Abstract

Background
Irritable Bowel Syndrome (IBS) is a common bio-psycho-social disorder, and is a disorder of gastrointestinal dysfunction. The pathogenic factors of the traditional Chinese medicine are complex and comprise genetic factors, mental factors, dietary factors, infection factors and the like. According to the characteristics of the stool, IBS is divided into four clinical types of diarrhea type, constipation type, mixed type and indeterminate type, and IBS patients in China are more common in diarrhea IBS.
The diarrhea-predominant irritable bowel syndrome is clinically manifested by persistent or intermittent diarrhea, but the diarrhea-predominant irritable bowel syndrome lacks exact basis such as morphological and laboratory abnormal changes and can occur in various states such as emotional stress, staying up night, dietary stimulation and the like. Any slight stimulation in the onset period may cause obvious clinical symptoms, which seriously affect the quality of life of the affected people.
At present, the treatment method for diarrhea-predominant irritable bowel syndrome is mainly symptomatic diarrhea checking, but the antidiarrheal drug only has temporary effect, short action time and poor treatment effect, and the antidiarrheal drug can aggravate the functional disorder of intestinal peristalsis if not used properly.

Preferably, the liver-softening pain-relieving traditional Chinese medicine is selected from at least one of angelica sinensis, ligusticum wallichii, fructus evodiae, fried white paeony root and fructus toosendan, and further preferably at least one of ligusticum wallichii, fructus evodiae and fried white paeony root;
the liver-regulating and spleen-soothing traditional Chinese medicine is selected from at least one of divaricate saposhnikovia root, costustoot and green tangerine orange peel, and is further preferably selected from at least one of divaricate saposhnikovia root and green tangerine orange peel;
the traditional Chinese medicine for soothing liver-qi stagnation is selected from at least one of radix curcumae, rhizoma cyperi, radix bupleuri, akebia seed and fingered citron, and further preferably at least one of radix curcumae, rhizoma cyperi and radix bupleuri;
the middle-warmer warming and pain relieving traditional Chinese medicine is selected from at least one of perilla, baked ginger, galangal and pepper, and is further preferably selected from at least one of baked ginger and galangal;
the traditional Chinese medicine for cooling blood and stopping dysentery is at least one selected from eclipta, purslane, Chinese pulsatilla root, indigo naturalis and brucea javanica, and at least one selected from eclipta, purslane and Chinese pulsatilla root is further preferably adopted;
the traditional Chinese medicine for removing food stagnation is selected from at least one of fructus Aurantii, endothelium corneum Gigeriae Galli, rhizoma Zingiberis recens, parched Raphani semen, fructus crataegi preparata, fructus Hordei Germinatus, parched Massa Medicata Fermentata, herba Agastaches, semen Alpiniae Katsumadai and semen Myristicae, and preferably at least one of fructus Aurantii, rhizoma Zingiberis recens, parched Raphani semen and semen Myristicae;
the traditional Chinese medicine for relieving spasm and pain is selected from at least one of trogopterus dung, honey-fried licorice root and turmeric, and further preferably at least one of honey-fried licorice root and turmeric.
In a second aspect, the embodiment of the present invention also provides a preparation method of the above traditional Chinese medicine composition for irritable bowel syndrome, which includes the following operations: weighing the traditional Chinese medicines according to the weight part ratio in the traditional Chinese medicine composition for irritable bowel syndrome, mixing, decocting with water for extraction, filtering, and collecting the obtained filtrate.
In a third aspect, an embodiment of the present invention further provides another preparation method of the above traditional Chinese medicine composition for irritable bowel syndrome, including the following operations: weighing the traditional Chinese medicines according to the weight part ratio in the traditional Chinese medicine composition for irritable bowel syndrome, respectively decocting and extracting the traditional Chinese medicines with water, filtering, collecting obtained filtrate, and mixing.
According to the actual needs of clinical use, the filtrates obtained by the two preparation methods can be further concentrated, dried and the like, and then processed into oral dosage forms such as syrup, oral liquid, powder, granules, capsules, tablets, big honeyed pills and the like.

18g of codonopsis pilosula, 5g of grifola, 10g of rhizoma alismatis, 10g of angelica sinensis, 20g of ligusticum wallichii, 12g of pericarpium citri reticulatae viride, 15g of radix curcumae, 4g of perilla, 15g of herba ecliptae, 15g of ginger and 8g of trogopterus dung.
The preparation method comprises the following steps: weighing the above Chinese medicinal materials, mixing, decocting in water for 2 times, each for 20min, filtering, mixing filtrates, concentrating, and drying.
Example 2
The embodiment of the invention provides a traditional Chinese medicine composition for irritable bowel syndrome, and each traditional Chinese medicine composition comprises the following traditional Chinese medicines in parts by weight:
15g of radix pseudostellariae, 20g of semen coicis, 25g of fructus evodiae, 7g of elecampane, 10g of rhizoma cyperi, 8g of galangal, 10g of Chinese pulsatilla root, 12g of endothelium corneum gigeriae galli and 8g of turmeric.
The preparation method comprises the following steps: weighing the above Chinese medicinal materials, mixing, decocting in water for 2 times, each for 20min, filtering, mixing filtrates, concentrating, and drying.
Example 3
The embodiment of the invention provides a traditional Chinese medicine composition for irritable bowel syndrome, and each traditional Chinese medicine composition comprises the following traditional Chinese medicines in parts by weight:
18g of astragalus, 18g of fried bighead atractylodes rhizome, 25g of fructus evodiae, 12g of green tangerine peel, 10g of rhizoma cyperi, 8g of galangal, 15g of eclipta, 5g of ginger, 5g of fried radish seed and 8g of turmeric.
The preparation method comprises the following steps: weighing the above Chinese medicinal materials, mixing, decocting in water for 2 times, each for 20min, filtering, mixing filtrates, concentrating, and drying.

Effect example 1
The effect example provides the effect of the traditional Chinese medicine composition for irritable bowel syndrome on diarrhea-predominant irritable bowel syndrome in mice.
1. Material
1.1 animals
ICR mice, male, body weight at the start of the experiment: 18-23 g. Purchased from Beijing Weitonglihua laboratory animal technology Co., Ltd, license number: SCXK (Kyoto) 2016-.
1.2 medicine
The Chinese medicinal composition for irritable bowel syndrome obtained in examples 1 to 10.
The clinical dose of the adult is 1.95g crude drug/kg calculated according to the body weight of 60 kg.
1.3 feed
The SPF-level rat maintenance feed is provided by Australian cooperative feed limited company of Beijing Ke, and the license number of the feed production is Kyoho feed (2018) 06073. Drinking clean water through the ROA50 experimental animal drinking system every day, filling the drinking bottle for animals to drink freely, flushing the drinking bottle every day and changing water once, and changing the water bottle and the water nozzle 2 times every week.
2. Method of producing a composite material
2.1 grouping and administration of Experimental animals
Animals were randomly divided into 12 groups, i.e., normal group, model group, and examples 1-10 groups. The dose administered in each of the groups of examples 1 to 10 was 19.5g crude drug/kg (ICR mouse clinical equivalent dose was about 10 times the adult clinical dose). The administration volume of each group is 1ml/100g body weight, and the administration is performed by intragastric administration every day for 7 days.
2.2 Experimental animal modeling method
And (3) performing intragastric administration on the 8% senna leaf powder according to the dose of 1g crude drug/kg to establish an animal model of diarrhea-predominant irritable bowel syndrome.
2.3 Experimental procedures
Newly received experimental animals, numbered, adaptively raised for 5 days, and randomly divided into the above 12 groups according to body weight. After the quarantine is completed, the administration is started, the traditional Chinese medicine composition for irritable bowel syndrome obtained in examples 1 to 10 is respectively administered to groups of examples 1 to 10 by intragastric administration every day, and deionized water with the same volume is administered to a normal group and a model group 1 time a day for 7 days continuously. After 30min of the last administration, the model group and the mice of the groups of the examples 1 to 10 were administered 8% senna powder by gavage at a dose of 1g crude drug/kg, and the normal group was administered deionized water of the same volume. The mice in each group were observed for loose stool rate, loose stool grade, and diarrhea index over 6 h.
2.4 statistical analysis
Performing One-Way ANOVA (One-Way ANOVA) by using SPSS 22.0 statistical software, firstly performing homogeneous variance test by using Leven's, performing homogeneous variance test (P > 0.05), and performing comparative test between groups by using Dunnett t test; if the variance is not uniform (P.ltoreq.0.05), a comparison test between groups is carried out with Dunnett T3. All statistical tests were two-sided t-tests with significance levels set at 0.05 and 0.01.
3. Results
Compared with the model group, the mice of the examples 1-10 groups have obviously reduced index of loose stool rate, level of loose stool and diarrhea (P<0.05) as shown in table 1.
4. Conclusion
The traditional Chinese medicine composition for treating irritable bowel syndrome provided by the invention can effectively improve diarrhea-type irritable bowel syndrome of an ICR mouse.

Claims (10)
1. The traditional Chinese medicine composition for irritable bowel syndrome is characterized by comprising the following traditional Chinese medicines in parts by weight: 15-20 parts of qi-tonifying and spleen-tonifying traditional Chinese medicine, 15-20 parts of dampness-eliminating and spleen-tonifying traditional Chinese medicine, 20-30 parts of liver-softening and pain-relieving traditional Chinese medicine, 7-12 parts of liver-regulating and spleen-soothing traditional Chinese medicine, 10-15 parts of liver-soothing and depression-relieving traditional Chinese medicine, 4-8 parts of middle-warming and pain-relieving traditional Chinese medicine, 10-15 parts of blood-cooling and dysentery-stopping traditional Chinese medicine, 10-15 parts of stagnation-removing and stagnation-removing traditional Chinese medicine and 4-8 parts of acute pain-relieving traditional Chinese medicine.
2. The traditional Chinese medicine composition for irritable bowel syndrome according to claim 1, wherein the qi and spleen invigorating herbs are at least one selected from the group consisting of codonopsis pilosula, pseudostellaria root, astragalus root, ginseng, dioscorea opposita thunb, jujube and poria cocos.
3. The traditional Chinese medicine composition for irritable bowel syndrome according to claim 2, wherein the qi and spleen invigorating class of traditional Chinese medicines is selected from at least one of codonopsis pilosula, astragalus membranaceus, ginseng and poria cocos.
4. The traditional Chinese medicine composition for irritable bowel syndrome according to claim 1, wherein the dampness-drying and spleen-invigorating herbs are at least one selected from the group consisting of coix seed, polyporus umbellatus, alisma orientale, white atractylodes rhizome, amomum villosum, chaenomeles sinensis, dried orange peel, areca peel and white mulberry root-bark.
5. The traditional Chinese medicine composition for irritable bowel syndrome according to claim 4, wherein the dampness-drying and spleen-invigorating herbs are at least one selected from coix seeds, polyporus umbellatus, roasted white atractylodes rhizome and pawpaw.
6. The composition of claim 1, wherein the liver-soothing and pain-relieving herbs are selected from at least one of Dang Gui, Chuan Xiong, Wu Zhu Yu, Shi Zhu, Bai Shao and Chuan Lian Zi; and/or the liver-regulating and spleen-soothing traditional Chinese medicine is selected from at least one of divaricate saposhnikovia root, costustoot and green tangerine orange peel; and/or the traditional Chinese medicine for soothing liver-qi stagnation is selected from at least one of radix curcumae, rhizoma cyperi, radix bupleuri, akebia seed and fingered citron; and/or the middle-warmer warming and pain relieving Chinese medicinal materials are selected from at least one of Perillae herba, rhizoma Zingiberis Preparata, rhizoma Alpiniae Officinarum and fructus Zanthoxyli; and/or the blood-cooling dysentery-stopping traditional Chinese medicine is selected from at least one of eclipta, purslane, pulsatilla root, indigo naturalis and brucea javanica; and/or the traditional Chinese medicines with effects of resolving food stagnation and removing food stagnation are selected from at least one of fructus Aurantii, endothelium corneum Gigeriae Galli, rhizoma Zingiberis recens, parched Raphani semen, fructus crataegi preparata, fructus Hordei Germinatus, parched Massa Medicata Fermentata, herba Agastaches, semen Alpiniae and semen Myristicae; and/or the traditional Chinese medicine for relieving spasm and pain is selected from at least one of trogopterus dung, honey-fried licorice root and turmeric.

The method for preparing a traditional Chinese medicine composition for irritable bowel syndrome according to any one of claims 1 to 6, comprising the following steps: weighing the traditional Chinese medicines according to the weight part ratio in the traditional Chinese medicine composition for irritable bowel syndrome, mixing, decocting with water for extraction, filtering, and collecting the obtained filtrate.

The method of preparing the traditional Chinese medicine composition for irritable bowel syndrome according to any one of claims 1 to 6, comprising the following operations: weighing the traditional Chinese medicines according to the weight part ratio in the traditional Chinese medicine composition for irritable bowel syndrome, respectively decocting and extracting the traditional Chinese medicines with water, filtering, collecting obtained filtrate, and mixing.
9. Use of the Chinese medicinal composition for irritable bowel syndrome according to any one of claims 1 to 6 for the preparation of a medicament for treating diarrhea-predominant irritable bowel syndrome.
10. A medicament for treating diarrhea-predominant irritable bowel syndrome, which comprises the Chinese medicinal composition for treating the diarrhea-predominant irritable bowel syndrome according to any one of claims 1 to 6.
