背景技术

对于合成肉,严格的环境控制和组织监测可以从一开始就防止肉类培养物感染,并且在运输给消费者之前可以检测到任何潜在的感染。除了预防和避免疾病,以及不使用抗生素或任何其他化学物质外,合成肉还可以利用许多生物技术进步,包括增加营养强化、单独定制的细胞和分子成分,以及最佳营养状况,所有这些都使它比传统肉类健康得多。
有报道称,功能性重组肌肉肌球蛋白的过表达具有挑战性,因为其体积大,且肌球蛋白折叠所需的细胞机制复杂。迄今为止,功能性肌球蛋白表达仅在小鼠C2C12细胞培养成肌细胞和果蝇(Drosophila melanogaste)中有报道,但从未在低等真核生物或微生物宿主中有报道。肌球蛋白由两条大的重链和两对轻链组装而成,它们排列成一个不对称的分子,两个疏水的球状头部连接在一个长的螺旋杆状亲水尾巴上。肌球蛋白重链已被充分研究,由三个结构域组成:球状头部结构域(重肌球蛋白亚片段-1)和结合肌动蛋白并依靠ATP酶发力的轻链结合颈部结构域(重肌球蛋白亚片段-2),以及通过卷曲螺旋基序对纤维进行组装至关重要的尾部结构域(轻酶解肌球蛋白)。虽然头部区域对肌肉收缩至关重要,但众所周知,它很难过表达。酶解肌球蛋白(LMM)显示了一种由四个七肽组成的28个残基重复序列的模式,这些七肽被称为skip残基的四个间隔很宽的额外氨基酸打断,这些氨基酸为杆状结构域提供了灵活性。本发明通过表达肌球蛋白的卷曲螺旋结构域来绕过具有挑战性的肌球蛋白表达问题。

其中一种产品是富含肌肉蛋白的酵母抽提物。酵母发酵过程将停止,并进行自发细胞裂解。富含内源性和肌肉蛋白及肽的可溶物将从细胞壁碎片中分离并干燥以供进一步加工。酵母细胞壁的分离不仅能提高蛋白质含量,还能提高产品的消化率。
细胞内或细胞外产生肌肉蛋白的另一个主要宿主家族包括丝状真菌和蘑菇。这些丝状真菌包括可食物种,如Fusarium venenatum、米曲霉(Aspergillus oryzae)、紫红曲霉(Monascus purpureus)、米根霉(Rhizopus oryzae)、间型脉孢菌(Neurosporaintermedia)、木霉属(Trichoderma sp)。这些蘑菇包括小型褐菇(Cremini mushroom)、大型顶蘑菇(Portobello mushroom)和纽扣蘑菇(Button mushroom,Agaricus bisporus)、羊肚菌(Morchella esculenta)、香菇(Lentinula edodes)、平菇(Pleurotus ostreatus)、金针菇(Flammulina velutipes),和牛肝菌(Boletus edulis)。这些重组蘑菇(孢子)将接种在真菌琼脂平板上,如马铃薯葡萄糖琼脂(PDA)或麦芽提取物琼脂(MEA)以形成菌丝体。这些菌丝体将在谷物和其他富含纤维素物质的培养基上进行固态发酵或深层发酵。菌丝体的生物量将被收获,制成肉制品。通过这种方式,肌球蛋白和肌动蛋白的肉味将与蘑菇的肉质相结合,以模拟肉类的感官效果。
为了改善肉类替代品的质构和味道,可通过化学或生化过程将表达的肌肉蛋白进一步交联成纤维,包括转谷氨酰胺酶和赖氨酸单加氧酶的共表达,以及使用上述酶进行体外交联。这些过程包括以下额外的方法及其组合:
(1)pH调节和预热处理,展开三级结构并影响蛋白质结构域的电荷,以产生排斥力,使凝胶网络保持膨胀状态。
(2)高压过程(HPP),广泛用于通过变性、聚集或凝胶化来改变蛋白质结构。HPP条件,如压力强度、压力梯度、持续时间、温度等,将针对动物蛋白和菌丝蛋白之间的相互作用进行优化。
(3)通过蛋白质氧化增加二硫键水平,其中蛋白质可以在羟基自由基生成系统中孵育。
(4)精氨酸和赖氨酸含氮侧链上的甲基化,或羟脯氨酸残基修饰的非肌肉蛋白上的羟基化。羰基含量和表面疏水性将发生显著变化,这对凝胶和乳化性能至关重要。
(5)干或湿挤压技术,其是商业上可获得的,并广泛用于豆类(pulse)蛋白。它可以将非凝胶蛋白转化为肉质产品。
(6)亲水胶体,如海藻酸钠(可与钙形成凝胶)、魔芋葡甘聚糖(可与碱形成凝胶)和黄原胶/天然淀粉(可提高乳液稳定性和结合强度)。
这些含有菌丝体或蘑菇的肌肉蛋白随后将与胶原蛋白、水和脂肪混合,和/或与植物蛋白质混合,形成各种肉制品,如热狗、肉干、肉糕、块(nugget)、香肠、培根、意大利腊肠等。
或者,含有酵母或酵母抽提物的肌肉蛋白可以被配制成宠物食品。本发明提供了一种模拟用于宠物食品应用的预备的Fybraworks蛋白质成分(FPI)的组成、功能性和营养价值的新方法。本发明方法涉及用鸡肌原纤维蛋白和/或水解豌豆蛋白模拟蛋白质质构,所述水解豌豆蛋白用L-蛋氨酸和L-组氨酸强化,氨基酸组成与肌肉蛋白相似。Fybraworks配料的蛋白质含量可高达重量基准的50%,可配制成多种不同类别的营养良好的宠物食品,包括处于不同生长阶段的宠物、狗和猫的主食或零食(见表2)。
Fybraworks的蛋白质成分可以由全肌肉蛋白或部分蛋白质和肽组成,它们与全蛋白质具有相同的营养价值,但具有更好的消化率,可以满足低过敏级/兽医处方食品的要求。
Fybraworks的蛋白质成分在整个宠物食品基质中都有独特的风味。提高宠物食品适口性的传统做法是添加动物消化物或适口剂,方法是在干宠物食品表面涂覆涂层,或混入湿宠物食品的汁液部分。这些宠物食品的适口性分布通常不均匀。Fybraworks的蛋白质成分的适口性因肽、氨基酸和糖之间的美拉德反应产生的令人愉悦的肉味以及酵母的鲜味而得到加强。样品的感官测试(图2)证实了一种独特且同质的肉味,这是本地美味宠物食品所没有的。

实施例
以下示例仅是说明本发明的一些方面的一种方式,不应将其解释为限制本发明的范围。
实施例1肌原纤维蛋白提取
从新鲜鸡胸肉中制备肌肉肌原纤维(洗涤肌原纤维)蛋白。在用Stephan混合器切碎之前去除可见的结缔组织和脂肪。将切碎的肉与三份水混合,搅拌,并通过直径为2.5mm的螺旋式进料(screw-fed)过滤器(Bibun Machine,广岛,日本)。然后每次用有机玻璃布将多余的水从肌原纤维提取物中分离出来。除过滤步骤外,所有步骤都再重复两次,但最后一次水洗含有0.5%(w/v)NaCl,以促进脱水。加工结束时,这种富含肌原纤维的洗涤部分的蛋白质含量达到20%w/w,用于所有后续产品开发(PD)。
实施例2由肌肉蛋白和蘑菇粉制成的“鸡”块中肌肉蛋白与蘑菇的比例(样品制备和烹饪程序保持不变)的评估
“鸡”块由肌原纤维蛋白、蘑菇粉、调味料、椰子油、水、磷酸钠制成。分别在3:1(对照)、1.5:1(Tl)、1:5(T2)的条件下测试了不同比例的肌原纤维蛋白(干基)/蘑菇粉。先将磷酸盐与水混合2分钟,然后加入盐、调味料,再混合2分钟,在碗中腌制腌料。将所得糊状物在冰箱中冷却至34°F,并转移至成型设备,以形成一份单一尺寸的块。最后,块要经过预涂粉、打浆和烘烤的步骤。它们将在365°F下预炸30秒,然后在烤箱中完全烤熟,直到产品中心温度达到165°F。在冷藏箱中过夜后进行产品评估。
确定了肌肉蛋白与蘑菇粉的最佳配比(1.5:1)。如图1所示,对照组和T1组均显示出无面包屑部分的弹性纹理。横截面显示出均匀且有弹性的凝胶特征(见图2)。当比例低于1.5:1时,质构变得不那么耐嚼/有弹性,更脆。此外,在T2中观察到大量烹饪损失,如图3所示,其中左侧代表T2(1:5=肌肉蛋白:蘑菇粉),右侧代表T1(1.5:1=肌肉蛋白:蘑菇粉)。显然,烹调后,油溢出到T2样品的表面,并且在每个非面包肉部分的底部发现了更多过度烹调的碎屑。此外,T2样品出现焦糊,因为配方中的蘑菇粉比T1和对照中的蘑菇粉多。
这组实验的结果表明,肌肉蛋白/蘑菇粉的比例对成品的质构很重要,这将指导我们的产品,并为发酵蛋白质含量设定蛋白质含量目标。
实施例3蘑菇粒径和甲基纤维素水平对烹调产品影响的评估(配方、样品制备和烹调程序保持不变)
这个“鸡块”样品的制作程序与前一节所述相同。本实施例中的变量不同:1)在配方中加入甲基纤维素,以进一步改善低蛋白质含量产品的质构(肌肉蛋白/干蘑菇的比例增加至1:9w/w)。在加入其他成分之前,先将其与水高速混合。2)评估了不同粒度的干蘑菇对产品质构的影响。
优选更大粒径(约0.5-1cm)的干蘑菇,因为它产生了更好的质构(见图4),具有强烈的咬合力和弹性,而较小的粒径(约0.1-0.5cm)产生了脆性凝胶,尽管外观均匀。如图5所示,两种尺寸的蘑菇粉在烹饪时都表现出良好的成型性。
这些发现将有助于改善肉类替代产品的质构和外观,以满足客户的不同需求。
实施例4肌肉蛋白对产品质构和功能影响的评估。
“鸡”块样品的制作程序与前一节中所述的相同。通过包括无肌肉蛋白对照品(用等量的豌豆蛋白代替)评估肌肉蛋白对产品性能的影响。试验配方模拟了一些现成产品,除了干蘑菇试验中使用了大颗粒(见上一节)作为TVP(即基于大豆的)替代品。主要目的是确定肌原纤维蛋白是否会影响产品的外观、质构和面包粘性。
综上所述,煎炸后含有9%肌肉蛋白的产品显示出比对照样品更高的成型性、更强的咬合力和更致密的结构,尽管两个样品在非面包部分显示出相似的凝胶结构,如图6所示。还观察到,该产品的面包粘性略好于对照品。
这些结果表明,与不含肌肉蛋白的对照组相比,含有肌肉肌原纤维蛋白的对照组具有更好的性能。
最后但并非最不重要的一点是,研究发现,由于蘑菇和肌原纤维蛋白之间的协同风味增强了鲜味,至少1/3的盐可以从原配方中去除。这证明了在食品中结合蘑菇和肌原纤维蛋白时,潜在的钠减少作用的额外益处。
实施例5由肌肉蛋白和平菇粉制成的碎肉干
碎肉干(groundjerkey)由肌原纤维蛋白、蘑菇粉、红烧调味料、腌料(cure)、磷酸钠、椰子油、焦糖和水制成。分别以1.5:1、2:1、2.5:1、3:1、3.5:1的比例测试了不同比例的干基肌原纤维蛋白与蘑菇粉。首先,将肌原纤维蛋白与腌料和磷酸钠在食品处理器中以高剪切速度混合1分钟。然后将水和油添加到混合器中,再混合1分钟。接下来,将调味料和蘑菇粉加入混合物中,并在高剪切下混合30秒。最后的面糊在冷却机中固化一整夜后,通过干枪挤压成条状。最后,在165°F的商用脱水机中充分煮熟6小时。产品从脱水机中取出后,将进行真空包装以进行感官评估。
研究发现,肌肉蛋白/蘑菇粉的比例至少为2:1(干基为w/w)对于实现良好的成型性(即无破损)至关重要(见图7)。研究发现,至少2.5:1的比例对于获得良好的肉质干品的嫩度和咀嚼性至关重要(见图7)。如图9所示,在脱水过程中,产品还表现出诸如凝胶和收缩特性的真正的肉特性,这归因于蛋白质变性和聚集。
这些发现将有助于改善肉类替代产品的质构和外观,以满足客户的不同需求。
实施例6将肌肉蛋白配制成宠物食品
将提取的肌原纤维蛋白或用L-蛋氨酸和L-组氨酸强化的水解豌豆蛋白与面包酵母混合,以选定的干重基准比模拟酵母发酵材料。蛋白质物质和酵母的优选干重基准比从5:95到6:94、7:93、8:92、9:91、10:90、11:89、12:88、13:87、14:86、15:85到16:84、17:83、18:82、19:81或20:80之中选择。
本发明提供了使用FPI生产各种宠物食品的新应用方法。FPI首先被彻底水合,以促进离子相互作用、氢键相互作用、范德华相互作用和疏水相互作用。然后加入植物油和甘油,以及调味品和色素成分。然后将混合物充分混合,形成浆。慢慢加入碳水化合物成分,形成均匀的混合物。将材料制成理想形状,在68℃至95℃的温度范围内加热,持续通风1至9小时。宠物食品经过冷却和包装,以尽量减少水分和香气的迁移(见图8)。
优选的植物油选自但不限于菜籽油、葵花籽油、花生油、大豆油、菜籽油、橄榄油、棉籽油、椰子油、玉米油、棕榈油、红花油、芝麻油、杏仁油、腰果油、榛子油、核桃油。
优选的调味品和色素成分选自但不限于盐、各种烟和天然香料、糖蜜、葡萄糖、蔗糖、焦糖色素、磷酸钠、三磷酸钠、海藻酸钠、硫酸钙、磷酸、迷迭香提取物、混合生育酚。
优选的碳水化合物成分选自但不限于大米淀粉、米粉、土豆淀粉、土豆粉、木薯淀粉、木薯粉、豌豆淀粉、豌豆粉、玉米淀粉、玉米粉、燕麦粉、亚麻籽粉、小麦淀粉、小麦粉。
优选的混合过程形式选自泵送、混合、预处理和均质。
加热过程的优选形式是烘焙、脱水、挤压和干馏。
预计用FPI生产宠物食品的方法可以通过连续、分批或两种处理方式的组合。
实施例7肌肉肌球蛋白卷曲螺旋结构域在大肠杆菌中的表达

肌肉肌动蛋白卷曲螺旋结构域的表达量达到35mg/L,溶解度小。肌动蛋白缺乏溶解性可能是由于自发聚合,除了43kDa单体,86kDa二聚体和129kDa三聚体也可以从蛋白质印迹中看到(见图10)。
实施例9肌肉肌球蛋白卷曲螺旋结构域在毕赤酵母中的表达
在AOX启动子的控制下将与α因子信号肽和6×His标签(SEQ ID NO:15)融合的肌球蛋白卷曲螺旋结构域序列克隆到整合载体pPICZalphaA,转化到毕赤酵母X-33表达菌株中,在BMGY/BMMY上生长,并通过在28℃以200rpm添加甲醇96h来诱导表达。表达水平较低,但可在细胞颗粒中检测到(见图11)。
实施例10肌肉肌动蛋白卷曲螺旋结构域在毕赤酵母中的表达
在AOX启动子的控制下将与α因子信号肽和6×His标签(SEQ ID NO:14)融合的肌动蛋白编码序列,克隆到整合载体pPICZalphaA,转化到毕赤酵母X-33表达菌株中,在BMGY/BMMY上生长,并通过在28℃和200rpm下添加甲醇诱导表达96h。克隆#6的表达水平最高(见图12),并且易于自发聚合,这与它在大肠杆菌中的表达类似。

Claims (19)

1.一种通过在可食用生物宿主中重组表达肌肉蛋白来生产合成肉的新方法包括以下步骤:
a)将重组源材料接种到生长培养基中;
b)在酵母或细菌或其他可食用生物宿主存在下,在基础生长培养基上在发酵过程中培养上述重组源材料,以形成结构肌肉蛋白;
c)繁殖酵母菌和细菌,使其大量生长;
d)使用生化和化学方法将上述结构肌肉蛋白交联成用于生产蛋白质的纤维,用于生产蛋白质纤维;
e)将上述交联纤维束缚在一起,以提供强度和硬度;
f)将上述获得的蛋白质纤维与胶原蛋白、水和脂肪混合,以生产所需的合成肉;
其中,所得动物肉的特征在于由胶原蛋白、水、肌肉蛋白、结缔组织脂肪和血球蛋白组成,并且除了用其他必需营养素强化之外,还能进一步与基于植物的蛋白质混合。
2.根据权利要求1所述的新方法,其中,可食用生物宿主选自丝状真菌,所述丝状真菌包括可食物种,诸如Fusarium venenatum、米曲霉、紫红曲霉、米根霉、间型脉孢菌、木霉属;这些蘑菇包括小型褐菇、大型顶蘑菇和纽扣蘑菇、羊肚菌、香菇、平菇、金针菇,以及牛肝菌。
3.根据权利要求1所述的新方法,其中细胞包含编码含有多肽的肌肉蛋白的核酸,所述多肽选自肌动蛋白、肌球蛋白、截短肌动蛋白、截短肌球蛋白及其组合。
4.根据权利要求1所述的新方法,其中细胞包含编码截短肌肉肌球蛋白的核酸,所述肌肉肌球蛋白是卷曲螺旋。
5.根据权利要求1所述的新方法,其中细胞包含启动子序列,所述启动子序列是组成性启动子。
6.根据权利要求1所述的新方法,其中细胞包含所述启动子序列,所述启动子序列为组织特异性启动子。
7.根据权利要求1所述的新方法,其中细胞包含所述启动子序列,所述启动子序列为可诱导启动子。
8.根据权利要求1所述的新方法,其中细胞进一步包含转录终止区。
9.根据权利要求1所述的新方法,其中细胞包含信号肽,所述信号肽为分泌信号肽。
10.根据权利要求1所述的新方法,其中,所述酵母选自酿酒酵母、包括毕赤酵母、假丝酵母属、Komagataella属、乳克鲁维酵母、解脂耶氏酵母、东方伊萨酵母属在内的毕赤酵母属或其混合物。
11.根据权利要求1所述的新方法,其中,所述细菌选自链球菌、乳酸杆菌、芽孢杆菌、大肠杆菌、沙门氏菌、奈瑟氏菌、醋酸杆菌、节杆菌、曲霉、双歧杆菌、棒状杆菌、荧光假单胞菌或其混合物。
12.根据权利要求1所述的新方法,其中,所述结构肌肉蛋白来自动物种的肌球蛋白II和肌肉肌动蛋白,所述动物物种为诸如鸭子(鸭属)、兔子(真兔属)、鹅(雁属)、野牛和水牛(北美野牛、水牛属)、绵羊(绵羊属)、山羊(山羊属)、马(马属)、鱼类(太平洋鲑属、罗非鱼属、鳕属、红点鲑属,狼鲈属,银鲈属、鲶属、鰤属,鲈属、扇贝(扇贝属)、虾(滨对虾属,对虾属、毛虾属、鹰爪虾属、明对虾属、北极甜虾),螃蟹(滨蟹属、蓝蟹、首长黄道蟹、北太平洋雪蟹)、牡蛎和贻贝(翼形亚纲)、鸡(原鸡)、猪(野猪)和牛(普通牛)。
13.根据权利要求1所述的新方法,其中,产生的纤维可来自一组“长链”蛋白质,诸如在头发、肌肉或鞭毛中发现的蛋白质。
14.根据权利要求1所述的新方法,其中,基于植物的蛋白质的混合物可选自鹰嘴豆、花生、杏仁、扁豆等。
15.根据权利要求1所述的新方法,其中,所得动物肉可添加选自ω-3脂肪酸的营养素,以增加其营养价值。
16.根据权利要求1所述的新方法,其中,所得动物肉可用于培根、火腿、热狗、意大利熏火腿、香肠、宠物食品、动物饲料等;此外,基于植物的蛋白质可以是从植物获得的食物蛋白源。
17.根据权利要求1所述的新方法,其中,所得肉类在外观、味道、气味、质构或其他因素上可能与传统生产的肉类相似。
18.根据权利要求1所述的新方法,其中,与传统肉类相比,所述动物肉具有潜在的健康、环境、文化和经济考虑。
19.根据权利要求1所述的新方法,其中,改进的方法有助于显著减少屠宰动物。
