技术领域
本申请涉及消毒剂技术领域,更具体地说,它涉及一种低灰染邻苯二甲醛消毒剂及其制备方法与应用。
背景技术
邻苯二甲醛作为一种新型高效消毒剂,较传统戊二醛消毒剂具有气味低、毒性低、使用周期长、高效广谱、使用前无需活化、作用浓度低、刺激性弱、低腐蚀等优点,在消毒领域尤其是医疗器械(内窥镜,手术器械,牙科器械等等)消毒方面应用广泛。
相关技术中,使用邻苯二甲醛进行内窥镜等医疗器械消毒时,在水槽、操作台面、操作人员的皮肤和工作服等部位常常会出现深浅不一的淡灰色斑点,这是邻苯二甲醛特有的“灰染”现象。由于邻苯二甲醛使蛋白质快速变性,与蛋白质接触后显示出独特的着色提示,这种现象提示了内窥镜清洗不彻底,同时清洗过程中会使自动内窥镜清洗机着色。因此需要用水多次冲洗经邻苯二甲醛消毒的器械,才能减少消毒剂残留,以达到不危害患者或工作人员安全的水平,防止造成皮肤黏膜变色等。
此外,现有的邻苯二甲醛消毒液对光和空气敏感,易氧化,有效期短;其对芽孢的杀灭作用不理想,尤其是杀灭枯草杆菌黑色变种芽孢的作用微弱。基于上述陈述,本申请提供了一种低灰染邻苯二甲醛消毒剂及其制备方法与应用。
发明内容
为了解决相关技术中邻苯二甲醛消毒剂稳定性差,易氧化,杀菌作用不理想,“灰染”现象严重等问题,本申请提供了一种低灰染邻苯二甲醛消毒剂及其制备方法与应用。
第一方面,本申请提供了一种低灰染邻苯二甲醛消毒剂,采用如下的技术方案:一种低灰染邻苯二甲醛消毒剂,包括以下重量百分比的原料:邻苯二甲醛0.35-1%、增效剂3-8%、乙醇10-22%、复合双链季铵盐0.1-1%、渗透剂0.5-5%、消泡剂0.5-1%、缓冲剂1-7%、余量为纯化水;
所述增效剂为质量比3-11:1的阿魏酸和质量分数3%硼酸溶液的复配物。
优选的,所述低灰染邻苯二甲醛消毒剂,包括以下重量百分比的原料:邻苯二甲醛0.5-0.8%、增效剂4-7%、乙醇12-20%、复合双链季铵盐0.3-0.7%、渗透剂1-4%、消泡剂0.6-0.8%、缓冲剂2-6%、余量为纯化水。
优选的,所述低灰染邻苯二甲醛消毒剂,包括以下重量百分比的原料:邻苯二甲醛0.6%、增效剂5.5%、乙醇16%、复合双链季铵盐0.5%、渗透剂2.5%、消泡剂0.7%、缓冲剂4%、余量为纯化水。
通过采用上述技术方案,复合双链季铵盐的添加,能够增强邻苯二甲醛的耐酸碱、耐光热稳定性,同时累积邻苯二甲醛的杀菌效力;增效剂能够提高邻苯二甲醛的抗氧化性能、抗菌性,防止“灰染”现象的发生;渗透剂能改变脂质与蛋白结构,对细胞壁/膜的结构进行破坏,增加通透性,进而增强杀菌效果;本申请配方科学,通过控制原料配比,有效保证了各组分之间协同作用良好,所得邻苯二甲醛消毒剂,抗氧化性强,化学稳定性好,连续使用14天邻苯二甲醛的含量仍在0.35%以上,能够在快速彻底杀灭芽孢的同时,对医疗器械和物品不产生“灰染”或腐蚀。
采用阿魏酸和硼酸溶液复配作为增效剂,阿魏酸具有优异的抗氧化性、抗菌、抗病毒能力;同时具有防凝、保护皮肤的功效;硼酸溶液具有抗菌、抗炎、消毒、收敛的作用,两者复配能够在增强邻苯二甲醛的杀菌性和稳定性的同时,有效解决了邻苯二甲醛的“灰染”问题,降低其对医疗器械或皮肤黏膜的损伤。
优选的,所述渗透剂为1-3:5-9:1的乙二胺四乙酸、脂肪醇聚氧乙烯醚和甘草酸铵的复配剂。

将总质量1/2的纯化水加入到配料罐中,升温至48-55℃,加入邻苯二甲醛和增效剂,以100-300r/min的转速搅拌混合均匀得混合液;

优选的,所述医疗器械消毒前,先利用检测试纸检测邻苯二甲醛消毒剂的浓度,检测合格,即可应用于后续消毒工序。
优选的,所述检测试纸由滤纸浸泡吸收检测液后烘干而得,所述检测液由亚硫酸钠、氯化铵、琥珀酸、酚酞指示剂和去离子水混合复配而得。
优选的,所述亚硫酸钠、氯化铵、琥珀酸、酚酞指示剂、去离子水的质量比为16:7:3:1:50。
通过采用上述技术方案,本申请的检测试纸对浓度合格的邻苯二甲醛消毒液(浓度0.3%及以上)显色,对浓度不合格的邻苯二甲醛(浓度0.3%以下)不显色,较相关技术中的氨基酸显色法,具有无需通过比色卡比较,易于判断的特点。本申请检测试纸显色速度快,可随用随检,具有使用方便,保障性高的特点。

一种低灰染邻苯二甲醛消毒剂的制备方法,具体包括以下制备步骤:
称取邻苯二甲醛、增效剂、乙醇、复合双链季铵盐、渗透剂、消泡剂、缓冲剂和纯化水备用;
将总质量1/2的纯化水加入到配料罐中,升温至48℃,加入邻苯二甲醛和增效剂,以100r/min的转速搅拌混合均匀得混合液;
将剩余纯化水、乙醇和复合双链季铵盐预混合后加入到混合液中,保持转速不变,加入消泡剂、渗透剂和缓冲剂,搅拌至充分混匀,静置1h后过滤、分装,得到低灰染邻苯二甲醛消毒剂。
实施例8
一种低灰染邻苯二甲醛消毒剂,包括以下原料:邻苯二甲醛1kg、增效剂8kg、乙醇22kg、复合双链季铵盐1kg、渗透剂5kg、消泡剂1kg、缓冲剂7kg、纯化水55kg;
其中,增效剂为质量比11:1的阿魏酸和质量分数3%硼酸溶液的复配物;
渗透剂为3:5:1的乙二胺四乙酸、脂肪醇聚氧乙烯醚和甘草酸铵的复配剂;
消泡剂为BYK028;
缓冲剂为质量比7:1的磷酸三钾和碳酸钠的复配物。
一种低灰染邻苯二甲醛消毒剂的制备方法,具体包括以下制备步骤:
称取邻苯二甲醛、增效剂、乙醇、复合双链季铵盐、渗透剂、消泡剂、缓冲剂和纯化水备用;
将总质量1/2的纯化水加入到配料罐中,升温至55℃,加入邻苯二甲醛和增效剂,以300r/min的转速搅拌混合均匀得混合液;
将剩余纯化水、乙醇和复合双链季铵盐预混合后加入到混合液中,保持转速不变,加入消泡剂、渗透剂和缓冲剂,搅拌至充分混匀,静置6h后过滤、分装,得到低灰染邻苯二甲醛消毒剂。
为了验证本申请实施例1-8中制备的低灰染邻苯二甲醛消毒剂的综合性能,申请人设置了对比例1-10,具体如下:
对比例1
同实施例1,区别仅在于,增效剂选用质量比4:1的水杨酸和质量分数3%硼酸溶液的复配物。
对比例2
同实施例1,区别仅在于,增效剂选用质量比4:1的柠檬酸和质量分数3%硼酸溶液的复配物。
对比例3
同实施例1,区别仅在于,渗透剂选用乙二胺四乙酸。
对比例4
同实施例1,区别仅在于,渗透剂选用脂肪醇聚氧乙烯醚。
对比例5
同实施例1,区别仅在于,渗透剂选用甘草酸铵。
对比例6
同实施例1,区别仅在于,渗透剂选用1.5:6的乙二胺四乙酸和脂肪醇聚氧乙烯醚复配剂。
对比例7
同实施例1,区别仅在于,渗透剂选用1.5:1的乙二胺四乙酸和甘草酸铵的复配剂。
对比例8
同实施例1,区别仅在于,渗透剂选用6:1的脂肪醇聚氧乙烯醚和甘草酸铵的复配剂。
对比例9
同实施例1,区别仅在于,缓冲剂选用质量比4:1:1的磷酸氢二钾、三聚磷酸钾和碳酸钠的复配物。
对比例10
同实施例1,区别仅在于,邻苯二甲醛选用0.25kg,纯化水选用73.95kg。
取上述实施例1-8和对比例1-9中制得的消毒剂,分别对不锈钢片、碳钢片、铜片、铝片连续浸泡14天,均未见腐蚀现象。
采用悬液定量杀菌试验,在25℃条件下对细菌的杀灭效果见表1。加速稳定性试验结果见表2。

由上述表1显示数据可知:除实施例7外,本申请的实施例1-6、实施例8和对比例9中制备的邻苯二甲醛消毒剂均能在特定时间内达到彻底杀菌的效果,综合杀菌效果远优于对比例1-8和对比例10中制备的邻苯二甲醛消毒剂。

由上述表2显示数据可知:本申请实施例1-8和对比例9中制备的邻苯二甲醛消毒剂产品性质稳定,使用周期和保存周期长,且稳定性远优于对比例1-8和对比例10中制备的制备的邻苯二甲醛消毒剂。
由实施例1和对比例1、2可知:本申请选用阿魏酸和硼酸溶液复配作为增效剂,阿魏酸和硼酸溶液能够协同增效,提高消毒剂的产品稳定性,本申请中阿魏酸的选用较普通有机酸(水杨酸或柠檬酸),具有显著的进步。
由实施例1和对比例3-8可知:本申请实施例1选用乙二胺四乙酸、脂肪醇聚氧乙烯醚和甘草酸铵复配作为渗透剂,较对比例3-5单独选用乙二胺四乙酸、脂肪醇聚氧乙烯醚或甘草酸铵,或对比例6-8选用乙二胺四乙酸、脂肪醇聚氧乙烯醚、甘草酸铵中的任意两者复配,本申请实施例1的产品稳定性明显优于对比例3-8。
由实施例1和对比例9可知:本申请中缓冲剂优选质量比4:1:1的磷酸氢二钾、三聚磷酸钾和碳酸钠的复配物,最终制得的产品稳定性更好。
灰染实验
将15x15cm大小的棉布用稀释牛奶浸泡5min,简单清洗之后,分别浸入实施例1-8和对比例1-9中制备的邻苯二甲醛消毒剂中来回摆动涮洗,肉眼观察判定其灰染现象,发现本申请实施例1-8和对比例9制备的邻苯二甲醛消毒剂不易产生灰染现象;而对比例1-8制备的邻苯二甲醛消毒剂均出现不同程度的灰染现象,其中对比例1-5灰染现象较为严重。
由上述表1、表2和灰染实验结果可知:本申请中制备的邻苯二甲醛消毒剂产品的稳定性好、杀菌性强,且不发生“灰染”现象,其综合性能的产生是配方中各组分共同作用,协同增效产生的结果。本申请通过控制各组分的科学配比,以邻苯二甲醛为主要杀菌成分,复合双链季铵盐的添加,能够增强邻苯二甲醛的耐酸碱、耐光热稳定性,同时累积邻苯二甲醛的杀菌效力;增效剂能够提高邻苯二甲醛的抗氧化性能、抗菌性,防止“灰染”现象的发生;渗透剂能改变脂质与蛋白结构,对细胞壁或细胞膜的结构进行破坏,增加通透性,进而增强杀菌效果;所得的邻苯二甲醛消毒剂性能稳定,能够发挥高效的消毒性能,本申请制备方法简单,制备条件温和,所得的消毒剂产品均一稳定,使用范围广,使用周期长。
实验例1
将亚硫酸钠、氯化铵、琥珀酸、酚酞指示剂、去离子水按质量比16:7:3:1:50进行搅拌溶解,混合均匀制得检测液;
将滤纸充分浸泡在检测液中,使纸张充分吸收饱和后取出,轻轻刮去多余液体,置于50℃烘干后裁切即得检测试纸。
用检测试纸分别检测本申请实施例1-6和对比例10中制备的邻苯二甲醛消毒剂,观察显色情况及显色时间,得出结果见表3。

由上述表3显示数据可知:本申请的检测试纸对浓度0.3%及以上的邻苯二甲醛消毒剂显色,对浓度浓度0.3%以下的邻苯二甲醛消毒剂不显色;且显色速度快,浓度越大显色时间越短。

Claims (10)
1.一种低灰染邻苯二甲醛消毒剂,其特征在于,包括以下重量百分比的原料:邻苯二甲醛0.35-1%、增效剂3-8%、乙醇10-22%、复合双链季铵盐0.1-1%、渗透剂0.5-5%、消泡剂0.5-1%、缓冲剂1-7%、余量为纯化水;
所述增效剂为质量比3-11:1的阿魏酸和质量分数3%硼酸溶液的复配物。
2.根据权利要求1所述的低灰染邻苯二甲醛消毒剂,其特征在于,包括以下重量百分比的原料:邻苯二甲醛0.5-0.8%、增效剂4-7%、乙醇12-20%、复合双链季铵盐0.3-0.7%、渗透剂1-4%、消泡剂0.6-0.8%、缓冲剂2-6%、余量为纯化水。
3.根据权利要求1所述的低灰染邻苯二甲醛消毒剂,其特征在于,包括以下重量百分比的原料:邻苯二甲醛0.6%、增效剂5.5%、乙醇16%、复合双链季铵盐0.5%、渗透剂2.5%、消泡剂0.7%、缓冲剂4%、余量为纯化水。
4.根据权利要求1所述的低灰染邻苯二甲醛消毒剂,其特征在于,所述渗透剂为1-3:5-9:1的乙二胺四乙酸、脂肪醇聚氧乙烯醚和甘草酸铵的复配剂。
5.根据权利要求1所述的低灰染邻苯二甲醛消毒剂,其特征在于,所述消泡剂为BYK024、BYK011、BYK025、BYK028中的至少一种。
6.根据权利要求1所述的低灰染邻苯二甲醛消毒剂,其特征在于,所述缓冲剂为质量比4-7:1的磷酸盐和碳酸盐的复配物。
7.根据权利要求6所述的低灰染邻苯二甲醛消毒剂,其特征在于,所述磷酸盐为磷酸三钾、磷酸三钠、磷酸氢二钾、磷酸氢二钠、三聚磷酸钾、三聚磷酸钠中的至少一种;所述碳酸盐为碳酸钠、碳酸钾、碳酸氢钠、碳酸氢钾中的至少一种。
8.一种权利要求1-7任一项所述的低灰染邻苯二甲醛消毒剂的制备方法,其特征在于,包括以下制备步骤:
按重量百分比计,称取邻苯二甲醛、增效剂、乙醇、复合双链季铵盐、渗透剂、消泡剂、缓冲剂和纯化水,混合各组分,至完全溶解,静置后得到低灰染邻苯二甲醛消毒剂。
9.一种权利要求1-7任一项所述的低灰染邻苯二甲醛消毒剂的应用,其特征在于,将权利要求1-7任一项所述的低灰染邻苯二甲醛消毒剂应用于医疗器械消毒。
10.根据权利要求9所述的低灰染邻苯二甲醛消毒剂的应用,其特征在于,所述医疗器械消毒前,先利用检测试纸检测邻苯二甲醛消毒剂的浓度,检测合格,即可应用于后续消毒工序;
所述检测试纸由滤纸浸泡吸收检测液后烘干而得,所述检测液由亚硫酸钠、氯化铵、琥珀酸、酚酞指示剂和去离子水混合复配而得。
