Abstract

The transformation of the potted landscape rape into the sterile line can prolong the flowering time of potted landscape rape flowers, improve the ornamental value, reduce the pollen dispersion in the indoor and scenic spots and protect the health of allergic people.

The breeding method of the bonsai ornamental rape provided by the embodiment of the invention adopts the lodging-resistant rape variety with short full-growth period (180-210 days) as a parent, so that the bred short-stalk variety is easy to vernalize, and the cultivation of the four-season ornamental rape flower can be realized. In addition, the breeding method firstly creates short-stalk short-growth-period mutant plant types through a mutagenesis technology, and then the obtained short-stalk mutant strains are hybridized with rape varieties with different colors and large leaf color petals, so that a plurality of improved varieties can be obtained simultaneously in one cultivation period, and the breeding efficiency is greatly improved.
Example 1
The invention provides a method for breeding potted ornamental rape, which comprises the following steps:
1. creation of short-stalk rape
1.1 mutagenesis of oilseed rape varieties
Selecting double No. 11 in the medium-maturing variety with strong disease resistance and lodging resistance and short full-growth period (180-210 days) as a parent, and performing mutagenesis treatment on the seed of the parent by adopting an EMS solution, wherein the method specifically comprises the following steps: soaking the parent seeds for 8 hours at 30 ℃, and then soaking the parent seeds in 1.5 mass percent Ethyl Methane Sulfonate (EMS) solution for 24 hours;
all mutagenic seed resources are planted in the test field with isolated conditions according to the conventional cultivation technology, and are generally sown in the last 9-10 months. The fertilizer application in the field is carried out according to the principle of heavy base fertilizer application, increased phosphorus-potassium fertilizer application and necessary boron fertilizer application, wherein 10-12kg of pure nitrogen and phosphorus (P) are applied per mu2O5)5-6kg, potassium (K)2O)5-6kg, borax 1.0-1.5 kg; wherein, nitrogen fertilizer is applied at 60% base, and seedling fertilizer and bolting fertilizer are respectively applied at 20%; completely applying the phosphate fertilizer at the bottom; 70% of potassium fertilizer is applied on the bottom, and 30% of potassium fertilizer is used as a bolting fertilizer; 0.9-1.35kg of borax is applied at the bottom, and 150g of leaves at the bolting stage are sprayed. After the base fertilizer is applied to the field, the rape seeder (model: 2BYD-6, no seeds are added) is used for completing the operations of plowing, ditching, compartment making and the like of the test field at one time, the width of the compartment is 1.5m, and the uniform seeding and the tidy seedling emergence are ensured. After flowering, all the single plants are bagged by sulfuric acid paper bags or non-woven fabric bags according to the bagging requirement of rape breeding, and after the seeds are mature, all the single plants M1 generation seeds are harvested.
1.2 Material identification
The harvested M1 generation seeds are all sowed according to the cultivation method, a single plant with the plant height of 30-60cm is selected in the flowering period to be bagged and selfed, and a weak winter early-maturing short-stalk mutation single plant with the height of 30-100cm and the full growth period of less than 180 days is selected in the maturation period to harvest M2 generation seeds.
And (3) carrying out identification on the seeds of the M2 generation in the second year according to the method, selecting a dwarf line with stable phenotype as shown in figure 1, harvesting seeds of the M3 generation, and applying the seeds to the breeding of potted ornamental rape varieties.
2. Polymerization of purple leaf large-petal safflower and dwarf stalk
2.1 selecting purple-leaf large-petal safflower rape variety YC102 and short-stem (30-100cm) rape resource hybridization with short full-growth period (less than 180 days), and continuously backcrossing for 2-3 generations.
2.2 selecting short stalk single plant polymerized with purple leaf, safflower and large petal, selfing for 5-6 generations until breeding into short stalk homozygous plant polymerized with purple leaf, large petal and safflower.
3. Transformation of sterile line
3.1 selecting cytoplasmic sterile plant with large petals (the diameter of the flower is more than 16mm) as female parent, and using the short-stalk homozygous single plant of purple leaf, large petal and safflower obtained in the step 2 as male parent, and making hybridization to obtain hybrid F1 generation sterile plant.
3.2 taking the sterile plant of the hybrid F1 generation as a female parent, taking the short-stalk homozygous single plant of the purple leaves, the big petals and the safflower obtained in the step 2 as a male parent, and hybridizing again for 5 generations until the short-stalk big-flower sterile line of the purple leaves, the big petals and the safflower is selected and bred.
Example 2
The contents are the same as those of example 1 except for the following.
The purple-leaf large-petal white-flower rape variety YC103 and the created short-stalk (30-100cm) rape resource with short full-growth period (less than 180 days) are selected for hybridization.
Example 3
The contents are the same as those of example 1 except for the following.
The green leaf big petal safflower rape variety YC104 is selected to be hybridized with the rape resource which is created with short stalk (30-100cm) and short whole growth period (less than 180 days).
Example 4
The contents are the same as those of example 1 except for the following.
The green-leaf large-petal white-flower rape variety YC105 is selected to be hybridized with the rape resource which is created with short stalk (30-100cm) and short whole growth period (less than 180 days).
The data of the obtained dwarf mutant strain ornamental rape and the Zhongshuang No. 11 parent strain are compared and shown in the table 1.

Claims (10)
1. A method for breeding ornamental rape bonsai is characterized by comprising the following steps:
(1) selecting an anti-lodging rape variety with a full growth period of 180-210 days as a parent, and carrying out mutagenesis treatment on the seed of the parent to obtain a mutagenized seed; planting the mutagenized seeds, selfing the single plants after flowering, and harvesting M1 generation seeds after maturation;
(2) planting the M1 seeds, selecting a single plant with the plant height of 30-60cm for selfing in the flowering period, selecting a single plant with the plant height of 30-100cm in the mature period and the full growth period of less than 180 days, and harvesting M2 seeds;
(3) planting the M2 generation seeds, and harvesting M3 generation seeds according to the method in the step (2);
(4) planting seeds of the M3 generation, hybridizing the seeds with rape varieties with different flower colors and leaf colors and large petals respectively in the flowering period, backcrossing for 2-3 generations to obtain short-stalk single plants with different flower colors and leaf colors and large petals, planting seeds of the short-stalk single plants, selfing for 5-6 generations to obtain short-stalk homozygous lines with different leaf colors and flower colors and large petals.
2. The selective breeding method of claim 1, wherein the parents are double 11 rape or early-maturing rape Ganxiao oil No 1 in the medium-maturing variety.
3. A selective breeding method according to claim 1 or 2, characterized in that the mutagenic treatment is carried out by means of radiation, the radiation source including but not limited to one or more of X-rays, gamma-rays, 60Co gamma-rays, cosmic rays, atmospheric pressure room temperature plasma based on the atmospheric pressure injection frequency glow discharge principle.
4. The breeding method according to claim 1 or 2, characterized in that the mutagenic treatment is carried out using a mutagenizing agent, in particular: soaking the parent seeds for 8 hours at 30 ℃, and then soaking the parent seeds in a solution with the mass percentage of ethyl methanesulfonate of 1.5% for 24 hours.
5. The breeding method according to any one of claims 1 to 4, wherein the mutagenized seeds are planted in a test field with segregating conditions.
6. The breeding method according to claim 5, wherein crucifers cannot be present around the test field.
7. The breeding method according to any one of claims 1 to 6, wherein the rape variety in step (4) is a purple-leaf and large-petal rape variety YC102, a purple-leaf and large-petal white-flower rape variety YC103, a green-leaf and large-petal safflower rape variety YC104, or a green-leaf and large-petal white-flower rape variety YC105, which is bred by crossing the red-flower rape CS3 and the purple-flower rape CS4 provided by Zhejiang academy of agricultural sciences.
8. The breeding method according to any one of claims 1 to 7, further comprising transforming the dwarf homozygous lines with different leaf colors and flower colors and large petals obtained in step (4) into sterile lines.
9. The breeding method of claim 8, wherein the transformation method of the sterile line comprises: taking the short stalk homozygous lines with different leaf colors and flower colors and large petals obtained in the step (4) as male parents, selecting cytoplasmic sterile plants with large petals as female parents, and hybridizing to obtain F1 generation sterile plants; and backcrossing for a plurality of generations by taking the F1 generation sterile plant as a female parent until a short-stalk sterile line with different leaf colors and flower colors and large petals is bred.
10. A potted canola plant comprising canola seeds obtained by the selective breeding method of any one of claims 1-9.
