the preparation method of the sandalwood essential oil comprises the following steps: accurately weighing a certain amount of dry sandalwood, pulverizing into coarse powder, adding distilled water 10 times the weight of the dry sandalwood, soaking at room temperature for 72h, then adding the medicinal powder and the soaking solution into a distillation kettle for normal pressure steam distillation, wherein the distillation extraction time is 4h, receiving essential oil by an essential oil extractor, obtaining crude sandalwood extract after distillation, condensation and oil-water separation, adding anhydrous sodium sulfate with the crude extract oil quality of 10% for dehydration refining, filtering to obtain sandalwood essential oil, sealing and placing in a shade place, and aging for 30 days;
the preparation method of the aromatic south medicinal essential oil comprises the following steps: adding oleum Santali albi into oleum Simmondsia chinensis, stirring in the same direction for 5min, mixing, diluting, mixing, filtering with 5 μm pore size filter, packaging, and sealing to obtain aromatic south medicinal essential oil.
Effect examples
1. Sensory evaluation
50 volunteers 18 to 60 years old are randomly selected as test groups, and sensory evaluation (including appearance, smell and the like) is carried out on the aromatic south drug compound essential oils prepared in examples 1 to 13 and comparative examples 1 to 5. Appearance indexes: the essential oil liquid appearance is presented: clear, pale yellow transparent liquid, no turbidity and no delamination. Odor index: the smell of the essential oil is pleasant, loved, happy and comfortable. The testing method comprises the following steps: the aromatic south medicine compound essential oil is smeared on the palm center to smell fragrance, each person scores the smell according to the happy mood and happy degree, the highest score is 10, the lowest score is 0, and finally the average score is taken.
The test results are shown in table 1, all the prepared aromatic south drug compound essential oils are clear, pale yellow, light brown or nearly colorless liquid, are not turbid and layered, but have certain difference in smell, the aromatic south drug compound essential oils prepared in examples 1-5 are round and pleasant in smell, have costustoot and fresh fruit smell, have slightly medicinal smell, and the aromatic south drug compound essential oils prepared in comparative examples 1-5 have defects of different degrees in smell, such as pungency, nose flushing, fishy smell, and insufficient round smell and the like. The pleasant degree of the smell indicates the acceptance of the smell of different combined essential oils by the crowd, and the score of the pleasant degree of the smell can be seen, the scores of examples 1-5 are highest, when fresh patchouli, the patchouli medicinal material and the sandalwood medicinal material are not adopted in the preparation process of the patchouli essential oil and the patchouli essential oil, the sandalwood medicinal material is not soaked for more than 72 hours in the preparation process of the patchouli essential oil and the sandalwood essential oil, and the score of the pleasant degree of the smell of the prepared aromatic south medicinal compound essential oil is lower when the aging time of the essential oil is not more than 30 days, so that the flavor acceptance of each example is higher than that of the comparative example, and the compatibility relation between the smell characteristics and various essential oils and the preparation process are obvious.

2. Determination of bacteriostasis
The antibacterial effect of the aromatic south medicine compound essential oil of the examples 1-3 is measured by using a filter paper sheet method, and experimental results show that the aromatic south medicine compound essential oil of the examples 1-3 has better inhibition effect on escherichia coli, staphylococcus aureus, staphylococcus albus and candida albicans, and the antibacterial circle sizes of all the aromatic south medicine compound essential oils on all bacteria are close without obvious difference, wherein the diameter of the antibacterial circle on candida albicans is larger than 20mm, and the inhibition effect is most obvious and is strong; the diameter of the inhibition zone for the escherichia coli, staphylococcus aureus and staphylococcus albus is 10-20 mm, and the bacteria are moderately inhibited.

3. Thermal and ultraviolet stability detection
In order to examine the stability of the aromatic south drug compound essential oil of examples 1 to 3, experiments were performed on the thermal stability and ultraviolet stability of the aromatic south drug compound essential oil.
Thermal stability experiments: the aromatic south drug compound essential oil of examples 1 to 3 was placed under different temperatures (20 ℃, 30 ℃, 40 ℃, 50 and 60 ℃) for 25 minutes, the properties and the smell thereof were observed, and the antibacterial effect of the aromatic south drug compound essential oil on escherichia coli, staphylococcus aureus, staphylococcus albus and candida albicans at different temperatures was measured by using a filter paper sheet method.
Ultraviolet stability experiment: the aromatic south drug compound essential oil of examples 1 to 3 was irradiated with ultraviolet rays for different times (5 min, 10min, 15min, 20min and 25 min), the properties and the smell thereof were observed, and the antibacterial effect of the aromatic south drug compound essential oil after the ultraviolet irradiation treatment for different times on escherichia coli, staphylococcus aureus, staphylococcus albus and candida albicans was measured by a filter paper sheet method.
By observing the properties and the smell of the aromatic south drug compound essential oil of examples 1-3 after being treated at different temperatures, no obvious difference between the properties and the smell of the aromatic south drug compound essential oil and the properties and the smell of the aromatic south drug compound essential oil before being treated is found. The antibacterial effect of each aromatic south drug compound essential oil is further detected, and the result shows that the aromatic south drug compound essential oil treated at different temperatures has no obvious difference from the aromatic south drug compound essential oil before being treated (table 3), and the temperature has no obvious influence on the antibacterial effect of the aromatic south drug compound essential oil in examples 1-3, namely, the antibacterial components of the aromatic south drug compound essential oil are not obviously influenced along with the temperature rise, and the aromatic south drug compound essential oil in examples 1-3 has better heat stability.
By observing the properties and the smell of the aromatic south drug compound essential oil in examples 1-3 after being subjected to ultraviolet irradiation for different times, no obvious difference between the properties and the smell of the aromatic south drug compound essential oil and the properties and the smell of the aromatic south drug compound essential oil before being treated is found. The antibacterial effect of each aromatic south drug compound essential oil is further detected, and the result shows that the aromatic south drug compound essential oil subjected to ultraviolet irradiation treatment for different time has no obvious difference from the aromatic south drug compound essential oil before the treatment (table 4), and the result shows that ultraviolet light has no obvious influence on the antibacterial effect of the aromatic south drug compound essential oil in examples 1-3, and the aromatic south drug compound essential oil in examples 1-3 has good ultraviolet stability.

4. Sleep-aiding effect
The aromatic south drug compound essential oil of examples 1 to 3 was applied to the pentobarbital sodium-induced ICR mice, the sleep latency and sleep duration of the mice were recorded, and the sleeping-aiding effect of the aromatic south drug compound essential oil of examples 1 to 3 was examined by observing whether the aromatic south drug compound essential oil of examples 1 to 3 can prolong the sleeping time of pentobarbital sodium.
Male ICR mice of 4 weeks of age are selected and randomly divided into 9 groups, 13 groups are specifically a blank control group, an aromatic south drug compound essential oil one day group (further divided into three groups, namely an aromatic south drug compound essential oil one day group of example 1, an aromatic south drug compound essential oil one day group of example 2, an aromatic south drug compound essential oil one day group of example 3), an aromatic south drug compound essential oil three day group (further divided into an aromatic south drug compound essential oil three day group of example 1, an aromatic south drug compound essential oil three day group of example 2, an aromatic south drug compound essential oil three day group of example 3), a positive control one day group and a positive control three day group.
Before the experiment, the physiological saline is respectively injected into the abdominal cavity of the mice in the blank control group and the aromatic south drug compound essential oil groups according to the ratio of 10 mL/kg. Adding 20g/L aromatic south drug compound essential oil into an aromatherapy atomizer, placing the ultrasonic atomizer into an animal aromatherapy chamber, opening the ultrasonic atomizer for 5min to fill the whole aromatherapy chamber with the aromatic south drug compound essential oil, placing the mice of each group of aromatic south drug compound essential oil into the aromatherapy chamber, closing a box cover after the mice completely enter the aromatherapy chamber, allowing the mice to move in the aromatherapy chamber for 60min, and sequentially placing the mice into a mouse cage after the inhalation is completed. Wherein, for each group of mice of the aromatic south drug compound essential oil three-day group, the treatment is carried out for 3 days continuously according to the method. The mice of each positive control group were given 3mg/kg (this dose was an effective dose measured by pre-experiments to enable 100% of the mice to fall asleep but not to sleep for too long) by intraperitoneal injection, and diazepam was formulated as a 0.05% solution with distilled water. Wherein the positive control three-day group of mice was treated as described above for 3 consecutive days.
The mice of the last treated group were injected with pentobarbital sodium by intraperitoneal injection at a dose of 45mg/kg (this dose was an effective dose measured by a preliminary experiment, which could make the mice fall asleep 100% but not make the sleep time too long), and the pentobarbital sodium was formulated into a 0.5% solution with physiological saline. The treatment of each group was observed for prolonged sleep time of sodium pentobarbital and the sleep latency and sleep time of each group of mice were recorded. Judging the basis: the phenomenon that the front turning reflection is in a sleep state does not occur within 1 min; the period from the time when pentobarbital sodium is injected into the abdominal cavity to the time when the eversion and the reflection disappear is a sleep latency period, and the time from the time when the eversion and the reflection disappear to the time when the reflection is restored is a sleep time.
The experimental results of the pentobarbital-induced sleep latency and the sleep time after the ICR mice inhale the compound essential oil are shown in table 5, and compared with the mice in the blank control group, the sleep latency of the mice in the positive control group has significant differences (P < 0.05) from the sleep latency of the mice in the positive control group for one day and the sleep latency of the mice in the positive control group for three days, so that the sleep latency of the mice can be significantly shortened; the sleep latency of each group of mice in one day group of aromatic south drug compound essential oil and the sleep latency of mice in the blank group of mice are not significantly different (P is more than 0.05), but the data still show that the aromatic south drug compound essential oil of the examples 1-3 can shorten the sleep latency to a certain extent; compared with mice in a blank control group, the sleep latency of the mice is obviously shortened (P < 0.05) in all the mice in the three-day group of aromatic south drug compound essential oil, which shows that the aromatic south drug compound essential oil of examples 1-3 has a trend of reducing the sleep latency of the mice along with the extension of the administration time and the increase of the administration times, and meanwhile, the aromatic south drug compound essential oil of examples 1-3 has no obvious difference in shortening the sleep latency.
From the aspect of sleep duration, compared with mice in a blank control group, the sleep duration of mice in a positive control group with one day and mice in a positive control group with three days is obviously prolonged (P < 0.05); the sleep duration of mice in one day group of aromatic south drug compound essential oil is obviously increased (P < 0.05), and the sleep duration of mice in three days group of aromatic south drug compound essential oil is obviously longer than the sleep duration of mice in one day group of aromatic south drug compound essential oil, which indicates that the aromatic south drug compound essential oil of examples 1-3 can obviously prolong the sleep duration of mice, and has the same change trend as the mice in diazepam group, and meanwhile, the aromatic south drug compound essential oil of examples 1-3 has no obvious difference in prolonging the sleep duration of mice.

Claims (6)

1. The compound essential oil is characterized by comprising the following components in parts by volume: 2-6 parts of butyl essential oil, 10-18 parts of cinnamon essential oil, 15-25 parts of wrinkled giant hyssop essential oil, 25-35 parts of sandalwood essential oil, 10-20 parts of fingered citron essential oil and 300-500 parts of base oil; the raw material of the wrinkled giant hyssop essential oil is fresh wrinkled giant hyssop; the fingered citron essential oil is prepared from fresh fingered citron; the preparation method of the sandalwood essential oil comprises the following steps: soaking and extracting sandalwood serving as a raw material and water serving as a solvent to obtain sandalwood essential oil; the soaking time is not less than 72 hours;
the compound essential oil is prepared by the following preparation method: mixing oleum Caryophylli, oleum Cinnamomi, herba Agastaches essential oil, lignum Santali albi essential oil and fructus Citri Sarcodactylis essential oil, aging, and mixing with base oil to obtain compound essential oil; the aging time is not less than 30 days.
2. The compound essential oil according to claim 1, wherein the preparation method of the patchouli essential oil is characterized in that fresh patchouli is used as a raw material, water is used as a solvent, and the patchouli essential oil is obtained by extraction.
3. The compound essential oil of claim 2, wherein the preparation method of the fingered citron essential oil comprises the following steps: fresh fingered citron is taken as a raw material, water is taken as a solvent, and fingered citron essential oil is obtained by extraction.
4. The compound essential oil according to any one of claims 1 to 3, wherein the base oil is at least one of jojoba oil, evening primrose seed oil, sweet almond oil, apricot kernel oil, avocado oil, wheat germ oil, macadamia nut oil and grape seed oil.
