Abstract
本发明公开了一种人参皂苷组合物、协同美白活性及制备美白化妆品的用途。本发明提供的人参皂苷组合物中的人参皂苷Rk3和拟人参皂苷Rh2具有协同抑制酪氨酸酶的活性,因此可以用作美白活性成分用于制备美白面霜、美白面膜、美白乳液、美白爽肤水或美白洗颜泥等具有美白功效的化妆品。本发明人参皂苷组合物的这种协同作用可以降低使用人参皂苷作为美白活性因子的成本。

技术领域
本发明属于化学和化妆品领域,具体涉及一种人参皂苷组合物、协同美白活性及制备美白化妆品的用途。
背景技术
近年来,随着人们生活水平的提高,对生活质量的要求也不断提高,以各种天然植物提取物为美白添加剂的化妆品越来越受到人们的青睐。东方女性历来对肌肤崇尚白皙美,具有美白功效的化妆品成了研究机构和化妆品公司的重要研究领域,使得美白类化妆品发展迅速。
皮肤变黑暗与皮肤细胞中的黑色素含量升高有关。黑色素是由酪氨酸经酪氨酸酶催化形成多巴,再由酪氨酸酶继续催化形成多巴醌,然后重新排列成5,6-醌吲哚,其发生聚合后与黑素体内的结构蛋白结合形成黑素蛋白即黑色素。酪氨酸酶在黑色素合成过程中起着关键作用。抑制酪氨酸酶的活性可以有效阻断黑色素的产生和聚集。
人参皂苷为人参的特征性化学成分,具有改善记忆障碍、延缓衰老、提高免疫功能、抗氧化、改善心血管系统功能、抗应激、壮阳等作用,一直是国内外医药领域的研究热点。人参皂苷既可抑制自由基的产生,也可直接对抗自由基对组织及细胞的损伤作用,或直接清除自由基,还可增强机体本身抗氧化系统的功能,从多个环节阻断自由基的损伤作用。人参通过提高体内及皮肤表皮细胞内超氧化物歧化酶的活性,来加强活性氧化自由基的消除,阻止体内有害物质的产生,改善机体的代谢,减少或消除已经产生并积滞于体内的脂褐质。人参还具有抗紫外线护肤、美白的功能。
由于人参皂苷的原料成本较高,应用于化妆品的成本较高。为了提高人参皂苷的美白效果进而减少添加量降低成本,研究人员进行了很多研究。其中,寻找发现具有协同作用的组合物为这种研究提供了一种路径。
发明内容
本发明的目的在于提供一种人参皂苷组合物、协同美白活性及制备美白化妆品的用途。
本发明目的通过如下方案实现:
一种人参皂苷组合物,该组合物由人参皂苷Rk3和拟人参皂苷Rh2组成。具体实施例的合用计算结果表明,人参皂苷Rk3和拟人参皂苷Rh2单体均具有较为优异的抑制酪氨酸酶的活性,且二者在抑制酪氨酸酶活性方面存在明显的协同作用,并且可以发现协同强度随着作用剂量的提高而增强。
上述人参皂苷组合物作为酪氨酸酶抑制剂用于制备化妆品的用途;其中,所述化妆品为美白化妆品。
优选地,所述美白化妆品为美白面霜、美白面膜、美白乳液、美白爽肤水或美白洗颜泥。
有益效果:
本发明提供的人参皂苷组合物中的人参皂苷Rk3和拟人参皂苷Rh2具有协同抑制酪氨酸酶的活性,因此可以用作美白活性成分用于制备美白面霜、美白面膜、美白乳液、美白爽肤水或美白洗颜泥等具有美白功效的化妆品。本发明人参皂苷组合物的这种协同作用可以降低使用人参皂苷作为美白活性因子的成本。
具体实施方式
实施例1:
参考云南省药物研究所吴德松老师公开的方法测定人参皂苷单体及其组合对酪氨酸酶的抑制活性(查雨锋,等:白梅花提取物抗氧化及美白功效评价;日用化学工业第52卷第2期)。参考中国中医科学院中药研究所李建荣老师公开的方法计算人参皂苷组合是否存在协同作用(王晶晶,等:筋骨草和茯苓配伍合用抑制乳腺癌细胞侵袭转移作用及初步机制研究;中国药理学通报第33卷第4期)。
具体方法如下:
称取人参皂苷适量,用PBS(含1%DMSO,下同)配制成不同浓度的人参皂苷溶液(浓度见表1,人参皂苷Rk3、拟人参皂苷Rh2均购自源叶,货号为B21067-20mg、B20903-5mg,均为分析标准品,HPLC≥98%),另以曲酸作为阳性对照(曲酸购自源叶,纯度≥98%),在96孔板中加入100μL样品溶液或曲酸溶液。每孔加入50μL酪氨酸酶溶液(酪氨酸酶购自源叶,货号S10171-25KU,酪氨酸酶溶液中的酶活为5U/mL)室温避光反应10min,加入50μLL-酪氨酸溶液(品牌为Perfemiker,纯度≥98%,溶液中L-酪氨酸浓度为1.0mg/mL)。空白对照用PBS代替样品,样品对照用PBS代替酪氨酸酶,其余操作均相同。充分混匀反应液,37℃避光反应30min。于492nm波长处测定吸光度OD值,按照公式计算酪氨酸酶抑制率:酪氨酸酶抑制率(%)=[1-(样品OD值-样品对照OD值)/空白对照组OD值]×100%。数据采用SPSS 19.0统计软件分析,以均值±标准差表示。

按文献方法用金氏Q值法计算合用效应,公式为:Q=EA+B/(EA+EB-EA×EB);
其中:EA+B代表两药合用的实际效应值,EA代表A药单用时的效应,EB代表B药单用时的效应。以Q<0.85为拮抗,0.85≤Q≤1.15为相加,Q>1.15为协同。
合用计算结果见表2。

合用计算结果表明,人参皂苷Rk3和拟人参皂苷Rh2单体均具有较为优异的抑制酪氨酸酶的活性,且二者在抑制酪氨酸酶活性方面存在明显的协同作用,并且可以发现协同强度随着作用剂量的提高而增强。

一种美白面霜,在常规面霜基材基础上添加人参皂苷Rk3和拟人参皂苷Rh2制成。

一种美白面膜,在常规面膜基材基础上添加人参皂苷Rk3和拟人参皂苷Rh2制成。

一种美白乳液,在常规乳液基材基础上添加人参皂苷Rk3和拟人参皂苷Rh2制成。

一种美白爽肤水,在常规爽肤水基材基础上添加人参皂苷Rk3和拟人参皂苷Rh2制成。
实施例6:
一种美白洗颜泥,在常规洗颜泥基材基础上添加人参皂苷Rk3和拟人参皂苷Rh2制成。

1.一种人参皂苷组合物,其特征在于:由人参皂苷Rk3和拟人参皂苷Rh2组成。
2.权利要求1所述的人参皂苷组合物作为酪氨酸酶抑制剂用于制备化妆品的用途;其中,所述化妆品为美白化妆品。
3.根据权利要求2所述的用途,其特征在于:所述美白化妆品为美白面霜、美白面膜、美白乳液、美白爽肤水或美白洗颜泥。
