Abstract
本发明属于营养与疾病健康领域;公开了一种具有调节糖脂代谢功能的复配杂粮,其特征在于复配杂粮由荞麦、燕麦、玉米淀粉或面粉组合而成;荞麦与燕麦经多功能粉碎机粉碎后过60目筛,制成粒径小于0.3mm的荞麦粉和燕麦粉,复配杂粮粉由以下重量的组分组合而成:荞麦2~3克,燕麦1~2克,玉米粉(或面粉)22~97克,优选荞麦2克,燕麦12克;本发明涉及的杂粮包括但不限于荞麦和燕麦;本发明所述的复配杂粮具有改善糖尿病等特殊人群体质和糖脂代谢的功能;本发明公开的复配杂粮的组合和比例为杂粮深入开发提供了科学依据。
Description
技术领域
本发明涉及一种用于糖尿病等特殊人群膳食干预的复配杂粮,能够预防和改善糖尿病的血糖水平以及对于体重和肠道菌群进行有益改善,属于营养与疾病健康领域。
背景技术
糖尿病是一种由于胰岛素分泌绝对或相对不足,或外周组织对胰岛素不敏感,引起的以糖代谢紊乱为主,包括脂肪、蛋白质代谢紊乱的内分泌代谢性疾病。糖尿病的流行与社会和经济水平密切相关,随着社会的快速发展和城市化所引起的不健康饮食和缺乏运动等,糖尿病的患病人数急剧增加。我国目前对糖尿病的防治措施以预防为主,强调要强化生活方式对糖尿病的干预和预防,其中,饮食干预占着极为重要的地位。
早在唐朝时期,“药王”孙思邈便在《千金食治》一书中强调了疾病食疗的重要性,记载了多种对于消渴[1](糖尿病的古称)有食疗功效的杂粮。现代研究表明:燕麦中丰富的可溶性膳食纤维有利于控制血糖、血脂及血压;β-葡聚糖也对糖脂代谢起到很大的帮助[2,3];多酚有很强的自由基清除能力[4,5];燕麦多肽能够改善糖尿病低体重及多饮多食的症状[6,7]。荞麦中的荞麦多糖以及丰富的黄酮类物质,如芦丁、槲皮素等,能够改善脂代谢[8,9],显著降低血脂、肝脂水平[10],膳食中荞麦的摄入能够显著降低餐后血糖水平和胰岛素抵抗[11],能够通过调节胃肠道饱腹感激素水平增加饱腹感,并能够改善糖尿病病人的肾功能障碍等症状[12]。
杂粮是我国广泛食用的医药同源类食物。但杂粮加工途径少,多是作为原料或简单分选、包装的初级加工食品,这些单一品种杂粮往往因为适口性差、可食性欠佳以及营养成分不足等原因,导致我国人均杂粮摄入量远低于2016年版《中国居民膳食指南》全谷物和杂豆类的成人推荐摄入量。杂粮的复配可改善上述单一品种杂粮的不足。近年来,国内外有相关研究证明:多种杂粮复配后,其各类蛋白质、矿物质(如钙、铁、锌)、维生素(如B1、B2)、膳食纤维、总黄酮和皂甙等物质的含量均高于普通主食(如米粉、面粉)。因此,复配杂粮有望对于特殊人群,例如糖尿病患者的膳食干预起到积极的作用。
尽管目前市场上出现了一些五谷杂粮食品,但这些杂粮的复配仅从可加工性、适口性等角度进行搭配,尚缺乏相应的医学数据支持其对于特殊人群膳食干预的效果。本发明以我国产量较高的燕麦和荞麦进行复配,通过细胞水平研究筛选燕麦和荞麦复配比例,并经糖尿病模型动物验证,得到一种能够显著改善糖尿病血糖及其相关生化指标的复配杂粮,对糖尿病的防治具有重要的意义。
发明内容
本发明的目的是提供一种具有调节糖脂代谢功能的复配杂粮,其特征在于复配杂粮由荞麦、燕麦、玉米淀粉或面粉组合而成。本发明涉及的杂粮包括但不限于燕麦、荞麦,主粮包括但不限于玉米淀粉和面粉。
本发明所述的复配杂粮,具体特征在于:
(1)荞麦与燕麦经多功能粉碎机粉碎后过60目筛,制成粒径小于0.3mm的荞麦粉和燕麦粉;
(2)荞麦和燕麦复配杂粮粉由以下重量的组分组合而成:荞麦2~3克,燕麦1~2克,玉米粉(或面粉)22~97克;
(3)优选荞麦荞麦2克,燕麦1克;
(4)本发明提出的复配杂粮有利于改善特殊人群的体质。
(5)本发明提出的复配杂粮有利于改善特殊人群的糖脂代谢功能。
本发明所公布的复配杂粮,具有改善特殊人群糖脂代谢功能,为社区居民选择杂粮以及我国杂粮品种的深入开发提供了科学依据,具有一定的指导意义。
本发明与现有技术相比,创新性在于:
经文献调研,目前用于糖尿病膳食干预的杂粮专利有两个,分别为《一种糖尿病患者食用杂粮面粉及其制备方法》(CN 111418763 A)和《一种藜麦复配的功能性杂粮预混粉及其制备工艺和应用》(CN 109820136 A)。这两个专利主要公开的杂粮面粉的配方组成和加工工艺。经查阅文献,大多数杂粮的复配仅从品种多样性和适口性等角度进行选择,无实验数据支持其对特殊人群血脂和血糖干预的效果,更没有组分和配比的相关研究。因此目前所公开的复配杂粮并不能指导我国杂粮复配品种的选择和配方组成。
对比分析已公布的专利信息和目前发表的文章所涉及的复配杂粮,本发明所提出这种改善糖脂代谢的复配杂粮的创新性体现在:
(1)本发明所提出的一种改善糖脂代谢的复配杂粮的组合经过科学创造性研究,在杂粮逐级程序化提取工艺的基础上,通过应用胰岛素抵抗细胞模型筛选优化得到了两种能够显著改善胰岛素抵抗的两种杂粮:荞麦和燕麦,并以此为基础进行了全范围复配比例对胰岛素抵抗功效的影响,优化得到了这两种杂粮的配方范围,类似的研究未见报道;
(2)本发明提出这种复配杂粮的组合和配方经过动物实验验证,结果表明能够显著改善糖尿病模型大鼠的体质和糖脂代谢,具有广阔的开发前景。

具体实施方式
下面结合实施例来具体说明本发明内容。应当理解,下面的实施例是用于说明本发明内容而非限定本发明内容,任何形式上的变动和/或变通都将落入本发明的保护范围之内。
下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径得到。
燕麦10kg(坝莜1号);荞麦10kg(大三棱),山西东方亮生命科技有限公司,杂粮均经多功能粉碎机粉碎后过60目筛,制成粒径小于0.3mm的杂粮粉储存备用。
实施例1改善胰岛素抵抗功效的荞麦及燕麦复配比例的优化
按照前期我们申请的中国专利《一种杂粮逐级程序化提取方法及其应用》(申请号201910902335.5)中所述方法,对荞麦和燕麦分别进行逐级程序化提取,得到这二种杂粮单品的水提物、醇提物和正己烷提取物。按照我们前期申请的中国专利《一种筛选优化杂粮复配组成和比例的方法及其应用》所述,荞麦和燕麦的醇提物能够显著改善胰岛素抵抗。因此,本发明以Hep G2胰岛素抵抗细胞模型为基础,考察荞麦和燕麦醇提物对葡萄糖利用的影响,由此筛选优化得到能够显著改善胰岛素抵抗的荞麦和燕麦复配比例范围。
一、实验方法
1.Hep G2胰岛素抵抗模型的建立
取100μLHepG2细胞(浓度为1×104个/mL)接种于96孔板中,加入DMEM高糖培养基于37℃5%CO2培养箱中培养24h,更换为含有10μmol/L胰岛素的DMEM高糖培养液诱导36h,采用葡萄糖检测试剂盒测定上清液中的葡萄糖含量。以不加入胰岛素组为空白对照。
2.荞麦和燕麦的复配比例对改善胰岛素抵抗功效的影响
称取适量燕麦醇提物及荞麦醇提物,以DEME高糖培养液配制荞麦∶燕麦比例分别为 100∶0,90∶10,80∶20,70∶30,60∶40,50∶50,40∶60,30∶70,20∶80,10∶90,0∶100受试样品溶液,其中总杂粮浓度为100μg/mL。
取100μLHepG2细胞(浓度为1×104个/mL)接种于96孔板中,加入DMEM高糖培养基于37℃5%CO2培养箱中培养24h,更换为含有10μmol/L胰岛素的DMEM高糖培养液诱导36h。弃去培养液,PBS磷酸盐缓冲液清洗模型细胞2遍,加入上述受试样品溶液,37℃下干预24h,采用葡萄糖检测试剂盒测定上清液中的葡萄糖含量。以不加入胰岛素组为空白对照,以含有10μmol/L的罗格列酮的DMEM高糖培养基为阳性对照组。受试样品的相对葡萄糖消耗量被定义为样品组与空白对照组葡萄糖消耗量的比值,比较样品组和胰岛素抵抗模型组的相对葡萄糖消耗量,用SPSS 21.0进行单因素方差分析,*P<0.05被认为差异具有统计学意义。
二、结果
1.胰岛素抵抗模型的建立
用DMEM培养液的含糖量减去上清液中所测的葡萄糖含量即为细胞的葡萄糖消耗量,如附图1所示,HepG2细胞经高胰岛素(10μmol/L)诱导36h后,上清液中的葡萄糖消耗量显著低于空白对照组(P<0.05),表明HepG2胰岛素抵抗细胞模型建立成功。
2.荞麦和燕麦的复配比例对改善胰岛素抵抗功效的影响
如附图2所示:与胰岛素抵抗模型组相比,阳性对照组罗格列酮显著提高了葡萄糖消耗量。荞麦与燕麦复配比例为7∶3和6∶4组改善胰岛素抵抗能力虽然低于罗格列酮,但也能够明显提高HepG2细胞的葡萄糖消耗量(p<0.05),且其功效显著高于较纯荞麦或燕麦,说明荞麦和燕麦复配具备改善胰岛素抵抗的潜力,这可能与两者营养成分的互补有关。
实施例2荞麦与燕麦杂粮复配方法
荞麦与燕麦经多功能粉碎机粉碎后过60目筛,制成粒径小于0.3mm的荞麦粉和燕麦粉。荞麦和燕麦复配杂粮粉由以下重量的组分组合而成:荞麦2克,燕麦1克,玉米粉(或面粉)22克~97克。
实施例3荞麦与燕麦复配调节糖尿病大鼠糖脂代谢的验证
根据原国家食品药品监督管理局(CFDA)发布的辅助降血糖功能评价方法,采用高热能饲料联合链脲佐菌素注射的方法建立糖尿病大鼠模型。在维持各组饲料高热能的基础上,用受试样品替换饲料中的玉米淀粉并调整营养成分,通过5周膳食干预评价荞麦与燕麦复配改善糖尿病大鼠糖脂代谢的功效,并通过统计学分析,确定最佳复配比例,为特殊人群尤其糖尿病患者的膳食干预提供研究基础。
一、实验方法
1.糖尿病大鼠模型的建立:
76只健康SPF级雄性SD大鼠(200±20g),适应性喂养1周。根据原国家食品药品监督管理局(CFDA)辅助降血糖评价方法,采用高热能饲料联合链脲佐菌素(STZ)腹腔注射的方法建立糖尿病大鼠模型。具体方法:(1)以高热能饲料喂养SD大鼠6周,诱导胰岛素抵抗的出现;(2)第6周末,大鼠隔夜禁食,自由饮水。(3)次日大鼠腹腔注射1%STZ溶液,剂量为30mg/kg·bw(即3.5mL/kg·bw),注射30min后大鼠自由饮水饮食;(4)注射72 h后采大鼠尾静脉血测量血糖,血糖值为10~25mmol/L的大鼠为糖尿病大鼠模型。
2.复配杂粮功效考察:
(1)分组及膳食干预
糖尿病模型大鼠适应性喂养5天后,禁食3~4h,自由饮水。禁食结束后取血测量模型大鼠空腹血糖,根据大鼠空腹血糖随机分为7组,如表1所示。实验流程如附图3所示。

(2)检测指标
一般指标:每日称量饲料及水量并计算出大鼠进食量和饮水量,每周称量大鼠体重。
糖代谢指标:实验结束后禁食5h,股动脉集采大鼠血液,血液采集后静置2h,而后于4℃低温冷冻离心机中3000r/min离心15min,分离血清,检测空腹血糖;口服葡萄糖耐量试验(oral glucose tolerance test,OGTT),根据血糖变化曲线计算血糖曲线下面积(glucose area under curve,GAUC);糖化血清蛋白(glycated serum protein,GSP);空腹胰岛素(insulin, INS);胰岛素敏感指数(insulin sensitive index,ISI)。
脂代谢指标:血清总胆固醇(TC);血清总甘油三酯(TG);血清高密度脂蛋白胆固醇(HDL-C);血清低密度脂蛋白胆固醇(LDL-C)。
(4)数据处理
符合正态分布的数据以均数±标准差(mean±SD)表示,使用SPSS 21.0软件(SPSS,Inc.,Chicago,IL,USA)进行单因素方差分析(one-way analysis of variance,ANOVA),在分析之前先检验方差齐性,方差齐时采用LSD检验比较各实验组与对照组之间的差异,非正态分布数据采用均数(四分位数间距)表示,采用秩和检验(Kruskal-Wallis H检验)。 P<0.05被认为差异具有统计学意义。
二、结果
1.复配杂粮对糖尿病大鼠体重的影响
从附图4结果中可见,第8周开始糖尿病模型组、面粉组大鼠饮水量显著高于正常大鼠(P<0.05)。第9~11周,与糖尿病模型组相比,杂粮低剂量组及中剂量组大鼠饮水量显著降低(P<0.05),高剂量组无显著差异(P>0.05)。与面粉组相比,杂粮低剂量组及中剂量组大鼠饮水量显著降低(P<0.05),高剂量组无显著差异(P>0.05)。
从附图5结果中可见,第7周造模成功后,糖尿病模型组、面粉组、二甲双胍组、杂粮中剂量组大鼠体重均显著低于正常组大鼠(P<0.05)。第8~11周,与模型组相比,杂粮低剂量组大鼠体重显著增加(P<0.05)。第10周,杂粮高剂量组大鼠体重显著高于模型组(P<0.05)。第7~9周,与面粉组相比,杂粮低剂量组及高剂量组大鼠体重显著增加(P<0.05)。
结论:复配杂粮对于改善糖尿病大鼠的体质具有积极影响。
2.复配杂粮粉对糖尿病大鼠糖代谢的影响
复配杂粮低剂量组及中剂量组大鼠空腹血糖显著低于模型组及面粉组大鼠(附图6);,复配杂粮低剂量组大鼠GAUC显著低于糖尿病模型组及面粉组(附图7);二甲双胍组及各杂粮干预组的糖化血清蛋白均显著低于糖尿病模型组(附图8);二甲双胍组及各杂粮干预组大鼠INS均显著低于糖尿病模型组大鼠,杂粮高剂量组血清胰岛素显著低于面粉组(附图9);二甲双胍组及各杂粮干预组大鼠胰岛素敏感指数ISI均有显著高于糖尿病模型组(表2所示),各杂粮干预组大鼠ISI均有显著高于面粉组(P<0.05),结果表明复配杂粮改善了糖尿病大鼠的糖代谢。

3.复配杂粮粉对糖尿病大鼠血脂的影响
如附图10所示,二甲双胍组大鼠及杂粮中、高剂量组大鼠血清TC显著低于糖尿病模型组,杂粮高剂量组大鼠血清TC显著低于面粉组(P<0.05);各组大鼠间血清TG无统计学差异(P>0.05);各杂粮干预组及二甲双胍组血清HDL-C均显著高于糖尿病模型组及面粉组(P<0.05)。面粉组及各杂粮干预组大鼠LDL-C均显著低于糖尿病模型组(P<0.05),表明复配杂粮有利改善糖尿病大鼠脂代谢。
上述复配杂粮复配对于糖尿病大鼠的体质、糖代谢和脂代谢均体现出良好的改善作用,说明荞麦与燕麦按照2∶1进行复配所获得的杂粮粉对于改善糖尿病患者膳食干预是一种优良的选择,其中杂粮粉与玉米淀粉(面粉)的搭配比例在3∶100~12∶100范围内均可有效提高糖尿病患者的糖代谢和脂代谢,该复配杂粮可作为今后杂粮食品的深入开发的提供了配方指导。

1.一种具有调节糖脂代谢功能的复配杂粮,其特征在于复配杂粮粉由以下重量的组分组合而成:荞麦2~3克,燕麦1~2克,玉米粉(或面粉)22~97克。
2.权利要求1所述荞麦与燕麦经多功能粉碎机粉碎后过60目筛,制成粒径小于0.3mm的荞麦粉和燕麦粉。
3.权利要求1所述杂粮包括但不限于燕麦、荞麦。
4.权利要求1所述复配杂粮粉中荞麦含量优选2克,燕麦质量优选1克。
5.根据权利要求1所述的复配杂粮具有显著改善糖尿病等特殊人群的体质和糖脂代谢功能。
6.根据权利要求1所述复配杂粮可用于馒头、面条、月饼等食品的加工。
