Abstract
本发明公开了一种抑制细胞损伤组合物及其应用,涉及化妆品技术领域。本发明提供了一种抑制细胞损伤组合物,包括以下组分:库拉索芦荟叶汁粉、芦苇茯苓提取物、库拉索芦荟花提取物。本发明的抑制细胞损伤组合物具有显著的保护细胞免受紫外线及蓝光损伤的功效;组合物具有天然安全、组分简单、各成分相容性好、稳定性高、配制工艺简单、成本可控的特点;该组合物可便捷的应用于化妆水、乳液、面霜、洁面产品等各种类型的化妆品中。
Description
技术领域
本发明涉及化妆品技术领域,尤其是一种抑制细胞损伤组合物及其应用。
背景技术
太阳辐射根据波长进行分类:紫外线(UVR)的波长小于400nm。可见光波长范围为400-700nm,红外线波长大于700nm不同波长的光谱对肌肤的影响是不同的:UVB是大部分晒伤的罪魁祸首,短波UVA也可能在较小程度上引发红斑,UVB和UVA会单独或共同改变免疫反应;可见光主要通过产生氧化损伤来影响皮肤;而红外则诱导热损伤,并改变皮肤包线粒体的完整性,导致ROS的产生。外植体皮肤模型(使用了一系列的防晒剂)暴露在天然午间的日光下,结果表明,所诱导的ROS中,UVB占4%,UVA占46%,可见光占50%,可见日光防护,尤其是UVR及可见光防护的重要性。
随着人们对日光防护意识的逐渐增强,日光造成的皮肤损伤也日益受到关注,市面上光损伤修护的化妆品组合物种类繁多,但原料组合方式、功效参差不齐,目前未见具有改善紫外线和蓝光引起的细胞损伤的组合物。
发明内容
基于此,本发明的目的在于克服上述现有技术的不足之处而提供一种抑制细胞损伤组合物及其应用。
为实现上述目的,本发明所采取的技术方案为:一种抑制细胞损伤组合物,包括以下组分:库拉索芦荟叶汁粉、芦苇茯苓提取物、库拉索芦荟花提取物。
本发明提供了一种含库拉索芦荟叶汁粉、库拉索芦荟花提取物、芦苇茯苓提取物的组合物,这些成分具有天然、易使用、安全性高的优势,该组合物各成分之间具有显著的协同增效作用,能抑制紫外线及蓝光引起的细胞损伤,该组合物在化妆水、乳液、面霜、洁面产品等化妆品中具有较好的应用性。
优选地,所述库拉索芦荟叶汁粉、芦苇茯苓提取物、库拉索芦荟花提取物的重量比为:库拉索芦荟叶汁粉:芦苇茯苓提取物:库拉索芦荟花提取物=1:(10-30):(10-30)。
本发明首次将库拉索芦荟叶汁粉、库拉索芦荟花提取物、芦苇茯苓提取物及其协同浓度应用于光损伤,尤其是紫外线及蓝光引起的细胞损伤而开发的化妆品组合物,目前该领域现有技术的组合物大多只关注紫外线或蓝光单一辐射波长对皮肤的损伤,未见从改善紫外线和蓝光引起的细胞损伤多方面来实现光损伤修护的组合物。发明人经过大量实验探究后发现,库拉索芦荟叶汁粉:芦苇茯苓提取物:库拉索芦荟花提取物=1:(10-30):(10-30)时,制备得到的抑制细胞损伤组合物具有显著的保护细胞免受紫外线及蓝光损伤的功效。
优选地,所述库拉索芦荟叶汁粉、芦苇茯苓提取物、库拉索芦荟花提取物的重量比为:库拉索芦荟叶汁粉:芦苇茯苓提取物:库拉索芦荟花提取物=1:30:30。
发明人经过大量实验探究后发现,库拉索芦荟叶汁粉:芦苇茯苓提取物:库拉索芦荟花提取物=1:30:30时,制备得到的抑制细胞损伤组合物保护细胞免受紫外线及蓝光损伤的功效达到最佳。
此外,本发明提供了所述的抑制细胞损伤组合物在化妆品中的应用。优选地,所述化妆品为化妆水、乳液、面霜、洁面泡沫中的至少一种。
进一步地,本发明提供了一种化妆水,包括以下重量百分比的组分:
A相:羟乙基纤维素0.05%、去离子水余量;
B相:甘油3%、丁二醇2%、甘油聚醚-26 4%、透明质酸钠0.01%、EDTA二钠0.02%、聚乙二醇-32 2%、羟苯甲酯0.2%;
C相:PEG-40氢化蓖麻油0.5%、苯氧乙醇0.36%、乙基己基甘油0.04%、香精0.05%;
D相:库拉索芦荟叶汁粉0.1%、芦苇提取物和茯苓提取物2%、库拉索芦荟花提取物2%。
本发明提供了所述化妆水的制备方法,包括如下步骤:(1)将A相投入真空乳化锅中,开启加热至80-85℃,保温搅拌至分散均匀;(2)将B相投入真空乳化锅中,混合搅拌均匀;(3)开启降温至50℃,将预先混合均匀的C相缓慢加入乳化锅中,开启均质2分钟;(4)继续降温至45℃,将D相原料加入乳化锅中,开启均质2分钟,搅拌降温至35℃,出料。
本发明提供了一种乳液,包括以下重量百分比的组分:
A相:丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物0.1%、卡波姆0.1%、黄原胶0.05%、去离子水20%;
B相:甘油3%、丁二醇3%、氢化卵磷脂0.5%、对羟基苯乙酮0.5%、羟苯甲酯0.2%、EDTA二钠0.02%、乙基己基甘油0.05%;
C相:鲸蜡硬脂醇2%、山嵛醇2%、低芥酸菜子油3%、辛酸/癸酸甘油三酯5%、聚甘油-6硬脂酸酯和聚甘油-6山嵛酸酯1%;
D相:氢氧化钠0.05%、去离子水余量;
E相:香精0.1%;
F相:库拉索芦荟叶汁粉0.1%、芦苇提取物和茯苓提取物3%、库拉索芦荟花提取物3%。
本发明提供了所述乳液的制备方法,包括如下步骤:(1)将A相原料缓慢撒入水锅中,开启搅拌至无明显白色颗粒物;(2)将B相原料依次投入水锅中,开启搅拌至分散均匀;(3)将C相原料依次投入油锅中,开启搅拌至分散均匀,开启加热至80-85℃;(4)将水锅物料抽入真空乳化锅中,搅拌均质3分钟,开启加热至80-85℃;(5)将油锅原料缓慢抽入真空乳化锅中,开启真空,搅拌均质8分钟;(6)降温至50℃,投入D相原料,开启真空,搅拌均质3分钟;(7)降温至45℃,投入E相和F相原料,搅拌均质2分钟后继续搅拌15分钟,降温至35℃,排真空,出料。
本发明提供了一种面霜,包括以下重量百分比的组分:
A相:甘油5%、丁二醇3%、对羟基苯乙酮0.6%、EDTA二钠0.02%、去离子水余量;
B相:丙烯酰二甲基牛磺酸铵/VP共聚物0.2%、汉生胶0.02%、丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物0.25%;
C相:鲸蜡硬脂醇橄榄油酸酯和山梨坦橄榄油酸酯2%、鲸蜡硬脂醇0.7%、牛油果树果脂5%、碳酸二辛酯3%、低芥酸菜子油3%、辛酸/癸酸甘油三酯3%、生育酚乙酸酯0.2%;
D相:去离子水2%、氢氧化钠0.0625%;
E相:苯氧乙醇0.36%、乙基己基甘油0.04%、香精0.1%;
F相:库拉索芦荟叶汁粉0.1%、芦苇提取物和茯苓提取物2%、库拉索芦荟花提取物2%。
本发明提供了所述面霜的制备方法,包括如下步骤:(1)将A相投入水锅中,混合搅拌至分散均匀;(2)将B相原料缓慢撒入水锅中,混合搅拌分散均匀;(3)将C相原料依次投入油锅中,开启搅拌至分散均匀,开启加热至80-85℃,搅拌至完全溶解;(4)将水锅物料抽入真空乳化锅中,搅拌均质5分钟,开启加热至80-85℃;(5)将油锅原料缓慢抽入真空乳化锅中,开启真空,搅拌均质8分钟;(6)降温至50℃,投入D相原料,开启真空,搅拌均质3分钟;(7)降温至45℃,投入E相和F相原料,搅拌均质2分钟后继续搅拌15分钟,降温至35℃,排真空,出料。
本发明提供了一种洁面泡沫,包括以下重量百分比的组分:
A相:甘油2%、丁二醇4%、1,2-己二醇0.5%、EDTA二钠0.1%、透明质酸钠0.1%、去离子水余量;
B相:聚甘油-10月桂酸酯0.5%、甲基椰油酰基牛磺酸钠(含量30%)3%、椰油酰甘氨酸钾(含量30%)15%、月桂酰两性基乙酸钠(含量35%)8%、聚季铵盐-39 1%;
C相:苯氧乙醇0.36%、乙基己基甘油0.04%、香精0.1%;
D相:库拉索芦荟叶汁粉0.1%、芦苇提取物和茯苓提取物3%、库拉索芦荟花提取物3%。
本发明提供了所述洁面泡沫的制备方法,包括如下步骤:(1)将A相原料投入真空乳化锅中,混合搅拌均匀;(2)依次加入B相原料,混合搅拌均匀;(3)依次加入C相原料,混合搅拌均匀;(4)依次加入D相原料,混合搅拌均匀,出料。
相对于现有技术,本发明的有益效果为:本发明的组合物具有显著的保护细胞免受紫外线及蓝光损伤的功效;组合物具有天然安全、组分简单、各成分相容性好、稳定性高、配制工艺简单、成本可控的特点;该组合物可便捷的应用于化妆水、乳液、面霜、洁面产品等各种类型的化妆品中。
具体实施方式
为更好的说明本发明的目的、技术方案和优点,下面将结合具体实施例对本发明作进一步说明。实施例中,所使用的实验方法如无特殊说明,均为常规方法,所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
库拉索芦荟叶汁粉,含量>96%,生产商为云南万绿生物股份有限公司;库拉索芦荟花提取物:含量为97.4%,生产商为云南万绿生物股份有限公司;芦苇和茯苓提取物:其中芦苇提取物的含量为0.1-1.0%;茯苓提取物的含量为0.1-1.0%;生产商为CLR公司(Chemisches Laboratorium Dr.Kurt Richter GmbH)
其他实验材料包括皮肤表皮细胞HaCaT、MTT、细胞培养基、FBS、IL-8 ELISA Kit等实验试剂材料购自美国ATCC、Sigma-Aldrich、Thermal Scientific、abcam和Gibco等公司。
试验例1细胞毒性测试
试验过程:样品细胞毒性实验采用MTT法进行,在没有蓝光照射的情况下检测样品对细胞活性的影响。实验步骤为:HaCaT细胞以5×103个/孔的密度接种于96孔板中孵育24小时后,加入系列浓度的样品作用24h。移去培养孔中培养液,每孔加MTT溶液(5mg/ml溶于PBS缓冲液中)100μl.继续孵育4小时,终止培养,小心吸弃孔内培养上清液,每孔加150μlDMSO溶液,振荡10分钟。在酶标仪上选择490nm波长测定各孔吸光值,记录结果。
试验结果:结果如表1所示;

样品对细胞的毒性检测结果如上表所示,细胞培养液中样品的浓度在600μg/ml以下对细胞无抑制作用。样品浓度在1000μg/ml时细胞活性有较明显的降低,但细胞活性均在90%以上,提示无细胞毒性。
试验例2抑制蓝光引起的细胞损伤实验和抑制紫外光引起的细胞损伤实验
试验方法1:蓝光皮肤细胞损伤模型采用人表皮细胞株HaCaT细胞,经蓝光照射后用MTT法检测细胞存活情况。
具体实验方法如下:正常的人表皮细胞HaCaT用DMEM完全培养基(含10%血清和1%双抗),放置于37℃和5%CO2细胞培养箱孵育培养及传代。实验开始时,在96孔板中以5×103个/孔的密度将HaCaT细胞接种孵育24小时后,加入系列浓度的样品。样品作用24h后,弃去培养基,用PBS洗两次,再往每孔加入50μL PBS,用波长λ=450nm的LED-BL(蓝光),以9000±500LUX的强度下照射4小时,空白对照组用锡箔纸包住遮光。照射后弃去PBS,往每孔加入100μL完全培养基,置于培养箱中培养。24小时后,用MTT进行细胞活力测定。
试验方法2:紫外光皮肤细胞损伤模型采用人表皮细胞株HaCaT细胞,经紫外光照射后用MTT法检测细胞存活情况。
具体实验方法如下:正常的人表皮细胞HaCaT用DMEM完全培养基(含10%血清和1%双抗),放置于37℃和5%CO2细胞培养箱孵育培养及传代。实验开始时,在96孔板中以5×103个/孔的密度将HaCaT细胞接种孵育24小时后,加入系列浓度的样品。样品作用24h后,弃去培养基,用PBS洗两次,再往每孔加入50μL PBS,紫外光照射剂量为30mJ/cm2(注1),空白对照组用锡箔纸包住遮光。照射后弃去PBS,往每孔加入100μL完全培养基,置于培养箱中培养。24小时后,用MTT进行细胞活力测定。
注1:在预实验中用不同剂量的紫外光照射HaCaT细胞,结果显示细胞存活随剂量增加而降低。不同剂量下细胞存活情况为:76.2%(20mJ/cm2),63.0%(30mJ/cm2),47.0%(50mJ/cm2),19.1%(100mJ/cm2)。选取30mJ/cm2进行样品实验。
试验方法3:统计学分析方法
差异性分析:差异性分析采用方差分析法(ANOVA)进行,分析各组细胞之间的显著性差异。(p<0.05为有统计学差异;p<0.01为有显著统计学差异;p<0.001为有极其显著的统计学差异)。
抑制蓝光引起的细胞损伤实验和抑制紫外光引起的细胞损伤实验的单组分试验结果如表2所示;
抑制蓝光引起的细胞损伤实验和抑制紫外光引起的细胞损伤实验的多组分试验结果如表3、表4所示;

通过上表可以看出,单独使用芦苇和茯苓提取物、库索芦荟花提取物、库拉索芦荟叶汁粉,对蓝光及紫外光引起的细胞损伤具有一定程度的抑制作用,且单独使用芦苇和茯苓提取物、库索芦荟花提取物在较高浓度的抑制效果相比模型组,具有统计学差异。在测试的不同组分的系列浓度中,当芦苇和茯苓提取物浓度为300μg/ml时,对蓝光及紫外光引起的细胞存活率相比其他系列浓度下最高,即效果最好。但芦苇和茯苓提取物的原料成本较高,对成本控制具有较高要求的化妆品开发项目而言,不具有最佳的应用性,故考虑多组分复配,以获得功效及应用性的最佳方案,以期更便捷的应用于不用类型化妆品的开发。

通过上表可以看出,当库拉索芦荟叶汁粉:库拉索芦荟花提取物:芦苇和茯苓提取物=1:10-30:10-30时,对蓝光及紫外光引起的细胞损伤具有抑制作用,且相比模型组均具有极其显著的统计学差异。通过上表可以看出,对比例1-5制备得到的组合物,对蓝光及紫外光引起的细胞损伤具有一定的抑制作用,但协同增效的效果显著差于本发明特定组合物的效果。
最后所应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。
Claims (8)
1.一种抑制细胞损伤组合物,其特征在于,包括以下组分:库拉索芦荟叶汁粉、芦苇茯苓提取物、库拉索芦荟花提取物。
2.如权利要求1所述的抑制细胞损伤组合物,其特征在于,库拉索芦荟叶汁粉、芦苇茯苓提取物、库拉索芦荟花提取物的重量比为:库拉索芦荟叶汁粉:芦苇茯苓提取物:库拉索芦荟花提取物=1:(10-30):(10-30)。
3.如权利要求2所述的抑制细胞损伤组合物,其特征在于,库拉索芦荟叶汁粉、芦苇茯苓提取物、库拉索芦荟花提取物的重量比为:库拉索芦荟叶汁粉:芦苇茯苓提取物:库拉索芦荟花提取物=1:30:30。
4.如权利要求1-3任一项所述的抑制细胞损伤组合物在化妆品中的应用。
5.一种化妆水,其特征在于,包括以下重量百分比的组分:
A相:羟乙基纤维素0.05%、去离子水余量;
B相:甘油3%、丁二醇2%、甘油聚醚-26 4%、透明质酸钠0.01%、EDTA二钠0.02%、聚乙二醇-32 2%、羟苯甲酯0.2%;
C相:PEG-40氢化蓖麻油0.5%、苯氧乙醇0.36%、乙基己基甘油0.04%、香精0.05%;
D相:库拉索芦荟叶汁粉0.1%、芦苇提取物和茯苓提取物2%、库拉索芦荟花提取物2%。
6.一种乳液,其特征在于,包括以下重量百分比的组分:
A相:丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物0.1%、卡波姆0.1%、黄原胶0.05%、去离子水20%;
B相:甘油3%、丁二醇3%、氢化卵磷脂0.5%、对羟基苯乙酮0.5%、羟苯甲酯0.2%、EDTA二钠0.02%、乙基己基甘油0.05%;
C相:鲸蜡硬脂醇2%、山嵛醇2%、低芥酸菜子油3%、辛酸/癸酸甘油三酯5%、聚甘油-6硬脂酸酯和聚甘油-6山嵛酸酯1%;
D相:氢氧化钠0.05%、去离子水余量;
E相:香精0.1%;
F相:库拉索芦荟叶汁粉0.1%、芦苇提取物和茯苓提取物3%、库拉索芦荟花提取物3%。
7.一种面霜,其特征在于,包括以下重量百分比的组分:
A相:甘油5%、丁二醇3%、对羟基苯乙酮0.6%、EDTA二钠0.02%、去离子水余量;
B相:丙烯酰二甲基牛磺酸铵/VP共聚物0.2%、汉生胶0.02%、丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物0.25%;
