凝胶眼贴。
实施例2
步骤一、按照质量份数,将野菊花15份、透骨草10份、决明子10份、 珍珠10份、丹参10份、石菖蒲10份、人参5份、红景天5份、黄连5份和 番红花0.5份分别进行预处理后获得野菊花提取物、透骨草提取物、决明子 提取物、珍珠提取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提 取物、黄连提取物和番红花提取物;
步骤二、将所述野菊花提取物、透骨草提取物、决明子提取物、珍珠提 取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提取物、黄连提取 物和番红花提取物混合均匀,45℃真空干燥至水分含量小于5%,冷却至室温 后粉碎获得提取物干粉;
步骤三、按照质量份数,取冰片0.5份和薄荷脑0.5份用无水乙醇溶解后, 喷加到所述提取物干粉上,混合均匀并充分挥散乙醇后,获得药物组合物, 将所述药物组合物填充至凝胶眼贴中获得中药凝胶眼贴。
实施例3
步骤一、按照质量份数,将野菊花10份、透骨草8份、决明子8份、珍 珠8份、丹参8份、石菖蒲8份、人参3份、红景天4份、黄连4份和番红 花0.2份分别进行预处理后获得野菊花提取物、透骨草提取物、决明子提取 物、珍珠提取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提取物、 黄连提取物和番红花提取物;
步骤二、将所述野菊花提取物、透骨草提取物、决明子提取物、珍珠粉 提取物、丹参提取物、石菖蒲提取物、黄连提取物、人参提取物、红景天提 取物和番红花提取物混合均匀,45℃真空干燥至水分含量小于5%,冷却至室 温后粉碎获得提取物干粉;
步骤三、按照质量份数,取冰片0.3份和薄荷脑0.2份用无水乙醇充分溶 解,喷加到提取物干粉,混合均匀并充分挥散乙醇后,获得药物组合物,将 所述药物组合物填充至凝胶眼贴中获得中药凝胶眼贴。
针对眼睛疲劳、眼睛干涩需要治疗的601例人员随机使用实施例1、实 施例2或实施例3制备的中药凝胶眼贴,每天临睡前1次,每次15分钟,并 且在基于实施例1和实施例2制备的中药凝胶药贴进行前期少量观察的基础 上,对于实施例3制备的中药凝胶眼贴的使用上可以增加观察人数,其使用 效果如表1所示:

本发明设计开发的一种中药凝胶眼贴,由野菊花、透骨草、决明子、珍 珠粉、丹参、石菖蒲、番红花、黄连、人参、红景天、薄荷、冰片组成的具 有缓解视疲劳、明目、改善眼部肌肤作用的组合物,本组合物可以是以上中 药提取物按照一定比例组成的组合物,也可以是含有以上化合物的其他多种 原料组成。该组合物是由有效成分创新组合成天然产物组合物,从几种不同 作用机制上入手缓解眼部疲劳,改善眼部肌肤,达到长短结合,标本兼治的效果,以期从疗效和安全性上有显著的突破和提高。
尽管本发明的实施方案已公开如上,但其并不仅仅限于说明书和实施方 式中所列运用,它完全可以被适用于各种适合本发明的领域,对于熟悉本领 域的人员而言,可容易地实现另外的修改,因此在不背离权利要求及等同范 围所限定的一般概念下,本发明并不限于特定的细节和这里示出与描述的实 施例。

1.一种中药凝胶眼贴,其特征在于,所述中药凝胶眼贴的组分及各组分的质量份数包括:
野菊花5-15份、透骨草5-10份、决明子5-10份、珍珠1-10份、丹参1-10份、石菖蒲1-10份、人参1-5份、红景天1-5份、黄连1-5份、番红花0.1-0.5份、冰片0.05-0.5份、薄荷脑0.05-0.5份。
2.如权利要求1所述的中药凝胶眼贴,其特征在于,所述中药凝胶眼贴的组分及各组分的质量份数为:
野菊花5份、透骨草5份、决明子5份、珍珠1份、丹参1份、石菖蒲2份、人参1份、红景天1份、黄连1份、番红花0.1份、冰片0.05份、薄荷脑0.05份。
3.如权利要求2所述的中药凝胶眼贴,其特征在于,所述中药凝胶眼贴的组分及各组分的质量份数为:
野菊花15份、透骨草10份、决明子10份、珍珠10份、丹参10份、石菖蒲10份、人参5份、红景天5份、黄连5份、番红花0.5份、冰片0.5份、薄荷脑0.5份。
4.一种中药凝胶眼贴的制备方法,其特征在于,包括如下步骤:
步骤一、按照质量份数,将野菊花5-15份、透骨草5-10份、决明子5-10份、珍珠1-10份、丹参1-10份、石菖蒲1-10份、人参1-5份、红景天1-5份、黄连1-5份和番红花0.1-0.5份分别进行预处理后获得野菊花提取物、透骨草提取物、决明子提取物、珍珠提取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提取物、黄连提取物和番红花提取物;
步骤二、将所述野菊花提取物、透骨草提取物、决明子提取物、珍珠提取物、丹参提取物、石菖蒲提取物、人参提取物、红景天提取物、黄连提取物和番红花提取物混合均匀,45℃真空干燥至水分含量小于5%,冷却至室温后粉碎获得提取物干粉;
步骤三、按照质量份数,取冰片0.05-0.5份和薄荷脑0.05-0.5份用无水乙醇溶解后,喷加到所述提取物干粉上,混合均匀直至挥散乙醇,获得药物组合物,将所述药物组合物填充至凝胶眼贴中获得中药凝胶眼贴。
5.如权利要求4所述的中药凝胶眼贴的制备方法,其特征在于,所述野菊花的预处理为:
将所述野菊花以75%乙醇作溶剂,按照料液比1:20g/ml回流提取2小时,提取温度为60℃,回收乙醇提取液,过滤后减压蒸馏至干获得野菊花提取物;
所述透骨草的预处理为:
将所述透骨草置于烧杯中,加入65%乙醇后,置于700W微波炉中提取4分钟,再将提取液以转速为3500rpm离心8分钟,取上清液置于45℃干燥箱中,浓缩至膏状获得透骨草提取物。
6.如权利要求5所述的一种中药凝胶眼贴的制备方法,其特征在于,所述决明子的预处理为:
将决明子粉碎后以70%乙醇提取2次,第1次1.5h,第2次1.5h,合并提取液,过滤,滤液回收乙醇,水浴蒸干得干膏,以45℃真空低温干燥至恒定质量获得决明子提取物;
所述珍珠的预处理为:
将珍珠研磨成粉后置于锥形瓶中,加入纯净水,煮沸13分钟后降至室温,加入中性蛋白酶,将锥形瓶至于摇床,调节温度为43℃,转速为110rpm,酶解190分钟后,再次加入中性蛋白酶,继续酶解200分钟,酶解结束后,将提取液煮沸340分钟,冷却后在转速为3800rpm下离心15分钟,取上清液后继续离心15分钟,再次取上清液,真空浓缩40℃下浓缩至干获得珍珠提取物。
7.如权利要求6所述的中药凝胶眼贴的制备方法,其特征在于,所述丹参的预处理为:
将丹参切段后至于锥形瓶中,加入55%乙醇,超声提取20分钟,过滤,滤液置水浴锅蒸干获得丹参提取物;
所述石菖蒲的预处理为:
将石菖蒲研磨成粉末后置于锥形瓶中,加丙酮溶解后封口,放入30℃,150r/min的恒温振荡器上混合振荡10h,振荡后配置料液比为1:18,将其放入35℃,超声功率为700W的超声器中,工作1.5h后取出,对得到的提取液抽滤浓缩后获得石菖蒲提取物。
8.如权利要求7所述的中药凝胶眼贴的制备方法,其特征在于,所述番红花的预处理为:
将番红花置于55℃烘箱烘干,粉碎后,置于锥形瓶中,加入95%乙醇,室温下磁力搅拌24h后抽滤,滤渣重复提取3次,将所得滤液合并,置于旋转蒸发仪中浓缩至深棕色膏状物获得番红花提取物;
所述黄连的预处理为:
将黄连研磨成粉末,按料液比1:8加入75%乙醇,于50℃下超声提取30分钟,提取3次,过滤,合并提取液,蒸干获得黄连提取物。
9.如权利要求8所述的中药凝胶眼贴的制备方法,其特征在于,所述人参的预处理为:
将人参切段后至于锥形瓶中,加入60%乙醇,超声提取30分钟,过滤,滤液置于水浴锅中蒸干获得人参提取物;
所述红景天的预处理为:
将干燥红景天用粉碎机粉碎后过40目筛,加入8倍量的水煎煮1.5h,抽滤后收集滤液,将分离后得到的滤渣加入6倍量的水再次进行煎煮1.5h,抽滤后合并2次所得滤液并旋转蒸发,浓缩到原来体积的1/3获得红景天提取物。
