Abstract
本发明涉及一种抗氧化缓解帕金森病的手性纳米酶组装体及其制备方法和应用,属于生物技术领域。本发明以2‑甲基咪唑为有机配体,Zn2+为无机配体,通过仿生矿化途径以Pt纳米酶为核心形成ZIF‑8外层,同时将手性组氨酸分子嵌入ZIF‑8保护层,构建共载手性组氨酸和Pt纳米酶的手性纳米酶组装体。本发明提供的制备方法条件温和,操作简单,制备得到的手性纳米酶颗粒粒径小,分散性好,稳定性强,经尾静脉注射给帕金森病小鼠后,在小鼠体内半衰期较长,且在6.7 h仍然保持较高的SOD酶样活性,该组装体可以穿透血脑屏障进入小鼠大脑,有效清除MPTP刺激在脑部产生的过量的ROS,从而缓解帕金森病行为障碍。

本发明属于生物技术领域,具体涉及一种抗氧化缓解帕金森病的手性纳米酶组装体及其制备方法。
背景技术
帕金森病是一种严重且常见的神经退行性疾病,研究发现,脑中过量产生的活性氧得不到及时清除,是导致帕金森疾病的一个重要原因。利用抗氧化酶体系来降低胞内活性氧水平,缓解由氧化应激造成的神经元变性损伤,有望成为基于抗氧化系统缓解帕金森病的有效策略。抗氧化酶要想发挥其催化清除氧自由基的功效,必须保证其在药品和载体中都保持活性且能稳定穿透生物膜屏障到达病理部位。然而,如果直接使用天然抗氧化酶,其制备、运输和给药过程中,往往因为本身易降解、半衰期短造成活性降低,以及在经过血脑屏障时受到各类生物膜的阻碍,很难确保天然抗氧化酶到达靶向部位时还具有高效的活性去发挥清除自由基的药理效用。针对天然游离酶易失活和跨膜能力差等缺点,开发一种体内半衰期较长,稳定且能高效地穿透血脑屏障,对人体低毒性和生物安全性较高的人工模拟酶,是本技术领域亟待研究和解决的问题。
金属有机框架(MOF)是由金属节点和有机配体通过配位键结合而成的一类杂化材料,具有比表面积高、易调节和修饰、稳定性强、合成条件温和等优点,也是常见的固定游离酶的良好载体。仿生矿法是利用共沉淀方式进行固定游离酶的一种类型,具有反应条件温和、适用性高、合成路径简单等优点。因此,利用仿生矿技术将人工模拟酶固载在金属有机框架中有望为天然酶的活性替代研究提供一种有效方法。
发明内容
针对现有技术中存在的问题和不足,本发明的目的旨在提供一种抗氧化缓解帕金森病的手性纳米酶组装体及其制备方法。
基于上述目的,本发明采用如下技术方案:
本发明第一方面提供一种抗氧化缓解帕金森病的手性纳米酶组装体的制备方法,具体方法为:将2-甲基咪唑、十六烷基三甲基溴化铵和手性组氨酸加入水中,搅拌混匀,向混合溶液中依次加入Pt纳米酶溶液和Zn(NO3)2溶液,室温反应5~180 min,反应结束后,将反应液离心,收集沉淀物,用水重悬沉淀物后进行超声分散,再次收集沉淀物,将沉淀物洗涤、冷冻、干燥后即得手性纳米酶组装体。
优选地,所述2-甲基咪唑、十六烷基三甲基溴化铵和手性组氨酸的摩尔比为(400~2000):1:(10~100)。
更为优选地,所述2-甲基咪唑、十六烷基三甲基溴化铵和手性组氨酸的摩尔比为(1400~2000):1:(40~100)。
优选地,所述Pt纳米酶溶液的浓度为1~10 mg/mL,所述Zn(NO3)2溶液的浓度为40~300 mM,2-甲基咪唑与Zn(NO3)2的摩尔比为4~160:1,Pt纳米酶溶液与Zn(NO3)2溶液的体积比为1:1~5。
更为优选地,所述Zn(NO3)2溶液的浓度为40~100 mM,2-甲基咪唑与Zn(NO3)2的摩尔比为40~100:1。

(1)本发明以2-甲基咪唑为有机配体,Zn2+为无机配体,通过仿生矿化途径以Pt纳米酶为核心形成ZIF-8外层,同时将手性组氨酸分子嵌入ZIF-8保护层,构建共载手性组氨酸和Pt纳米酶的共组装体系,与现有的技术相比,本发明提供的制备方法操作简单且合成条件温和,能够很好地维持酶分子催化性能的稳定性。
(2)本发明中手性组氨酸和十六烷基三甲基溴化铵的加入能够调节ZIF的尺寸大小,控制引入金属有机骨架包裹Pt纳米酶后的组装体粒径尺寸小于200 nm,有助于手性纳米酶组装体通过血脑屏障顺利到达脑部,发挥抗氧化酶活性,调节氧化应激水平。
(3)动物研究发现,向帕金森病小鼠尾静脉注射本发明制备的手性纳米酶组装体,该手性纳米酶组装体在小鼠体内的半衰期较长,6.7 h后仍然保持较高的SOD酶样活性,且该手性纳米酶组装体24 h可以进入小鼠大脑,实现小鼠机体内ROS的长效清除,有效缓解帕金森病小鼠的行为障碍,对帕金森疾病有着明显的治疗作用。
(4)本发明为天然酶的活性替代研究提供了行之有效的方法,可以解决天然酶制剂在生产、使用过程中活性、数量及效价损失严重的问题,具有较强的研究价值和商业指导意义,前景广阔,竞争力强,经济效益高。

具体实施方式
为使本领域的技术人员更好地理解本发明的技术方案,以下结合附图通过具体的实施例对本发明作进一步说明,但并不限制本发明的范围。
实施例1:
一种抗氧化缓解帕金森病的手性纳米酶组装体的制备,具体包括以下步骤:
(1)将133 mg聚乙烯吡咯烷酮(分子量30 KDa)溶于180 mL甲醇中,然后加入20 mL溶有48 mg H2PtCl6的去离子水溶液,充分混合,于70℃,转速400 rpm下磁力搅拌反应3 h,反应结束后,将反应液旋蒸除去多余甲醇后超声震荡,收集沉淀物,沉淀物经正己烷多次洗涤至中性后放入真空干燥箱干燥,得到Pt纳米酶;
(2)称量331.65 mg 2-甲基咪唑,1 mg十六烷基三甲基溴化铵和 19.15 mg D–组氨酸溶于5 mL去离子水中,转速400 rpm搅拌混匀,再向混合溶液中依次滴入1 mL溶有1 mgPt纳米酶的去离子水溶液和1 mL溶有29.06 mg Zn(NO3)2•6H2O的去离子水溶液,室温持续搅拌20 min,反应结束后,将反应液以转速5000 rpm离心5 min,收集沉淀物,使用去离子水重悬沉淀物后进行超声分散,再次收集沉淀物,将沉淀物洗涤2次、冷冻、干燥后即得手性纳米酶组装体Ptzyme@D-ZIF-8。
实施例2:
一种抗氧化缓解帕金森病的手性纳米酶组装体的制备,具体包括以下步骤:
(1)将133 mg聚乙烯吡咯烷酮(分子量30 KDa)溶于180 mL甲醇中,然后加入20mL溶有48 mg H2PtCl6的去离子水溶液,充分混合,于70℃,转速400 rpm下磁力搅拌反应3 h,反应结束后,将反应液旋蒸除去多余甲醇后超声震荡,收集沉淀物,沉淀物经正己烷多次洗涤至中性后放入真空干燥箱干燥,得到Pt纳米酶;

实施例3:
一种抗氧化缓解帕金森病的手性纳米酶组装体的制备,具体包括以下步骤:
(1)将133mg聚乙烯吡咯烷酮(分子量30 KDa)溶于180 mL甲醇中,然后加入10 mL溶有48 mg H2PtCl6的去离子水溶液,充分混合,于70℃,转速400 rpm下磁力搅拌反应3 h,反应结束后,将反应液旋蒸除去多余甲醇后超声震荡,收集沉淀物,沉淀物经正己烷多次洗涤至中性后放入真空干燥箱干燥,得到Pt纳米酶;
(2)称量331.65 mg 2-甲基咪唑,1 mg十六烷基三甲基溴化铵和19.15mgD–组氨酸溶于5 mL去离子水中,转速400 rpm搅拌混匀,再向混合溶液中依次滴入1 mL溶有1 mg Pt纳米酶的去离子水溶液和1 mL溶有14.60 mg Zn(NO3)2•6H2O的去离子水溶液,室温持续搅拌20 min,反应结束后,将反应液以转速5000 rpm离心5 min,收集沉淀物,使用去离子水重悬沉淀物后进行超声分散,再次收集沉淀物,将沉淀物洗涤2次、冷冻、干燥后即得手性纳米酶组装体Ptzyme@D-ZIF-8。
实施例4:
一种抗氧化缓解帕金森病的手性纳米酶组装体的制备,具体包括以下步骤:
(1)将133mg聚乙烯吡咯烷酮(分子量30 KDa)溶于180 mL甲醇中,然后加入10 mL溶有24 mg H2PtCl6的去离子水溶液,充分混合,于70℃,转速400 rpm下磁力搅拌反应3 h,反应结束后,将反应液旋蒸除去多余甲醇后超声震荡,收集沉淀物,沉淀物经正己烷多次洗涤至中性后放入真空干燥箱干燥,得到Pt纳米酶;
(2)称量331.65 mg 2-甲基咪唑,1mg十六烷基三甲基溴化铵和38.30 mg D–组氨酸溶于5 mL去离子水中,转速400 rpm搅拌混匀,再向混合溶液中依次滴入1 mL溶有1mg Pt纳米酶的去离子水溶液和1 mL溶有29.06 mg Zn(NO3)2•6H2O的去离子水溶液,室温持续搅拌20 min,反应结束后,将反应液以转速5000 rpm离心5 min,收集沉淀物,使用去离子水重悬沉淀物后进行超声分散,再次收集沉淀物,将沉淀物洗涤2次、冷冻、干燥后即得手性纳米酶组装体Ptzyme@D-ZIF-8。
对比例1:
对比例1的内容与实施例1基本相同,不同之处在于:步骤(2)中未加入D–组氨酸,最终制得纳米酶组装体Ptzyme@ZIF-8。
结构表征和性能测试:
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF、Ptzyme@ZIF-8进行性能表征。
1. 红外光谱表征:
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Ptzyme@ZIF-8进行红外光谱分析,结果如图1所示。
由图1可知,D-组氨酸和L-组氨酸修饰的金属有机骨架Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8在1650~1690出现的吸收峰为游离态氨基峰,说明本发明成功对金属有机骨架进行组氨酸修饰,能够为金属有机骨架提供手性结构。
2. 圆二色谱表征:
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Ptzyme@ZIF-8进行圆二色谱分析,结果如图2所示。
由图2可知,D-组氨酸和L-组氨酸修饰的金属有机骨架Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8具有对称圆二色谱峰,未经组氨酸修饰的金属有机骨架Ptzyme@ZIF-8未检测到圆二色谱峰,说明本发明成功制备了具有手性结构的金属有机骨架。
3. 水合粒径检测:
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Ptzyme@ZIF-8进行水合粒径检测。
具体方法为:分别将实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8和Ptzyme@ZIF-8分散到超纯水中,进行无限稀释,防止浓度过高引起纳米颗粒之间相互作用力增大,粒径增大,在将稀释后的溶液加入四通的皿池中,37℃条件下进行水合粒径的检测,结果如图3所示。
由图3可知,实施例1制备的Ptzyme@D-ZIF-8的平均水合粒径为120 nm,实施例2制备的Ptzyme@L-ZIF-8平均水合粒径为188 nm,与对比例1制备的Ptzyme@ZIF-8相比,说明加入的手性氨基酸能够调节ZIF尺寸大小,控制引入金属有机骨架包裹的高效Pt纳米酶达到纳米级大小,有利于增强酶的催化效率,有效地提高组织的摄取率,有助于组装体通过血脑屏障顺利到达脑部。
4. 半衰期测定:
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Ptzyme@ZIF-8进行半衰期测定。
半衰期测定的具体操作为:将实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Ptzyme@ZIF-8分别单次尾静脉注射给正常的C57BL/6小鼠(商购于北京斯贝福生物计数有限公司),三组给药剂量均为5 mg/kg/次,三组分别在不同的时间点(10min、0.5h、1h、2h、4h、8h、24h、48h和72 h)静脉取血,采用WST-法对血浆中的SOD样活性进行检测,检测结果如图4所示。
从图4中可以看出,Ptzyme@ZIF-8的半衰期为2.2 h。两种手性纳米酶组装体的酶活性衰减速率不同,Ptzyme@L-ZIF-8在血液中的酶活性衰减速率明显下降,尾静脉注射10min后的活性减少了20%,其半衰期为1.2 h,说明Ptzyme@L-ZIF-8会快速被血液中蛋白酶降解;而Ptzyme@D-ZIF-8则相对稳定,半衰期延长至6.7 h。体内药物清除实验表明,相比Ptzyme@L-ZIF-8,Ptzyme@D-ZIF-8在血液中更加稳定,半衰期显著增加,可能在体内清除ROS,治疗帕金森疾病具备更优良的作用。
5. 血脑屏障透过性测定:
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Ptzyme@ZIF-8进行血脑屏障透过性测定:
(1)以MPTP(1-甲基-4-苯基-1,2,3,6-四氢吡啶)腹腔注射C57BL/6J小鼠的方式构建帕金森疾病动物模型,MPTP注射的剂量为35 mg/kg/day。MPTP注射24小时后,以尾静脉给药方式治疗,给药组的含量均为5 mg/kg,实验分组有以下五组:
①Control组(不注射MPTP,生理盐水给药)
②MPTP组(注射MPTP,生理盐水给药)
③MPTP+Ptzyme@D-ZIF-8组(注射MPTP,实施例1合成的Ptzyme@D-ZIF-8给药)
④MPTP+Ptzyme@L-ZIF-8组(注射MPTP,实施例2合成的Ptzyme@L-ZIF-8给药)
⑤MPTP+Ptzyme@ZIF-8(注射MPTP,对比例1合成的Ptzyme@ZIF-8给药)
(2)实验方法:
对给药24 h后的小鼠脑部的黑质致密部进行冷冻切片,利用冷冻透射电镜观察Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8的分布情况,结果如图5所示。
(3)实验结果:
如图5,在拍摄视野中可以看到,相比Control组和MPTP组,MPTP+Ptzyme@D-ZIF-8组的Ptzyme@L-ZIF-8纳米粒子和MPTP+Ptzyme@L-ZIF-8组的Ptzyme@D-ZIF-8纳米粒子均存在于小鼠脑组织,说明Ptzyme@L-ZIF-8和Ptzyme@D-ZIF-8纳米粒子能穿过小鼠血脑屏障,到达脑部位,Ptzyme@D-ZIF-8纳米粒子分布更多的原因可能是具有较低的粒径大小。而Ptzyme@ZIF-8并没有在脑部的富集。以上实验结果说明,Ptzyme@L-ZIF-8和Ptzyme@D-ZIF-8由于具有纳米级别的粒径和合适的形貌,能够成功跨越血脑屏障进入脑组织实质;并且Ptzyme@D-ZIF-8具有更优的脑部富集效率。
6. 体内清除ROS的能力测定
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Ptzyme@ZIF-8进行小鼠体内清除ROS的能力测定:
(1)以MPTP腹腔注射C57BL/6J小鼠的方式构建帕金森疾病动物模型,MPTP注射的剂量为35 mg/kg/day,注射时间为2 min。MPTP注射24 h后,尾静脉给药方式治疗,给药组的含量均为5 mg/kg,实验分组有以下五组:
①Control组(不注射MPTP,生理盐水给药)
②MPTP组(注射MPTP,生理盐水给药)
③MPTP+Ptzyme@D-ZIF-8组(注射MPTP,实施例1合成的Ptzyme@D-ZIF-8给药)
④MPTP+Ptzyme@L-ZIF-8组(注射MPTP,实施例2合成的Ptzyme@L-ZIF-8给药)
⑤MPTP+Ptzyme@ZIF-8(注射MPTP,对比例1合成的Pt@ZIF-8-8给药)
(2)实验方法:
根据总ROS含量检测试剂盒检测鼠脑部的ROS含量,对脑黑质部ROS水平进行检测,ROS水平检测结果如图6所示。
(3)实验结果:
从图6可以看出,Ptzyme@D-ZIF-8和Ptzyme@L-ZIF-8可以有效降低MPTP刺激在脑部产生的过量的ROS,且Ptzyme@D-ZIF-8效果优于Ptzyme@L-ZIF-8。而Ptzyme@ZIF-8对脑部ROS没有清除效果。Ptzyme@D-ZIF-8清除氧自由基效率更高的原因可能是具有较低的粒径大小,比表面积更大,具有更高效的催化抗氧化酶能力。以上实验结果说明,Ptzyme@D-ZIF-8能够更好的清除闹病灶部位ROS,从而在治疗PD方面展现出应用潜能。
7. 对帕金森模型鼠游泳能力的影响
对实施例1、实施例2、对比例1合成的Ptzyme@D-ZIF-8、Ptzyme@L-ZIF-8、Pt@ZIF-8进行帕金森模型鼠游泳能力的影响实验:
(1)以MPTP腹腔注射C57BL/6J小鼠(北京斯贝福)的方式构建帕金森疾病动物模型,MPTP注射的剂量为35 mg/kg/day,注射时间为2 min。MPTP注射24 h后,尾静脉给药方式治疗,给药组的含量均为5 mg/kg,实验分组有以下五组:
①Control组(不注射MPTP,生理盐水给药)
②MPTP组(注射MPTP,生理盐水给药)
③MPTP+Ptzyme@D-ZIF-8组(注射MPTP,实施例1合成的Ptzyme@D-ZIF-8给药)
④MPTP+Ptzyme@L-ZIF-8组(注射MPTP,实施例2合成的Ptzyme@L-ZIF-8给药)
⑤MPTP+Pt@ZIF-8组(注射MPTP,对比例1合成的Ptzyme@ZIF-8给药)
(2)实验方法:
采用Morris水迷宫实验测试,将每只小鼠单独放到直径为120 cm的黑色圆形水池中,其水深为60 cm,水温恒定为22 ℃。水池壁上标有东、南、西、北四个方位入水点,将水池划分为四个象限,在东南象限正中央处放一直径为10 cm的平台,且池水没过平台1 cm,由上方的图像自动采集记录小鼠游泳活动的路程轨迹以便后续分析。将4组小鼠置于Morris水箱中进行游泳能力的测试,结果如图7所示。
Morris水迷宫的基本思路为:鼠类擅长游泳却厌恶处于水中沉浸的状态,它们会为了脱离水中的环境,本能地搜索水中的躲避场所,这种过程涉及到大脑对空间信息进行整理、记忆等复杂高级处理,在一定程度上反映了空间记忆学习能力。
(3)实验结果:
从图7可以看出,最终只有Control组Ptzyme@D-ZIF能找到目标平台。定位航行过程中,MPTP损伤组的游泳路线相比Control组,路线变得曲折复杂,搜索策略主要是随机式或边缘式,说明MPTP对C57BL/6小鼠学习记忆能力具有损伤作用,小鼠伴有焦虑的症状。而经过引入Ptzyme@D-ZIF-8治疗后,路线变得较为简单,搜索策略倾向于直线式或者趋向式。表明Ptzyme@D-ZIF-8能对MPTP损害小鼠的空间学习功能有修复作用。然而,Ptzyme@L-ZIF-8和Ptzyme@ZIF-8治疗后,小鼠在行为学方面的表现明显差于Ptzyme@D-ZIF-8治疗后的小鼠,路线仍然曲折复杂,搜索策略主要以随机式或边缘式为主,类似于MPTP损伤组小鼠的行为学模式。造成这一差异的原因可能是Ptzyme@D-ZIF-8在体内的半衰期较长,脑内富集性较好,从而较为显著改善帕金森病小鼠的行为特征。
上述实施例为本发明的具体实施方式,但本发明的实施方式并不受上述实施例的限制,其它任何不超出本发明设计思路组合、改变、修饰、替代、简化,均落入本发明的保护范围之内。
Claims (10)
1. 一种抗氧化缓解帕金森病的手性纳米酶组装体的制备方法,其特征在于,所述制备方法为:将2-甲基咪唑、十六烷基三甲基溴化铵和手性组氨酸加入水中,搅拌混匀,得到混合溶液;向混合溶液中依次加入Pt纳米酶溶液和Zn(NO3)2溶液,室温反应5~180 min,反应结束后,将反应液离心,收集沉淀物,用水重悬沉淀物后进行超声分散,再次收集沉淀物,将沉淀物洗涤、冷冻、干燥后即得手性纳米酶组装体。
2.如权利要求1所述的制备方法,其特征在于,所述2-甲基咪唑、十六烷基三甲基溴化铵和手性组氨酸的摩尔比为(400~2000):1:(10~100)。
3.如权利要求2所述的制备方法,其特征在于,所述2-甲基咪唑、十六烷基三甲基溴化铵和手性组氨酸的摩尔比为(1400~2000):1:(40~100)。
4. 如权利要求1所述的制备方法,其特征在于,所述Pt纳米酶溶液的浓度为1~10 mg/mL,所述Zn(NO3)2溶液的浓度为40~300 mM,2-甲基咪唑与Zn(NO3)2的摩尔比为4~160:1,Pt纳米酶溶液与Zn(NO3)2溶液的体积比为1:1~5。
5. 如权利要求4所述的制备方法,其特征在于,所述Zn(NO3)2溶液的浓度为40~100mM,2-甲基咪唑与Zn(NO3)2的摩尔比为40~100:1。
6.如权利要求1所述的制备方法,其特征在于,所述手性组氨酸为D-组氨酸或L-组氨酸。
7. 如权利要求1~6任一所述的制备方法,其特征在于,所述Pt纳米酶溶液是将Pt纳米酶溶于水中配制而成,所述Pt纳米酶的制备方法为:将聚乙烯吡咯烷酮溶解在溶剂中,得到聚乙烯吡咯烷酮溶液,再向其中加入H2PtCl6溶液,于60 ℃~90 ℃搅拌反应3 h~24 h,反应结束后,将反应液超声震荡,收集沉淀物,再将沉淀物洗涤、干燥,即得Pt纳米酶。
8. 如权利要求7所述的制备方法,其特征在于,H2PtCl6溶液的浓度为1~10 mg/mL,聚乙烯吡咯烷酮与H2PtCl6的质量比为2~20:1。
9.一种利用权利要求1~8任一所述方法制备的抗氧化缓解帕金森病的手性纳米酶组装体。
10.权利要求9所述的一种抗氧化缓解帕金森病的手性纳米酶组装体在制备治疗帕金森病药物中的应用。
