Example 9 antidiarrheal Effect experiment
The effect of the massage cream prepared by the traditional Chinese medicine composition on the number of grains of diarrhea-induced mice caused by senna leaves is examined, so that the antidiarrheal effect of the traditional Chinese medicine composition is tested.
Experimental animals: healthy Kunming mice, five weeks old, weighing 18-22g, clean grade, 105, randomized into 7 groups: normal control group, model control group, experimental group 1-experimental group 5, 15 per group. The above experimental animals were all supplied by the university of south medical science laboratory center (license number: SCXK (Guangdong) 2011-0015). The indoor temperature is 24+/-1 ℃, and the test is carried out after the adaptive feeding is carried out for 5 days.
The diarrhea model of the mice is established by using the senna leaf decoction, and the preparation method of the senna leaf decoction is as follows: soaking 15g of folium sennae in 100ml of water for 15 min, boiling with medium fire for 15 min, stopping fire, and filtering to obtain decoction.
Test drug: the test drug of experiment group 1 was the massage cream prepared in example 6-1 (the Chinese medicinal composition prepared in example 1 was added); the test drugs for experimental groups 2-5 were: the Chinese medicinal compositions of examples 2-5 were added respectively, and the massage cream prepared by the method of example 6-1 was used; namely, the massage cream of experimental group 1-5 was different in that the added Chinese medicinal composition was different, and the other components and the preparation method were the same as those of example 6-1.
Normal control mice were not treated at all; taking a model control group and an experimental group, and fasted for 24 hours, and pouring the senna leaf decoction into the stomach according to the ratio of 0.15mL/10g by using a 12-number 55mm small-head curved stomach-pouring device; then, mice in a model control group are not treated, corresponding massage cream is smeared on Guanyuan acupoint, zusanli acupoint and nearby areas of the mice in an experimental group 1-5, the smearing amount is 0.1mg, each time of smearing is 1 minute, the administration is carried out 2 times, and the interval time is 2 hours; after administration, each group of mice was allowed to rest and eat normally, the mice were observed and the total particle count of wet feces discharged from each group of mice was calculated for 24 hours. The test results are shown in Table 5.
TABLE 5
Group of Number of animals Wet manure grain number
Normal control group 15 15.23±1.79
Model control group 15 25.87±1.68
Experiment group 1 15 16.47±1.57
Experiment group 2 15 20.67±1.49
Experiment group 3 15 20.28±1.69
Experiment group 4 15 21.79±1.59
Experiment group 5 15 22.78±1.64
From the above results, it can be seen that: the number of wet manure particles discharged by the mice in the model control group for 24 hours is obviously higher than that of the mice in the normal group, which indicates that the diarrhea model of the mice is successfully established. The mice in experimental groups 1-5 have a certain reduction in the number of wet feces discharged in 24 hours compared with the model control group, which indicates that the massage cream prepared by the traditional Chinese medicine composition can reduce the number of feces discharged by mice with diarrhea induced by senna leaves, and has a better antidiarrheal effect, wherein the massage cream prepared by the traditional Chinese medicine composition of example 1 has the best antidiarrheal effect.
