Description
Technical Field
The invention belongs to the technical field of aromatic south medicine essential oil, and particularly relates to aromatic south medicine compound essential oil and a preparation method thereof.
Background
Aromatic Chinese herbs refer to Chinese herbs that can emit strong aromatic odor from medicinal parts or generate aromatic odor after being burned and heated, and can be used for treating and preventing diseases, and are important parts of Chinese medicine clinical practice. The aromatic Chinese herbs have strong aromatic odor because they are rich in essential oils, which are also called volatile oils, and are a general term for a kind of volatile oily liquid that can be distilled with water vapor and is immiscible with water.
The essential oil of Chinese herbs usually has unique effects, such as relieving exterior syndrome, eliminating dampness, promoting qi circulation, inducing resuscitation, etc., and can act on local or whole body to prevent and treat diseases. The essential oil has mild action and small side effect, and has the functions of preventing infectious diseases, inhibiting bacteria, resisting inflammation, preventing corrosion, preventing and treating depression, resisting oxidation, promoting cell metabolism, cell regeneration and the like. The Chinese medicinal essential oil combines the pharmacological action of Chinese medicines with aromatherapy of Chinese medicine, and has wide application in medical treatment, health promotion, food, articles for daily use, etc. However, most of single essential oil has weak pharmacological action and single action, and may be more pungent and uncomfortable, and the single essential oil often cannot achieve the ideal effect. The compatibility of various essential oils can enhance the drug effect, play a synergistic role, cover and reduce the bad smell, and even generate the pharmacological action and specific smell which are not available in single essential oil, thereby expanding the application range of the essential oil.
The aromatic Chinese herbs in south medicine are called aromatic south herbs. Aromatic south herbs are a great treasury. In the south of Lingnan of China, in tropical and subtropical regions, the medicine is hot and rainy all the year round, the geothermal and hydrothermal resources are rich, and the produced medicine is hot and aromatic. Aromatic south herbs are numerous in quantity and excellent in quality and are mostly genuine medicinal materials. In the first 8 Lingnan Chinese medicinal materials protected in Lingnan Chinese medicinal material protection regulation of Guangdong province, 6 medicinal materials are aromatic south Chinese medicinal materials, which are exocarpium Citri Grandis, pericarpium Citri Reticulatae Chachiensis, fructus Amomi, herba Agastaches, lignum Aquilariae Resinatum, and fructus Citri Sarcodactylis. In addition, famous local south herbs such as cinnamon, sandalwood, dalbergia wood, galangal, star anise, fructus alpiniae oxyphyllae, rhizoma kaempferiae, round cardamom, katsumadai seed, tsaoko cardamon, turmeric, sappan wood and the like are aromatic south herbs. The aromatic south Chinese herbs have the advantages of numerous aromatic south herbs, definite drug effect, wide application and higher economic value.

The aromatic south medicine compound essential oil of examples 1 to 3 was applied to an ICR mouse induced by pentobarbital sodium, the sleep latency and the sleep duration of the mouse were recorded, and the sleep-aiding effect of the aromatic south medicine compound essential oil of examples 1 to 3 was examined by observing whether the aromatic south medicine compound essential oil of examples 1 to 3 can prolong the sleep time of the pentobarbital sodium.
The 4-week-old male ICR mice were selected and randomly divided into 9 groups, each of which was 13, specifically, a blank control group, a one-day group of aromatic south pharmaceutical compound essential oil (further subdivided into three groups, i.e., the one-day group of aromatic south pharmaceutical compound essential oil of example 1, the one-day group of aromatic south pharmaceutical compound essential oil of example 2, and the one-day group of aromatic south pharmaceutical compound essential oil of example 3), a three-day group of aromatic south pharmaceutical compound essential oil (further subdivided into the three-day group of aromatic south pharmaceutical compound essential oil of example 1, the three-day group of aromatic south pharmaceutical compound essential oil of example 2, and the three-day group of aromatic south pharmaceutical compound essential oil of example 3), a one-day group of positive control, and a three-day group of positive control.
Before the experiment, normal saline is injected into the abdominal cavity of mice of an empty control group and aromatic south medicine compound essential oil groups according to the proportion of 10mL/kg respectively. Adding aromatic south medicine compound essential oil with the concentration of 20g/L into an aromatherapy atomizer, placing the ultrasonic atomizer in an animal aromatherapy chamber, opening the ultrasonic atomizer for 5min to enable the aromatic south medicine compound essential oil to fill the whole aromatherapy chamber, placing mice of each group of the aromatic south medicine compound essential oil into the aromatherapy chamber, closing a box cover after the mice completely enter the aromatherapy chamber, enabling the mice to move in the aromatherapy chamber for 60min, and sequentially placing the mice back into a mouse cage after the completion of the inhalation. Wherein, the mice of the three-day group of the aromatic south medicine compound essential oil are continuously treated for 3 days according to the method. Mice in each group of positive control were administered (diazepam) by intraperitoneal injection at a dose of 3mg/kg (the dose was determined to be an effective dose in preliminary experiments to allow 100% of mice to sleep but not to allow too long sleep time) according to the weight of each mouse, and the diazepam was prepared in distilled water as a 0.05% solution. Mice in the positive control three-day group were treated continuously for 3 days as described above.
Pentobarbital sodium is injected into each group of mice after the last treatment in an intraperitoneal injection mode, the administration dose is 45mg/kg (the administration dose is the effective administration dose measured by a preliminary experiment, the mice can fall asleep 100 percent, but the sleeping time is not too long), and the pentobarbital sodium is prepared into a 0.5 percent solution by using normal saline. And (5) observing whether the treatment of each group can prolong the sleep time of the sodium pentobarbital, and recording the sleep latency and the sleep time of the mice of each group. The judgment basis is as follows: the falling asleep state is achieved without occurrence of turning reflection within 1 min; the time period from the intraperitoneal injection of the pentobarbital sodium to the disappearance of the righting reflex is the sleep latency period, and the time from the disappearance of the righting reflex to the restoration of the reflex is the sleep time.
The experimental results of the sleep latency and the sleep time induced by pentobarbital after the ICR mice inhale the compound essential oil are shown in Table 5, and compared with the blank control group mice, the sleep latency of the positive control one-day group mice and the sleep latency of the positive control three-day group mice are obviously different (P is less than 0.05) from the aspect of the sleep latency, so that the sleep latency of the mice can be obviously shortened; the sleep latency of each group of mice in one day group of the compound essential oil of aromatic south medicine has no significant difference (P is more than 0.05) with the sleep latency of mice in a blank control group, but the data still show that the compound essential oil of aromatic south medicine in examples 1-3 can shorten the sleep latency to a certain extent; compared with mice in a blank control group, the sleep latency of the mice in each group of three days of the compound essential oil of the aromatic south medicine is obviously shortened (P is less than 0.05), which shows that the sleep latency of the mice tends to be reduced along with the prolonging of the administration time and the increasing of the administration times of the mice in the embodiments 1 to 3, and meanwhile, the compound essential oil of the aromatic south medicine in the embodiments 1 to 3 has no obvious difference in the shortening of the sleep latency.
