Description

Traditional Chinese medicine composition and medicine for irritable bowel syndrome and preparation method and application thereof
Technical Field
The invention relates to the technical field of traditional Chinese medicines, in particular to a traditional Chinese medicine composition for irritable bowel syndrome, a medicine, a preparation method and application.
Background
Irritable Bowel Syndrome (IBS) is a common bio-psychological-social disorder, which is a gastrointestinal dysfunction disease. The pathogenic causes are complex, including genetic factors, mental factors, dietary factors, infectious factors and the like. IBS is classified into four clinical types of diarrhea type, constipation type, mixed type and amorphous type according to the characteristics of stool, and IBS patients in China are more frequently seen with diarrhea IBS.
The diarrhea type irritable bowel syndrome clinically manifests as continuous or intermittent diarrhea, but lacks definite basis of morphological and laboratory abnormal changes and the like, and can occur in various states such as emotional stress, stay up, dietary stimulation and the like. Any minor irritation during the period of onset may cause significant clinical symptoms, severely affecting the quality of life of the affected population.
The existing treatment method for diarrhea type irritable bowel syndrome mainly aims at symptomatic diarrhea, but antidiarrheal medicines only have temporary effects, have short acting time and poor treatment effect, and can aggravate intestinal motility dysfunction if being improperly used.
Disclosure of Invention
Aiming at the technical problems that the existing antidiarrheal medicine for diarrhea type irritable bowel syndrome has poor treatment effect and possibly aggravates intestinal peristalsis dysfunction, the invention provides a traditional Chinese medicine composition for diarrhea type irritable bowel syndrome, a medicine and a preparation method and application thereof.
In order to achieve the above purpose, the embodiment of the invention adopts the following technical scheme:
On one hand, the embodiment of the invention provides a traditional Chinese medicine composition for treating irritable bowel syndrome, which comprises the following traditional Chinese medicines in parts by weight: 15-20 parts of qi-tonifying spleen-strengthening traditional Chinese medicine, 15-20 parts of dampness-eliminating spleen-strengthening traditional Chinese medicine, 20-30 parts of liver-soothing pain-relieving traditional Chinese medicine, 7-12 parts of liver-regulating spleen-soothing liver-qi depression-relieving traditional Chinese medicine, 10-15 parts of middle-warming pain-relieving traditional Chinese medicine, 4-8 parts of blood-cooling dysentery-relieving traditional Chinese medicine, 10-15 parts of stagnation-removing traditional Chinese medicine and 4-8 parts of acute pain-relieving traditional Chinese medicine.
The invention takes qi-tonifying spleen-strengthening traditional Chinese medicines as monarch medicines, and has the effects of invigorating primordial qi and strengthening spleen and stomach; taking traditional Chinese medicines with the functions of eliminating dampness and strengthening spleen as ministerial medicines, strengthening spleen to prevent diarrhea by taking the wood as a guide, and eliminating dampness to stop diarrhea; the liver-softening and pain-relieving traditional Chinese medicine is used for relieving abdominal pain, the liver-regulating and spleen-smoothing traditional Chinese medicine is used for preventing liver qi from being taken by spleen, and the dampness-eliminating and spleen-invigorating traditional Chinese medicine is used for eliminating dampness and relieving diarrhea, the liver-soothing and depression-relieving traditional Chinese medicine is used for relieving emotion, and the liver-soothing and pain-relieving traditional Chinese medicine, the blood-cooling and diarrhea-relieving traditional Chinese medicine and the stagnation-removing traditional Chinese medicine are used together as adjuvant drugs; takes the traditional Chinese medicines of the group of slow-release pain-relieving medicines as guiding drugs. The whole formula is prepared by combining the traditional Chinese medicines with the functions through monarch, minister, assistant and guide, and the obtained traditional Chinese medicine composition can correct the intestinal motility dysfunction, has good effect of improving the clinical symptoms of diarrhea type irritable bowel syndrome, and can avoid recurrence.
In actual use, 1g of the medicine is used in an amount of 1-2 doses per day according to the illness state.

The traditional Chinese medicine composition for treating irritable bowel syndrome obtained in the examples 1-10.
The clinical dosage of the adult is 1.95g crude drug/kg calculated by 60kg of body weight.
1.3 Feed stuff
SPF-class rats maintained feed, supplied by Australian feed Co., beijing, feed production license number was Beijing feed (2018) 06073. The clean water of the ROA50 experimental animal drinking system is drunk daily, the drinking bottle is filled for animals to drink freely, the drinking bottle is washed and changed once daily, and the water bottle and the water nozzle are changed 2 times per week.
2. Method of
2.1 Grouping and administration of laboratory animals
Animals were randomly divided into 12 groups, namely, normal group, model group, and examples 1-10. The dose of the group of examples 1 to 10 was 19.5g crude drug/kg (ICR mice had a clinical equivalent dose of about 10 times the clinical dose of adults). Each group had a dosage volume of 1ml/100g body weight and was administered by intragastric administration daily for 7 consecutive days.
2.2 Experimental animal modeling method
8% Senna leaf powder is administrated by lavage according to the dosage of 1g crude drug/kg, and an animal model of diarrhea type irritable bowel syndrome is established.
2.3 Experimental procedure
The newly received experimental animals are numbered and adaptively fed for 5 days, and are randomly divided into the above 12 groups according to the weight. Administration is started after quarantine is completed, the traditional Chinese medicine composition for irritable bowel syndrome obtained in the examples 1-10 is respectively administered by stomach irrigation in the examples 1-10 each day, deionized water with the same volume is administered in the normal group and the model group, and the administration is carried out 1 time each day for 7 continuous days. After the last administration for 30min, 8% senna powder was administered by gavage to the model group and the mice of examples 1 to 10 according to the dose of 1g crude drug/kg, and the same volume of deionized water was administered to the normal group. The rare rate, rare grade and diarrhea index of each group of mice were observed within 6 hours.
2.4 Statistical analysis
Single factor analysis of variance (One-Way ANOVA) was performed using SPSS 22.0 statistical software, first with level's for variance alignment (P > 0.05), with Dunnett t test for group comparison test; if the variance is not uniform (P.ltoreq.0.05), then the inter-group comparison test is performed with Dunnett T3. All statistical tests were double-sided t-test, with significance levels set at 0.05 and 0.01.
3. Results
The rare rate, rare grade and diarrhea index were significantly reduced (P < 0.05) in the mice of examples 1-10 as compared to the model group, as shown in table 1.
