Efficacy evaluation method and results:
(1) Method for evaluating protection effect of product on zebra fish drug-induced eye injury by using zebra fish disease model
Benzalkonium bromide and ethylparaben are preservatives widely used in ophthalmic formulations, and their accumulation on the ocular surface is an important factor causing drug toxicity of ophthalmic formulations. Experiments randomly select 150-tail 3dpf zebra fish with normal development in six-hole plates, 30-tail each hole is treated by taking 0.1% DMSO as a control group, and the model group is placed in a mixed liquid medicine of benzalkonium bromide (5 mug/ml) and ethyl hydroxy benzoate (20 mug/ml) to induce zebra fish eye injury. Administration group example 5 was given instant granule concentrations of 10, 20 and 40 μg/mL with a 3mL per well capacity. Culturing at 28.5deg.C for 72 hr, changing liquid once every 24 hr, and observing the eye morphology change of the young zebra fish under a microscope. Iris bandwidth and pupil diameter were measured using Image J software and the ratio of the two was calculated.
As shown in fig. 1, compared with the normal control group, the control group of the eye injury model has the advantages that the ratio of the iris bandwidth to the pupil diameter of the zebra fish is obviously reduced, the iris edge is irregular, and the result is similar to the tearing or fracture symptoms of the human iris sphincter, so that the model is successfully established; the instant particles 10, 20 and 40 mug/mL mass concentration groups of the embodiment 5 have an improvement effect on the iris bandwidth and pupil diameter of the zebra fish, namely the low dose group, the medium dose group and the high dose group in the figure, wherein the ratio of the low dose group to the medium dose group to the high dose group is 4.65, 5.53 to 7.62, and the p is less than 0.01. The instant granules for nourishing the liver and protecting the eyes of children are proved to have the effect of repairing the damage of the eyes of the young zebra fish induced by benzalkonium bromide and ethyl hydroxybenzoate.
(2) Evaluation of product's protective action on zebra fish liver using zebra fish disease model
180 3d zebra fish were randomly selected in six well plates, 30 tails per well, and acetaminophen was used to induce liver damage in the zebra fish. The instant particle concentrations of example 5 were given at 10, 20 and 40. Mu.g/mL, respectively, with a positive control N-acetylcysteine (NAC) concentration of 1.5. Mu.g/mL, while setting the normal control and model control at 3mL per well. Example 5 after 48h of co-treatment of instant particles with acetaminophen, 10 zebra fish were randomly selected from each experimental group, photographed under a microscope and data were collected, and zebra fish liver area (a), liver brightness average (S) and yolk sac absorption delay area (a) were calculated for quantitative analysis, and statistics were performed, and the statistical treatment results were expressed in mean±se. Example 5 the calculation formula for the instant granule liver injury protection is as follows:
wherein A1 to A3, S1 to S3 and A4 to A6 are respectively the liver area, liver brightness and yolk sac absorption delay area of the normal control group, the model control group and the instant granule experimental group of example 5. The experimental results are shown in table 1 below and fig. 2.
As shown in table 1 and fig. 2, the significant differences of the liver area, the liver brightness average value and the yolk sac absorption delay area of the zebra fish are p < 0.01 compared with the normal control group, which indicates that the model establishment is successful; the positive control drug NAC1.5 mug/mL mass concentration group has an improvement effect (the improvement effect is 40 percent, and p is less than 0.01) on the liver degeneration of the zebra fish, but has no obvious improvement effect on the liver atrophy of the zebra fish and the absorption delay of the yolk sac; example 5 instant particles 10, 20 and 40. Mu.g/mL mass concentration groups all had an improvement in liver degeneration in zebra fish (low, medium and high dose groups improved by 37%, 52%, respectively),
Table 1 liver protection of zebra fish after treatment (n=30)
Note that: in comparison with the control group of the model, * p<0.01。
74%, p < 0.01). The instant granules for children to nourish liver and protect eyes are proved to have the effect of protecting liver.
