本发明公开了一种基于纤维素的可降解抗菌肽及其制备方法。包括以下步骤:步骤1:将纤维素加入硫酸溶液中,酸处理,乙醇终止反应;离心、洗涤、冷冻干燥,得到纤维素A;步骤2:将纤维素A分散在去离子水中,得到纤维素溶液;调节pH=9,加入1‑(3‑二甲氨基丙基)‑3‑乙基卡宾酰亚胺、N‑羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌10~12小时,加入甲氧基聚乙二醇胺,反应一段时间;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理2~4小时,洗涤、透析、冷冻干燥,得到可降解抗菌肽。

天然纤维素是一种来源广泛、生物降解容易的碳水化合物聚合物;但其抗菌活性差,限制了其在生物领域的应用。基于此,纤维素的改性已成为一个热点。而其衍生物羧甲基纤维素,具有良好的水溶性,继承了纤维素无污染、易降解的优点,在随机甲基纤维素的侧链上通过羧基活化改性可以酰胺化接枝,模拟制备基于羧甲基纤维素的可降解抗菌肽。如现有专利CN202011539085.2中公开的一种羧甲基纤维素仿生抗菌肽,但其虽在不影响羧甲基纤维素可降解的特性下,提高了基于羧甲基纤维素的改性效率,增加了抗菌效果;但是其抗菌效果仍然较低,同时由于其由于耐蛋白水解酶性差,其在血浆中的稳定性低,实际使用中会降低抗菌肽的活性。此外,增强抗菌肽的生物利用度,在不降低其抗菌效率基础上实现可控释放,也是需要解决的问题。
综上,解决上述问题,制备一种基于纤维素的可降解抗菌肽具有重要意义。
发明内容
本发明的目的在于提供一种基于纤维素的可降解抗菌肽及制备方法,以解决上述背景技术中提出的问题。
为了解决上述技术问题,本发明提供如下技术方案:
一种基于纤维素的可降解抗菌肽的制备方法,包括以下步骤:
步骤1:将纤维素加入硫酸溶液中,酸处理,乙醇终止反应;离心、洗涤、冷冻干燥,得到纤维素A;
步骤2:将纤维素A分散在去离子水中,得到纤维素溶液;调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌10~12小时,加入甲氧基聚乙二醇胺,反应一段时间;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理2~4小时,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
较为优化地,所述纤维素包括羧甲基纤维素、羧基化微晶纤维素、羧基化细菌纤维素中一种或多种。
较为优化地,步骤1中,硫酸溶液浓度为40~60%,纤维素与硫酸溶液的固液比为1g:10mL,酸处理温度为室温,酸处理时间为1~6小时;离心的速度为6000rmp,离心时间为10分钟。
较为优化地,步骤2中,所述二氨基化合物包括1,4-二氨基丁烷、1,5-二氨基戊烷,1,6-己基乙二胺、1,7-二氨基庚烷、1,8-二氨基辛烷、1,10-二氨基癸烷中一种或多种。
较为优化地,步骤2中,所述二氨基化合物的加入量占纤维素质量的18~22%。
较为优化地,步骤2:将纤维素A分散在去离子水中,得到0.1g/mL的纤维素溶液,调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺(EDC)、N-羟基硫代琥珀酰亚胺钠盐(NHS),搅拌2小时;加入过量的二氨基化合物,搅拌6~8小时;加入甲氧基聚乙二醇胺,加入0.1M的盐酸调节pH=5,搅拌8~10小时;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理2~4小时,滴加四丁基氢氧化铵溶液,继续搅拌2~4小时;反应结束后,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
较为优化地,所述甲氧基聚乙二醇胺的加入量是纤维素质量的5%~8%。
较为优化地,所述四丁基氢氧化铵溶液的浓度为10%;四丁基氢氧化铵溶液的滴加量为纤维素溶液质量的4%~7%。
较为优化地,将步骤2中可降解抗菌肽使用胆固醇改性,具体步骤为:将可降解抗菌肽与氯甲酸胆固醇酯按照质量比为3:1在氯仿溶液中室温搅拌24小时,使用乙醚沉淀,过滤、洗涤、冷冻干燥得到胆固醇改性的可降解抗菌肽。
较为优化地,一种基于纤维素的可降解抗菌肽的制备方法制备得到的可降解抗菌肽。
本技术方案中,基于纤维素模拟天然抗菌肽,通过酸解、酰胺化接枝过程在纤维素上接枝二胺和甲氧基聚乙二醇胺;利用酸解使得纤维素分子量适宜,羧基丰度适宜,用以增强接枝率,提高抗菌活性;同时利用接枝聚乙二醇链,提高可降解抗菌肽的抗菌活性,同时增加等离子体稳定性,增加抗菌肽的耐蛋白酶性,降低溶血活性,降低对细胞的毒性。
(1)方案中限定了纤维素的种类,因为羧基活化后与氨基酰胺化接枝是主反应过程,因此,羧基化影响接枝,因此需要较高具有较高羧基丰度的纤维素。同时,方案中限定了酸处理的条件,因为酸处理过程是纤维素降解过程,处理时间过短,分子量大,链长,接枝率下降,抗菌活性下降;处理时间过程,产量下降,因此需要限定酸解时间,从而得到产量较高,抗菌效果、抗菌效率高的抗菌肽。
(2)方案中选取二胺化合物中含有支链烷烃,可以与两亲性的抗菌肽中的脂溶性物质协同,与细菌细胞壁上的脂溶性成分结合,穿透细胞膜,从而提高抗菌性;但是,疏水性直链烷烃会增加溶血性能,存在细胞毒性;因此,方案中引入接枝聚乙二醇链段,利用由于更长亲水性链段的接枝,降低溶血活性,以此协同互补。此外,聚乙二醇链段可以提高耐蛋白水解酶性,提高其在生理环境中的应用。且可以增加抗菌肽的空间稳定性,抑制纤维素在后续反应过程中的具体,影响反应均匀性。
方案中,由于甲氧基聚乙二醇胺和二胺化合物一样通过偶联反应接枝,两者之间存在竞争性,同时,由于甲氧基聚乙二醇胺分子量更大,接枝性差,且会阻碍二胺化合物影响接枝率,因此,先接枝二胺化合物。又因为含有羧基的纤维素A在酸性条件下的聚集性会影响接枝率和接枝均匀性,因此,方案中通过NHS激活羧基官能团,增加反应性和反应稳定性,使得酰胺化可以在较高pH下进行,在EDC的介导下使得羧基和氨基产生酰胺化反应。由于羧基的消耗,增加了甲氧基聚乙二醇胺反应过程中的胶体稳定性,由于已经接枝了一部分,存在接枝阻力,因此,调节降低pH值,进一步活化剩余羧基,增强甲氧基聚乙二醇胺的接枝反应性;同时为了增加接枝反应性,限定了其加入量,一般来说加入量较高会增加接枝的反应性,但是在此方案中,加入量过多,会影响增加反应液的粘性,增加粘性阻力,不利于接枝。
(3)此外,聚乙二醇的接枝由于增加了支链长度,增加了抗菌肽分子之间的空间稳定性,因此,在盐酸季铵盐化过程中,可以进一步降低盐酸pH为3.5作用,同时,还可以在该酸性环境下,利用酸碱中和反应,引入四丁基铵盐,进一步增加阳离子电荷中心,增加抗菌性,由于季铵盐化过程促进了酸碱中和的正向反应,抑制了逆向反应。因此增加了阳离子中心。
(4)一般来说,通常都会使用脂质体进行对抗菌肽进行封装,但是封装会缓释活性物质,这样虽增加了生物利用度,但是会降低抗菌效率;因此方案中,并未全包封,而是采用胆固醇改性,利用聚乙二醇和胆固醇的共同作用,增加缓释性能,提高利用度,胆固醇与可降解抗菌肽通过酯键偶联;其可以显著提高改性后的可降解抗菌肽的稳定性,无溶血活性,降低毒性;同时进一步提高了抗菌效果。
具体实施方式
下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1:
步骤1:将2g羧甲基纤维素加入20mL的浓度为50%的硫酸溶液中,室温下酸处理2小时,乙醇终止反应;在离心速度为6000rmp下离心10分钟、洗涤、冷冻干燥,得到纤维素A;
步骤2:将纤维素A分散在去离子水中,得到纤维素溶液;调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的5-二氨基戊烷,搅拌12小时,加入甲氧基聚乙二醇胺,反应一段时间;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理4小时,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
本技术方案中,5-二氨基戊烷的加入量占纤维素质量的20%;甲氧基聚乙二醇胺的加入量是羧甲基纤维素质量的6%。
实施例2:
步骤1:将2g羧甲基纤维素加入20mL的浓度为50%的硫酸溶液中,室温下酸处理1~6小时,乙醇终止反应;在离心速度为6000rmp下离心10分钟、洗涤、冷冻干燥,得到纤维素A;
步骤2:将纤维素A分散在去离子水中,得到0.1g/mL的纤维素溶液,调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌8小时;加入甲氧基聚乙二醇胺,加入0.1M的盐酸调节pH=5,搅拌6小时;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理4小时,滴加四丁基氢氧化铵溶液,继续搅拌3小时;反应结束后,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
本技术方案中,5-二氨基戊烷的加入量占纤维素质量的20%;甲氧基聚乙二醇胺的加入量是纤维素质量的6%。所述四丁基氢氧化铵溶液的浓度为10%;四丁基氢氧化铵溶液的滴加量为羧甲基纤维素溶液质量的5%。
实施例3:
步骤1:将2g羧甲基纤维素加入20mL的浓度为40%的硫酸溶液中,室温下酸处理6小时,乙醇终止反应;在离心速度为6000rmp下离心10分钟、洗涤、冷冻干燥,得到纤维素A;
步骤2:将纤维素A分散在去离子水中,得到0.1g/mL的纤维素溶液,调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌6小时;加入甲氧基聚乙二醇胺,加入0.1M的盐酸调节pH=5,搅拌10小时;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理4小时,滴加四丁基氢氧化铵溶液,继续搅拌4小时;反应结束后,洗涤、透析、冷冻干燥,得到可降解抗菌肽。

实施例4:
步骤1:将2g羧甲基纤维素加入20mL的浓度为60%的硫酸溶液中,室温下酸处理1小时,乙醇终止反应;在离心速度为6000rmp下离心10分钟、洗涤、冷冻干燥,得到纤维素A;
步骤2:将纤维素A分散在去离子水中,得到0.1g/mL的纤维素溶液,调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌8小时;加入甲氧基聚乙二醇胺,加入0.1M的盐酸调节pH=5,搅拌8小时;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理2小时,滴加四丁基氢氧化铵溶液,继续搅拌2小时;反应结束后,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
本技术方案中,5-二氨基戊烷的加入量占纤维素质量的22%;甲氧基聚乙二醇胺的加入量是羧甲基纤维素质量的8%。所述四丁基氢氧化铵溶液的浓度为10%;四丁基氢氧化铵溶液的滴加量为纤维素溶液质量的7%。
实施例5:
步骤1:将2g羧甲基纤维素加入20mL的浓度为50%的硫酸溶液中,室温下酸处理1~6小时,乙醇终止反应;在离心速度为6000rmp下离心10分钟、洗涤、冷冻干燥,得到纤维素A;
步骤2:将纤维素A分散在去离子水中,得到0.1g/mL的纤维素溶液,调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌8小时;加入甲氧基聚乙二醇胺,加入0.1M的盐酸调节pH=5,搅拌6小时;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理4小时,滴加四丁基氢氧化铵溶液,继续搅拌3小时;反应结束后,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
步骤3:将3g可降解抗菌肽与1g氯甲酸胆固醇酯在50mL三氯甲烷中室温搅拌24小时,使用乙醚沉淀,过滤、洗涤、冷冻干燥得到胆固醇改性的可降解抗菌肽。
本技术方案中,5-二氨基戊烷的加入量占纤维素质量的20%;甲氧基聚乙二醇胺的加入量是纤维素质量的6%。所述四丁基氢氧化铵溶液的浓度为10%;四丁基氢氧化铵溶液的滴加量为羧甲基纤维素溶液质量的5%。
对比例1:按照专利CN202011539085.2的实施例1的制备方法,制备得到可降解抗菌肽;
对比例2:不加入甲氧基聚乙二醇胺,其余与实施例2相同;
对比例3:将甲氧基聚乙二醇胺的加入量调整为是羧甲基纤维素质量的15%,其余与实施例2相同。
对比例4:甲氧基聚乙二醇胺的加入时,不调节pH,其余与实施例2相同。
实验:取实施例和对比例进行体外抗菌活性实验和溶血性实验。(1)体外抗菌活性实验:使用微量肉汤稀释法测定最低抑菌浓度,用以表征抑菌活性;使用含0.2%BSA的浓度为0.01%的乙酸作为稀释液,以二倍法配置梯度的抗菌肽溶液,将其置于96孔细胞培养板中,加入等体积的终浓度为105CFU的细胞悬液,37℃恒温培养20小时;使用酶标仪在测定光吸收值,得到最低抑菌浓度。(2)将1mL血液离心,将离心所得的下层溶液使用PBS清洗,配成8%的细胞悬浮液,将梯度抗菌肽溶液添加至96孔细胞培养板中,加入等体积细胞悬浮液,37℃恒温培养1小时,离心后,上层溶液使用酶标仪测试光吸收值;以PBS作为阴性对照,以0.1%TritonX-100作为阳性对照;根据公式计算得到溶血率(红细胞抗菌肽浓度为2mg/mL),用以评价耐蛋白酶性质。所有数据如下表所示:
结论:由实施例1~4的数据可知:本方案中制备的基于纤维素的可降解抗菌肽具有优异的抗菌效果和低溶血率。最优溶血率低至5.8%,在6%之下,红细胞的渗透脆性影响较小,具有较好的细胞稳定性,细胞毒性小。同时,实施例1的数据与实施例2~3的数据对比可知,在后续通过酸碱中和过程,增加阳离子可以显著增加抗菌活性,原因在于:抗菌肽的抗菌性与其二级结构关系较低,与其两亲性和正电荷相关。
将实施例2与实施例5的数据对比可知:使用胆固醇改性,其抗菌活性并未降低,同时溶血率进一步下降,原因在于:胆固醇与可降解抗菌肽通过酯键偶联,与聚乙二醇协同提高了可降解抗菌肽的稳定性,因为胆固醇、聚乙二醇是常用于包封活性物质的聚合物,可以增强活性成分的缓释性和稳定性。对比例1的数据与实施例1、2的数据对比可知:聚乙二醇链段不仅能防止正电荷抗菌肽被负电荷白蛋白的吸引,同时可以提高耐蛋白水解酶性,增加血浆稳定性提高其在生理环境中的应用;而阳离子改性,进一步提高的抗菌肽的抗菌活性。再与对比例2~4的数据可知,对比例2中甲氧基聚乙二醇胺不加入不仅影响了耐蛋白酶性,而且还影响了抗菌活性;因为该物质的接枝,影响了后续利用酸碱中和反应引入四丁基铵盐;聚乙二醇链段未接枝,降低了抗菌肽与抗菌肽之间的空间稳定性,使得后续酸碱中和时,聚集,阳离子改性不均匀,正反应性降低;从而使得抗菌活性下降。对比例3中甲氧基聚乙二醇胺加入量过多,对比例4中不调节pH,由于影响聚乙二醇链段接枝的稳定性和反应性,使得接枝率下降,从而影响后续反应,使得抗菌活性下降、溶血率升高。

Claims (10)
1.一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:包括以下步骤:
步骤1:将纤维素加入硫酸溶液中,酸处理,乙醇终止反应;离心、洗涤、冷冻干燥,得到纤维素A;
步骤2:将纤维素A分散在去离子水中,得到纤维素溶液;调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌10~12小时,加入甲氧基聚乙二醇胺,反应一段时间;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理2~4小时,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
2.根据权利要求1所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:所述纤维素包括羧甲基纤维素、羧基化微晶纤维素、羧基化细菌纤维素中一种或多种。
3.根据权利要求1所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:步骤1中,硫酸溶液浓度为40~60%,纤维素与硫酸溶液的固液比为1g:10mL,酸处理温度为室温,酸处理时间为1~6小时;离心的速度为6000rmp,离心时间为10分钟。
4.根据权利要求1所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:步骤2中,所述二氨基化合物包括1,4-二氨基丁烷、1,5-二氨基戊烷,1,6-己基乙二胺、1,7-二氨基庚烷、1,8-二氨基辛烷、1,10-二氨基癸烷中一种或多种。
5.根据权利要求1所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:步骤2中,所述二氨基化合物的加入量占纤维素质量的18~22%。
6.根据权利要求1所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:步骤2:将纤维素A分散在去离子水中,得到0.1g/mL的纤维素溶液,调节pH=9,加入1-(3-二甲氨基丙基)-3-乙基卡宾酰亚胺、N-羟基硫代琥珀酰亚胺钠盐,搅拌2小时;加入过量的二氨基化合物,搅拌6~8小时;加入甲氧基聚乙二醇胺,加入0.1M的盐酸调节pH=5,搅拌8~10小时;乙醇沉淀、离心、洗涤,冷冻干燥;将其超声分散在水中,使用0.1M的盐酸调节pH=3.5,搅拌处理2~4小时,滴加四丁基氢氧化铵溶液,继续搅拌2~4小时;反应结束后,洗涤、透析、冷冻干燥,得到可降解抗菌肽。
7.根据权利要求6所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:所述甲氧基聚乙二醇胺的加入量是纤维素质量的5%~8%。
8.根据权利要求6所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:四丁基氢氧化铵溶液的滴加量为纤维素溶液质量的4%~7%。
9.根据权利要求1所述的一种基于纤维素的可降解抗菌肽的制备方法,其特征在于:将步骤2中可降解抗菌肽使用胆固醇改性,具体步骤为:将可降解抗菌肽与氯甲酸胆固醇酯按照质量比为3:1在氯仿溶液中室温搅拌24小时,使用乙醚沉淀,过滤、洗涤、冷冻干燥得到胆固醇改性的可降解抗菌肽。
10.根据权利要求1~9中任意一项所述的一种基于纤维素的可降解抗菌肽的制备方法制备得到的可降解抗菌肽。
