背景技术
植物根结线虫病是一类全球性的病害,根结线虫主要侵染植物根系,形成白色球状根结,随着病害逐渐加重,根结变大,表面产生褶皱且颜色加深变为棕褐色,导致植物营养吸收受阻,植株地上部萎蔫,叶片黄化、脱落,后期巨细胞空洞化,根部腐烂,严重时导致植株整株死亡。目前根结线虫病的防治以化学防治为主,主要药剂包括噻唑膦、阿维菌素等,但是化学药剂的施用存在着农药残留,使用安全性等问题,因此我们不断寻求更加绿色环保的新型防治方法与思路。
近年来蔬菜产业发展迅速,蔬菜种植面积不断扩大,随着人们生活质量的提高,对蔬菜的品质也提出了更高的要求。而生产中通过盲目增施肥料,尤其是化肥,获得高产已成为一种普遍现象,过量施用化肥会造成生产成本大幅度增加、土壤质量退化及环境污染严重等诸多问题,严重影响我国农产品安全及农业生产的可持续发展。
利用有益微生物来控制病害的发生以及诱导提高作物抗逆能力、促进生长、提升果实品质是近些年来研究的热点,尤其这些微生物对环境无污染,引起了科学家们利用生物防治手段控制病害、以及应对不良环境的兴趣。
专利CN106399131B《一株产红青霉菌及其应用》提供了一株产红青霉菌菌株(Penicillium rubens)PR1,该菌株已保藏于中国微生物菌种保藏管理委员会普通微生物中心,简称CGMCC,地址为:北京市朝阳区北辰西路1号院3号,中国科学院微生物研究所,菌种保藏号为CGMCC NO.13189;并公开了所述菌株提取物在促进农作物生长或增产中的应用。

针对上述现有技术,本发明的目的在于,提供产红青霉菌(Penicillium rubens)PR1菌丝提取物的应用。
本发明所述产红青霉菌PR1菌丝提取物的应用,具体为提高植物对根结线虫的抗性及提升果实品质;本申请所述产红青霉菌PR1菌丝提取物参照专利CN106399131B中公开的方法制得。
优选地,所述植物为番茄,所述果实品质为番茄果实和黄瓜果实。
优选地,提升果实品质具体指的是提高番茄果实中维生素C、番茄红素、游离氨基酸及可溶性糖的含量。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,本发明用以下具体实施例进行说明,但绝非仅限于此。以下所述为本发明较好的实施例,仅仅用于描述本发明,不能理解为对本发明的限制,应当指出的是在本发明的精神和原则之内所做的任何修改、等同替换和改进,均应包含在本发明的保护范围之内。
本发明所述产红青霉菌PR1菌丝提取物是根据专利CN106399131B实施例2中植物诱抗剂的制备方法制得,具体如下:
1、发酵培养:
将4℃下保存在试管斜面上的产红青霉(Penicillium rubens)PR1的菌种,接到平板PDA培养基上,25℃培养6天,用打孔器琼脂挖块接种于装有50 mL种子培养液(PDA液体培养基)的250mL三角瓶,于25℃,180r/min下旋转摇床上培养3天作为种子,以10%量接种入装有150mL发酵培养基的500mL三角瓶中,同样条件下培养5天,终止发酵,得种子液。
PDA培养基配方:马铃薯200g,葡萄糖20g,琼脂20g,蒸馏水1000mL。
发酵培养基配方:马铃薯提取液1.0L,酵母膏1.0g,蛋白胨3.0g,葡萄糖15.0g,琼脂17.0g。
马铃薯提取液的制备:取去皮马铃薯200g,切成小块,加蒸馏水1000mL煮沸30min,滤去马铃薯块,将滤液补足至1000mL。
2、乙醇提取:
将发酵液离心过滤,得菌丝体,将菌丝体在60℃下干燥、粉碎,加入乙醇冷浸24h,然后超声提取1h;重复提取3次,合并滤液,即得本发明所述菌丝提取物。所述乙醇的加入量为菌丝体重量的20%;每次提取加入乙醇的量相同。
实施例1 产红青霉菌PR1菌丝提取物对植物根结线虫的防效
1、基质准备:将基质灭菌,加水充分混匀,装入育苗盘,表面铺平,点入番茄种子,每穴孔1颗,播种深度0.5cm;于温度23℃,湿度50%-60%,光照6000-7000 lux,16/8h人工气候室内培养,期间定期浇水,待长至两叶一心期选择长势均匀一致的幼苗移栽至小花盆中。
2、移栽与处理:试验土壤取自山东省泰安市岱岳区大汶口镇东大吴村(117°8'21"E, 35°59'7" N)常年发生根结线虫病的日光温室,田间取土样过筛(2.5mm),除去大颗粒物,过筛后土壤与基质混合均匀(比例约2:1),装盆,移入番茄植株。
3、处理液准备:将产红青霉菌PR1菌丝提取物溶于清水,分别配制成浓度为5ng/mL、10ng/mL、20ng/mL、33ng/mL、50ng/mL、100ng/mL的溶液。
4、试验处理:移栽后缓苗3天进行试验处理,试验设灌施处理和叶面喷施处理,每7天处理一次,各处理设5盆重复;
其中灌施处理设4组,分别为:CK1:灌施清水,G5:5ng/mL溶液灌施处理;G10:10ng/mL溶液灌施处理;G20:20ng/mL溶液灌施处理;灌施用量为每株100mL。
其中叶面喷施处理设4组,分别为:CK2:喷施清水;Y20:20ng/mL溶液叶喷处理;Y33:33ng/mL溶液叶喷处理;Y50:50ng/mL溶液叶喷处理,Y100:100ng/mL溶液叶喷处理;叶喷处理具体为,将溶液加入小喷壶中均匀喷洒至叶片表面,液滴在叶片表面均匀分布,悬而不落,叶喷用量为每株4mL。
5、试验统计:各处理液处理30天后,进行结果检查:
(1)表型测定:用直尺测量番茄地上部分株高,用剪刀剪去地上部分,天平测定地上部分鲜重;测定结果如表1所示:

由表1可知,产红青霉菌PR1菌丝提取物处理组在灌施条件和喷施条件下的株高和鲜重与CK1和CK2组相比均有提升,且差异显著;另外,灌施处理组的株高和鲜重优于喷施处理组。
(2)根系测定:将番茄根部取出,流动水冲洗干净,吸水纸吸干根系表面的水分,天平测定地下部分根重,并将番茄根系置于解剖镜下进行观察计录根结数,具有明显分界记为一个根结,番茄根重及根结数如表2所示;

由表2可知,产红青霉菌PR1菌丝提取物处理组番茄在灌施条件下和喷施条件下的根重均高于CK1和CK2,且差异显著;单株根结数及每克根结数与CK1和CK2相比明显减少,且差异显著;可见,两种处理方式均可以提高番茄根重并降低番茄根结数量,且灌施效果优于喷施。
(3)病情指数及防治效果测定:根据根结发生情况进行防治效果统计;其中根结分级标准:0级,无根结;1级,0-10%根结;2级,11-20%根结;3级,21-30%根结;4级,31-40%根结;5级,41-50%根结;6级,51-60%根结;7级,61-70%根结;8级,71-80%根结;9级,81-90%根结;10级,91-100%根结。
根结指数=Ʃ(病株数×对应根结级数)/(总株数×最高根结级数)× 100
防治效果(%)= [(对照组的根结指数-处理组的根结指数)/ 对照组的根结指数]× 100
番茄植株的病情指数及防治效果如表3所示;

由表3结果可知,灌施处理组中,溶液浓度为10ng/mL时,防治效果最佳,而在溶液浓度为20ng/mL时,防治效果反而降低;喷施处理组中,溶液浓度为50ng/mL时,防治效果最佳,当溶液浓度增加至100ng/mL时,防治效果反而降低,说明产红青霉菌PR1菌丝提取物在高浓度下会引起植物免疫系统的过度反应,导致番茄生长受到抑制,抵抗病害的能力减弱;另外,由表中数据可知,灌施防效优于喷施。
(4)土壤虫口密度测定:浅盘法测定土壤虫口密度,称取各处理土壤50 g,用卫生纸包好,置于塑料穴盘中,卫生纸与塑料穴盘底部有5 mm空隙,沿盘边缘缓慢加入去离子水直至没过卫生纸包裹好的土壤,将其放置于28℃培养箱中培养,每间隔24 h将各处理穴盘中的水溶液取出,记录溶液体积,同时取水样进行线虫观察计数,直至水中无线虫孵化,统计线虫总数,试验重复三次;测定结果如表4所示:

由表4可知,灌施处理组和喷施处理组的虫口密度均高于CK1和CK2,由此推测,施用PR1菌丝提取物后,土壤中线虫不易侵染番茄根系,徘徊在根外土壤中;说明施用PR1菌丝提取物后,植物体内与防御反应相关的路径被激活,防御相关酶与信号物质如水杨酸等大量表达,植株根系分泌物发生改变,对根结线虫具有趋避作用的化合物作用增强,导致根系土壤虫口密度增加而植株病情指数降低;且灌施处理组的虫口密度高于喷施处理组。
实施例2 产红青霉菌PR1菌丝提取物对番茄果实品质的影响
按照实施例1所述方法中的步骤1和2准备盆栽基质并移栽番茄幼苗至大花盆,按照实施例1所述方法中的步骤3和4进行试验处理,灌施处理每株500ml,喷施处理每株约20ml,每7天处理一次,各处理5盆重复;果实成熟后,选取同一成熟度的果实进行品质测定,测定结果如表5所示;

由表5可知,灌施和喷施条件下,PR1菌丝提取物处理组番茄维生素C、番茄红素、游离氨基酸、可溶性糖含量增加,且与CK1、CK2组差异显著;说明施用产红青霉菌PR1菌丝提取物能够显著提升番茄果实品质。
实施例3 产红青霉菌PR1菌丝提取物对黄瓜果实品质的影响
按照实施例1所述方法中的步骤1和2准备盆栽基质并移栽黄瓜幼苗至大花盆,按照实施例1所述方法中的步骤3和4进行试验处理,灌施处理每株500ml,喷施处理每株约20ml,每7天处理一次,每个处理5盆重复;果实成熟后,选取同一成熟度的果实进行品质测定,测定结果如表6所示;

由表6可知,灌施与喷施条件下,产红青霉菌PR1菌丝提取物处理组黄瓜维生素C、游离氨基酸、可溶性糖含量增加,硝酸盐含量降低,说明施用产红青霉菌PR1菌丝提取物能够显著提升黄瓜果实品质。
以上所述实施例仅表达了本申请的具体实施方式,其描述较为具体和详细,但并不能因此而理解为对本申请保护范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本申请技术方案构思的前提下,还可以做出若干变形和改进,这些都属于本申请的保护范围。
Claims (4)
1.产红青霉菌PR1菌丝提取物的应用,其特征在于,用于提高番茄对根结线虫的抗性、提升番茄果实和黄瓜果实的品质,其中,提升番茄和黄瓜果实品质包括提高维生素C、番茄红素、游离氨基酸及可溶性糖的含量,降低硝酸盐含量;且所述产红青霉菌PR1菌丝提取物在提高番茄对根结线虫的抗性及提升番茄果实和黄瓜果实品质时的使用浓度为5ng/mL~100ng/mL。
2.根据权利要求1所述的产红青霉菌PR1菌丝提取物的应用,其特征在于,所述产红青霉菌PR1菌丝提取物的施用方式选自灌施、喷施中的一种。
3.根据权利要求2所述的产红青霉菌PR1菌丝提取物的应用,其特征在于,所述产红青霉菌PR1菌丝提取物灌施时的使用浓度为5ng/mL~20ng/mL。
4.根据权利要求2所述的产红青霉菌PR1菌丝提取物的应用,其特征在于,所述产红青霉菌PR1菌丝提取物喷施时的使用浓度为20ng/mL~100ng/mL。
