1. Test drugs
Example 6-1 massage cream for tonifying fire, supporting yang, warming the middle-jiao and dispelling cold
2. Test animal
20 healthy white rabbits (single skin irritation test) were used for both males and females, weighing (2.0+0.2) kg; 50 Kunming mice (multiple skin irritation tests) are used for both male and female, and the weight is 18-22 g. The experimental animals are provided by southern medical university laboratory center (license number: SCXK (Yue) 2011-. The animals are all bred in a single cage, fed freely and drunk freely, and are adapted to the environment of an animal house by feeding the animals regularly for 5 days before experiments.
3. Test methods and results
3.1 methods
The skin irritation test method and procedure of single administration and multiple administration were performed according to the skin irritation test method and procedure of "cosmetic safety specification 2015", wherein the test group was applied with the massage cream for tonifying fire and supporting yang, warming middle-jiao and dispelling cold of example 6-1 of the present invention, and the control group was not applied with any substance.
3.2 results
3.2.1 Single-dose skin irritation test, in which all rabbits were coated with 0.2g/kg of the test substance on the depilated skin area on one side and the skin on the other side was used as a control. The skin irritation intensity of the massage cream of example 6-1 was judged by applying the skin irritation cream for 2 hours in a closed test, removing the test substance, observing the skin reaction at the applied part for 1, 24, 48 and 72 hours, respectively, and observing the skin irritation intensity of the massage cream of example 6-1 according to the observation for 1, 24, 48 and 72 hours. The results are shown in Table 3.
TABLE 3
Figure BDA0003488069240000181
The results show that: 1. the skin reaction of the smearing part is observed for 24, 48 and 72 hours, and no erythema or edema, pimple, congestion and ulcer are observed. The total evaluation score and the average score of the skin irritation response are both 0, and the average response irritation intensity is less than 0.5, which indicates no irritation.
3.2.2 skin irritation test with multiple administrations, in the test process, 0.5g of the test substance is smeared on the unhaired skin area of one side of all Kunming mice, and the skin of the other side is used as a control. Applied 1 time per day for 14 days. From the next day, the hair should be cut and the residual test substance removed with water or a non-irritating solvent before each application. The results were observed for one hour. The results of judging the skin irritation intensity of the massage cream of example 6-1 by a plurality of times are shown in Table 4.
TABLE 4
Figure BDA0003488069240000182
Figure BDA0003488069240000191
The results show that 50 tested Kunming mice, 1 of which showed erythema and 1 of which showed papules, were scored on average less than 0.5 per animal per day on the first day; the next day 50 test Kunming mice, one of which developed pimples with an average score per animal of less than 0.5 per day; on days 3-14, all of the Kunming mice tested did not develop papules, erythema, congestion, ulcers. Multiple skin irritation levels were non-irritating according to standard evaluation criteria.
Example 9 antidiarrheal Effect test
In this example, the antidiarrheal effect of the Chinese medicinal composition of the present invention was tested by examining the effect of the massage cream prepared from the Chinese medicinal composition of the present invention on the number of defecation granules of mice with senna leaf-induced diarrhea.
Experimental animals: healthy Kunming mice, five weeks old, 18-22g in weight, clean grade, 105 mice, randomly divided into 7 groups: normal control group, model control group, experimental group 1-experimental group 5, 15 of each group. The experimental animals are provided by southern medical university laboratory center (license number: SCXK (Yue) 2011-. The indoor temperature is 24 +/-1 ℃, and the test is carried out after 5 days of adaptive feeding.
Establishing a mouse diarrhea model by using senna leaf decoction, wherein the preparation method of the senna leaf decoction comprises the following steps: soaking 15g folium sennae in 100ml water for 15 min, boiling with middle fire for 15 min, stopping fire, and filtering to obtain folium sennae decoction).
The tested drugs are: the test drug of experimental group 1 was the massage cream prepared in example 6-1 (the Chinese medicinal composition prepared in example 1 was added); the tested drugs of experimental groups 2-5 were: the massage creams prepared by the method of the embodiment 6-1 are added with the Chinese medicinal composition of the embodiment 2-5 respectively; namely, the massage creams of the experimental groups 1 to 5 are different in the added Chinese medicinal composition, and the other components and the preparation method are the same as those of the massage cream of the example 6-1.
The normal control group mice were not treated at all; taking model control group and experimental group 1-5, fasting for 24 hr, and performing intragastric administration with a 12 # 55mm small-head bent intragastric administration device according to 0.15mL/10g folium sennae decoction; then, the mice of the model control group are not treated, and the experimental groups 1-5 coat corresponding massage creams on the Guanyuan acupoint, the Zusanli acupoint and the nearby area of the mice, wherein the coating amount is 0.1mg, the coating is carried out for 1 minute each time, the administration is carried out for 2 times, and the interval time is 2 hours; after the administration, each group of mice was normally rested and fed with a diet, the mice were observed and the total number of wet feces excreted for 24 hours from each group of mice was calculated. The test results are shown in table 5.
TABLE 5
Group of Number of animals Number of wet manure particles
Normal control group 15 15.23±1.79
Model control group 15 25.87±1.68
Experimental group 1 15 16.47±1.57
Experimental group 2 15 20.67±1.49
Experimental group 3 15 20.28±1.69
Experimental group 4 15 21.79±1.59
Experimental group 5 15 22.78±1.64
From the above results it can be seen that: the number of wet feces discharged by the mice in the model control group in 24 hours is obviously higher than that of the mice in the normal group, which indicates that the diarrhea model of the mice is successfully established. The number of wet feces grains discharged by the mice in the experimental groups 1-5 in 24 hours is reduced to a certain extent compared with that of a model control group, which shows that the massage cream prepared by the traditional Chinese medicine composition can reduce the number of the feces grains of the mice with senna leaf-induced diarrhea and has a good antidiarrheal effect, wherein the massage cream prepared by the traditional Chinese medicine composition in the embodiment 1 has the best antidiarrheal effect.
