技术领域
本发明属于食品技术领域,具体涉及一种具有抗氧化功能的复方饮料及其制备方法。
背景技术
自由基是一类具有氧化活性的带负电的离子,化学性质相当活跃,内源产生包括线粒体,过氧化酶,NADPH氧化酶以及细胞色素P450等,外源引起其增多的有紫外辐射,电离辐射,环境毒素等因素。人体内自由基的数量过多,会破坏细胞结构,干扰人体的正常代谢活动,引起疾病并加速人体衰老进程。
为了维持正常代谢活动,机体主要通过终止自由基链反应而清除自由基等抗氧化防御系统以保护机体。生物体应对抗氧化应激主要涉及两种机制:一是以酶类为主的,涉及到三种关键酶,依次是超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px),主要是对体内氧化还原平衡产生影响;二是以抗氧化剂为主的,包括维生素,多酚等物质,影响体内氧化还原状态。
鉴于之前研究者对衰老机制的研究,清除过多的自由基成为减缓衰老的主要办法,而摄取足够的天然抗氧化剂成为防止肌肤衰老以及延缓身体老化的方便可行方式。
在日常生活中,常见的维生素C和E以及β-胡萝卜素可以通过稳定自由基而达到抗氧化的作用。天然产物作为重要的活性成分因其高效、低毒倍受关注,借助其还原能力、抑制与氧化有关的酶的活性、抑制脂质氧化降解、清除自由基、螯合过渡金属等来发挥抗氧化作用。而利用传统中药配伍结合现代加工技术,最大限度地保留天然产物活性成分,研发适合长期服用且具有抗氧化功能的复方饮料,具有重要意义。
发明内容
本发明的目的是克服上述背景技术中的不足,提供一种具有抗氧化功能的复方饮料及其制备方法。
本发明的技术方案是:
一种具有抗氧化功能的复方饮料,由下列重量份数原料的提取液组成:西红花1-3份、金银花10-30份、菊花10-30份、线叶金雀花1-3份、金花茶1-3份、裸藻0.5-1.5份、球状念珠藻0.5-1.5份。
该复方饮料由下列重量份数原料的提取液组成:西红花1.5-2.5份、金银花15-25份、菊花15-25份、线叶金雀花1.5-2.5份、金花茶1.5-2.5份、裸藻0.8-1.2份、球状念珠藻0.8-1.2份。
该复方饮料由下列重量份原料的提取液组成:西红花2份、金银花20份、菊花20份、线叶金雀花2份、金花茶2份、裸藻1份、球状念珠藻1份。
该复方饮料还添加适量的柠檬酸、柠檬酸钠、白糖与蜂蜜进行调味。
一种具有抗氧化功能的复方饮料的制备方法,按以下步骤进行:
(1)按照配方比例称取西红花,加入投料重量2800-3200倍的水,55-65℃水浴提取28-32min,过滤,得A液,避光保存;
(2)按照步骤(1)的配方比例称取金银花、菊花、线叶金雀花、金花茶,加入投料重量2800-3200倍的水,煎煮28-32min,过滤,得B液,避光保存;
(3)按照步骤(1)的配方比例称取裸藻、球状念珠藻,加入投料重量2800-3200倍的水,进行超声-微波协同提取,过滤,得C液,避光保存;
(4)按重量份数取A液1份,B液1份,C液1份,加水2300-2700份,混合均匀,添加适量的柠檬酸、柠檬酸钠、白糖、蜂蜜进行调味,即得具有抗氧化功能的复方饮料。
所述步骤(3)中所述的超声波功率为50W,微波功率为150-250W,提取时间为180-300s。
本发明的有益效果是:
本发明是运用传统中医药理论和现代加工工艺以及经过科学组方研制出的复方饮料,不仅具有显著的抗氧化功能,而且具有清热解毒、泻火除烦、解郁安神等保健功能;同时,本品口感较好、功效缓和、无副作用,可作日常保健长期服用;另外,本品携带、服用方便,便于运输储存。

实施例1
本发明提供的一种具有抗氧化功能的复方饮料,由下列重量份数的原料组成:西红花提取物2份、金银花提取物20份、菊花提取物20份、线叶金雀花提取物2份、金花茶提取物2份、裸藻提取物1份、球状念珠藻提取物1份。
该复方饮料的制备方法,按以下步骤进行:
(1)按照配方比例称取西红花,加入投料重量3000倍的水,60℃水浴提取30min,过滤,得A液,避光保存;
(2)称取金银花,菊花,线叶金雀花,金花茶加入投料重量3000倍的水,煎煮30min,过滤,得B液,避光保存;
(3)称取裸藻,球状念珠藻,加入投料重量3000倍的水,进行超声-微波协同提取,过滤,得C液,避光保存;
所述超声波功率为50W,微波功率为200W,提取时间为180s;
(4)按重量份数取A液1份,B液1份,C液1份,加水2500份,混合均匀,添加适量的柠檬酸、柠檬酸钠、白糖与蜂蜜调味,即得具有抗氧化功能的复方饮料。
实施例2
本发明提供的一种具有抗氧化功能的复方饮料,由下列重量份数的原料组成:西红花提取物1.5份、金银花提取物30份、菊花提取物30份、线叶金雀花提取物1.5份、金花茶提取物1.5份、裸藻提取物0.5份、球状念珠藻提取物0.5份。
该复方饮料的制备方法,按以下步骤进行:
(1)按照配方比例称取西红花,加入投料重量2900倍的水,65℃水浴提取32min,过滤,得A液,避光保存;
(2)按照步骤(1)的配方比例称取金银花、菊花、线叶金雀花、金花茶加入投料重量3100倍的水,煎煮31min,过滤,得B液,避光保存;
(3)按照步骤(1)的配方比例称取裸藻、球状念珠藻,加入投料重量2900倍的水,进行超声-微波协同提取,过滤,得C液,避光保存;
所述超声波功率为50W,微波功率为250W,提取时间为300s;

该复方饮料的制备方法,按以下步骤进行:
(1)按照配方比例称取西红花,加入投料重量3200倍的水,55℃水浴提取28min,过滤,得A液,避光保存;
(2)按照步骤(1)的配方比例称取金银花、菊花、线叶金雀花、金花茶加入投料重量3200倍的水,煎煮32min,过滤,得B液,避光保存;
(3)按照步骤(1)的配方比例称取裸藻、球状念珠藻,加入投料重量3200倍的水,进行超声-微波协同提取,过滤,得C液,避光保存;
所述超声波功率为50W,微波功率为150W,提取时间为210s;

该复方饮料的制备方法,按以下步骤进行:
(1)按照配方比例称取西红花,加入投料重量2800倍的水,57℃水浴提取29min,过滤,得A液,避光保存;
(2)按照步骤(1)的配方比例称取金银花、菊花、线叶金雀花、金花茶加入投料重量2800倍的水,煎煮28min,过滤,得B液,避光保存;
(3)按照步骤(1)的配方比例称取裸藻、球状念珠藻,加入投料重量2800倍的水,进行超声-微波协同提取,过滤,得C液,避光保存;
所述超声波功率为50W,微波功率为180W,提取时间为240s;
(4)按重量份数取A液1份,B液1份,C液1份,加水2700份,混合均匀,添加适量的柠檬酸、柠檬酸钠、白糖与蜂蜜调味,即得具有抗氧化功能的复方饮料。
以下通过试验进一步说明对本发明的有益效果:
(1)DPPH自由基清除能力的测定
将供试品(分别由实施例1-4制得)以及VC标准品分别配制成不同梯度浓度的样品液和标准液,浓度分别为12.5μg/mL、25μg/mL、50μg/mL、100μg/mL、200μg/mL、400μg/mL、600μg/mL,每个浓度取三个试管,标记为A、B、C组。
实施例组包括,A组:2mL样品液和2mL DPPH溶液;B组:2mL样品液和2mL无水乙醇;C组:2mL 50%乙醇和2mL DPPH溶液。
VC对照组包括,A组:2mL标准液和2mL DPPH溶液;B组:2mL标准液和2mL无水乙醇;C组:2mL 50%乙醇和2mL DPPH溶液。
将上述反应液置于室温下暗反应30min,在517nm处测定吸光度,重复3次。按下列公式计算清除率。

式中W1为DPPH自由基清除率(%);A0为C组的吸光度;A1为A组的吸光度;A2为B组的吸光度。
测定结果参见图1。
结果表明:所制备的复方饮料具有较好的抗氧化能力。
(2)ABTS自由基清除能力的测定
分别制备ABTS标准溶液(7mmol·L-1)和过硫酸钾溶液(140mmol·L-1),取ABTS标准液5mL加入88μL过硫酸钾溶液,放在黑暗中反应12~16h。取上述溶液用乙醇稀释并调整吸光度值在734nm下为0.7,制成ABTS工作液。
将供试品(分别由实施例1-4制得)以及VC标准品分别配制成不同梯度浓度为的样品液和标准液,浓度分别为50μg/mL、100μg/mL、200μg/mL、400μg/mL、600μg/mL,每个浓度取三个试管,标记为A、B、C组。
实施例组包括,A组:1mL样品液和3mL ABTS工作液;B组:1mL样品液和3mL无水乙醇;C组:1mL 50%乙醇加入3mL ABTS工作液。
VC对照组包括,A组:1mL标准液和3mL ABTS工作液;B组:1mL标准液和3mL无水乙醇;C组:1mL 50%乙醇加入3mL ABTS工作液。
将上述反应液置于室温下暗反应20min,在734nm处测定吸光度,重复3次。按下列公式计算清除率。

测定结果参见图2。
结果表明:所制备的复方饮料具有较好的抗氧化能力。

2.根据权利要求1所述的一种具有抗氧化功能的复方饮料,其特征在于:该复方饮料由下列重量份数原料的提取液组成:西红花1.5-2.5份、金银花15-25份、菊花15-25份、线叶金雀花1.5-2.5份、金花茶1.5-2.5份、裸藻0.8-1.2份、球状念珠藻0.8-1.2份。
3.根据权利要求2所述的一种具有抗氧化功能的复方饮料,其特征在于:该复方饮料由下列重量份原料的提取液组成:西红花2份、金银花20份、菊花20份、线叶金雀花2份、金花茶2份、裸藻1份、球状念珠藻1份。
4.权利要求1或2或3所述的一种具有抗氧化功能的复方饮料,其特征在于:该复方饮料还添加适量的柠檬酸、柠檬酸钠、白糖与蜂蜜进行调味。
5.权利要求1所述的一种具有抗氧化功能的复方饮料的制备方法,按以下步骤进行:
(1)按照配方比例称取西红花,加入投料重量2800-3200倍的水,55-65℃水浴提取28-32min,过滤,得A液,避光保存;
(2)按照步骤(1)的配方比例称取金银花、菊花、线叶金雀花、金花茶,加入投料重量2800-3200倍的水,煎煮28-32min,过滤,得B液,避光保存;
(3)按照步骤(1)的配方比例称取裸藻、球状念珠藻,加入投料重量2800-3200倍的水,进行超声-微波协同提取,过滤,得C液,避光保存;
(4)按重量份数取A液1份,B液1份,C液1份,加水2300-2700份,混合均匀,添加适量的柠檬酸、柠檬酸钠、白糖、蜂蜜进行调味,即得具有抗氧化功能的复方饮料。
6.根据权利要求5所述的一种具有抗氧化功能的制备方法,其特征在于:所述步骤(3)中所述的超声波功率为50W,微波功率为150-250W,提取时间为180-300s。
