技术领域
本发明涉及青蒿提取技术领域,具体为一种青蒿精油的提取及精油抑菌性能研究方法。
背景技术
青蒿精油含有丰富的单萜和倍半萜类,是一种具有药用价值的副产品。现代研究证明青蒿挥发油具有抗菌、抗癌、抗病毒、抗炎、驱蚊、杀虫、抗氧化等活性。主要用途有治疗痤疮、抑制流感病毒、消炎、抗溃疡、局部刺激和强心、解热、止咳、祛痰、平喘和镇痛,并能作为保健食品,还可用于研制驱蚊贴。
青蒿在进行提取时,通过单一提取方法,要不提取时间过长,要不就是提取量较少,较为浪费时间,或者对青蒿进行浪费,而且对细菌进行抑制实验时,均通过一次进行添加,使得实验数据不准。

针对现有技术的不足,本发明提供了一种青蒿精油的提取及精油抑菌性能研究方法,解决了提取浪费时间或提取不多的问题。
(二)技术方案
为实现以上目的,本发明通过以下技术方案予以实现:一种青蒿精油的提取,包括以下步骤:
S1、材料处理
选取重庆涪陵地区9~10月青蒿植株的叶和花作为材料,进行清洗、浸泡,留取待用;
S2、进行蒸煮处理
将浸泡后的青蒿进行粉碎,然后放入蒸煮锅中进行蒸煮,且蒸煮锅上方设置有冷凝管,对蒸馏出的精油进行收集;
S3、进行采油
将蒸煮后的青蒿放入压榨机中进行压榨,然后将压榨后的残渣,放入蒸煮炉中进行蒸馏提纯,然后将三次得到的精油进行混合;
S4、进行提纯
将混合的精油通过过滤膜进行过滤,然后将过滤后的溶液对精油与水进行分离,得到相应的青蒿精油。
优选的,所述S3中压榨的次数为三次,所述蒸馏的时间为2小时。
优选的,所述S4中对精油与水分离的方法为离心法、反渗透膜法、有机溶剂提取法、分液器法中的一种。

S2、对细菌进行培养
将挑选的细菌在其相对的培养基中进行培养,然后对细菌进行配置溶液、稀释,得到相同数量的菌群;
S3、进行实验
将处理后的滤纸放置在培养基中,然后在滤纸的中间孔放置细菌,进行培养,并且每天在滤纸的边缘进行滴入相同质量的青蒿精油,并对细菌的数量进行统计;
S4、进行总结
对细菌的变化进行总结,从而得到相应的抑菌效果。
优选的,所述S1中培养孔的直径为2厘米,所述浸泡的时间为5分钟。
优选的,所述S2中进行培养时的培养条件为置于恒温培养箱中培养,如:细菌在37℃下培养24h,真菌在28℃下培养48~72h,测定抑菌圈直径。
优选的,所述S3中放入空白滤纸作为空白组对照,且每日在滤纸边缘滴入相同体积的水。
