Detailed Description

The preparation method of the mullein extract comprises the following steps: extracting herba Verbasci Thapsi with 50% methanol at room temperature for 1h, at a material-to-liquid ratio of 1: 10, heating the extracting solution in water bath at 60 ℃ until the extracting solution is concentrated to 1/5 of the original volume, and obtaining the mullein extract for later use.
Example 1 preparation of the composition
(1) Adding glycerol and bisabolol in the formula amount into water, and performing ultrasonic treatment until the glycerol and the bisabolol are completely dissolved to obtain a mixture 1; the ultrasonic temperature is 55 ℃, and the ultrasonic power is 100W;
(2) cooling the mixture 1 obtained in the step (1) to 35-40 ℃, adding the centella asiatica extract, the verbena extract and the tea extract according to the formula dosage, stirring for 40min, and homogenizing for 1h to obtain the composition with the whitening effect.

Result detection
1. Safety test
The human skin closed patch test was performed according to the human skin patch test specified in the technical standards for cosmetic safety (2015 edition). 35 volunteers were selected, and no suspicious reaction or positive reaction was found in all of the products A to N of the formula of the present invention, which proved that the product of the present invention is safe and has no adverse reaction.
2. And (3) stability detection:
the detection method comprises the following steps: cold and hot test in turn, seal the storage 24h at 45 ℃, 25 ℃, 15 ℃ with the gained product in proper order, circulate 5 times after, place 12h at 25 ℃, thin scribble the product on the black observation board with the corresponding product of the new preparation of the same method, compare the outward appearance, the classification is as follows:

The results in Table 3 show that the formulas A to N have good stability, and have no problems of precipitation, discoloration, delamination and the like after cold and hot alternation experiments.
3. Test of whitening Effect
3.1 detection method: tyrosinase inhibition assay
The enzymatic reaction converts L-dopa to a red dopaquinone, which has a maximum absorption at 475 nm. When tyrosinase and whitening active ingredients exist simultaneously, the catalytic activity of the tyrosinase can be inhibited to a certain degree, so that the generation of dopaquinone is reduced. Therefore, the absorbance at 475nm is measured by a spectrophotometer, the concentration of the generated dopaquinone can be reflected, and the inhibition degree of the composition on the tyrosinase activity can be further judged.
Group I: 0.5mL of L-dopa solution (1.0mg/mL) +3.0mL of phosphate buffer (pH 6-7) +0.2mL of tyrosinase (250U/mL) was placed in a 32 ℃ water bath.
Group II: 0.2mL tyrosinase (250U/mL) +3.5mL phosphate buffer (pH 6-7).
Group III: 0.5mL of L-dopa solution +3.0mL of the formulation composition solution (one set for each formulation separately) +0.2mL of tyrosinase (250U/mL) was placed in a 32 ℃ water bath.
Group IV: 3.0mL of the formulation solution +0.5mL of phosphate buffer (pH 6-7) +0.2mL of tyrosinase (250U/mL).

The results in table 4 show that the formula G-I, L-N has a higher inhibitory effect on tyrosinase activity, and compared with the case where only the tea extract group B and only the mullein extract J are added, the case where both the tea extract and the mullein extract are added can significantly improve the inhibitory effect on tyrosinase. Meanwhile, on the premise of adding 5% of tea extract, the addition amount of the mullein extract is stably improved from 0.3%, and has a synergistic improvement effect when the addition amount is 1%, and the addition of the 5% of tea extract and the 1% of mullein extract is most suitable for batch production and application by comprehensively considering the raw material cost and the product effect.
3.2 detection method: melanin inhibition assay
140 volunteers aged 25-35 and women with obvious blotches on their faces were selected and randomly divided into 14 groups of 10 each using the products prepared according to formulas A-N. The product is applied on face uniformly, and is flapped to be absorbed completely, and is used once every day, 1.5mL each time, and continuously for three weeks without other skin care products. The skin melanin value was measured with a skin melanin measuring instrument (Courage Khazaka) before and after one week of use (average value was taken after selecting three parts of eye tail, cheek and forehead). The test of the same subject is carried out by the same instrument and the same tester. Before each measurement, the probe is cleaned, and meanwhile, the tested person washes the face with clear water, keeps relaxing, sits still for 10min, and receives the test.

The melanin inhibition was calculated for each individual group and averaged, and the results are given in the following table:

2. The composition with whitening efficacy according to claim 1, characterized by comprising the following components in percentage by weight: 10-25% of glycerin, 5-15% of tea extract, 0.3-2% of mullein extract, 2-10% of centella extract, 0.5-2% of bisabolol and water.
3. The composition with whitening efficacy according to claim 1 or 2, wherein the weight amount of the tea extract is more than five times of that of the mullein extract.
4. The composition with whitening effect according to claim 3, characterized by comprising the following components in percentage by weight: 20% of glycerin, 5% of tea extract, 1% of mullein extract, 4% of centella asiatica extract, 1% of bisabolol and water.

The composition with whitening efficacy according to claim 1, wherein the preparation method of the tea extract comprises: extracting folium Camelliae sinensis with ethanol for 1-3 times, centrifuging, collecting supernatant, and removing solvent to obtain folium Camelliae sinensis extract; the preparation method of the mullein extract comprises the following steps: extracting herba Verbasci Thapsi with methanol, and heating and concentrating the extractive solution to 1/5 to obtain herba Verbasci Thapsi extract.
6. The method for preparing the composition having whitening efficacy according to any one of claims 1 to 5, comprising the steps of:
(1) adding glycerol and bisabolol in the formula amount into water, and performing ultrasonic treatment until the glycerol and the bisabolol are completely dissolved to obtain a mixture 1;
