Abstract
本发明公开了一种复合藻粉片及其制备方法,原料包括螺旋藻粉0‑100份、小球藻粉0‑100份、裸藻粉0‑100份、雨生红球藻粉0‑100份、盐藻粉0‑100份或迦得拟微绿球藻粉0‑100份中的两种或多种藻粉的组合物。本发明产品具有抗肿瘤、调节血脂、增强人体免疫力、抗氧化等多种作用。本发明的藻片包含多种营养物质,弥补单一藻片营养不足的弱点,制得的藻片营养成分高、方便食用、便于保存、利于吸收,具有良好的保健效果。

技术领域
本发明涉及藻类及加工工艺,特别涉及一种复合藻粉片及其制备方法。
背景技术
随着社会的发展,人们对营养健康生活的追求逐渐提高,膳食成分和饮食结构通常对人们的健康颇有影响。微藻是海洋食物链的基础,具有营养丰富、医药价值高、易大规模培养等特点。
螺旋藻系颤藻科、螺旋藻属,螺旋藻富含蛋白质、藻蓝蛋白、多种维生素等多种营养物质,被联合国粮农组织誉为“21世纪最佳的理想食物”,同时被世界卫生组织评为“21世纪的最佳保健品”,螺旋藻多糖能增强人体免疫力、抗肿瘤的作用,不饱和脂肪酸能够降低胆固醇、抗疲劳。小球藻是绿藻门、小球藻属,蛋白核小球藻富含蛋白质,包含多种人体必需氨基酸、小球藻多糖、脂肪酸、小球藻生长因子,具有降血压、降血脂、增强免疫力、抗氧化,作为食品添加剂具有广阔的前景。裸藻没有细胞壁,光合作用强,具有59种人体每天必须的营养元素,裸藻多糖的多孔结构可以吸附人体中的胆固醇、重金属、酒精,具有抗病毒、清除自由基的作用。雨生红球藻被认为自然界生产天然虾青素最好的生物之一,虾青素的抗氧化功能是维生素E的500倍,在化妆品和保健品具有良好的发展前景。盐藻目前发现的唯一能在高浓度盐水中生存的奇特生命,由于富含独特而丰富的生命元素,被世界科学界誉为“细胞的动力源”,具有预防和治疗冠心病、糖尿病、癌症等作用。紫菜在我国食用历史较长,且其含碘量高、富含胆碱、钙、铁,具有提高免疫功能、抗衰老等作用。菊芋又名洋姜,其地下块茎富含淀粉、菊糖含量高,有较好的食用价值。
藻片是微藻产品形式之一,目前大部分藻片为一种藻种加以辅料制得所制得,有效成分低、利用率差,且一些保健品作用单一、毒副作用大、不宜服用,寻找综合高效的保健品是研究的热点。
发明内容
发明目的:本发明目的是提供一种具有营养价值高、增强免疫力、抗氧化等功能的复合藻粉片。
本发明另一目的是提供所述复合藻粉片的制备方法。
技术方案:所述的复合藻粉片,原料包括螺旋藻粉0-100份、小球藻粉0-100份、裸藻粉0-100份、雨生红球藻粉0-100份、盐藻粉0-100份或迦得拟微绿球藻粉0-100份中的两种或多种藻粉的组合物。
进一步地,所述螺旋藻粉包括钝顶螺旋藻粉、极大螺旋藻粉或盐泽螺旋藻粉;小球藻粉包括蛋白核小球藻粉、普通小球藻粉或原壳小球藻粉;盐藻粉包括杜氏盐藻粉。还
进一步地,包括辅料:菊芋粉0-100份、紫菜粉0-100份、紫球藻粉0-100份、异麦芽酮糖0-10份、菊芋多糖0-10份、刺参低聚肽0-10份、虾肽螯合钙0-10份、改善口感或鲜味的食品添加剂0-10份、微晶纤维素0-10份、维生素0-5份、羧甲基纤维素0-4份、硬脂酸镁0-4份或二氧化硅0-4份。

所述的复合藻粉片的制备方法,按照配比称量原辅料,混合后加入压片机进行压片得到复合藻粉片。
有益效果:本发明与现有技术相比,具有如下优势:本发明产品具有抗肿瘤、调节血脂、增强人体免疫力、抗氧化等多种作用。本发明的藻片包含多种营养物质,弥补单一藻片营养不足的弱点,制得的藻片营养成分高、方便食用、便于保存、利于吸收,具有良好的保健效果。

具体实施方式
实施例1
本实施例的复合藻粉片及其制备方法,按照重量份进行配置:
钝顶螺旋藻粉49份、蛋白核小球藻粉49份、二氧化硅2份。
具体制备方法:
按照上述要求配比称取质量合格的钝顶螺旋藻粉49份、蛋白核小球藻粉49份,加入二氧化硅混合1h,待均匀后倒入压片机进行压片得到复合藻粉片。
实施例2
本实施例的复合藻粉片及其制备方法,按照重量份进行配置:
钝顶螺旋藻粉49份、裸藻粉49份、二氧化硅2份。
具体制备方法:
按照上述要求配比称取质量合格的钝顶螺旋藻粉49份、裸藻粉49份,加入二氧化硅混合1h,待均匀后倒入压片机进行压片得到复合藻粉片。
实施例3
本实施例的复合藻粉片及其制备方法,按照重量份进行配置:
蛋白核小球藻粉49份、裸藻粉49份、二氧化硅2份。
具体制备方法:
按照上述要求配比称取质量合格的蛋白核小球藻粉49份、裸藻粉49份,向两种藻粉分别加入2份二氧化硅搅拌1h,待均匀后依次加入压片机进行压片,得到双色藻粉片。
实施例4
本实施例的复合藻粉片及其制备方法,按照重量份进行配置:
钝顶螺旋藻粉45份、蛋白核小球藻粉45份、微晶纤维素8份、羧甲基纤维素1.6份、硬脂酸镁0.4份。
具体制备方法:
按照上述要求配比称取质量合格的钝顶螺旋藻粉45份、蛋白核小球藻粉45份,加入微晶纤维素8份、羧甲基纤维素1.6份、硬脂酸镁0.4份。混合1h,待均匀后倒入压片机进行压片得到复合藻粉片。
以上仅为本发明的优选实施案例,并不限制本发明保护的范围本。发明中所提及的藻粉均可根据实际需求进行组合制作复合藻粉片,在本发明的基础上,本领域的技术人员所做的任何非实质性更改均属于本发明所要求的保护的范围。
实施例5混合后的藻粉提取物与单独的藻粉提取物活性对比
(1)活性物质提取方法:
钝顶螺旋藻、蛋白核小球藻、雨生红球藻、裸藻的提取物分别用X、Q、Y、L来表示,其中QX表示钝顶螺旋藻粉和蛋白核小球藻粉1∶1混合后的提取物,QLY为蛋白核小球藻、裸藻和雨生红球藻1∶1∶1混合后的提取物,其它组合同上,均为两种或三种藻粉的等份混合物,11组样品均用二氯甲烷与甲醇混合溶剂提取,并用超声波细胞破碎仪Sicenta-IID在600W下超声破碎30分钟。
(2)活性筛选方法:基于人结肠癌细胞HCT116和肝癌细胞HepG2的存活率检测
本研究采用基于人结肠癌细胞HCT116和肝癌细胞HepG2进行抗肿瘤药物的筛选,具有快速、重复性高等特点。HCT116、HepG2都是癌细胞,具有癌细胞的基本特征,通过测定细胞的存活率来检测化合物的杀肿瘤作用,加入不同浓度的化合物,检测化合物对细胞活性的影响。
将人结肠癌细胞HCT116和肝癌细胞HepG2接种在96孔板中,每孔细胞密度为2×103个,将孔板放进培养箱中培养。培养24h,然后加入含化合物稀释液的培养基,总体积为200μl(保留3~6个孔仍为纯培养基)。培养48h后,每孔加入20μl的MTT溶液(4-5mg/m1),培养4h,取出96孔板离心,待离心结束后,吸出96孔板中的培养基,加入200μl的DMSO,振荡(350r×5-10min)溶解板底紫色固体,酶联免疫检测仪在570nm处检测。所得结果利用GraphPad Prism软件处理。每个化合物浓度均设3个复孔。
(3)筛选结果

其中,钝顶螺旋藻粉和蛋白核小球藻粉1∶1混合后的提取物QX对人结肠癌细胞HCT116的抑制率比单独的钝顶螺旋藻粉提取物(X)、蛋白核小球藻粉提取物(Q)分别高出12.7%、8.9%;裸藻粉和钝顶螺旋藻粉1∶1混合后的提取物LX对人结肠癌细胞HCT116的抑制率比单独的钝顶螺旋藻粉提取物(X)、裸藻粉提取物(L)分别高出7.3%、17.2%;裸藻粉和蛋白核小球藻粉1∶1混合后的提取物QL对人结肠癌细胞HCT116的抑制率比单独的裸藻粉提取物(L)高出13.8%。

其中,钝顶螺旋藻粉和蛋白核小球藻粉1∶1混合后的提取物QX对肝癌细胞HepG2的抑制率比单独的钝顶螺旋藻粉提取物(X)、蛋白核小球藻粉提取物(Q)分别高出17.4%、28.1%;裸藻粉和钝顶螺旋藻粉1∶1混合后的提取物LX对肝癌细胞HepG2的抑制率比单独的钝顶螺旋藻粉提取物(X)、裸藻粉提取物(L)分别高出7.3%、24%;裸藻粉和蛋白核小球藻粉1∶1混合后的提取物QL对肝癌细胞HepG2的抑制率比单独的裸藻粉提取物(L)、蛋白核小球藻粉提取物(Q)分别高出15.5%、9.5%;蛋白核小球藻粉和雨生红球藻粉1∶1混合后的提取物QY对肝癌细胞HepG2的抑制率比单独的雨生红球藻粉提取物(Y)、蛋白核小球藻粉提取物(Q)分别高出13.73%、10.73%;蛋白核小球藻粉、裸藻粉和雨生红球藻粉1∶1∶1混合后的提取物QLY对肝癌细胞HepG2的抑制率比单独的雨生红球藻粉提取物(Y)、裸藻粉提取物(L)、蛋白核小球藻粉提取物(Q)分别高出10.6%、13.6%和7.6%。
由此可见,复合藻粉的提取物表现出了对肿瘤细胞更好的抑制效果。

通过GraphPad Prism软件测得QX与LX对人结肠癌细胞HCT116存活抑制的IC50值分别为39.46、59.17μg/mL(图1、图2);QX对肝癌细胞HepG2存活抑制的IC50值为41.74μg/mL(图3)。
Claims (10)
1.一种复合藻粉片,其特征在于:原料包括螺旋藻粉0-100份、小球藻粉0-100份、裸藻粉0-100份、雨生红球藻粉0-100份、盐藻粉0-100份或迦得拟微绿球藻粉0-100份中的两种或多种藻粉的组合物。
2.根据权利要求1所述的复合藻粉片,其特征在于:所述螺旋藻粉包括钝顶螺旋藻粉、极大螺旋藻粉或盐泽螺旋藻粉;小球藻粉包括蛋白核小球藻粉、普通小球藻粉或原壳小球藻粉;盐藻粉包括杜氏盐藻粉。
3.根据权利要求1所述的复合藻粉片,其特征在于:还包括辅料:菊芋粉0-100份、紫菜粉0-100份、紫球藻粉0-100份、异麦芽酮糖0-10份、菊芋多糖0-10份、刺参低聚肽0-10份、虾肽螯合钙0-10份、改善口感或鲜味的食品添加剂0-10份、微晶纤维素0-10份、维生素0-5份、羧甲基纤维素0-4份、硬脂酸镁0-4份或二氧化硅0-4份。
4.根据权利要求1或2所述的复合藻粉片,其特征在于:所述钝顶螺旋藻粉49份、蛋白核小球藻粉49份。
5.根据权利要求1或2所述的复合藻粉片,其特征在于:所述蛋白核小球藻粉49份、裸藻粉49份。
6.根据权利要求1或2所述的复合藻粉片,其特征在于:所述钝顶螺旋藻粉45份、蛋白核小球藻粉45份、微晶纤维素8份、羧甲基纤维素1.6份、硬脂酸镁0.4份。
7.根据权利要求1或2所述的复合藻粉片,其特征在于:所述蛋白核小球藻粉45份、裸藻粉45份、微晶纤维素8份、羧甲基纤维素1.6份、硬脂酸镁0.4份。
8.根据权利要求1或2所述的复合藻粉片,其特征在于:所述钝顶螺旋藻粉50份、雨生红球藻粉46份、食用香料1份、维生素C1份。
9.根据权利要求1或2所述的复合藻粉片,其特征在于:所述钝顶螺旋藻粉33份、雨生红球藻粉33份、裸藻粉32份;钝顶螺旋藻粉33份、蛋白核小球藻粉33份、裸藻粉32份;钝顶螺旋藻粉41份、菊芋粉41份、紫菜粉5份、异麦芽酮糖1份。
10.权利要求1-9所述的复合藻粉片的制备方法,其特征在于:按照配比称量原辅料,混合后加入压片机进行压片得到复合藻粉片。
