Abstract

本发明公开了一种用于抗病无核葡萄胚挽救培养及成苗培养的培养液,它是由有机营养、无机营养溶解配制而成,它是用于对杂交授粉后剥取的胚珠进行离体胚珠内胚培养,以获得大量杂交后代的胚挽救幼苗,使用本发明可使胚发育率达到20%,成苗率达到12%,且胚发育整齐,畸形胚少,加速了抗病无核葡萄的育种进程,使其达到世界领先地位,本发明将为培育抗病无核葡萄新品种开创广阔的前景。

一种用于抗病无核葡萄胚挽救培养及成苗培养的培养液
                          技术领域
本发明涉及一种农用化肥,特别是一种用于抗病无核葡萄胚挽救培养及成苗培养的培养液。
                          背景技术
葡萄因其含有丰富的营养成份已成为人们喜爱食用的主要水果之一,其栽培面积及产量在世界范围内成上升趋势,而无核葡萄无论鲜食、制干和加工均已成为世界水果消费的重要方向。在美国鲜食葡萄的80%是无核葡萄,澳大利亚、南非及欧洲等葡萄生产大国对无核葡萄的研究与开发也极为重视,而无核葡萄的育种方法是为生产提供优质品种的重要环节,美国《园艺科学》杂志1990年25卷第4期393~398页报道的以欧洲葡萄无核品种间杂交后进行胚挽救的技术是一种经济、高效、快速的无核葡萄育种方法,该方法近10多年来得到迅猛发展,但该方法培养的多数无核葡萄品种其突出缺点是缺乏抗病性。国外葡萄育种者从一个世纪前就试图将欧洲葡萄无核基因与圆叶葡萄抗病基因有机结合在一起,但成效不大,美国《植物细胞》杂志1994年37卷第1期55~59页发表的胚挽救技术是利用将无核品种作母本与圆叶葡萄作父本进行杂交而生成无核葡萄胚珠进行胚挽救培养,但其成苗率极低,仅为1.25%,且植株可育性极低。目前,在世界范围内,其葡萄育种科学家都在为缺乏适宜有效的葡萄抗性材料和培养基而困惑。
中国野生葡萄没有美洲葡萄种群的狐臭味,且抗病抗逆性强,产量高,又与欧亚葡萄种群无核品种亲和性好,易于杂交,是抗病无核育种的宝贵资源,西北农林科技大学园艺学院于2000年借助胚挽救技术用已有的培养基对欧洲无核葡萄作母本、中国野生葡萄作父本杂交的胚珠进行培养,其成苗率也仅达到2.5%,其根本原因是缺少对胚珠进行挽救培养的培养基。
                          发明内容
本发明的目的是解决欧洲葡萄无核品种与抗病的中国野生葡萄杂交后,进行葡萄胚挽救过程中因缺少适宜的培养基而使胚发育率及成苗率极低的问题,而发明了一种适宜抗病无核葡萄胚挽救培养及成苗培养的培养液。
本发明的组分及浓度配比为:

本发明组分合理,制作简单,使用本发明所获得的抗病无核葡萄胚发育整齐,畸形胚少,胚发育率高,成苗率高。以西北农林科技大学葡萄种质资源圃的中国野生葡萄杂种“北醇”为父本,以新疆葡萄瓜果开发研究中心葡萄品种资源圃的无核品种“底来特”作母本杂交,授粉48天,无菌剥取胚珠接种于本发明中进行胚珠培养,胚发育率为15.88%,成苗率达到12.38%。另以“爱莫无核”作母本,“北醇”作父本杂交,授粉48天-52天,接种于本发明中进行胚珠培养,胚发育率达20.02%,成苗率达10.68%,处于目前世界领先水平。
本发明的配制方法是:首先将各组分(除活性炭外)分别用水溶解,然后混合,再加入活性炭,制成培养液。
本发明的使用方法是:在培养液中加0.55~0.7%的琼脂粉,将其加热至沸腾使琼脂粉彻底溶解形成混合液,再趁热将混合液分装于培养瓶中,待其冷却后形成培养基。将无核葡萄胚珠或剥取的胚接种于培养基上进行培养。
本发明主要用于抗病无核葡萄胚挽救培养,若加入一定量的激素后可用于成苗培养。
本发明可用于抗病无核葡萄胚挽救培养及成苗培养;无核葡萄胚挽救培养及成苗的培养;极早熟葡萄胚的培养;葡萄无性胚状体诱导的培养;转基因植株的获得;多倍体葡萄胚培养;部分核果类果树胚的培养。
