Inoculating clinically isolated Pasteurella multocida to cottonInoculating Escherichia coli into Maiconk agar medium, culturing at 37deg.C for 18 hr, selecting typical colony, inoculating into 5mL broth, culturing at 37deg.C for 18 hr, and diluting to 10 with 0.5 McUK standard turbidimetric tube 6 CFU/mL E.coli diluted to 10 5 CFU/mL was ready for use.
Drug sensitivity test: the drug sensitivity test was performed using the oral liquid of the Chinese medicinal composition prepared in example 1 of the present invention as a test group and penicillin, ampicillin, and ciprofloxacin as a control group. Inoculating Pasteurella multocida to sheep fresh blood agar medium, inoculating Escherichia coli to Maiconkai agar medium, punching the culture medium by plate punching method, and adding 1g/mL of the Chinese herbal medicine oral liquid of the invention into the holes respectively, preferably to fill up but not overflow the holes. Paper sheets of three antibiotics of 10 IU/tablet of penicillin, 10 mug/mL of ampicillin and 5 mug/mL of ciprofloxacin are taken as drug sensitivity test control, and are placed in a constant temperature incubator at 37 ℃ for culturing for 24 hours, the diameter of a bacteriostasis circle is measured, the drug sensitivity test of each drug is repeated three times, and the drug sensitivity test results are shown in table 1:
TABLE 1 results of drug sensitivity test of different drugs against two bacteria
Determination criteria: according to the standards of traditional Chinese medicine pharmacology and drug sensitivity test standardization related to 2005 edition of American clinical laboratory standardization institute, namely, the antibacterial diameter is more than or equal to 20mm and is extremely sensitive, and the antibacterial diameter is more than or equal to 15mm and is highly sensitive; 15mm antibacterial diameter is equal to or larger than 10mm, and is middle sensitization, and lower sensitization below 10 mm.
Table 1 shows that the oral liquid of example 1 is extremely sensitive to Pasteurella multocida and Escherichia coli, and has antibacterial diameters of 23mm and 24mm, respectively, and the ciprofloxacin, ampicillin and penicillin are inferior to the invention. The Chinese herbal medicine oral liquid has better inhibition effect on two pathogenic bacteria of the beef cattle transportation stress syndrome than three antibiotics of penicillin, ampicillin and ciprofloxacin, and is the first choice in the practice of preventing and treating the beef cattle transportation stress syndrome.
2. Example 1 test of the therapeutic Effect of oral liquid on beef cattle Transit stress syndrome
The transport stress syndrome naturally occurs in beef cattle, which are respectively provided by farmers in Changli county, 28390 can county and Letin county in Hebei province, and is confirmed to be the transport stress syndrome according to clinical symptoms and laboratory examination results. The Chinese medicinal composition of the invention is used as a test group, ciprofloxacin is used as a control group, the sick cattle are treated, the cure rate is counted, and the treatment results are shown in tables 2 and 3.
Standard of medication: the Chinese herbal medicine oral liquid is mixed into water according to the dosage of 50 ml/head for the sick cattle, and the administration is carried out for 5-7 days for a treatment course, wherein the administration is carried out 1 time a day. Ciprofloxacin administration standard: orally taking 2.5-5 mg/kg body weight.
Cure rate criterion: after the medicine is used, the patient is continuously observed for 3 to 5 days, the patient is judged to be cured by the disappearance of clinical symptoms, the drinking water, the ingestion and the recovery of mental state, and then the prevention and treatment effect is determined by combining the pathological section detection sampling detection result, so that the cure rate is counted.
Table 2: example 1 Effect of oral liquid on treating naturally occurring beef cattle transportation stress syndrome
Table 3: effect of ciprofloxacin control group on treating naturally-occurring beef cattle transportation stress syndrome
The oral liquid of the embodiment 1 has obvious treatment effect on the transport stress syndrome of the beef cattle by treating 157 naturally-occurring transport stress syndromes of the beef cattle, the average cure rate is 99 percent, and the symptoms generally disappear through 6 to 7 days, so that normal feeding is recovered; and the ciprofloxacin control group treats 152 beef cattle transportation stress syndromes with natural morbidity, the average cure rate is 96.71%, and the 7d symptoms disappear, so that normal feeding is recovered. The traditional Chinese medicine composition has better treatment effect on the beef cattle transportation stress syndrome than that of a ciprofloxacin control group.
3. Detection of hematological index before and after administration of drug for treating beef cattle transportation stress syndrome
And selecting 20 beef cattle (10 beef cattle in a traditional Chinese medicine treatment group and a ciprofloxacin group) with clinically confirmed diagnosis of the beef cattle transportation stress syndrome in a certain cattle farm of Changli, 8 beef cattle in a synchronous healthy control group (healthy control group), and 10 beef cattle in a diseased control group (diseased group, which is not treated by medication). The traditional Chinese medicine composition treatment group and ciprofloxacin beef cattle are respectively treated by the oral liquid of the example 1 and ciprofloxacin (oral liquid of 2.5-5 mg/kg body weight) after the patients are diagnosed, the dosage is 50mL each, 1 time a day, and 6d is 1 course of treatment. Example 1 oral liquid and ciprofloxacin-treated beef cattle were collected once for each jugular vein before and after administration of the confirmed diagnosis, and red blood cell count, hemoglobin content, and the like were performed. The healthy beef cattle control group and the diseased beef cattle control group are subjected to synchronous blood sampling, and erythrocyte counting and hemoglobin content measurement are also carried out. The measurement results of the number of erythrocytes, the amount of hemoglobin and the hematocrit in blood are shown in Table 4.
Table 4: measurement of the number of red blood cells, the amount of hemoglobin and the hematocrit in blood
As can be seen from table 2, the number of red blood cells, the content of hemoglobin and the hematocrit of healthy beef cattle are maintained at the normal level basically, and the number of red blood cells, the content of hemoglobin and the hematocrit of injured beef cattle are small, and the number of red blood cells, the content of hemoglobin and the hematocrit of beef cattle in the treatment group of example 1 are maintained at the normal level after the beef cattle are cured by the drug, which indicates that both the oral liquid of example 1 and the ciprofloxacin can increase the number of red blood cells, the content of hemoglobin and the hematocrit of beef cattle, but the effect of the oral liquid of example 1 is better than that of the treatment group of ciprofloxacin. Erythrocytes and hemoglobin play an important role in hematopoiesis and oxygen transport in the body, are important for animal survival, growth and production, and have a partial immune function. After the oral liquid of the embodiment 1 treats the beef cattle transportation stress syndrome, the three indexes are obviously increased to reach normal values, which shows that the Chinese herbal medicine oral liquid of the invention has the function of increasing the number of red blood cells and the content of hemoglobin in blood.
The experimental result shows that the Chinese herbal medicine oral liquid for treating the beef cattle transportation stress syndrome has the effects of clearing heat and detoxicating, resisting bacteria and diminishing inflammation, and improving the immunity and disease resistance of organisms.
