技术领域
本发明涉及一种洗涤剂组合物,特别是包含脂肪酶和皂苷的洗涤剂组合物。
背景技术
脂肪酶也是用于洗涤剂制剂,特别是用于洗衣洗涤剂的有用组分。然而,脂肪酶是一种昂贵的组分,并且期望增强脂肪酶活性以使得用量减少或提高所得制剂的清洁潜力。
因此,需要具有增强的脂肪酶活性的洗涤剂组合物。
然而,脂肪酶通常与许多有用的洗涤剂制剂组分不相容,例如去污聚合物,特别是聚酯基去污聚合物,因为脂肪酶会降解去污聚合物。该问题在液体洗涤剂组合物中特别显著。
还需要其中组合物中的脂肪酶可以被抑制的洗涤剂组合物,并且因此允许制剂包括对脂肪酶降解敏感的组分。
发明内容
我们发现,在洗涤剂组合物中以一定含量掺入皂苷抑制洗涤剂制剂中的脂肪酶活性和/或当在洗涤条件下稀释时增强脂肪酶活性。
在本发明的第一方面,提供了一种在洗涤剂组合物中抑制脂肪酶活性的方法,所述方法涉及将0.25至40重量%,优选地0.5至30重量%,更优选地0.5至25重量%,更优选地1至20重量%,最优选地1至15重量%的皂苷掺入到所述包含脂肪酶的组合物中,其中所述洗涤剂组合物优选地为家庭护理洗涤剂组合物,更优选地洗衣洗涤剂组合物。
在本发明的第二方面,提供了皂苷在洗涤剂组合物,优选地洗衣洗涤剂组合物中抑制脂肪酶活性的用途,其中所述皂苷0.25至40重量%,优选地0.5至30重量%,更优选地0.5至25重量%,更优选地1至20重量%,最优选地1至15重量%的皂苷的含量存在于洗涤剂制剂中,其中所述洗涤剂组合物优选地为家庭护理洗涤剂组合物,更优选地洗衣洗涤剂组合物。
优选地,在所述方法或用途中,所述洗涤剂组合物包含:
(i)0.25至40重量%,优选地0.5至30重量%,更优选地0.5至25重量%,更优选地1至20重量%,最优选地1至15重量%的皂苷;和,

图7是突出显示建议在该制剂中最大最终皂苷浓度1mM(在稀释至洗涤液之后)以确保保持对牛脂肪污渍(CS61)的完全脂肪酶清洁性能的图。在30℃下在FH32水中使用1g/L制剂洗涤30分钟,其含有:LAS 5.8%、SLES 4.5%、NI(Neodol 25-7)4.5%、脂肪酸0.9%、TEA8.8%、甘油2%、柠檬酸1%、聚合物/香料/防腐剂(5%)和水(至100%)。

洗涤剂组合物可以施用于任何合适的基材,包括但不限于施用家庭护理组合物的任何基材,例如纺织品、陶器和餐具。特别优选的基材是纺织品。特别优选的洗涤剂组合物是洗衣洗涤剂组合物。优选地,洗衣洗涤剂组合物是液体、凝胶或粉末,更优选地,洗涤剂是液体洗涤剂。
洗衣洗涤剂组合物可以采取任何合适的形式。优选地形式是液体或粉末,最优选地是液体。
脂肪酶
所述组合物包含0.0005至2.5重量%,优选地0.001至2重量%,更优选地0.005至1重量%的脂肪酶。
优选地,所述脂肪酶属于酶分类EC 3.1.1.3。
优选地,所述脂肪酶是细菌或真菌来源的,更优选地所述脂肪酶是真菌来源的。

皂苷
所述组合物包含0.25至40重量%,优选地0.5至30重量%,更优选地0.5至25重量%,更优选地1至20重量%,最优选地1至15重量%的皂苷。
皂苷是包含与非芳香族4、5和6元环的稠合体系结合的糖部分的天然化合物。所述环体系优选地选自三萜类化合物,例如羊毛脂甾烷、达玛烷、羽扇豆烷、齐墩果烷、乌苏烷和何帕烷。R.A.Hill等人的Natural Product Reports 27(2010),79-132中提供了这些环体系的概述。

皂苷还可以由细菌(例如糖基化的藿烷类(hopanoids),例如核糖基藿烷(ribosylhopane))和海洋生物(包括海参、海星和海绵)产生(Bahrami,Y.,Zhang,W.&Franco,C.M.(2018)Marine Drugs 16:423-453)。皂苷还可以通过生物技术,或通过体外酶促生物合成或通过微生物细胞工厂的工程生产(Moses,T.等人(2014)PNAS 28:1634-1639)。
所述皂苷优选地为茶皂苷(例如优选地源自山茶属物种)、无患子(Soapnut)皂苷(例如优选地源自无患子属物种)、皂树树皮(Quillaja Bark)皂苷或七叶皂苷(例如优选地源自七叶树属物种)。
优选地,所述皂苷具有包含三萜骨架和一个或多个连接至三萜骨架的糖部分的结构。
皂苷列举在Chemical Entities of Biological Interest(ChEBI)数据库(Hastings,J.,de Matos,P.,Dekker,A.,Ennis,M.,Harsha,B.,Kale,N.,Muthukrishnan,V.,Owen,G.,Turner,S.,Williams,M.和Steinbeck,C.(2013)The ChEBI referencedatabase and ontology for biologically relevant chemistry:enhancements for2013.Nucleic Acids Res.)中。例如,CHEBI:61778-三萜皂苷。
收获后剩余的农业残余物可以适用于提取和供应皂苷。例如,甜菜叶和皮可以进一步提取以获取有用量的皂苷。在中国,从油茶籽提取茶籽油后剩余的籽饼中提取的茶皂苷的生产和供应十分成熟。或者,皂苷可以从野外(例如,来自蒺藜的原薯蓣皂苷)或通过管理的种植园(例如来自皂树树皮皂苷)采集的植物部分中提取。
优选的组分
表面活性剂
洗涤剂组合物优选地包含表面活性剂(其包括两种或更多种表面活性剂的混合物)。所述组合物包含1至60重量%,优选地2.5至50重量%,更优选地4至40重量%的表面活性剂。甚至更优选地表面活性剂含量为6至40重量%,更优选地8至35重量%。
可以使用的合适的阴离子洗涤剂化合物通常是具有含约8至约22个碳原子的烷基的有机硫酸和磺酸的水溶性碱金属盐,术语烷基用于包括较高级烷基的烷基部分。
合适的阴离子洗涤剂化合物的实例是鼠李糖脂、烷基硫酸钠和烷基硫酸钾,特别是通过使例如由牛油或椰油制得的较高级C8至C18醇硫酸化而获得的那些,烷基C9至C20苯磺酸钠和钾,特别是直链仲烷基C10至C15苯磺酸钠;以及烷基甘油醚硫酸钠,特别是衍生自牛油或椰油的较高级醇和衍生自石油的合成醇的那些醚。

最优选的阴离子表面活性剂选自:鼠李糖脂、直链烷基苯磺酸盐;烷基硫酸盐;烷基醚硫酸盐及其混合物。优选地,烷基醚硫酸盐是具有平均1至3个EO(乙氧基化物)单元的C12-C14正烷基醚硫酸盐。
月桂基醚硫酸钠(SLES)是特别优选地。优选地,直链烷基苯磺酸盐是C11至C15烷基苯磺酸钠。优选地,烷基硫酸盐是直链或支链C12至C18烷基硫酸钠。十二烷基硫酸钠是特别优选地(SDS,也称为伯烷基硫酸盐)。鼠李糖脂可以优选地富含单-鼠李糖脂(超过60%)、富含二-鼠李糖脂(超过60%),或是单-和二-鼠李糖脂的40/60至60-40混合物。
在液体制剂中,优选地存在两种或更多种阴离子表面活性剂,例如直链烷基苯磺酸盐连同烷基醚硫酸盐。
在液体制剂中,优选地,除了阴离子表面活性剂之外,洗衣组合物还包含烷基乙氧基化非离子表面活性剂,优选地2至8重量%的烷基乙氧基化非离子表面活性剂。
可以使用的合适的非离子洗涤剂化合物包括,特别是,具有脂肪族疏水基团和反应性氢原子的化合物(例如,脂肪醇、酸或酰胺),尤其是环氧乙烷(单独或与环氧丙烷一起)的反应产物。优选的非离子洗涤剂化合物是脂肪族C8至C18直链或支链伯或仲醇与环氧乙烷的缩合产物。烷基多糖苷(APG)也是优选地。
最优选地,非离子洗涤剂化合物为烷基乙氧基化非离子表面活性剂,其为具有7EO至9EO单元的平均乙氧基化的C8至C18伯醇。

去污聚合物
优选地包括去污聚合物。
洗衣洗涤剂组合物优选地包含0.1至8重量%的去污聚合物。
去污聚合物的优选地含量为0.2-6重量%,更优选地0.5-5重量%,最优选地1-5重量%。
优选地,所述去污聚合物是聚酯去污聚合物。
更优选地,所述聚酯去污聚合物是聚对苯二甲酸乙二醇酯和/或丙二醇酯基去污聚合物,最优选地聚对苯二甲酸丙二醇酯基去污聚合物。
合适的聚酯基去污聚合物描述于WO 2014/029479和WO 2016/005338中。
烷氧基化聚胺
当洗涤剂组合物是洗衣组合物的形式时,优选地是包括烷氧基化聚胺。
洗衣洗涤剂优选地包含0.1至8重量%的烷氧基化聚胺。
烷氧基化聚胺优选含量为0.2至6重量%,更优选地0.5至5重量%。另一个优选含量为1至4重量%。
烷氧基化聚胺可以是直链或支链的。其可以支化至其是树枝状大分子的程度。烷氧基化通常可以是乙氧基化或丙氧基化,或两者的混合。优选地,烷氧基化聚胺包含烷氧基化聚乙烯亚胺和/或烷氧基化聚丙烯亚胺,更优选地烷氧基化是乙氧基化或丙氧基化或两者的混合。当氮原子被烷氧基化时,优选的平均烷氧基化度为10-30,优选地15-25。
优选的材料是烷氧基化聚乙烯亚胺,最优选地乙氧基化聚乙烯亚胺,其平均乙氧基化度为10-30,优选地15-25,其中氮原子被乙氧基化。
另外的酶
除了所需的脂肪酶之外,另外的酶可以存在于清洁组合物中。优选的是除脂肪酶以外的另外的酶存在于优选的洗衣洗涤剂组合物中。
如果存在,则每种另外的酶在本发明的组合物中的含量为0.0001重量%至0.1重量%。
在该组合物中存在的酶的含量优选地与作为纯蛋白的酶的含量相关。
优选的酶包括由以下组成的组中的那些:蛋白酶、纤维素酶、α-淀粉酶、过氧化物酶/氧化酶、果胶酸裂解酶和/或甘露聚糖酶。所述优选的酶包括这些酶中的两种或更多种的混合物。
优选地,所述酶选自:蛋白酶、纤维素酶和/或α-淀粉酶。

本发明可以在分类为EC3.1.1.4和/或EC3.1.1.32的磷脂酶的存在下进行。如本文所用,术语磷脂酶是对磷脂具有活性的酶。
磷脂,例如卵磷脂或磷脂酰胆碱,由在外部(sn-1)和中间(sn-2)位用两个脂肪酸酯化并且在第三位用磷酸酯化的甘油组成;磷酸进而可以酯化成氨基醇。磷脂酶是参与磷脂水解的酶。可以区分几种类型的磷脂酶活性,包括水解一个脂肪酰基(分别在sn-1和sn-2位)以形成溶血磷脂的磷脂酶A1和A2;和可以水解溶血磷脂中的剩余脂肪酰基的溶血磷脂酶(或磷脂酶B)。磷脂酶C和磷脂酶D(磷酸二酯酶)分别释放二酰基甘油或磷脂酸。
该组合物可使用分类于EC3.1.1.74中的角质酶。根据本发明使用的角质酶可以是任何来源的。优选地角质酶是微生物来源的,特别是细菌、真菌或酵母来源的。

酶稳定剂
存在于组合物中的任何酶可以使用常规稳定剂来稳定化,例如多元醇如丙二醇或甘油、糖或糖醇、乳酸、硼酸或硼酸衍生物,例如芳族硼酸酯,或苯基硼酸衍生物,例如4-甲酰基苯基硼酸,并且组合物可以如例如WO92/19709和WO 92/19708中所述配制。
另外的材料
可以包含在洗涤剂组合物(优选地,洗衣洗涤剂组合物)中的进一步任选的但优选的材料包括助洗剂、螯合剂、荧光剂、香料、调色染料和聚合物。
助洗剂或络合剂
所述组合物可以包含助洗剂。
助洗剂材料可选自1)钙螯合剂材料,2)沉淀材料,3)钙离子交换材料和4)其混合物。
钙螯合剂助洗剂的实例包括碱金属多磷酸盐如三聚磷酸钠,和有机螯合剂如乙二胺四乙酸。
沉淀助洗剂材料的实例包括正磷酸钠和碳酸钠。
钙离子交换助洗剂材料的实例包括各种类型的水不溶性结晶或无定形硅铝酸盐,其中沸石是其众所周知的代表,例如沸石A、沸石B(也称为沸石P)、沸石C、沸石X、沸石Y以及沸石P型,如EP-A-0384070中所述。
组合物还可以含有0-65重量%的助洗剂或络合剂如乙二胺四乙酸、二亚乙基三胺-五乙酸、烷基或烯基琥珀酸、次氮基三乙酸或下文提及的其它助洗剂。许多助洗剂由于其络合金属离子的能力也是漂白稳定剂。
沸石和碳酸盐(碳酸盐(包括碳酸氢盐和倍半碳酸盐))是优选的助洗剂,其中碳酸盐是特别优选的。
所述组合物可含有结晶硅铝酸盐作为助洗剂,优选地碱金属硅铝酸盐,更优选地硅铝酸钠。这通常以小于15重量%的含量存在。

这些材料含有一些结合水,并且需要具有至少50mg CaO/g的钙离子交换容量。优选地铝硅酸钠在上式中含有1.5-3.5个SiO2单元。它们可以容易地通过硅酸钠和铝酸钠之间的反应制备,如文献中充分描述的。表面活性剂与铝硅酸盐(当存在时)的比率优选地大于5:2,更优选地大于3:1。
作为对铝硅酸盐助洗剂的替代或补充,可以使用磷酸盐助洗剂。在本领域中,术语“磷酸盐”包括二磷酸盐、三磷酸盐和膦酸盐种类。其它形式的助洗剂包括硅酸盐,如可溶性硅酸盐、偏硅酸盐、层状硅酸盐(例如来自Hoechst的SKS-6)。
如果是洗衣洗涤剂,则优选地洗衣洗涤剂制剂为非磷酸盐助洗的洗衣洗涤剂制剂,即含有小于1重量%的磷酸盐。最优选地洗衣洗涤剂制剂不是助洗的,即含有小于1重量%的助洗剂。
螯合剂
螯合剂可以存在或不存在于洗涤剂组合物中。
如果存在,则螯合剂的存在量为0.01至5重量%。
膦酸(或其盐)螯合剂的实例为:1-羟基亚乙基-1,1-二膦酸(HEDP);二亚乙基三胺五(亚甲基膦酸)(DTPMP);六亚甲基二胺四(亚甲基膦酸)(HDTMP);氨基三(亚甲基膦酸)(ATMP);乙二胺四(亚甲基膦酸)(EDTMP);四亚甲基二胺四(亚甲基膦酸)(TDTMP);和膦酰基丁烷三羧酸(PBTC)。
荧光剂
所述组合物优选地包含荧光剂(荧光增亮剂)。荧光剂是众所周知的,且许多这样的荧光剂可商购获得。通常,这些荧光剂以其碱金属盐例如钠盐的形式提供和使用。
用于组合物中的一种或多种荧光剂的总量通常为0.0001至0.5重量%,优选地0.005至2重量%,更优选地0.01至0.1重量%。
优选的荧光剂的类型是:二苯乙烯基联苯化合物,例如Tinopal(商标)CBS-X,二胺芪二磺酸化合物,例如Tinopal DMS pure Xtra和Blankophor(商标)HRH,以及吡唑啉化合物,例如Blankophor SN。

最优选的荧光剂是:2-(4-苯乙烯基-3-磺基苯基)-2H-萘并(napthol)[1,2-d]三唑钠、4,4'-双{[(4-苯胺基-6-(N-甲基-N-2-羟乙基)氨基-1,3,5-三嗪-2-基)]氨基}芪-2-2'-二磺酸二钠、4,4'-双{[(4-苯胺基-6-吗啉基-1,3,5-三嗪-2-基)]氨基}芪-2-2'-二磺酸二钠以及4,4'-双(2-磺基苯乙烯基)联苯二钠。
该方法中使用的水性溶液具有荧光剂存在。荧光剂优选地在该方法中使用的水性溶液中以0.0001g/L至0.1g/L,更优选地0.001至0.02g/L的范围存在。
香料
组合物优选地包含香料。在由CFTA Publications出版的CTFA(Cosmetic,Toiletry and Fragrance Association)1992International Buyers Guide和由SchnellPublishing Co.出版的OPD 1993Chemicals Buyers Directory 80th Annual Edition中提供了香料的许多合适实例。

香料头香可用于提示本发明的白度和亮度益处。

可以与本发明一起应用的另一组香料是所谓的“芳香疗法”材料。这些包括也用于香水中的许多组分,包括精油如鼠尾草、桉树、天竺葵、薰衣草、肉豆蔻干皮(Mace)提取物、橙花油、肉豆蔻、留兰香、甜紫罗兰叶和缬草的组分。
优选的是洗衣处理组合物不含过氧漂白剂,例如过碳酸钠、过硼酸钠和过酸。
调色染料
优选地,当组合物是洗衣洗涤剂组合物时,则其包含调色染料。优选地,调色染料以组合物的0.0001至0.1重量%存在。
染料在Color Chemistry Synthesis,Properties and Applications ofOrganic Dyes and Pigments,(H Zollinger,Wiley VCH,Zürich,2003)和IndustrialDyes Chemistry,Properties Applications.(K Hunger(ed),Wiley-VCH Weinheim 2003)中描述。
用于洗衣组合物的调色染料优选地在可见光范围(400-700nm)中的最大吸收处具有大于5000L mol-1cm-1,优选地大于10000L mol-1cm-1的消光系数。染料的颜色是蓝色或紫色。
优选地调色染料发色团是偶氮、吖嗪、蒽醌和三苯甲烷。
偶氮、蒽醌、酞菁和三苯甲烷染料优选地带有净阴离子电荷或不带电荷。吖嗪优选地带有净阴离子或阳离子电荷。在洗涤过程的洗涤或漂洗步骤期间,蓝色或紫色调色染料沉积到织物上,从而为织物提供可见的色调。在这方面,染料对白色布料赋予蓝色或紫色,色调角为240至345、更优选地250至320、最优选地250至280。该测试中使用的白色布料是经漂白的、非丝光处理的编织棉片。

单偶氮染料优选地含有杂环,并且最优选地是噻吩染料。单偶氮染料优选地是烷氧基化的,并且优选地在pH=7时不带电荷或带阴离子电荷。烷氧基化噻吩染料在WO/2013/142495和WO/2008/087497中讨论。噻吩染料的优选实例如下所示:

聚合物
组合物可包含一种或多种另外的聚合物。优选地洗涤剂组合物包含0.1至20重量%,优选地0.5至15重量%或一种或多种聚合物。实例为羧甲基纤维素、聚(乙二醇)、聚(乙烯醇)、聚羧酸酯如聚丙烯酸酯、马来酸/丙烯酸共聚物和甲基丙烯酸月桂酯/丙烯酸共聚物。
实施例
将通过以下非限制性实施例阐述本发明。
实验方法:
蛋白质浓度的测定
通过使用Sigma-Aldrich(二辛可宁酸)BCA测定试剂盒来估计酶样品的蛋白质总量,并且按照使用者手册中的指示制备工作试剂。通过以50:1(v/v)比率混合溶液A[1%(w/v)的钠盐形式的二辛可宁酸、2%(w/v)的碳酸钠、0.16%(w/v)的酒石酸钠、0.4%(w/v)的氢氧化钠、0.95%(w/v)的碳酸氢钠,pH11.5]与溶液B[4%(w/v)的硫酸铜]制备BCA试剂。在去离子水中进行牛血清白蛋白(2mg/mL)的连续稀释,以产生7个点的标准曲线。为了进行该测定,将BCA试剂(200μL)加入96孔板的孔中,随后样品蛋白稀释液(20μL)。将微量滴定板(MTP)密封并在37℃下孵育30分钟。在孵育之后,在分光光度计上测量540nm处的吸光度。
脂肪酶活性的生化测定
使用4-硝基苯基-戊酸酯(C5)和4-硝基苯基-十二烷酸酯(C12)作为底物,通过比色法测定脂肪酶活性。将4-硝基苯基-十二烷酸酯(25mg)或4-硝基苯基-戊酸酯(18mg)溶解在10mL溶剂(甲醇)中以制备8mM储备溶液。将1mL储备溶液加入7mL酸化水(pH 4.5)中,以得到最终浓度1mM(底物)。在96孔微量滴定板中加入以下试剂:60μL dH2O、80μL Tris-HCl缓冲液(pH 8.5、50mM)、20μL稀释酶溶液、20μL皂苷10x浓缩储备溶液。然后加入20μL底物开始反应。对于零酶对照,酶溶液用H2O代替。加入底物后,在Varioskan读板器中,在环境温度下于405nm处监测产物(4-硝基苯酚)的释放15分钟。
微量滴定板清洁方法
将用牛肉脂肪(CS61)(Centre for Testmaterials-荷兰)染污的机织棉织物切割至空96孔微量滴定板中,并读取洗涤前污渍强度的读数。在FH32水中制备脂肪酶/皂苷洗涤溶液,然后在30℃下孵育之前使用多通道移液器将(200μL)转移到污渍上,以200rpm的速度振摇30分钟。洗涤后,立即使用多通道移液器去除洗涤液,并用200μL dH2O将污渍盘洗涤3次,然后在橱柜中放置过夜至干。干燥后,对污渍板进行数字扫描,并测量它们的△E。该值用于表示清洁效果,并定义为白色布和洗涤后的染污布之间的色差异。在数学上,△E的定义是:

本文中,清洁效果以去污指数(SRI)的形式表示:

SRI越高,布越清洁,SRI=100(白色)。
Tergotometer实验:
通过在tergotometer中洗涤标准污渍样本来测定含有皂苷的制剂的清洁性能。水硬度为13FH或26FH。洗涤时间为30分钟,然后在与洗涤液硬度相同的水中漂洗3次。洗涤温度为25℃。
污渍样本是包含猪油污渍的机织棉。在洗涤前后使用分光光度计测量每个污渍的吸光度以获得△E值。使用与微量滴定板测定相同的程序计算所得的SRI。
酯酶活性的生化测定
代替使用C4-pNP酯底物(4-硝基苯基-丁酸酯),应用与测定脂肪酶活性相同的方法。
使用的材料
实施例1生化实验表明七叶皂苷活化脂肪酶
在生化测定中测试了各种脂肪酶对皂苷的剂量响应。在这些实验中脂肪酶的浓度为10μg/L。选择该水平以确保底物水解和由此产生的吸光度测量值增加落在分光光度计的灵敏度范围内。表1表明包含选择的脂肪酶的酶活性增加百分比和七叶皂苷的浓度增加。在所有情况下,当七叶皂苷浓度达到阈值以上时,脂肪酶活性增加,该阈值因所讨论的酶而异,但在0.01(Resinase HT)至0.10μM(Lipex,100L、Lipex Evity和Novozyme 51032)范围内。随着七叶皂苷浓度进一步高于该值直至测试的最大浓度,酶活性也增加。

实施例2生化实验表明三萜骨架和一个或多个连接至三萜骨架的糖部分对获得脂肪酶的良好活化至关重要
使用生化测定测试了多种天然皂苷和通过代谢工程衍生的皂苷对单一脂肪酶(Lipex 100L)的活化。还测试了未糖基化的萜类齐墩果酸(OA)以确定脂肪酶活化是否是萜类部分的结果或需要萜类骨架被糖基化。为了直接比较这一点,通过包含在三萜类化合物的C28位置被3个或4个糖苷残基糖基化的齐墩果酸(齐墩果酸-28-O-三葡萄糖苷(OA-28x3Gly)和齐墩果酸-28-O-四葡萄糖苷(OA-28x4Gly)的皂苷的酶活化直接与齐墩果酸获得的活化进行比较。
图3表明,如实施例1所预期的,在10μM下测试的所有皂苷(七叶皂苷、燕麦根皂苷、OA-28x3Gly和OA-28x4Gly)活化Lipex 100L。虽然齐墩果酸对脂肪酶的活化确实很小,这明显低于所有皂苷大于15倍活化且OA-28x3Gly最大地大于20倍的皂苷所获得的活化。因此,我们可以得出结论,虽然萜类骨架是活化所必需的,但骨架上糖残基的存在对于脂肪酶的显著地和商业相关活化是必需的。
图4表示与图3相同的数据,但这次叠加在Lipex 100L活性vs.浓度的图上。这清楚地表明,将脂肪酶浓度从10增加至50ng/ml,10uM皂苷对脂肪酶活性的影响可以相同。鉴于酶在洗衣制剂中通常是最昂贵的组分,包含皂苷可以显著减少所需的酶的量,并且因此降低制剂的总原材料成本。
实施例3清洁研究表明皂苷在制剂中的作用
在tergotometer实验中证明了在存在和不存在脂肪酶的情况下,在25℃下用七叶皂苷替代洗衣洗涤剂的30%或10%的表面活性剂组合物的作用。
研究了两表面活性剂的共混物:
1.70%LAS、15%SLES和15%Neodol 25-7(70/15/15)
2.85%LAS和15%SLES(85/15/0)
总表面活性剂的使用浓度是0.58g/L。
Tergotometer实验在两种水硬度以及使用和不使用25mg/L的Lipex100L下进行。用机织棉上的猪油/结晶紫污渍(来自CFT的测试布)监测清洁性能。结果如图5所示。
图5表明在酶不存在的情况下,七叶皂苷对清洁有不利影响。然而,在脂肪酶存在的情况下,包含皂苷的制剂始终优于不包含皂苷的制剂。
最终表明皂苷活化脂肪酶,因为制剂中的皂苷本身(无脂肪酶)对清洁产生轻微的负面影响。本实施例表明制剂含量的效果。在本实施例中,证明在皂苷使用浓度0.0058%至0.0174%,相当于产品中浓度5.8重量%至17.4重量%下,皂苷对脂肪酶有效活化。
洗涤剂制剂中脂肪酶活性的抑制
在研究脂肪酶的皂苷活化的同时,还观察到高于给定的皂苷浓度下,进一步加入皂苷导致酶活性降低,而不是继续活化。这可以潜在应用于防止瓶子中的脂肪酶活性,其中皂苷以高浓度存在,且否则可能攻击脂肪酶易分解的键,例如酯键。同时,在稀释产品时,一旦皂苷浓度被稀释,在洗涤中将使实现完全酶活化。
实施例4表明皂苷作为脂肪酶活性的天然抑制剂-一种防止脂肪酶催化的去污聚合物降解的方法
测量脂肪酶活性的生化测定最初用于高通量微量滴定板形式,以使不同剂量的不同皂苷能够作为Lipex 100L脂肪酶(Novozymes)的潜在抑制剂进行筛选。寻求有效的抑制剂来防止通常观察到的脂肪酶对去污聚合物的水解活性。表2显示四种作为Lipex 100L脂肪酶活性抑制剂的皂苷。茶皂苷、皂树树皮皂苷和七叶皂苷特别值得注意的是它们能够将脂肪酶活性降低到可忽略不计的水平。当在这种情况下使用10ng/mL脂肪酶(0.3μM)时(选择该水平是为了确保底物水解和由此产生的吸光度测量值增加落在分光光度计的灵敏度范围内),抑制脂肪酶所需的通常皂苷浓度约为50-125μM。

观察到有效皂苷抑制剂结构之间的相似性,具有一个或多个糖部分与其连接的三萜骨架的共同结构特征。
这种抑制的效果是可逆的。图6表明储存的脂肪酶/皂苷样品的稀释(复制洗涤中稀释)如何解除皂苷的抑制作用,从而恢复活性。在含有5mM茶皂苷的溶液中使用洗衣制剂相关水平的Lipex 100L(0.4%w/v),这表现为从含有5mM茶皂苷的样品的1/400稀释液中获得样品中脂肪酶活性的显著提高。洗衣制剂包含0.4%w/v Lipex和5mM QBS。
实施例5表明皂苷作为脂肪酶活性的天然抑制剂-一种防止脂肪酶催化的去污聚合物降解的方法
进一步的研究表明,皂苷可用于抑制另一种真菌脂肪酶(Lipex Evity),从而将该技术的潜在适用性扩展到未来几代洗衣脂肪酶。

实施例6
之前的生化测定测量表明该技术可扩展到洗衣相关的脂肪酶水平。随后进行了洗涤研究,以确定建议在该制剂中最高的最终皂苷浓度1mM(稀释到洗涤液中之后)以确保观察到完全的脂肪酶清洁性能(见图7)。因此,可以在储存的制剂中使用过量的皂苷(考虑到溶解度和成本),以防止针对SRP的脂肪酶活性。

测试了各种皂苷抑制Lipex 100L脂肪酶(最终为10ng/mL)的能力。使用缓冲液(50mM Tris pH 8.5)并在25℃下进行实验。在500μM的固定剂量下,测试的那些皂苷的多种可有效降低脂肪酶活性。用于测试的底物为C12 pNP酯(最终为0.1mM)。这些包括齐墩果酸、燕麦根皂苷、茶树皂苷和七叶皂苷的糖基化衍生物(表4)。奥利司他作为一种已知的脂肪酶抑制剂也被包括用于比较(表4)。
测试的皂苷(OA 28-x4Gly、茶皂苷、七叶皂苷、燕麦根皂苷和OA 28-x3Gly)均显著抑制脂肪酶,范围从80%脂肪酶抑制(OA 28-4xGly)到>90%抑制(茶皂苷、七叶皂苷)到完全抑制(燕麦根皂苷,OA 28-x3Gly)。齐墩果酸(其不是皂苷,因为它不存在糖部分)不能抑制脂肪酶。

这些实施例充分表明,洗涤液中相对低水平的皂苷可以活化脂肪酶。洗涤剂制剂中存在的相对高水平的皂苷可以抑制脂肪酶,从而允许在制剂中包含对脂肪酶敏感的材料。可以单独或共同地观察到这些作用。高度优选的皂苷水平可以抑制洗涤剂制剂(即市场上出售给消费者的洗涤剂)中的脂肪酶,但当产品在洗涤液中稀释时仍能活化脂肪酶。这些作用在许多不同的脂肪酶和许多不同的皂苷中得到证明。
Claims (12)
1.一种在洗涤剂组合物中抑制脂肪酶活性的方法,所述方法涉及将0.25至40重量%,优选地0.5至30重量%,更优选地0.5至25重量%,更优选地1至20重量%,最优选地1至15重量%的皂苷加入到所述包含脂肪酶的组合物中,其中所述洗涤剂组合物优选地为家庭护理洗涤剂组合物,更优选地洗衣洗涤剂组合物。
2.皂苷在洗涤剂组合物,优选洗衣洗涤剂组合物中抑制脂肪酶活性的用途,其中所述皂苷以0.25至40重量%,优选地0.5至30重量%,更优选地0.5至25重量%,更优选地1至20重量%,最优选地1至15重量%的皂苷的含量存在于洗涤剂制剂中,其中所述洗涤剂组合物优选地为家庭护理洗涤剂组合物,更优选地洗衣洗涤剂组合物。
3.根据权利要求1所述的方法,或根据权利要求2所述的用途,其中所述洗涤剂组合物包含:
(i)0.25至40重量%,优选地0.5至30重量%,更优选地0.5至25重量%,更优选地1至20重量%,最优选地1至15重量%的皂苷的皂苷;和,
(ii)0.0005至2.5重量%,优选地0.001至2重量%,更优选地0.005至1重量%的脂肪酶。
4.根据权利要求1至3中任一项所述的方法或用途,其中所述脂肪酶是细菌或真菌来源的。
5.根据权利要求1至4中任一项所述的方法或用途,其中所述皂苷具有三萜骨架和一个或多个连接至所述三萜骨架的糖部分。
6.根据权利要求1至5中任一项所述的方法或用途,其中所述皂苷具有三萜骨架,和两个或更多个连接至所述三萜骨架的糖部分。
7.根据权利要求1至6中任一项所述的方法或用途,其中所述洗涤剂组合物包含1至60重量%,优选地2.5至50重量%,更优选地4至40重量%,最优选地8至35重量%的表面活性剂,所述表面活性剂不包括皂苷。
8.根据权利要求1至7中任一项所述的方法或用途,其中所述洗涤剂组合物包含阴离子和/或非离子表面活性剂,优选地包含阴离子和非离子表面活性剂两者。
9.根据权利要求1至8中任一项所述的方法或用途,其中所述组合物是洗衣洗涤剂组合物,优选地所述洗衣洗涤剂组合物是液体、凝胶或粉末,更优选地所述洗衣洗涤剂是液体洗涤剂。
10.根据权利要求9所述的方法或用途,其中所述洗衣洗涤剂组合物包含烷氧基化聚胺,其含量为优选地0.1至8重量%,更优选地0.2至6重量%,最优选地0.5至5重量%,优选地所述烷氧基化聚胺是烷氧基化聚乙烯亚胺和/或烷氧基化聚丙烯亚胺,更优选地所述烷氧基化是乙氧基化或丙氧基化或两者的混合。
11.根据权利要求9或10所述的方法或用途,其中所述洗衣洗涤剂组合物包含去污聚合物,其含量为优选地0.1至8重量%,更优选地0.2至6重量%,甚至更优选地0.5至5重量%,最优选地1至5重量%,最优选地所述去污聚合物是聚酯去污聚合物。
12.根据权利要求1至11中任一项所述的方法或用途,进一步地包含一种或多种另外的酶,其选自:蛋白酶、纤维素酶、α-淀粉酶、过氧化物酶/氧化酶、果胶酸裂解酶和/或甘露聚糖酶,及其混合物。
