Abstract
The invention discloses a pharmaceutical composition for treating gastric ulcer and a preparation method thereof, wherein the pharmaceutical composition comprises the following components in parts by weight: 8-30 parts of fructus amomi, 5-20 parts of sea buckthorn, 2-5 parts of cistanche, 2-5 parts of hawthorn, 4-9 parts of pseudo-ginseng, 2-5 parts of mugwort leaf, 3-8 parts of black nightshade, 3-9 parts of galangal, 5-8 parts of radix angelicae, 2-9 parts of raw radix aconiti, 2-9 parts of fructus kadsurae, 3-9 parts of semen vaccariae and 1-3 parts of speranskia herb. The pharmaceutical composition has remarkable effect of treating gastric ulcer. In addition, the preparation method of the invention adopts a supercritical technology, so that the effective components in the medicinal materials can be fully extracted, and the maximum efficacy is exerted.
Description
Technical Field
The invention relates to the technical field of gastric ulcer treatment, in particular to a pharmaceutical composition for treating gastric ulcer and a preparation method thereof.
Background
Gastric ulcers are ulcers that occur in the corners of the stomach, antrum, cardia, hiatal hernias, etc., and are one type of peptic ulcers. The outline of materia medica is described in: chun Sha ren, zingiberaceae, fructus Amomi, wen Weixin, has the actions of activating qi-flowing and regulating the function of the stomach and activating spleen, and is mainly used for treating chest and gastric distention, stomach and fruit food stagnation, dysphagia and vomiting. The Chinese medicinal composition is advocated for fructus amomi.

Fructus Amomi 20, fructus Hippophae 10, herba cistanches 3, fructus crataegi 4, radix Notoginseng 6, folium Artemisiae Argyi 3, herba Solani Nigri 5, rhizoma Alpiniae Officinarum 5, radix Angelicae Dahuricae 6, radix Aconiti 6, fructus Schisandrae Sphenantherae 5, semen Vaccariae 5, and herba speranskiae tuberculatae 2.
The preparation method of the pharmaceutical composition for treating gastric ulcer comprises the following steps:
(1) Mixing the components, drying, pulverizing, and sieving to obtain a mixture;
(2) Placing the mixture into a closed extraction kettle for supercritical carbon dioxide fluid extraction under the conditions of extraction pressure of 35-40 MPa, extraction temperature of 45-55 ℃, analysis pressure of 8-10 MPa and analysis temperature of 55-65 ℃ for 2-4 hours, and collecting the extract.
Cistanche salsa is a parasitic plant parasitic on the roots of the haloxylon ammodendron of desert trees, and absorbs nutrients and moisture from the haloxylon ammodendron host. The ginseng-ginseng compound wine has the reputation of 'desert ginseng', has extremely high medicinal value, and is a traditional rare Chinese medicinal material in China. Sweet in taste; salty; warm nature. The main functions are as follows: tonify kidney yang, benefit essence and blood, moisten intestine.
Hawthorn fruit, of the genus Hawthorn of the family Rosaceae, has hard core, thin pulp and slightly sour and astringent taste. The dried fruit can be eaten raw or used as a preserved fruit cake, can be used as a medicine after being dried, is a special tree seed for Chinese medicinal fruits, has the effects of reducing blood fat, blood pressure, strengthening heart, resisting arrhythmia and the like, is also a good medicine for strengthening spleen, stimulating appetite, resolving food stagnation, promoting blood circulation and reducing phlegm, and has good curative effects on chest, diaphragm, spleen fullness, hernia, blood stasis, amenorrhea and the like. The flavonoid vitexin in haw is one kind of anticancer medicine with high anticancer effect and its extract has certain effect in inhibiting cancer cell growth, proliferation and infiltration metastasis.
Notoginseng radix is plant of Araliaceae, and the part to be treated is dry root. Has effects of removing blood stasis, stopping bleeding, promoting blood circulation, and relieving pain. It is used for treating hemorrhage, traumatic injury, and swelling and pain due to blood stasis.
The folium Artemisiae Argyi is a Compositae plant, and the part to be used as medicine is the dry leaf of the plant. Pungent and bitter in nature and warm in nature. Is mainly used for treating hematemesis, epistaxis, metrorrhagia, menorrhagia, metrostaxis, lower abdomen cold pain, cold menstruation, infertility due to cold womb; it is used for treating skin pruritus.
The herba Solani Nigri is whole plant of Solanaceae herb herba Solani Nigri. Bitter, slightly sweet and cold. The chemical components comprise saponin, solanine and steroid alkaloid. Has expectorant and antitussive effects, and can inhibit Staphylococcus aureus, bacillus dysenteriae, bacillus typhi, bacillus proteus, escherichia coli, and Pseudomonas aeruginosa.
Radix Angelicae Dahuricae is an Umbelliferae plant, and the part to be treated is dry root. Pungent and warm in nature. It is used for treating common cold due to wind-cold, headache, toothache, rheumatalgia, nasosinusitis, leukorrhagia, sore, carbuncle, and toxic swelling.
Radix Aconiti is a plant of Ranunculaceae and has effects in dispelling pathogenic wind, removing dampness, warming channel, and relieving pain. Can be used for treating arthralgia due to wind-cold-dampness, arthralgia, psychroalgia of heart and abdomen, and cold hernia pain. Can be used for treating arthralgia due to wind-cold-dampness, and arthralgia.
Kadsura longepedunculata is a wrenching shrub, and is grown under the valley-Milin or in the shrub. Distribution: the southeast of China; drug-taking part: root, stem. Sex taste: slightly warm nature, slightly sweet in taste and fragrant in smell. The functions are as follows: dispelling wind and cold, and relieving pain.
Semen Vaccariae is a plant of Caryophyllaceae, and the part to be used as medicine is dry mature seed. Except for south China, the distribution is available in all regions of the country. Has effects of promoting blood circulation, dredging channels, promoting lactation, relieving swelling, inducing diuresis, and treating stranguria. Can be used for treating amenorrhea, dysmenorrhea, galactostasis, acute mastitis, swelling and pain, and stranguria.
The speranskia herb is root or leaf of Rhododendron, or Santali albi. Bitter and pungent in flavor and warm in nature. The functions of the medicine are mainly dispelling wind and removing dampness.
Galangal is a plant of the genus Alpinia of the family Zingiberaceae. The medicine is prepared from rhizome, has the functions of warming middle-jiao, dispelling cold, relieving pain and promoting digestion.

2. The preparation methods of examples 1-3 and comparative example 1 include the following steps:

Clinical diagnostic criteria for the efficacy of treatment of gastric ulcers are classified as efficacious, effective and ineffective, as follows:
(1) The effect is shown: by the fiber endoscope examination, congestion and edema around the mucosa disappear, the substrate roughness becomes smooth, and the ulcer periphery appears as nodular bump and disappears;

(3) Invalidation: by means of a fibre endoscope, all symptoms were unchanged.
After 4 treatment courses, 4 clinical teams were subjected to a fiber endoscope examination, the results of which are shown below:
(1) Experiment group 1: 26 effective cases (87%), 4 effective cases (13%), and 0 ineffective cases;
(2) Experiment group 2: 20 significant effects (67%), 9 effective (30%), 1 ineffective (3%);
(3) Experiment group 3: 21 significant effects (70%), 8 effective effects (27%), 1 ineffective (3%);
(4) Control group 1:1 (3%), 11 (37%), 18 (60%) in effect;
the 4 clinical experiment groups have no toxic or side effect.
From the above results, it can be seen that the composition of comparative example 1 has a very general therapeutic effect on gastric ulcer, and that each of experimental group 1, experimental group 2 and experimental group 3 has a better therapeutic effect on gastric ulcer, and that the therapeutic effect of experimental group 1 is optimal.
