Example 1
Culturing Phellinus igniarius strains and selecting the optimal culture conditions, culturing Phellinus igniarius strains by using potato basal medium (PDA), and culturing at different temperatures and different pH values, wherein the selection temperature range is 25-37 ℃, and the pH range is 4-8. The test result shows that the optimum growth temperature of the phellinus igniarius is 30 ℃, the optimum pH value is 6, and under the condition, the phellinus igniarius strain grows fastest and the yield is higher.
Comparative example 1
Pretreatment of corn straws: mixing the corn straw with water (10: 3) under high pressure and high temperature by an autoclave, treating for 24h, and centrifuging to obtain straw precipitate.
And cleaning the precipitate, and drying to obtain the corn carbonized straw carbon powder.
Mixing straw carbon powder and dry powder of potato basic solid culture medium (every 4.6g of dry powder of potato basic solid culture medium contains 2g of glucose, 0.6g of potato and 2g of agar) for mixed culture:
no. 1, potato basic solid culture medium 4.6g
No. 2, 2.47g of dry powder of potato basic solid culture medium (agar mass is not counted) and 0.13g of straw carbon powder;
no. 3, 2.34g of dry powder of potato basic solid culture medium (agar mass is not counted) and 0.26g of straw carbon powder;
no. 4, 2.21g of dry powder of potato basic solid culture medium (agar mass is not counted) and 0.39g of straw carbon powder;
no. 5, 1.95g of dry powder of a potato basic solid culture medium (not counting the mass of agar) and 0.65g of straw carbon powder;
no. 6, 1.69g of dry powder of potato basic solid culture medium (agar mass is not counted) and 0.91g of straw carbon powder;
no. 7, 1.43g of dry powder (not counting the mass of agar) and 1.17g of straw carbon powder of potato basic solid culture medium;
no. 8, 1.04g of dry powder of a potato basic solid culture medium (agar mass is not counted) and 1.56g of straw carbon powder;
no. 9, 0.52g of dry powder (not counting the mass of agar) and 2.08g of straw carbon powder of potato basic solid culture medium;
no. 10, 2g agar (control group)
Except No. 1 and No. 10, agar 2g is added to each group, and No. 1-10 is dissolved in 100ml water and sterilized at 121 deg.C for 30 min. Adjusting the pH value to 6, culturing at 37 ℃ for 16 days after inoculation, wherein only 5% and 10% of groups can grow the phellinus igniarius (as shown in B in figure 1), and comparing A in figure 1 (a picture shot when culturing for 5 days), the agar contained in the solid culture medium has certain influence on the experimental result, the phellinus igniarius can also grow on the culture medium (No. 10) only containing the agar, and in order to avoid the influence, the subsequent culture is carried out by using the liquid culture medium, and meanwhile, because the growth effect of the phellinus igniarius in the culture medium added with the straw carbon powder is poorer than that of the culture medium only containing the potato, the corn straw carbon powder sediment obtained after the high-pressure kettle contains substances for inhibiting the growth of fungi.
Example 2
The corn stalks are dried after being cleaned, crushed by a centrifugal crusher and screened by a 100-mesh screen. Obtaining corn stalk fine powder, preparing a culture medium by using the corn stalk fine powder and water, and adding potato basic liquid culture medium dry powder (2 g of glucose and 0.6g of potatoes) according to different proportions.

no. 5, 0.52g of dry powder of a potato basic liquid culture medium and 2.08g of straw powder;
no. 6, straw powder 2.6g
No. 7, 100ml water (control group)
Dissolving the No. 1-6 substances in 100ml of water, and sterilizing at 121 ℃ for 30 min. Adjusting pH to 6, inoculating, culturing at 37 deg.C for 14 days, as shown in FIG. 2, the No. 5 culture medium has good growth vigor, i.e. 0.52g potato and 2.08g straw powder, and Phellinus linteus can also grow in the presence of only straw powder (No. 6 culture medium), and the No. 6 culture medium is the best culture medium for Phellinus linteus culture according to comprehensive consideration of material usage, yield, Phellinus linteus quality and price.
Example 3
The dry weight of the phellinus igniarius strains growing in the culture medium No. 1-6 in the figure 2 is measured after the phellinus igniarius strains are dried, and the phellinus igniarius yield in different culture media is calculated, wherein the yield results are shown in the table 1:
TABLE 1 yield results
Medium number Phellinus igniarius yield (g)
Number 1 0.308
Number 2 0.438
No. 3 0.619
Number 4 0.790
Number 5 0.956
Number 6 0.886
The results of example 2 were further verified by weighing the dry weight of Phellinus linteus, and the experimental results were further determined by considering the material, yield, quality and price of Phellinus linteus comprehensively, with medium No. 6 being the best medium for Phellinus linteus culture.
And (4) conclusion: the invention discovers that the corn straws can also be used as a carbon source under appropriate conditions, the yield is higher in the same time compared with a commonly used potato culture medium, but the corn straws are easier to obtain and have economic benefits compared with potatoes, the culture medium is prepared by utilizing the corn straws, the problem of environmental pollution caused by difficulty in processing crop straws is solved, the resource recycling is realized, the prepared culture medium can be used as nutrient substances required by the growth of phellinus igniarius, the preparation method of the culture medium is simple, and a new path is provided for the culture of medicinal and edible fungi.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
