The method comprises the following steps: weighing the medicinal tea prepared in the embodiment 2, soaking and extracting in water bath, and freeze-drying; selecting transgenic zebra fish with 2dpf immune cell fluorescence labeling, establishing an immune hypofunction model by using vinorelbine, performing anesthesia photographing after medicinal tea treatment, and counting the fluorescence intensity of tail immune cells; selecting 5dpf wild AB line zebra fish, placing the zebra fish into an anoxic box, performing hypoxia treatment to cause a fatigue model, and detecting the total swimming distance and the average swimming speed after medicinal tea treatment.
Experimental results show that 50-200 mu g/mL of the polygonatum-maca medicinal tea extract has an immunoregulation effect on the zebra fish, and 25-200 mu g/mL of the polygonatum-maca medicinal tea extract has an anti-fatigue effect on the zebra fish.
Materials and instruments
1) Material
The zebra fish is provided for a zebra fish drug screening platform of biological research institute of academy of sciences in Shandong province; samples were provided by Shanxi Shadong pharmaceutical Co.
2) Instrument for measuring the position of a moving object
Model AXIO zoom.v16 fluorescence microscope (zeiss, germany), FSX100 fluorescence microscope (olympus, japan), zebra fish breeding system (eisheng technologies ltd, beijing).
(II) method
1) Extraction of herb tea
Weighing medicinal tea, adding 5 times of purified water, placing in a 95 deg.C water bath, extracting for 1h, filtering or centrifuging, collecting supernatant, concentrating under reduced pressure with a rotary evaporator, concentrating to a certain volume, and transferring to a freeze-drying machine for freeze-drying.
2) Zebrafish embryo harvest
And (4) separately feeding male and female zebra fishes, alternately lighting for 14 h/dark for 10h, and regularly feeding artificial granular bait and newly hatched artemia nauplius. And (3) putting healthy and mature zebra fish into a mating jar according to the female parent and the male parent by the ratio of 1:1 during egg collection, and obtaining fertilized eggs the next 9-10 days. Sterilizing and washing fertilized egg, transferring into water (containing 5.0mM NaCl, 0.1 mM NaCl) for culturing zebra fish embryo7mM KCl,0.4mM CaCl 2 ,0.16mM MgSO 4 ) Culturing at 28 deg.C under controlled light.
3) Evaluation of modulating immune Activity
Transgenic zebrafish (Tg (lyz: DSRED2)) fluorescently labeled with 2dpf immune cells were randomly transferred to 24-well plates, and 12 pieces of each well were divided into a blank control group, a model group and a herb tea treatment group. E3 culture water is given to the blank control group, 150 mu g/mL vinorelbine solution is respectively given to the other groups (model group and herb tea treatment group) to establish a zebra fish immune hypofunction model, and 100, 200 and 400 mu g/mL herb tea solution is respectively given to the herb tea treatment group. After the culture box is cultured for 24 hours, anesthesia and photographing are carried out, and the fluorescence intensity of tail immune cells is counted.
4) Evaluation of anti-fatigue Activity
5dpf of healthy AB-line zebra fish is randomly transferred to a 24-well plate, 16 zebra fish in each well are divided into a blank control group, a model group and a herb tea treatment group. The blank control group and the model group are given with normal oxygen culture water, and the herb tea treatment group is given with 100, 200, 400 and 800 mug/mL herb tea extract solution respectively. And placing the model group and the medicinal tea treatment group in an anaerobic condition to cause a fatigue model, culturing for 6 hours in an incubator, transferring each group of zebra fishes to a 48-hole plate, placing the zebra fishes in an ethology detection box, standing for 10 minutes, collecting 10-minute ethology data, and counting the total moving distance and the average moving speed.
(III) results of the experiment

As shown in fig. 1, the number of immune cells in zebra fish in the vinorelbine treatment group (model group) is significantly reduced compared with that in the blank control group, and the number of immune cells in zebra fish in the levamisole hydrochloride (positive drug group) and medicinal tea extract treatment group is significantly increased compared with that in the model group, which indicates that the polygonatum sibiricum and maca tea extract has immunoregulation activity on zebra fish at 50-200 μ g/mL.
2) Evaluation of anti-fatigue Activity

As shown in fig. 2, the total moving distance and the average moving speed of zebra fish in the hypoxia treatment group (model group) are both significantly reduced compared with those in the blank control group, the total moving distance and the average moving speed of rhodiola rosea in the treatment group (positive medicine group) are both significantly increased compared with those in the model group, and the total moving distance and the average moving speed of the herb tea extract treatment group are both significantly increased compared with those in the model group, which indicates that the polygonatum-maca herb tea extract of 25-200 mug/mL has an anti-fatigue effect on zebra fish.
(IV) conclusion
The experiment shows that 50-200 mu g/mL of the polygonatum sibiricum and maca tea extract has immunoregulation activity on a zebra fish immune hypofunction model caused by vinorelbine, and 25-200 mu g/mL of the polygonatum sibiricum and maca medicinal tea extract has an anti-fatigue effect on a zebra fish fatigue model caused by hypoxia.
