Abstract
本发明提供了一种富含脂质体纳米粒的美白敷料及其制备方法,该美白辅料由以下组分组成:水,脂质体纳米粒,甘油,苹果酸,维生素C,传明酸,烟酰胺,黄原胶,乙二胺四乙酸二钠,氢化卵磷脂。所述脂质体纳米粒是将玫瑰提取物包封于类脂质双分子层内而形成的微型球状载体,其中包封的膜材是一定比例改性磷脂酰胆碱、聚乙二醇枝连磷脂酰胆碱,制备得到的脂质体纳米粒即不易被氧化而使内容物泄露,也不会由于团聚而影响产品的质感以及吸收效率降低,具有良好的稳定性且易于被皮肤吸收。同时脂质体纳米粒还能促进皮肤对美白敷料中其他美白成分的吸收,配合脂质体纳米粒包埋的玫瑰提取物,具有良好的美白效果。

技术领域
本发明涉及化妆品领域,尤其涉及一种富含脂质体纳米粒的美白敷料及其制备方法。
背景技术
当代人以白为美,然而紫外线、作息等因素导致皮肤中黑色素增多聚集,致使面部色度降低影响形象,美白敷料作为功效面膜中最常见的一类,因其使用便利、对皮肤的副作用小、美白效果好等优点成为大家改善皮肤色泽的首要选择。美白敷料是指添加了一些可使黑色素代谢加快、抑制或者破坏黑色素的生成、复原黑色素中间体或者阻止酪氨酸酶活化等具有美白功效成分的敷料。因此具有抗氧化性以及可抑制酪氨酸酶活性的维生素C及其衍生物、烟酰胺、熊果苷、传明酸等成分常作为美白剂加入敷料中。
黑色素主要是由皮肤的表皮基底层的黑色素细胞生成,其生成机制为:位于黑色素细胞膜上的酪氨酸酶是一种具有双核铜离子的金属酶,在黑色素合成中起着限速酶的作用,可在正常生理或外界刺激如紫外线照射等条件下激活,黑色素细胞中的酪氨酸在酪氨酸酶的催化氧化作用下,经一系列转化生成褐色素及真黑素,两者统称为黑色素。根据黑色素的生成步骤,目前市面上美白面膜的作用机制可归纳为抑制氧化反应发生、抑制酪氨酸酶的活性,或加快皮肤新陈代谢,使黑色素细胞向角质层转移并随着角质层更新脱落而达到美白的作用。
脂质体纳米粒是指粒径为纳米级的脂质体结构,多为单室脂质体。由于其粒径处于纳米级范围,因此在稳定性、吸收和体内分布方面具有纳米粒子的特殊效应。因为这种纳米效应使其穿透细胞的能力增强,进入细胞的数量增多,能够携带有效成分进入靶组织,这种现象就是小尺寸效应。因为粒径极小,同一体积含有更多的粒子,使更多的活性物质可以与靶组织接触发挥作用。作为化妆品中活性成分的传递媒介,脂质体有许多优势,它与微乳和多乳状液一起被看作是最有前途的三种化妆品体系。
专利CN110090165B提供了一种小核酸纳米美白霜,由小核酸活性成分、纳米材料、乳膏基质组成。该发明采用TYR-siRNA作为小核酸活性成分,利用纳米材料复合TYR-siRNA,并且乳膏中还含有保湿因子,促进透皮的成分等制备,该发明所述小核酸纳米美白霜能有效抑制黑色素生成,抑制皮肤络氨酸酶的表达,具有美白保湿功效,可应用于化妆品领域,发挥美白作用。其问题在于,纳米材料粒径很小,容易发生团聚,致使产品的稳定性较难得到保证,同时通过单一有效成分的美白效果有限。
专利CN111728924A提供了一种美白面膜及其制备方法,该面膜由以下重量份的组分制成:紫甘薯花青素纳米脂质体2~20份、北美金缕梅提取物2~10份、丝瓜茎提取物1~5份、芦荟提取物2~5份、甘油2~10份、丁二醇5~10份、海藻糖0.2~1份、透明质酸钠0.1~0.5份、抗坏血酸磷酸酯钠1~5份、烟酰胺3~8份、玫瑰精油10~20份、苦橙花精油1~2份、pH调节剂0.05~0.5份,去离子水80~120份;按比例称取原料,分别制成水相和油相,混合乳化,调节pH值,将无纺布贴膜浸泡于面膜混合液中,然后抽真空使面膜混合液与无纺布贴膜完全结合,抽检,分装。其问题在于,纳米脂质体较易发生团聚,同时配方中并未添加有效的乳化剂,使得产品的稳定性较差,导致美白效果降低。
发明内容
有鉴于上述现有技术的缺陷,本发明提供了一种富含脂质体纳米粒的美白敷料及其制备方法,该方法制备的美白敷料不仅具有良好的美白效果,同时还具有贮藏稳定性好,易于被皮肤吸收的优点。
为了实现上述发明目的,本发明提供以下技术方案:
一种富含脂质体纳米粒的美白敷料,由以下组分组成:水,脂质体纳米粒,甘油,苹果酸,维生素C,传明酸,烟酰胺,黄原胶,乙二胺四乙酸二钠,氢化卵磷脂。
优选的,一种富含脂质体纳米粒的美白敷料,由以下质量份的组分组成:20~40份水,10~20份脂质体纳米粒,2~8份甘油,0.1~1份苹果酸,0.5~2份维生素C,1~2份传明酸,1~2份烟酰胺,0.2~0.6份黄原胶,0.1~0.5份乙二胺四乙酸二钠,2~5份氢化卵磷脂。
优选的,所述脂质体纳米粒的制备方法,包括以下步骤:
S1将吐温80溶解在磷酸缓冲液中得到A液;
S2将磷脂、胆固醇、玫瑰提取物分散在无水乙醇中,加热溶解得到B液;
S3将A液与B液混合,减压浓缩后超声,冷冻干燥得到脂质体纳米粒。
优选的,所述脂质体纳米粒的制备方法,包括以下步骤:
S1将1~3g吐温80溶解在200~600mL pH=7~9的0.1~0.2mol/L磷酸缓冲液中得到A液;
S2将2~6g磷脂、0.4~1g胆固醇、0.2~1g玫瑰提取物分散在30~100mL无水乙醇中,在50~70℃下充分溶解得到B液;
S3将A液与B液混合,在40~60℃下减压浓缩至原混合液体积的2/3~4/5,将混合液超声5~10min,最后在-40~-45℃下冷冻干燥24~48h,得到脂质体纳米粒。
玫瑰是集药用、食用、美化、绿化为一体的木本植物,由其提取制备的玫瑰提取物具有抑菌、抗氧化、抗敏感、保湿、促进细胞再生等功效,还是一种纯天然的香料,与此同时玫瑰中还有多种具有抗氧化、美白功效的多糖、多酚、黄酮类化合物,且相比化学合成的美白成分更加安全。但其缺点在于,水溶性差,现代美白类化妆品一般以水作为溶剂,如果添加量过多,玫瑰提取物中部分成分会析出,一方面局部浓度过大,影响美白效果,另一方面也影响产品的性状;此外,玫瑰提取物中美白活性成分不稳定,渗透性较差,有效成分不能到达皮肤深层发挥有效作用;同时在提取过程中,不可避免使用到部分化学试剂,其中可能会导致过敏反应。
本发明制备脂质体纳米粒是将玫瑰提取物包封于类脂质双分子层内而形成的微型球状载体。其中水溶性成分可包裹在磷脂的内水相,脂溶性成分可包裹在磷脂双分子层间。其表面的磷脂具有很强的水合作用,局部使用大量脂质体使皮肤水合作用增强,角质细胞间隙变大,脂质双层中疏水性尾部排列紊乱,最终脂溶性成分通过扩散和毛细吸力作用进入细胞间隙;另一方面,脂质体的磷脂与角质层中的脂质融合,其屏障作用发生逆转,角质层形成扁平的颗粒状结构,包封有玫瑰提取物的脂质体顺利通过脂质颗粒的间隙,促进玫瑰提取物透皮吸收;同时脂质体纳米粒具有长效缓释效果,与皮脂相似的类脂所形成的脂质体与皮肤角质层脂质有高度的相似性,为有效成分在皮肤内的储存提供空间,增加在局部的积累,通过包载也避免了有效成分发生降解或氧化失效。
优选的,所述磷脂为磷脂酰胆碱、改性磷脂酰胆碱、聚乙二醇枝连磷脂酰胆碱其中一种或两种或两种以上的混合。
优选的,所述改性磷脂酰胆碱的制备方法为:将磷脂酰胆碱溶解在正己烷中,再加入硬脂酸和脂肪酶,加热反应,降压蒸除溶剂,收集残留物,用丙酮洗涤后干燥得到改性磷脂酰胆碱。
优选的,所述改性磷脂酰胆碱的制备方法为:将20~60g磷脂酰胆碱溶解在100~300mL正己烷中,加入40~120g硬脂酸、4~12g脂肪酶,在40~60℃下反应30~50h,反应结束后减压蒸除溶剂,收集残留物,用丙酮洗涤1~3次后,在40~60℃下干燥12~24h得到改性磷脂酰胆碱。
磷脂酰胆碱含有亲水和疏水基团,水中能自发形成双分子层结构,是构建脂质体的主要膜材。但天然磷脂酰胆碱的酰基链上油酸和亚油酸等不饱和脂肪酸较多,不饱和脂肪酸双键在空气中易被氧化,易导致脂质体膜的破坏,导致内容物外泄。
本发明使用不含双键的硬脂酸与磷脂酰胆碱的酰基链上不饱和脂肪酸进行置换,制备得到的改性磷脂酰胆碱的酰基链上不饱和脂肪酸含量显著降低,使其具有良好的氧化稳定性,因此由改性磷脂酰胆碱作为膜材制备得到脂质体纳米粒稳定性更好,在脂质体纳米粒在制备、保存、运输过程中避免因为磷脂酰胆碱的氧化水解导致的内容物泄露,以饱和度高的改性磷脂为膜材构建的脂质体纳米粒从原料上降低脂质氧化,比制备过程中隔绝空气、使用抗氧化剂等举措更加有效。
优选的,所述聚乙二醇枝连磷脂酰胆碱的制备方法,包括以下步骤:
N1将聚乙二醇溶解在丙酮溶液中得到聚乙二醇丙酮溶液;将氧化铬溶解于水中,加入硫酸水溶液得到羧基化试剂;向聚乙二醇丙酮溶液加入羧基化试剂混合反应,除去溶剂得到端羧基聚乙二醇;
N2将端羧基聚乙二醇溶解在二氯甲烷中,加入二氯亚砜反应,得到聚乙二醇酰氯;
N3将磷脂酰胆碱、聚乙二醇酰氯、三乙胺溶解在三氯甲烷中,加热反应,除去溶剂得到聚乙二醇枝连磷脂酰胆碱。
优选的,所述聚乙二醇枝连磷脂酰胆碱的制备方法,包括以下步骤:
N1将5~10g聚乙二醇溶解在100~200mL丙酮中得到聚乙二醇丙酮溶液;将50~100g氧化铬溶解在500~1000mL水中,加入50~100mL90~98wt%硫酸水溶液得到羧基化试剂;向上述聚乙二醇丙酮溶液中加入5~10mL羧基化试剂,在20~30℃反应12~24h,反应结束后加入20~40mL异丙醇终止反应,加入50~100mL水,在40~60℃下减压浓缩至原体积的1/3~1/2,加入碳酸氢钠至无气泡产生,用0.1~1mol/L盐酸调节溶液pH=1~3,在60~80℃下真空干燥得到白色固体,用50~100mL三氯甲烷溶解白色固体,过滤,收集滤液,在40~60℃下减压浓缩后干燥得到端羧基聚乙二醇;
N2将2~5g端羧基聚乙二醇溶解在10~20mL二氯甲烷中,加入20~40mL二氯亚砜,在20~30℃反应6~12h,在40~60℃下减压浓缩至原体积的1/3~1/2,加入无水乙醚沉淀,过滤收集沉淀,在40~60℃干燥得到聚乙二醇酰氯;
N3将4~10g磷脂酰胆碱、2~5g聚乙二醇酰氯、3~5mL三乙胺溶解在50~100mL三氯甲烷中,在45~60℃下反应4~10h,在40~60℃下减压蒸除溶剂,收集残留物,用50~100mL四氢呋喃溶解沉淀,过滤,收集滤液,在40~60℃下减压浓缩后干燥得到聚乙二醇枝连磷脂酰胆碱。
由于制备的脂质体纳米粒的粒径很小,虽然粒径越小越有利于皮肤的吸收,但是也会使的脂质体纳米粒的稳定性变差,从而导致脂质体纳米粒发生团聚,影响产品的质感以及吸收效率。聚乙二醇具有水溶性、柔顺性、无毒、免疫原性低和可生物降解等优点,使用聚乙二醇枝连磷脂酰胆碱作为膜材制备得到的脂质体纳米粒,在其表面形成“构象云”和水化膜,为脂质体纳米粒提供了较大的空间位阻,并掩盖其表面疏水性结合位点,使得脂质体纳米粒的理化稳定性得到显著的改善。
进一步优选的,所述磷脂为质量比为1~5:1的改性磷脂酰胆碱和聚乙二醇枝连磷脂酰胆碱的混合。
虽然聚乙二醇枝连磷脂酰胆碱作为膜材制备得到的脂质体纳米粒的理化稳定性得到显著的改善,但是由于其表面枝连过多的聚乙二醇,聚乙二醇分子中存在长链极性基团乙氧基,能与水分子形成氢键,从而脂质体纳米粒表面形成一层水化膜,且聚乙二醇链在表面形成的毛刷状的“构象云”,会阻止皮肤对脂质体纳米粒的吸收。
因此,本发明使用一定比例改性磷脂酰胆碱、聚乙二醇枝连磷脂酰胆碱作为膜材制备脂质体纳米粒,制备得到的脂质体纳米粒即不易被氧化而使内容物泄露,也不会由于团聚而影响产品的质感以及吸收效率降低,具有良好的稳定性且易于被皮肤吸收。
其中甘油即丙三醇,具有良好的保湿效果,可以起到长效的补水和保湿的作用,强化美白的作用。
苹果酸是一种有机酸,其可以通过加速角质细胞的代谢来达到美白的目的。维生素C又名抗坏血酸,是一种具有抗氧化性的水溶性维生素,能够通过抑制酪氨酸酶的活性减缓酪氨酸的转化速度从而减少黑色素的形成,同时维生素C的抗氧化性可使已经形成的黑色素转化为黑色素前体物质,从而达到美白的作用。
传明酸有很多优点,其中美白与祛斑的特质广受人们喜爱,它可以让肌肤呈现白净晶透的状态,传明酸的主要作用是当皮肤受到紫外线照射后,抑制前列腺素在前端干扰黑色素生成;在后端干扰黑色素从黑色素细胞向周围细胞扩散。其与维生素C等美白活性成分的复配对色素沉着有明显的改善作用。
烟酰胺是维生素B3的一种衍生物,性质稳定不易被酸碱破坏,易透过角质层且美白祛斑效果好,烟酰胺的美白机制为抑制黑色素颗粒的形成,加快皮肤代谢,使含有黑色素的细胞脱落,淡斑美白。
黄原胶是由野油菜黄单胞杆菌以碳水化合物为主要原料经发酵工程生产的一种作用广泛的微生物胞外多糖,是目前国际上集增稠、悬浮、乳化、稳定于一体,性能最优越的生物胶,能够有效的提升护肤品的稳定性和使用质感。
乙二胺四乙酸二钠是一种螯合剂,其可以通过螯合酪氨酸酶中的金属离子,通过使酪氨酸酶失去催化活性,从而阻断黑色素的合成途径达到美白效果。
氢化卵磷脂使卵磷脂的部分水解物,氢化卵磷脂提高了它的亲水能力,有着一定的保湿效果,可以被人的皮肤吸收并能促进其他营养物质的渗透,加强护肤品中有效成分的作用效果,并且其乳化性能强,能稳定乳状液,同时具有一定的抗氧化功能,保证护肤品中的有效成分不因氧化而失效。
本发明还提供了所述富含脂质体纳米粒的美白敷料的制备方法,包括以下步骤:
M1按配方称取原料,将水、脂质体纳米粒、苹果酸、维生素C、传明酸、黄原胶、乙二胺四乙酸二钠、氢化卵磷脂混匀得到水相;
M2将甘油、烟酰胺混匀得到油相;
M3将水相和油相混合后均质得到富含脂质体纳米粒的美白敷料。
优选的,所述富含脂质体纳米粒的美白敷料的制备方法,包括以下步骤:
M1按配方称取原料,将水、脂质体纳米粒、苹果酸、维生素C、传明酸、黄原胶、乙二胺四乙酸二钠、氢化卵磷脂加入搅拌器中,在50~70℃下,搅拌25~40min,得到水相;
M2将甘油、烟酰胺加入搅拌器中,在50~70℃下,搅拌25~40min,得到油相;
M3将水相和油相混合后放入均质机中,在压力为150~200MPa,均质速度为均质3~5min,得到富含脂质体纳米粒的美白敷料。
与现有技术相比,本发明的有益效果:
1、本发明使用一定比例改性磷脂酰胆碱、聚乙二醇枝连磷脂酰胆碱作为膜材制备脂质体纳米粒,制备得到的脂质体纳米粒即不易被氧化而使内容物泄露,也不会由于团聚而影响产品的质感以及吸收效率降低,具有良好的稳定性且易于被皮肤吸收;
2、本发明使用多种美白成分协同作用,配合脂质体纳米粒包埋的玫瑰提取物,具有良好的美白效果。

下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
本发明实施例中部分物质和仪器的具体参数如下:
脂肪酶,诺维信435脂肪酶,10000PLU/g,杭州创科生物科技有限公司。
玫瑰提取物,提取来源为玫瑰的干燥花蕾,主要成分有香茅醇、牻牛儿醇、橙花醇、丁香油酚、苯乙醇、槲皮甙、苦味质、鞣质、脂肪油、有机酸(没食子酸)、红色素、黄色素、蜡质、β-胡萝卜素等,西安优铂汇生物科技有限责任公司。
聚乙二醇,分子量2000,食品级,山东优索化工科技有限公司。
实施例1
一种富含脂质体纳米粒的美白敷料的制备方法,包括以下步骤:
M1将30g水、10g脂质体纳米粒、0.5g苹果酸、1g维生素C、2g传明酸、0.5g黄原胶、0.2g乙二胺四乙酸二钠、3g氢化卵磷脂加入搅拌器中,在60℃下,搅拌30min,得到水相;
M2将8g甘油、1g烟酰胺加入搅拌器中,在55℃下,搅拌30min,得到油相;
M3将水相和油相混合后放入均质机中,在压力为150MPa,均质速度为均质4min,得到富含脂质体纳米粒的美白敷料。
所述脂质体纳米粒的制备方法,包括以下步骤:
S1将3g吐温80溶解在600mLpH=8的0.1mol/L磷酸缓冲液中得到A液;
S2将6g磷脂、1g胆固醇、0.8g玫瑰提取物分散在100mL无水乙醇中,在60℃下充分溶解得到B液;
S3将A液与B液混合,在50℃下减压浓缩至原混合液体积的2/3,再将混合液超声5min,最后在-45℃下冷冻干燥48h,得到脂质体纳米粒。
所述磷脂为质量比为5:1的改性磷脂酰胆碱和聚乙二醇枝连磷脂酰胆碱的混合。
所述改性磷脂酰胆碱的制备方法为:将20g磷脂酰胆碱溶解在100mL正己烷中,加入40g硬脂酸、4g脂肪酶,在50℃下反应40h,反应结束后减压蒸除溶剂,收集残留物,用丙酮洗涤3次后,在45℃下干燥24h得到改性磷脂酰胆碱。
所述聚乙二醇枝连磷脂酰胆碱的制备方法,包括以下步骤:
N1将10g聚乙二醇溶解在200mL丙酮中得到聚乙二醇丙酮溶液;将80g氧化铬溶解在500mL水中,加入70mL 98wt%硫酸水溶液得到羧基化试剂;向上述聚乙二醇丙酮溶液中加入5mL羧基化试剂,在25℃反应12h,反应结束后加入20mL异丙醇终止反应,加入50mL水,在55℃旋蒸至原体积的1/2,加入碳酸氢钠至无气泡产生,用1mol/L盐酸调节溶液pH=1,在70℃下真空干燥得到白色固体,用50mL三氯甲烷溶解白色固体,过滤,收集滤液,在50℃下减压浓缩后干燥得到端羧基聚乙二醇;
N2将5g端羧基聚乙二醇溶解在20mL二氯甲烷中,再加入40mL二氯亚砜,在25℃反应6h,在55℃减压浓缩至原体积的1/3,加入无水乙醚沉淀,过滤收集沉淀,在50℃干燥得到聚乙二醇酰氯;
N3将10g磷脂酰胆碱、5g聚乙二醇酰氯、4mL三乙胺溶解在80mL三氯甲烷中,在50℃下反应6h,在50℃减压蒸除溶剂,收集残留物,用50mL四氢呋喃溶解沉淀,过滤,收集滤液,在50℃下干燥减压浓缩后得到聚乙二醇枝连磷脂酰胆碱。
实施例2
一种富含脂质体纳米粒的美白敷料的制备方法,包括以下步骤:
M1将30g水、10g脂质体纳米粒、0.5g苹果酸、1g维生素C、2g传明酸、0.5g黄原胶、0.2g乙二胺四乙酸二钠、3g氢化卵磷脂加入搅拌器中,在60℃下,搅拌30min,得到水相;
M2将8g甘油、1g烟酰胺加入搅拌器中,在55℃下,搅拌30min,得到油相;
M3将水相和油相混合后放入均质机中,在压力为150MPa,均质速度为均质4min,得到富含脂质体纳米粒的美白敷料。

S3将A液与B液混合,在50℃下减压浓缩至原混合液体积的2/3,再将混合液超声5min,最后在-45℃下冷冻干燥48h,得到脂质体纳米粒。
所述磷脂为改性磷脂酰胆碱。
所述改性磷脂酰胆碱的制备方法为:将20g磷脂酰胆碱溶解在100mL正己烷中,加入40g硬脂酸、4g脂肪酶,在50℃下反应40h,反应结束后减压蒸除溶剂,收集残留物,用丙酮洗涤3次后,在45℃下干燥24h得到改性磷脂酰胆碱。
实施例4
一种富含脂质体纳米粒的美白敷料的制备方法,包括以下步骤:
M1将30g水、10g脂质体纳米粒、0.5g苹果酸、1g维生素C、2g传明酸、0.5g黄原胶、0.2g乙二胺四乙酸二钠、3g氢化卵磷脂加入搅拌器中,在60℃下,搅拌30min,得到水相;
M2将8g甘油、1g烟酰胺加入搅拌器中,在55℃下,搅拌30min,得到油相;
M3将水相和油相混合后放入均质机中,在压力为150MPa,均质速度为均质4min,得到富含脂质体纳米粒的美白敷料。
所述脂质体纳米粒的制备方法,包括以下步骤:
S1将3g吐温80溶解在600mLpH=8的0.1mol/L磷酸缓冲液中得到A液;
S2将6g磷脂、1g胆固醇、0.8g玫瑰提取物分散在100mL无水乙醇中,在60℃下充分溶解得到B液;
S3将A液与B液混合,在50℃下减压浓缩至原混合液体积的2/3,再将混合液超声5min,最后在-45℃下冷冻干燥48h,得到脂质体纳米粒。
所述磷脂为聚乙二醇枝连磷脂酰胆碱。
所述聚乙二醇枝连磷脂酰胆碱的制备方法,包括以下步骤:
N1将10g聚乙二醇溶解在200mL丙酮中得到聚乙二醇丙酮溶液;将80g氧化铬溶解在500mL水中,加入70mL 98wt%硫酸水溶液得到羧基化试剂;向上述聚乙二醇丙酮溶液中加入5mL羧基化试剂,在25℃反应12h,反应结束后加入20mL异丙醇终止反应,加入50mL水,在55℃旋蒸至原体积的1/2,加入碳酸氢钠至无气泡产生,用1mol/L盐酸调节溶液pH=1,在70℃下真空干燥得到白色固体,用50mL三氯甲烷溶解白色固体,过滤,收集滤液,在50℃下减压浓缩后干燥得到端羧基聚乙二醇;
N2将5g端羧基聚乙二醇溶解在20mL二氯甲烷中,再加入40mL二氯亚砜,在25℃反应6h,在55℃减压浓缩至原体积的1/3,加入无水乙醚沉淀,过滤收集沉淀,在50℃干燥得到聚乙二醇酰氯;
N3将10g磷脂酰胆碱、5g聚乙二醇酰氯、4mL三乙胺溶解在80mL三氯甲烷中,在50℃下反应6h,在50℃减压蒸除溶剂,收集残留物,用50mL四氢呋喃溶解沉淀,过滤,收集滤液,在50℃下减压浓缩后干燥得到聚乙二醇枝连磷脂酰胆碱。
测试例1
美白功效测试:选择60位皮肤健康、无过敏史的女性志愿者(年龄在25岁至60岁之间)参加测试,将志愿者分为4组,分别使用上述实施例1~4制备的美白敷料,每日晚上使用一次,使用方法为取2g美白敷料均匀涂抹在脸上,保持15min,然后再用清水冲洗,连续使用30天,对比其美白效果以及皱纹改善情况,结果如表1所示。

从表1的结果可知,实施例1具有良好的美白去皱效果,实施例2~4相对较差,其原因可能在于,实施例2中使用磷脂酰胆碱作为脂质体纳米粒的主要膜材,其在使用中易被空气中的氧气氧化,导致其结构被破坏,同时由于其粒径较小,易通过疏水作用发生团聚,形成较大的脂质体颗粒影响皮肤对其的吸收,实施例3虽然膜材不易发生氧化,但是同样存在易聚集的问题,实施例4使用聚乙二醇枝连磷脂酰胆碱通过空间位阻可以防止其发生团聚,但是表面的大量聚乙二醇会影响磷脂对皮肤角质层的渗透,从而影响吸收和利用,同时也存在脂肪酸链被氧化,导致内容物泄露的问题,实施例1使用一定质量比的改性磷脂酰胆碱和聚乙二醇枝连磷脂酰胆碱作为膜材,既能保证脂质体结构不因为氧化而破坏,也不会由于聚集而影响皮肤对脂质体纳米粒的吸收,因此其效果最好。同时脂质体纳米粒使皮肤水合作用增强,角质细胞间隙变大,脂质双层中疏水性尾部排列紊乱,可以促进美白敷料中其他有效成分通过扩散和毛细吸力作用进入细胞间隙,脂质体纳米粒穿越角质层,除可以缓释内容的玫瑰提取物外,还能填充角质层,达到改善皱纹的目的。
测试例2
稳定性测试:将实施例1~4制备的美白敷料分别置于4℃和30℃下保存,保存1个月后取样,分别对样品进行10000rpm离心25min,观察样品是否发生分层或有沉淀析出,结果如表2所示。

脂质体纳米粒对于皮肤具有良好的渗透效果,促进皮肤对有效成分的吸收,但是其易被氧化或通过疏水相互作用发生团聚,导致稳定性降低影响产品质量,因此稳定性是评价美白敷料好坏的重要指标。
从表2的结果可知,实施例1在4℃和30℃都具有良好的稳定性,实施例2~4则部分出现分层甚至沉淀析出,其原因可能在于,在4℃下,氧化反应速率降低,分子间热运动速率降低,相对来说比在30℃保存更加稳定,实施例2的稳定性最差,其原因在于其膜材易被氧化释放出内容物,同时也易发生团聚,因此在30℃下保存一个月后离心甚至有沉淀析出,实施例4制备的美白敷料在4℃下保存的稳定性很好,这是由于温度降低,氧化反应速率很低,其脂质体结构不易被氧化破坏,同时表面的聚乙二醇通过空间位阻的作用能有效防止其发生团聚,因此在低温下保存具有良好的稳定性,而在30℃的条件下,氧化反应速率急剧提高,脂质体结构被破坏,稳定性受到影响,离心后出现微分层,实施例1制备的美白敷料在4℃和30℃保存一个月后,离心后均不分层,说明其稳定性很好。
综上所述,本发明制备的富含脂质体纳米粒的美白敷料不仅具有良好的美白效果,还克服了脂质体纳米粒易被氧化、发生团聚的缺点,使得制备的美白敷料同时还具有良好的贮藏稳定性,保证其易被皮肤吸收的效果不受影响,具有良好的美白去皱效果。
Claims (2)
1.一种富含脂质体纳米粒的美白敷料,其特征在于,由以下质量份的组分组成:20~40份水,10~20份脂质体纳米粒,2~8份甘油,0.1~1份苹果酸,0.5~2份维生素C,1~2份传明酸,1~2份烟酰胺,0.2~0.6份黄原胶,0.1~0.5份乙二胺四乙酸二钠,2~5份氢化卵磷脂;
所述脂质体纳米粒的制备方法,包括以下步骤:
S1将1~3g吐温80溶解在200~600mL pH=7~9的0.1~0.2mol/L磷酸缓冲液中得到A液;
S2将2~6g磷脂、0.4~1g胆固醇、0.2~1g玫瑰提取物分散在30~100mL无水乙醇中,在50~70℃下充分溶解得到B液;
S3将A液与B液混合,在40~60℃下减压浓缩至原混合液体积的2/3~4/5,将混合液超声5~10min,最后在-45~-40℃下冷冻干燥24~48h,得到脂质体纳米粒;
所述磷脂为质量比为1~5:1的改性磷脂酰胆碱和聚乙二醇枝连磷脂酰胆碱的混合;
所述改性磷脂酰胆碱的制备方法为:将20~60g磷脂酰胆碱溶解在100~300mL正己烷中,加入40~120g硬脂酸、4~12g脂肪酶,在40~60℃下反应30~50h,反应结束后减压蒸除正己烷,收集残留物,用丙酮洗涤1~3次后,在40~60℃下干燥12~24h得到改性磷脂酰胆碱;
所述聚乙二醇枝连磷脂酰胆碱的制备方法,包括以下步骤:
N1将5~10g聚乙二醇溶解在100~200mL丙酮中得到聚乙二醇丙酮溶液;将50~100g氧化铬溶解在500~1000mL水中,加入50~100mL90~98wt%硫酸水溶液得到羧基化试剂;向上述聚乙二醇丙酮溶液中加入5~10mL羧基化试剂,在20~30℃反应12~24h,反应结束后加入20~40mL异丙醇终止反应,加入50~100mL水,在40~60℃下减压浓缩至原体积的1/3~1/2,加入碳酸氢钠至无气泡产生,用0.1~1mol/L盐酸调节溶液pH=1~3,在60~80℃下真空干燥得到白色固体,用50~100mL三氯甲烷溶解白色固体,过滤,收集滤液,在40~60℃下减压浓缩后干燥得到端羧基聚乙二醇;
N2将2~5g端羧基聚乙二醇溶解在10~20mL二氯甲烷中,加入20~40mL二氯亚砜,在20~30℃反应6~12h,在40~60℃下减压浓缩至原体积的1/3~1/2,加入无水乙醚沉淀,过滤收集沉淀,在40~60℃干燥得到聚乙二醇酰氯;
N3将4~10g磷脂酰胆碱、2~5g聚乙二醇酰氯、3~5mL三乙胺溶解在50~100mL三氯甲烷中,在45~60℃下反应4~10h,在40~60℃下减压蒸除溶剂,收集残留物,用50~100mL四氢呋喃溶解沉淀,过滤,收集滤液,在40~60℃下减压浓缩后干燥得到聚乙二醇枝连磷脂酰胆碱。
2.如权利要求1所述的富含脂质体纳米粒的美白敷料的制备方法,其特征在于,包括以下步骤:
M1按配方称取原料,将水、脂质体纳米粒、苹果酸、维生素C、传明酸、黄原胶、乙二胺四乙酸二钠、氢化卵磷脂混匀得到水相;
M2将甘油、烟酰胺混匀得到油相;
M3将水相和油相混合后均质得到富含脂质体纳米粒的美白敷料。
