(6) Eye indications: such as herniation, ptosis, conjunctival turbidity, iritis, conjunctivitis.
(7) Cardiovascular indications: the blood vessel expands.
(8) Gastrointestinal indications: fecal character, abdominal morphology, and the like.
(9) Urogenital indications: external genitalia condition.
At the end of the observation period, the surviving animals are sacrificed after weighing and are subjected to anatomical examination, the changes of organs such as heart, liver, kidney, spleen, lung, stomach and intestine are observed, if obvious changes exist, pathological examination is further performed, the weights of the organs are counted, and the organ coefficients are recorded.
3. Experimental results
After the mice are subjected to gastric lavage treatment for 14 days by using yin-deficiency capsules with different dosages, compared with a control group, the appearance of the mice is normal, no death phenomenon occurs, the appearance, the action and the posture of the mice are not obviously abnormal, which indicates that the LD50 of the yin-deficiency capsules is larger than the maximum tolerance dosage of the mice, and no obvious acute toxicity exists.
The maximum dose of the mice was converted to 1.5 g/kg body weight, which is about 87.5 times the normal dose (1.2 g/70 kg) for adults, showing good safety.
Because the test sample has no obvious toxicity, in order to make the experimental result simple and visual, only the normal control group and the yin deficiency capsule high-dose experimental group are analyzed later.
(1) Appearance observation of experimental animal
The animals have no obvious abnormality in diet and activity before and after administration, and the excreta has normal appearance. No animal death was observed throughout the experiment. Through continuous observation, the mice before and after administration have no obvious abnormality in appearance, clear mind, steady action gait, fresh hair color and no salivation and nasal discharge (tables 5 and 6).
(2) Recording of changes in body weight of mice before and after administration
Mice were weighed at different time points and found to have a weight change substantially consistent with the trend of the control group after receiving the intragastric administration treatment, and the weight of the yin-deficiency capsule-administered mice was significantly higher than that of the control group on days 11 and 14 of administration (table 7).
(3) Visceral change record of experimental animal after gastric lavage treatment
After the end of the experiment, animals of the administration group and the control group were roughly dissected, and no obvious abnormal changes were observed in organs such as heart, lung, liver, spleen, kidney and stomach of each group of mice (see tables 8 and 9). And weighing partial organs such as liver, spleen, lung, kidney and reproductive organs, calculating related organ coefficients (organ weight/weight), wherein compared with a control group, the yin deficiency capsule administration treatment group (yin deficiency capsule high dose group) has no obvious difference in indexes of main organs such as heart, lung, liver and kidney, and is interesting that the indexes of spleen and thymus of a mice in the yin deficiency capsule group are obviously increased, so that the yin deficiency capsule can promote the growth of immune organs of the mice and improve the immunity of the mice.
(4) Pathological examination of partial organs of mice
And (3) respectively taking partial viscera of each group of mice, performing histopathological examination by tissue section, wherein the viscera of each treatment group of mice are not found to have obvious abnormal lesions, as shown in figure 1, the viscera are uniform in texture, clear in nuclear mass edge, regular in shape, free of tissue necrosis phenomenon, complete in liver and spleen tissue morphology, and free of obvious abnormal in structure.
5. Conclusion(s)
After the mice are infused with different doses of yin-deficiency capsules within one day, continuous observation is carried out for 14 days, no abnormal reaction is seen in the animals during the whole observation period, and no death condition of the animals is seen. The weight measurement results show that the weight of the mice is significantly higher than that of the control group after one week of gastric lavage of the yin deficiency capsule. Through tissue observation and histopathological examination, the yin deficiency capsules are not found to have obvious adverse effects on main organs of mice, but the increase of spleen and thymus quality is obviously promoted.
The results show that the yin deficiency capsule has no obvious acute toxicity to experimental mice, and has obvious growth promoting effect on main immune organs of the mice.
Experimental example 2 therapeutic Effect of yin deficiency Capsule on mice with kidney yin deficiency

the K and alpha values of the drug group and the control group are counted, and t value test is carried out. The test data were analyzed using SPSS software and the results are shown in table 10.

Healthy Kunming mice were 30, 5-6 weeks old, and after one week of adaptive rearing, the mice were randomly divided into 3 groups of 10. The specific grouping is as follows:

Kidney yin deficiency group: hydrocortisone 50 mg/(kg body weight. Day) was injected intraperitoneally for 7 consecutive days;
treatment group with kidney yin deficiency: the dosage of the oral capsule solution for curing the deficiency of stomach-yin is 10 mL/kg body weight, and the oral capsule solution is continuously 7 days.
During the experiment, the mice drink water freely, the appearance form and the activity state of the mice are observed every day, relevant records are made, and after the 7-day experiment treatment period is ended, the blood of the mice is collected for relevant biochemical and biological index detection.
Experimental results:
(1) Morphological appearance observation
Normal mice have normal appearance and behavior, are bright in hair color, active in action, normal in diet and wet in feces in granular form. Mice with deficiency of kidney-yin have fluffy hair, inactive behaviors, dysphoria, irritability, biting, poor mental state, sparse stool and frequent extrados. After the yin-deficiency capsule is used for stomach irrigation, the hair color of the mice becomes bright, the activity of the mice is increased, and the feces are granular.
(2) Biochemical detection
By detecting the levels of FSH, testosterone and estrogen E2 in the serum of each group of mice, the levels of FSH and testosterone in the serum of the mice with kidney-yin deficiency are obviously lower than those of a normal control group (P < 0.05), but the change of the estrogen level is not obvious. After treatment with yin deficiency capsules, testosterone levels were significantly increased, the values were not significantly different from those of the normal group, FSH levels were also significantly increased, but there was still a large difference compared with the normal group; the yin deficiency capsule also reduces the estrogen level in the serum of mice with kidney yin deficiency to a certain extent, but the difference is not obvious, and the specific experimental results are shown in table 11.
Experimental example 4 Effect of yin deficiency Capsule on people with deficiency of Qi and Yin
With reference to the "clinical study guidelines for New Chinese medicine" (trial run), 60 candidates were selected in combination with the dialectical criteria of Chinese medicine, including compliance with the dialectical criteria of Chinese medicine with deficiency of qi and yin, age 40-78 years, informed consent of group personnel, etc. The individuals after the group were subjected to the differentiation of symptoms in the traditional Chinese medicine by a single blind method, and the individuals were classified into 6 grades according to the severity of symptoms according to asymptomatic symptoms (0 score), mild symptoms (1 score), lighter symptoms (2 score), moderate symptoms (3 score), heavier symptoms (4 score) and severe symptoms (5 score) and scored and counted.

As can be seen from the table 12, after the yin-deficiency capsule is taken, the symptoms of people with deficiency of qi and yin are obviously relieved, and the symptoms of palpitation and shortness of breath are mainly relieved, the phenomena of spontaneous perspiration and night sweat and dry mouth are obviously improved, the complexion, the lip color and the tongue characterization are obviously improved, and the pulse condition is stable. The level of each inflammatory factor is obviously reduced.
The results show that the yin deficiency capsules have remarkable improving effect on the symptoms of people with qi and yin deficiency.
