Abstract
本发明涉及兽用中药注射液技术领域,尤其涉及一种广谱抗病毒中药注射液及其制备方法,包括以下重量份的原料:金银花3‑5份、穿心莲4‑7份、黄芩2‑5份、连翘6‑10份、野菊花2‑6份、大蒜2‑4份、黄芪1‑3份、贯众2‑5份、西洋参0.5‑2份、甘草1‑3份、五味子1‑2份、淫羊藿0.5‑3份。本发明的目的是通过多种中药成份提取后制备的注射液,通过多种中药成份提取后制备的注射液,有广谱抗病毒作用,配方科学合理,且在提取的过程中,分别对寒性和甘性药材进行分别提取,再使用纯化颗粒进行处理,能够有效去除提取物中的杂质和未知颗粒,尽可能保证提取物的纯度,从而提高注射液的稳定性以及减少发生副作用的几率。

本发明涉及兽用中药注射液技术领域,尤其涉及一种广谱抗病毒中药注射液及其制备方法。
背景技术
病毒(virus)是由一个核酸分子(DNA或RNA)与蛋白质构成的非细胞形态的营寄生生活的生命体。由于环境恶化、气候反常、饲料伪劣导致畜禽多病原微生物的易感性增加,各种疾病的发生及危害也日益严重,并已成为严重影响养殖业发展和经济效益提升的主要因素。畜禽疾病的频发,不仅会造成畜禽大批死亡,造成养殖户的直接损失,且死亡的畜禽需要焚烧,进一步造成资源浪费。
目前,在畜禽防治疾病过程中,主要是使用大量抗生素、激素及化学合成药物。但使用大量的抗生素、激素及化学合成药物容易导致药物残留,影响畜禽的出栏品质,同时,在抗生素、激素等使用期间,也会造成畜禽食欲不振,体重下降。
发明内容
有鉴于此,本发明的目的是提供一种广谱抗病毒中药注射液及其制备方法,通过多种中药成份提取后制备的注射液,有广谱抗病毒作用,配方科学合理,且在提取的过程中,分别对寒性和甘性药材进行分别提取,再使用纯化颗粒进行处理,能够有效去除提取物中的杂质和未知颗粒,尽可能保证提取物的纯度,从而提高注射液的稳定性以及减少发生副作用的几率。
本发明通过以下技术手段解决上述技术问题:
一种广谱抗病毒中药注射液,包括以下重量份的原料:金银花3-5份、穿心莲4-7份、黄芩2-5份、连翘6-10份、野菊花2-6份、大蒜2-4份、黄芪1-3份、贯众2-5份、西洋参0.5-2份、甘草1-3份、五味子1-2份、淫羊藿0.5-3份。
进一步,包括以下重量份的原料:金银花4份、穿心莲5.5份、黄芩3.5份、连翘8份、野菊花4份、大蒜3份、黄芪2份、贯众3.5份、西洋参1.2份、甘草2份、五味子1.5份、淫羊藿1.5份。
金银花:性甘、寒,味芳香,甘寒泄降,清热解毒,芳香疏泄为花主散,为外解内清之良药;
穿心莲:味苦、性寒,具有清热解毒,凉血消肿的效果;
黄芩:性苦、寒,具有清热燥湿,泻火解毒;
连翘:性凉、味苦,具有清热,解毒,散结,消肿的效果,是一味具有广谱抗病毒作用的中草药;
野菊花:味苦、微寒,具有清热解毒,泻火平肝的效果,与金银花合用,可促进外解内和,具有排毒解热的功效;
大蒜:具有广谱抗菌灭菌和消炎作用,有很强的抗病毒能力,能够提高免疫功能,活化细胞,促进能量产生,加快新陈代谢,扩张血管,改善血液循环,缓解疲劳;
黄芪:味甘,性微温,可直接阻断病毒的核酸合成,从而杀灭病毒;同时具有免疫增强作用,可促进抗体的形成,增强巨噬细胞对病原体的吞噬作用,进而杀灭病原体,作用迅速持久,与黄芩合用,可促进内毒的分解、排出;
贯众:味苦、涩,性寒,具有杀虫、清热、解毒、凉血止血的效果;
甘草:味甘,性平,具有补脾益气、清热解毒、调和诸药的效果;
五味子:味酸、甘,性温,具有收敛固涩,益气生津,补肾宁心的效果;
淫羊藿:性辛、甘、温;具有补肾阳、强筋骨、增强免疫功能、抑菌、抗炎、抗病毒的效果;
西洋参:具有抗疲劳、调节内分泌、增强免疫力的功效;
本发明中注射液采用纯中药配方,诸药按君臣佐使原则配伍,安全可靠、无毒副作用,配方简单、材料易得,诸药合用可起到广谱杀菌、抗病毒的效果,同时,能够起到滋阴补阳,调和表里,提高畜禽的机体免疫力,促进畜禽的健康生长。
本发明还公开了一种广谱抗病毒中药注射液的制备方法,包括以下步骤:
S1:将金银花、穿心莲、黄芩、连翘、野菊花和贯众按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干,阴干后置于提取器中,加入碱性处理液,进行浸渍提取,浸渍提取过程中,温度保持在32-45℃,得到提取液一,对提取液一进行过滤,备用;
S2:将大蒜、黄芪、西洋参、甘草、五味子和淫羊藿按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干,阴干后置于提取器中,加入酸性处理剂和乙醇,进行浸渍提取,得到提取液二,对提取液二进行过滤,备用;
S3:将S1步骤和S2步骤中浸渍提取后的药材用去离子水清洗3-5次,清洗后烘干,烘干后粉碎,将粉碎后的药材置于提取器中,加入80-90%的乙醇和纯化颗粒,进行重渗漉提取,得到提取液三;
S4:将提取液一、提取液二和提取液三混合,再次加入纯化颗粒,静置10-24H,再将纯化颗粒去除,调节PH、瞬间冷冻,然后加入苯甲醇和注射用水进行灌封、灭菌得到注射液。
通过在中药材的提取过程中,对寒性药材进行碱性处理液浸泡后,再进行浸渍提取,一方面,碱性处理液能够增加寒性药材的细胞壁通透性,另一方面,能够使寒性药材中的生物酸充分提取,从而充分得到寒性的有效成分的提取液一;对性平及性甘的药材通过酸性处理剂以及乙醇进行浸渍提取,一方面,能够增强性平及性甘的药材的细胞壁通透性,另一方面,能够使性平及性甘的药材中的生物碱充分提取,进而得到甘性的有效成分的提取液二;再将两者药材混合,通过重渗漉提取,将所用中药材中的有效成分完全提取,并采用纯化颗粒对提取液三进行纯化,去除提取液三中的杂质和未知颗粒,再将三种提取液混合后,再次纯化,尽可能保证提取液的纯度,从而提高注射液的稳定性以及减少发生副作用的几率,再通过对提取液进行瞬间冷冻处理,降低提取液中有效成分的活性,避免在未使用时,失去活性。
进一步,所述S1步骤中,碱性处理液包括氨水、丙酮和水,所述氨水、丙酮和水的质量比为:5:80:15,过滤时,采用微滤膜过滤,过滤压力为0.5-1.5Mpa。
通过控制氨水、丙酮和水的比例,形成碱性缓冲体系,加快提取效率。通过采用加压的方式进行微滤,将提取液一种的杂质和位置颗粒去除。
进一步,所述S2步骤中,酸性处理剂包括:苹果酸、乙酸和蒙脱土,所述酸性处理剂的制备包括以下步骤:
S201:将蒙脱土置于去离子水中,加入0.2mol/L的稀盐酸,超声波分散5-10min,分散完成后、抽滤、干燥,得到去除杂质的蒙脱土;
S202:将经过S201步骤处理的蒙脱土置于去离子水中,加入十二烷基硫酸钠,超声波分散5-10min,加入苹果酸和乙酸,于35-50℃的条件下,继续超声波分散3-5min,抽滤、冷冻干燥,得到中间体;
S203:将S202步骤中制备的中间体制备成颗粒状,将颗粒状中间体置于带孔的胶囊皮中,得到酸性处理剂。
通过将蒙脱土除杂,再将苹果酸和乙酸插接在蒙脱土的间层中,再将蒙脱土制备成颗粒状,并用胶囊包覆,一方面,使蒙脱土可持续性释放酸性物质,另一方面,使蒙脱土能够重新吸附酸性物质,节省成本,且还能吸附中药材中存在的重金属离子,减轻中药材中提取液的重金属含量。
进一步,所述S203步骤中,中间体经过挤压制备成颗粒状,挤压的压力为3-5Mpa,所述颗粒状中间体的粒径为20-30mm,所述胶囊皮为无毒的聚乙烯塑料,所述胶囊皮上的孔径为1-3mm。
通过将中间体制备成颗粒状,在用带孔的胶囊皮将其包覆,便于酸性处理剂与中药材分离。
进一步,所述S3步骤中的纯化颗粒的制备包括以下步骤:将沸石置于稀盐酸溶液中,超声波震荡1-3min,超声波震荡后,取出沸石,钻孔,再机械打磨成颗粒状,将颗粒状沸石置于清水中,超声波震荡1-3min,超声波完成后,干燥,得到纯化颗粒。
通过对沸石的孔隙进行酸修饰,酸修饰后再进行钻孔,扩大孔径,使沸石能够充分吸附提取液中的杂质和未知颗粒,提高提取液的纯度,且沸石可再生,节约资源。
进一步,所述纯化颗粒的制备过程中,钻孔采用机械钻孔,孔数大于3,孔径为0.2-2um。
进一步,所述S3步骤中,粉碎后药材的粒径为200-320目,所述纯化颗粒的粒径为400-500目。
进一步,所述S4步骤中,采用氢氧化钾调节PH,所述PH值的范围为6.2-6.8。
本发明的有益效果:通过采用纯中药配方,并将中药提取形成注射液,使注射液可起到广谱杀菌、抗病毒的效果,同时,能够起到滋阴补阳,调和表里,提高畜禽的机体免疫力,促进畜禽的健康生长。在注射液的提取过程中,通过分别对不同性质的药材进行处理,一方面,能够充分提取药材中的有效成分,另一方面,提高提取效率,同时,采用纯化颗粒对提取液进行纯化,去除提取物中的杂质和未知颗粒,尽可能保证提取物的纯度,从而提高注射液的稳定性以及减少发生副作用的几率,并对纯化的提取液进行瞬间冷冻,降低提取液中有效成分的活性,避免在未使用时,失去活性。
具体实施方式
以下将结合具体实施例对本发明进行详细说明:
实施例1,广谱抗病毒中药注射液的制备一
包括以下重量份的原料:金银花3份、穿心莲4份、黄芩2份、连翘6份、野菊花2份、大蒜2份、黄芪1份、贯众2份、西洋参0.5份、甘草1份、五味子1份、淫羊藿0.5份。
其中,酸性处理剂的制备包括以下步骤:
S201:将10kg蒙脱土置于去离子水中,加入足量0.2mol/L的稀盐酸,超声波以80KHZ的功率分散3-5min,分散完成后、抽滤、干燥,得到去除杂质的蒙脱土;
S202:将经过S201步骤处理的10kg蒙脱土置于去离子水中,加入1.8kg十二烷基硫酸钠,超声波以120KHZ的功率分散5-10min,加入2.5L苹果酸和1.5L乙酸,于35-50℃的条件下,超声波以120KHZ的功率继续分散3-5min,抽滤、冷冻干燥,得到中间体;
S203:将S202步骤中制备的中间体经过挤压设备以3-5Mpa的压力挤压成粒径为20-30mm的颗粒,将颗粒状中间体置于的聚乙烯胶囊皮中,聚乙烯胶囊皮上通过钻孔,得到大量孔洞,孔洞的直径为1-3mm,从而得到酸性处理剂。
S1:将3重量份的金银花、4重量份的穿心莲、2质量份的黄芩、6重量份的连翘、2重量份的野菊花和2重量份的贯众按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干,阴干后置于提取器中,加入50重量份的氨水、丙酮和水的混合液,于32℃的条件下进行浸渍提取,得到提取液一,对提取液一进行过滤,过滤时,采用微滤膜进行过滤,过滤压力为0.5Mpa,得到去除杂质的提取液一,备用;
S2:将1重量份的黄芪、2重量份的大蒜、0.5重量份的西洋参、1重量份的甘草、1重量份的五味子和0.5重量份的淫羊藿按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干;阴干后置于提取器中,加入5重量份的酸性处理剂和足量的95wt%的乙醇,进行浸渍提取,得到提取液二,对提取液二进行过滤,过滤时,采用微滤膜进行过滤,过滤压力为0.5Mpa,得到去除杂质的提取液二,备用;
S3:将S1步骤和S2步骤中浸渍提取后的全部中药材采用去离子水清洗3-5次,清洗后烘干,烘干后机械粉碎,粉碎至200目,将粉碎后的药材置于提取器中,加入80-90%的乙醇和纯化颗粒,进行重渗漉提取,得到提取液三;其中,纯化颗粒采用沸石颗粒,沸石颗粒在制备时,将沸石置于0.2mol/ml的稀盐酸溶液中,超声波以20KHZ的频率震荡1min,超声波震荡后,取出沸石,机械钻孔,孔数随意分布在沸石表面,且为4个,孔径为2um,再机械打磨成粒径为400目的颗粒状,将颗粒状沸石置于清水中,超声波以25KHZ的频率震荡1min,超声波完成后,干燥,得到纯化颗粒;
S4:将S1步骤制备的提取液一、S2步骤制备的提取液二和S3步骤制备的提取液三混合均匀,再次加入足量的纯化颗粒,静置10-24H,静置完成后,将纯化颗粒捞出,采用氢氧化钾调节PH,使混合提取液的PH为6.2,然后将混合提取液瞬间冷冻至3℃,然后加入苯甲醇和注射用水进行灌封、灭菌得到注射液。
实施例2,广谱抗病毒中药注射液的制备二
包括以下重量份的原料:金银花4份、穿心莲5.5份、黄芩3.5份、连翘8份、野菊花4份、大蒜3份、黄芪2份、贯众3.5份、西洋参1.2份、甘草2份、五味子1.5份、淫羊藿1.5份。
其中,酸性处理剂的制备步骤与实施例1中的制备步骤相同。
S1:将4重量份的金银花、5.5重量份的穿心莲、3.5质量份的黄芩、8重量份的连翘、4重量份的野菊花和3.5量份的贯众按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干,阴干后置于提取器中,加入80重量份的氨水、丙酮和水的混合液,于40℃的条件下进行浸渍提取,得到提取液一,对提取液一进行过滤,过滤时,采用微滤膜进行过滤,过滤压力为1Mpa,得到去除杂质的提取液一,备用;
S2:将2重量份的黄芪、3重量份的大蒜、1.2重量份的西洋参、2重量份的甘草、1.5重量份的五味子和1.5重量份的淫羊藿按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干后后置于提取器中,加入8重量份的酸性处理剂和足量的95wt%的乙醇,进行浸渍提取,得到提取液二,对提取液二进行过滤,过滤时,采用微滤膜进行过滤,过滤压力为1Mpa,得到去除杂质的提取液二,备用;
S3:将S1步骤和S2步骤中浸渍提取后的全部中药材采用去离子水清洗3-5次,清洗后烘干,烘干后机械粉碎,粉碎至260目,将粉碎后的药材置于提取器中,加入80-90%的乙醇和纯化颗粒,进行重渗漉提取,得到提取液三;其中,纯化颗粒采用沸石颗粒,沸石颗粒在制备时,将沸石置于0.2mol/ml的稀盐酸溶液中,超声波以20KHZ的频率震荡2min,超声波震荡后,取出沸石,机械钻孔,孔数随意分布在沸石表面,且为6个,孔径为1um,再机械打磨成粒径为450目的颗粒状,将颗粒状沸石置于清水中,超声波以25KHZ的频率震荡2min,超声波完成后,干燥,得到纯化颗粒;
S4:将S1步骤制备的提取液一、S2步骤制备的提取液二和S3步骤制备的提取液三混合均匀,再次加入足量的纯化颗粒,静置10-24H,静置完成后,将纯化颗粒捞出,采用氢氧化钾调节PH,使混合提取液的PH为6.5,然后将混合提取液瞬间冷冻至3℃,然后加入苯甲醇和注射用水进行灌封、灭菌得到注射液。
实施例3,广谱抗病毒中药注射液的制备三
包括以下重量份的原料:金银花5份、穿心莲7份、黄芩5份、连翘10份、野菊花6份、大蒜4份、黄芪3份、贯众5份、西洋参2份、甘草3份、五味子2份、淫羊藿3份。
其中,酸性处理剂的制备步骤与实施例1中的制备步骤相同。
S1:将5重量份的金银花、7重量份的穿心莲、5质量份的黄芩、10重量份的连翘、6重量份的野菊花和5量份的贯众,按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干,阴干后置于提取器中,加入100重量份的氨水、丙酮和水的混合液,于50℃的条件下进行浸渍提取,得到提取液一,对提取液一进行过滤,过滤时,采用微滤膜进行过滤,过滤压力为1.5Mpa,得到去除杂质的提取液一,备用;
S2:将3重量份的黄芪、4重量份的大蒜、2重量份的西洋参、3重量份的甘草、2重量份的五味子和3重量份的淫羊藿按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干;阴干后置于提取器中,加入10重量份的酸性处理剂和足量的95wt%的乙醇,进行浸渍提取,得到提取液二,对提取液二进行过滤,过滤时,采用微滤膜进行过滤,过滤压力为1.5Mpa,得到去除杂质的提取液二,备用;
S3:将S1步骤和S2步骤中浸渍提取后的全部中药材采用去离子水清洗3-5次,清洗后烘干,烘干后机械粉碎,粉碎至320目,将粉碎后的药材置于提取器中,加入80-90%的乙醇和纯化颗粒,进行重渗漉提取,得到提取液三;其中,纯化颗粒采用沸石颗粒,沸石颗粒在制备时,将沸石置于0.2mol/ml的稀盐酸溶液中,超声波以20KHZ的频率震荡3min,超声波震荡后,取出沸石,机械钻孔,孔数随意分布在沸石表面,且为8个,孔径为0.2um,再机械打磨成粒径为500目的颗粒状,将颗粒状沸石置于清水中,超声波以25KHZ的频率震荡3min,超声波完成后,干燥,得到纯化颗粒;
S4:将S1步骤制备的提取液一、S2步骤制备的提取液二和S3步骤制备的提取液三混合均匀,再次加入足量的纯化颗粒,静置10-24H,静置完成后,将纯化颗粒捞出,采用氢氧化钾调节PH,使混合提取液的PH为6.8,然后将混合提取液瞬间冷冻至3℃,然后加入苯甲醇和注射用水进行灌封、灭菌得到注射液。
实施例4(对比实施例),广谱抗病毒中药注射液的制备四
包括以下重量份的原料:金银花5份、穿心莲7份、黄芩5份、连翘10份、野菊花6份、大蒜4份、黄芪3份、贯众5份、西洋参2份、甘草3份、五味子2份、淫羊藿3份。
S1:将3重量份的金银花、4重量份的穿心莲、2质量份的黄芩、6重量份的连翘、2重量份的野菊花、1重量份的黄芪、2重量份的贯众、2重量份的大蒜、0.5重量份的西洋参、1重量份的甘草、1重量份的五味子和0.5重量份的淫羊藿按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干;
S2:将S1步骤中的中药材置于提取器中,加入70-85%的乙醇,于35℃的条件下,进行浸渍提取,得到提取液一,对提取液一进行过滤,过滤时,采用微滤膜进行过滤,过滤压力为0.5Mpa,得到去除杂质的提取液一,备用;
S3:将S2步骤中浸渍提取后的全部中药材采用去离子水清洗3-5次,清洗后烘干,烘干后机械粉碎,粉碎至200目,将粉碎后的药材置于提取器中,加入80-90%的乙醇进行重渗漉提取,得到提取液二;
S4:将S2步骤制备的提取液一和S3步骤制备的提取液二混合均匀,在混合过程中,采用氢氧化钾调节PH,使混合提取液的PH为6.2,然后加入苯甲醇和注射用水进行灌封、灭菌得到注射液。
利用实施例2所得兽用中药抗病毒注射液进行药效试验,具体过程如下:
1、试验药物:上述处方所述的注射液,规格:10ml。
2、对照药物:黄芪多糖注射液,规格:10ml:1g黄芪多糖,批号20211026,由诸城市华特兽药有限公司生产;实施例4制备出的注射液,规格:10ml;
3、供试动物:市郊猪场20-30日乳猪,体重4-8kg,常规饲养,母猪饲料为不含抗菌药物的全价饲料,仔猪均有自然发病状况,出现激烈咳嗽、眼鼻有黏液、体温升高、腹泻,发热,精神沉郁,食欲下降,耳部和体表皮肤发紫症状等症状,初步诊断为圆环病毒、伪狂犬病、链球菌等引起的细菌与病毒混合感染的急性传染病。
4、试验方法:80头患病仔猪,随机分成4组,每组20头,分别为试验组、试验对照组1、试验对照组2和空白对照组。
5、治疗方法:试验组:肌肉注射本发明产品,0.1ml/kg体重,一日一次,连用5天;试验对照组1和试验对照组2均采用0.1ml/kg体重,一日一次,连用5天;空白对照组不用药。
疗效指标
死亡率:凡在试验期间,出现上述发病症状,剖检有典型病变特征,并分离出相应病原菌者,治疗无效,判定感染死亡,根据死亡头数算出死亡率。
有效率:凡在试验期间,经用药后临床症状明显减轻至消失,体重较用药前增加,剖杀后无可见的病理变化,分离无相应病原菌者,判为有效,按此计算有效率。
试验结果:
试验组应用实施例2所得兽用中药抗病毒注射液紧急处理后,发病猪只死亡2头,3头明显好转,其余15头治愈,有效率达90%,治愈率达75%,试验对照组1发病猪死亡5头,4头明显好转,其余11头治愈,有效率达75%,治愈率达55%,试验对照组2发病猪死亡4头,4头明显好转,其余12头治愈,有效率达80%,治愈率达60%,空白对照组发病猪死亡16头死亡,死亡率达到80%。具体见表1:
由表1可见,试验组因注射实施例2所得兽用中药抗病毒注射液,对圆环病毒、伪狂犬病与链球菌混合感染的临床疗效确定,其效果明显优于试验对照组1的药物,与对照组1的药物相比差异极显著(P<0.01)。
在死亡率及有效率上,注射本发明抗病毒注射液的试验组与对照组1相比,有效率高于对照组1,差异显著(p<0.06)。
由试验组和试验对照组2中的数据可知,通过对本方案中的中药材按照药材的寒性以及甘性分别进行处理,再通过对提取液进行纯化处理,能够得到药效更充足的注射液,说明本方案中按照药材的寒性以及甘性分别进行处理,再通过对提取液进行纯化处理能够提升注射液的稳定性和药效。
经上述药物临床试验表明,本发明中药抗病毒注射液对猪圆环病毒、伪狂犬病等引起的毒混合感染的急性传染病有良好的治疗效果,临床推荐剂量:每kg体重肌肉注射0.1ml。
以上实施例仅用以说明本发明的技术方案而非限制,尽管参照较佳实施例对本发明进行了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的宗旨和范围,其均应涵盖在本发明的权利要求范围当中。本发明未详细描述的技术、形状、构造部分均为公知技术。
Claims (10)
1.一种广谱抗病毒中药注射液,其特征在于,包括以下重量份的原料:金银花3-5份、穿心莲4-7份、黄芩2-5份、连翘6-10份、野菊花2-6份、大蒜2-4份、黄芪1-3份、贯众2-5份、西洋参0.5-2份、甘草1-3份、五味子1-2份、淫羊藿0.5-3份。
2.根据权利要求1所述的一种广谱抗病毒中药注射液,其特征在于,包括以下重量份的原料:金银花4份、穿心莲5.5份、黄芩3.5份、连翘8份、野菊花4份、大蒜3份、黄芪2份、贯众3.5份、西洋参1.2份、甘草2份、五味子1.5份、淫羊藿1.5份。
3.根据权利要求1-2任一所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,包括以下步骤:
S1:将金银花、穿心莲、黄芩、连翘、野菊花和贯众按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干,阴干后置于提取器中,加入碱性处理液,进行浸渍提取,浸渍提取过程中,温度保持在32-45℃,得到提取液一,对提取液一进行过滤,备用;
S2:将大蒜、黄芪、西洋参、甘草、五味子和淫羊藿按照比例置于清水中,清洗并去除杂质,烘干,烘干后,阴干,阴干后置于提取器中,加入酸性处理剂和乙醇,进行浸渍提取,得到提取液二,对提取液二进行过滤,备用;
S3:将S1步骤和S2步骤中浸渍提取后的药材用去离子水清洗3-5次,清洗后烘干,烘干后粉碎,将粉碎后的药材置于提取器中,加入80-90%的乙醇和纯化颗粒,进行重渗漉提取,得到提取液三;
S4:将提取液一、提取液二和提取液三混合,再次加入纯化颗粒,静置10-24H,再将纯化颗粒去除,调节PH、瞬间冷冻,然后加入苯甲醇和注射用水进行灌封、灭菌得到注射液。
4.根据权利要求3所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,所述S1步骤中,碱性处理液包括氨水、丙酮和水,所述氨水、丙酮和水的质量比为:5:80:15,过滤时,采用微滤膜过滤,过滤压力为0.5-1.5Mpa。
5.根据权利要求4所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,所述S2步骤中,酸性处理剂包括:苹果酸、乙酸和蒙脱土,所述酸性处理剂的制备包括以下步骤:
S201:将蒙脱土置于去离子水中,加入0.2mol/L的稀盐酸,超声波分散5-10min,分散完成后、抽滤、干燥,得到去除杂质的蒙脱土;
S202:将经过S201步骤处理的蒙脱土置于去离子水中,加入十二烷基硫酸钠,超声波分散5-10min,加入苹果酸和乙酸,于35-50℃的条件下,继续超声波分散3-5min,抽滤、冷冻干燥,得到中间体;
S203:将S202步骤中制备的中间体制备成颗粒状,将颗粒状中间体置于带孔的胶囊皮中,得到酸性处理剂。
6.根据权利要求5所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,所述S203步骤中,中间体经过挤压制备成颗粒状,挤压的压力为3-5Mpa,所述颗粒状中间体的粒径为20-30mm,所述胶囊皮为无毒的聚乙烯塑料,所述胶囊皮上的孔径为1-3mm。
7.根据权利要求6所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,所述S3步骤中的纯化颗粒的制备包括以下步骤:
将沸石置于稀盐酸溶液中,超声波震荡1-3min,超声波震荡后,取出沸石,钻孔,再机械打磨成颗粒状,将颗粒状沸石置于清水中,超声波震荡1-3min,超声波完成后,干燥,得到纯化颗粒。
8.根据权利要求7所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,所述纯化颗粒的制备过程中,钻孔采用机械钻孔,孔数大于3,孔径为0.2-2um。
9.根据权利要求8所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,所述S3步骤中,粉碎后药材的粒径为200-320目,所述纯化颗粒的粒径为400-500目。
10.根据权利要求9所述的一种广谱抗病毒中药注射液的制备方法,其特征在于,所述S4步骤中,采用氢氧化钾调节PH,所述PH值的范围为6.2-6.8。
