Abstract
本发明属于化妆品技术领域,具体涉及一种银杏外种皮抗衰祛痘膏及其制备方法。针对现有采用银杏制备的祛痘产品效果有待进一步提高的问题,本发明提供了一种银杏外种皮抗衰祛痘膏,组成包括:按重量份数计,银杏外种皮10‑30份,丁香花20‑40份,菖蒲根20‑40份,辅料90‑150份。本发明采用银杏外种皮、丁香花和菖蒲根为主要功能原料,通过微生物发酵获得发酵液,银杏、丁香花和菖蒲根经过发酵后得到富含异鼠李素的发酵液,能够显著抑制痤疮丙酸杆菌的生长繁殖,达到预防和治疗痤疮的效果。本发明为预防痤疮的药妆提供了一种新的选择,具有重要的意义。

技术领域
本发明属于化妆品技术领域,具体涉及一种银杏外种皮抗衰祛痘膏及其制备方法。
背景技术
“痘痘”是痤疮的俗称,又叫“青春痘”、“暗疮”或“粉刺”,是由于毛囊及皮脂腺阻塞、发炎所引发的一种皮肤病。痤疮主要好发于青少年,主要因为体阳热偏盛,血热外涌,气血忧郁,临床表现以好发于面部的粉刺、丘疹、脓疱、结节等多形性皮损为特点。痤疮主要是由于痤疮丙酸杆菌所引起的,这种细菌易传染易繁殖,因而常导致青春痘反反复复,不能彻底治愈,长期下去会导致肌肤变得敏感,使患者皮肤发红发痒,康复之后还会出现疤痕,容易给患者造成一定的精神压力。
目前,常见的祛痘方法主要有:激光治疗法、激素治疗法和药妆治疗法等。激光治疗青春痘方法是通过特定波长的高强度染料激光照射,能快速破坏丙酸青春痘杆菌,有效抑制细菌繁殖,消除其对组织的刺激与破坏作用,同时使扩张的毛细血管迅速萎缩,充血与红肿的炎症快速消退,阻断了青春痘对组织的进一步破坏作用,同时促使青春痘创面及早康复,避免日后出现不可逆转的凹坑及青春痘疤痕。该方法起效迅速,能使炎症性的丘疹、结节迅速消退。同时,能使毛囊口的白头、黑头易清洁排除。然而,激光治疗并不适用于所有患者,对一些皮肤较敏感的患者不适用,成本高,治疗后容易反复。
激素治疗法主要采用一些激素类药物比如乙烯雌酚、抗菌素如庆大霉素、大环内酯类抗生素、四环素等,这类治疗方法有一定的疗效,但副作用也比较大,治愈率低,愈后易复发,而且大多为化学制剂和激素类药物,使用后容易产生副作用。
药妆治疗法主要采用一些天然野生植物祛痘,祛痘温和不刺激的,尤其适用于敏感肌肤祛痘,且长期使用也不会造成皮肤负担,祛痘效果显著。目前,已有采用银杏叶制备祛痘产品的相关报道,主要是将银杏提取后制备成面霜、洗面奶等产品,祛痘效果还有待进一步增强。
丁香,为木犀科丁香属落叶灌木或小乔木,丁香花花序硕大、开花繁茂,花色淡雅、芳香,习性强健,栽培简易。目前,尚未见有将丁香用作祛痘产品的报道。
菖蒲,是一种多年生草本,根状茎粗壮,根茎可制香味料。菖蒲根具有镇静作用,现代研究发现菖蒲根提取的挥发油可以减弱麻黄碱导致的小鼠的中枢兴奋作用,所以有一定的镇静作用。菖蒲内服可以促进消化液的分泌,有松弛肠管平滑肌痉挛的作用,因此有一定治疗慢性胃炎疼痛的效果。但未见有将菖蒲根提取物用来制备祛痘的护肤品的报道。
综上可见,现有采用银杏制备的祛痘产品效果不好,需要进一步提升祛痘效果,亟待开发一种祛痘效果好、安全性高的祛痘膏。
发明内容
本发明要解决的技术问题为:现有采用银杏制备的祛痘产品效果有待进一步提高的问题。
本发明解决上述技术问题的技术方案为:提供了一种银杏外种皮抗衰祛痘膏。具体的,本发明所述的银杏外种皮抗衰祛痘膏,组成包括:按重量份数计,银杏外种皮10-30份,丁香花20-40份,菖蒲根20-40份,辅料90-150份。
优选的,上述银杏外种皮抗衰祛痘膏,组成包括:按重量份数计,银杏外种皮20份,丁香花30份,菖蒲根30份,辅料120份。
其中,上述银杏外种皮抗衰祛痘膏,组成还包括:3-5重量份的微生物,所述微生物为米曲霉或植物乳杆菌中的至少一种。
进一步的,上述银杏外种皮抗衰祛痘膏中,所述微生物为米曲霉与植物乳杆菌按活菌数比例1-5:3-8混合而成,微生物的浓度为106-108cfu/mL。
其中,上述银杏外种皮抗衰祛痘膏中,所述的辅料为:甘油、丙二醇、透明质酸钠、黄原胶、羧甲基纤维素、甘草酸二钾、己二醇、乳木果油和氨甲基丙醇。
进一步的,所述的辅料中,甘油为5-10份,丙二醇10-15份、透明质酸钠10-20份、黄原胶15-20份、羧甲基纤维素20-25份、甘草酸二钾5-15份、己二醇10-15份、乳木果油5-15份、氨甲基丙醇10-15份。
本发明还提供了一种上述银杏外种皮抗衰祛痘膏的制备方法,包括以下步骤:
a、将丁香花和菖蒲根粉碎后,加入超声萃取釜中,加入50-75%的乙醇进行萃取,萃取30-45min后过滤,弃去滤渣,得到提取物;
b、将银杏外种皮与步骤a所得提取物混合后,加入米曲霉和/或植物乳杆菌,进行酶转化反应,制得发酵液,过滤,得滤液;
c、取黄原胶和透明质酸钠在甘油中预先分散,加入甘草酸二钾、羧甲基纤维素、步骤b所得滤液和去离子水,搅拌溶解至分散均匀透明;
d、将甘草酸二钾和氨甲基丙醇在丙二醇和己二醇中溶解,加入到步骤c所得溶液中,加入乳木果油,混匀,得到银杏外种皮抗衰祛痘膏。
其中,上述银杏外种皮抗衰祛痘膏的制备方法中,步骤a所述粉碎后的粒径为200目以下。

本发明的有益效果为:
本发明提供了一种银杏外种皮抗衰祛痘膏,采用银杏外种皮、丁香花和菖蒲根为主要功能原料,通过微生物发酵获得发酵液,银杏、丁香花和菖蒲根经过发酵后得到富含异鼠李素的发酵液,能够显著抑制痤疮丙酸杆菌的生长繁殖,达到预防和治疗痤疮的效果;同时,发酵液还具有很强的清除氧自由基的能力,对预防衰老也有一定的积极效果。本发明将发酵液制备成祛痘膏,使用方便,安全性高,具有很好的祛痘抗衰老效果,适宜推广使用。
具体实施方式
本发明中采用了银杏外种皮,其含有丰富的银杏酸、黄酮、萜内脂和多糖,具有很好的祛痘效果,通过添加微生物后,发酵产物中的异鼠李素大为提高,祛痘功能显著增强。
本发明还配合使用了丁香花,其含有丰富的丁香油酚、乙酰丁香油酚、B-石竹烯,以及甲基正戊基酮、水杨酸甲酯、葎草烯、苯甲醛、苄醇、间甲氧基苯甲醛、乙酸苄酯、胡椒酚、a一衣兰烯等。本发明研究发现,添加丁香花发酵后,祛痘效果显著优于仅使用银杏外种皮的祛痘效果。
进一步的,本发明还采用了菖蒲根,其主要含有α-、β-细辛醚、顺甲基异丁香酚、甲基丁香酚、菖蒲稀二醇、菖蒲酮等,具有明显的抗炎活性及祛痘效果,能抑制促炎因子的产生,对急性、慢性和一些免疫炎症模型均有抗炎效果;对酵母菌和痤疮丙酸杆菌具有强烈的抑菌效果,制备祛痘产品效果进一步增强。
本发明通过研究发现,将银杏外种皮、丁香花和菖蒲根发酵后的提取液中,异鼠李素的含量显著增加,异鼠李素对痤疮丙酸杆菌引起给炎症具有较好的消炎作用,用作祛痘产品时效果显著。
下面将通过实施例对本发明的具体实施方式做进一步的解释说明,但不表示将本发明的保护范围限制在实施例所述范围内。
实施例和对比例中所使用的设备和原料均为普通市售产品。
实施例和对比例的抗衰祛痘膏的成分如下表1所示。

具体的制备步骤包括:
a、将丁香花和菖蒲根粉碎后,加入超声萃取釜中,加入50-75%的乙醇进行萃取,萃取的料液比为1kg∶20-25L,萃取30-45min后过滤,弃去滤渣,得到提取物;
b、将银杏外种皮与步骤a所得提取物混合后,加入米曲霉和/或植物乳杆菌,米曲霉和/或植物乳杆菌的菌体总浓度为106-108cfu/mL,进行酶转化反应,酶转化反应温度为30-40℃,时间为3-5h,制得发酵液,过滤,得滤液;
c、取黄原胶和透明质酸钠在甘油中预先分散,加入甘草酸二钾、羧甲基纤维素、步骤b所得滤液和去离子水,搅拌溶解至分散均匀透明;
d、将甘草酸二钾和氨甲基丙醇在丙二醇和己二醇中溶解,加入到步骤c所得溶液中,加入乳木果油,混匀,得到银杏外种皮抗衰祛痘膏。
实施例2制备本发明银杏外种皮抗衰祛痘膏
原料成分如表1所示。
具体的制备步骤包括:

a、将银杏外种皮中加入米曲霉和/或植物乳杆菌,米曲霉和/或植物乳杆菌的菌体总浓度为106-108cfu/mL,进行酶转化反应,酶转化反应温度为30-40℃,时间为3-5h,制得发酵液,过滤,得滤液;
b、取黄原胶和透明质酸钠在甘油中预先分散,加入甘草酸二钾、羧甲基纤维素、步骤a所得滤液和去离子水,搅拌溶解至分散均匀透明;
c、将甘草酸二钾和氨甲基丙醇在丙二醇和己二醇中溶解,加入到步骤b所得溶液中,加入乳木果油,混匀,得到银杏外种皮抗衰祛痘膏。
对比例2不加入丁香花制备祛痘膏
原料成分如表1所示。(不加入丁香花)

b、将银杏外种皮与步骤a所得提取物混合后,加入米曲霉和/或植物乳杆菌,米曲霉和/或植物乳杆菌的菌体总浓度为106-108cfu/mL,进行酶转化反应,酶转化反应温度为30-40℃,时间为3-5h,制得发酵液,过滤,得滤液;
c、取黄原胶和透明质酸钠在甘油中预先分散,加入甘草酸二钾、羧甲基纤维素、步骤b所得滤液和去离子水,搅拌溶解至分散均匀透明;
d、将甘草酸二钾和氨甲基丙醇在丙二醇和己二醇中溶解,加入到步骤c所得溶液中,加入乳木果油,混匀,得到银杏外种皮抗衰祛痘膏。
对比例4不发酵制备祛痘膏
原料成分如表1所示。(不加入米曲霉和/或植物乳杆菌)
具体的制备步骤包括:
a、将丁香花和菖蒲根粉碎后,加入超声萃取釜中,加入50-75%的乙醇进行萃取,萃取的料液比为1kg∶20-25L,萃取30-45min后过滤,弃去滤渣,得到提取物;将银杏外种皮加入提取物中;
b、取黄原胶和透明质酸钠在甘油中预先分散,加入甘草酸二钾、羧甲基纤维素、步骤a所得提取物和去离子水,搅拌溶解至分散均匀透明;
c、将甘草酸二钾和氨甲基丙醇在丙二醇和己二醇中溶解,加入到步骤b所得溶液中,加入乳木果油,混匀,得到银杏外种皮抗衰祛痘膏。
对实施例和对比例中制备的抗衰祛痘膏的功效进行验证。
(1)实施例和对比例的抗衰祛痘膏的抑菌实验
将实施例和对比例制备的抗衰祛痘膏加入去离子水,制备成浓度为2%的样品水溶液,备用。
各取7种样品水溶液1mL分别加到无菌玻璃试管中,分别倾倒8mL预先制备的改良GAM肉汤培养基。取出已分离提纯并在35℃恒温CO2培养箱培养48h的临床痤疮丙酸杆菌,分别取1mL菌悬液注入已加样品水溶液的改良GAM肉汤培养基试管中,以一个不含菌悬液的玻璃试管做阴性对照。每种样品培养基接种痤疮丙酸杆菌,接种后尽快置于35℃恒温CO2培养箱培养48h。培养后,先抽取改良GAM肉汤培养基中的液体进行初步验证实验,再通过VITEK全自动微生物分析系统、ANI厌氧菌鉴定卡确定痤疮丙酸杆菌的杀灭情况,抑菌率的计算公式为:X=(A-B)/A×100%,其中X为抑菌率,A为对照样品平均回收菌落,B为试验样品平均回收菌落。功效评价标准为:(1)抑菌率≥90%为显效;(2)抑菌率50~90%为有效;(3)抑菌率<50%为无效。
结果如下表2所示。

实验结果表明,本发明制备的抗衰祛痘膏对痤疮丙酸杆菌有明显抑制效果,组合物间的协同作用显著,对痤疮丙酸杆菌抑制能力明显高于对比例。另外,实施例和对比例1-3的抑菌效果明显优于对比例4,说明中药组合物经过发酵后,抑菌功效得到明显增强。
(2)清除自由基的效果实验
将实施例和对比例6制备的抗衰祛痘膏配成质量分数为度为1%的样品水溶液,进行下面实验。
超氧阴离子自由基清除实验:取0.05mol/L pH=8.2的Tris-HCl缓冲液4.5mL,于25℃水浴锅中预热20min。再加入1mL样品水溶液和0.4mL 25mmol/L的邻苯三酚溶液,混匀后,于25℃水浴中反应5min,加入8mol/L的HCl 1.0mL终止反应。以Tris-HCl缓冲液作参比,在299nm处测吸光度值。空白对照用1mL样品水溶液的溶剂来替代样品。计算清除率。超氧阴离子自由基清除率(%)=[1-(A2/A1)]×100%;其中A1为空白对照的吸光度值;A2为药物的吸光度值。
羟自由基的清除实验:在25mL比色管中依次加入2mmol/L FeSO4 3mL,1mmol/LH2O2 3mL,摇匀,接着加入6mmol/L水杨酸3mL,摇匀,于37℃水浴加热15min后取出,测其吸光度;分别加入一定浓度的待测液,摇匀,继续水浴加热15min,取出测其吸光度。羟自由基的清除率(%)=[A1-A2-(A1-A3)]/A1×100%;其中,A1为未加药品前反应体系的吸光度值;A2为药品清除羟自由基后体系的吸光度值;A3为空白对照清除羟自由基后体系的吸光度值。
实验结果如下表3所示。

(3)实施例和对比例的抗衰祛痘膏的祛痘实验
评价对象:选择面部有痤疮、年龄为15~30岁的年轻人70例,其中男性35例,女性35例,随机平均分成7组,每组10人。
评价方法:每组分别使用上述实施例1-3,对比例1-4制备的抗衰老祛痘膏,实验共进行28天,考察28天的效果。
参考《中国痤疮治疗指南》进行综合评价。痊愈:皮损全部消退,仅存在少量色素沉着及轻微疤痕;显效:皮损缓解程度≥70%,各项临床症状显著改善;有效:皮损缓解程度超过≥30%,各项临床症状有明显好转趋势,但未消退;无效:皮损缓解程度<30%,各项临床症状未见改善或存在明显加剧趋势。
28天后的试验结果统计表4如下。

由上述实验结果可知,本发明制备的祛痘膏抑菌能力更强,祛痘效果更好,还具有清楚自由基的作用,同时能够抗衰老,具有很好的效果。
Claims (10)
1.银杏外种皮抗衰祛痘膏,其特征在于,组成包括:按重量份数计,银杏外种皮10-30份,丁香花20-40份,菖蒲根20-40份,辅料90-150份。
2.根据权利要求1所述的银杏外种皮抗衰祛痘膏,其特征在于,组成包括:按重量份数计,银杏外种皮20份,丁香花30份,菖蒲根30份,辅料120份。
3.根据权利要求1所述的银杏外种皮抗衰祛痘膏,其特征在于,组成还包括:3-5重量份的微生物,所述微生物为米曲霉或植物乳杆菌中的至少一种。
4.根据权利要求3所述的银杏外种皮抗衰祛痘膏,其特征在于:所述微生物为米曲霉与植物乳杆菌按活菌数比例1-5:3-8混合而成,微生物的浓度为106-108cfu/mL。
5.根据权利要求1所述的银杏外种皮抗衰祛痘膏,其特征在于:所述辅料包括甘油、丙二醇、透明质酸钠、黄原胶、羧甲基纤维素、甘草酸二钾、己二醇、乳木果油和氨甲基丙醇。
6.根据权利要求5所述的银杏外种皮抗衰祛痘膏,其特征在于:所述辅料为甘油为5-10份,丙二醇10-15份、透明质酸钠10-20份、黄原胶15-20份、羧甲基纤维素20-25份、甘草酸二钾5-15份、己二醇10-15份、乳木果油5-15份、氨甲基丙醇10-15份。
7.权利要求1-6任一项所述的银杏外种皮抗衰祛痘膏的制备方法,其特征在于,包括以下步骤:
a、将丁香花和菖蒲根粉碎后,加入超声萃取釜中,加入50-75%的乙醇进行萃取,萃取30-45min后过滤,弃去滤渣,得到提取物;
b、将银杏外种皮与步骤a所得提取物混合后,加入米曲霉和/或植物乳杆菌,进行酶转化反应,制得发酵液,过滤,得滤液;
c、取黄原胶和透明质酸钠在甘油中预先分散,加入甘草酸二钾、羧甲基纤维素、步骤b所得滤液和去离子水,搅拌溶解至分散均匀透明;
d、将甘草酸二钾和氨甲基丙醇在丙二醇和己二醇中溶解,加入到步骤c所得溶液中,加入乳木果油,混匀,得到银杏外种皮抗衰祛痘膏。
8.根据权利要求7所述的银杏外种皮抗衰祛痘膏的制备方法,其特征在于:步骤a所述粉碎后的粒径为200目以下。
9.根据权利要求7所述的银杏外种皮抗衰祛痘膏的制备方法,其特征在于:步骤a所述萃取的料液比为1kg:20-25L。
10.根据权利要求7所述的银杏外种皮抗衰祛痘膏的制备方法,其特征在于:步骤b所述酶转化反应温度为30-40℃,时间为3-5h。
