Test animals and groups
Selecting 60 ternary sows (Duroc, Dabai and Changbai) with consistent gestation times and similar weight, randomly dividing the 60 sows into three groups which are respectively a blank group, a test group and a control group by adopting a single-factor influence design test; wherein the blank group of drinking water and feed maintains the original standard unchanged; test group 1000g of the powder prepared according to the formulation of example 5 was added to each 100kg of feed; in the control group, 1000g of Sijunzi powder is added into every 100kg of feed, namely the powder prepared by the formula in the comparative example 1; the preparation method of the powder comprises the following steps: cleaning and drying the raw materials, weighing the raw materials according to the weight parts of each formula, crushing, sieving by a 80-mesh sieve, and uniformly mixing to obtain the powder. Each group of test pigs eat and drink water freely, the breeding standards are kept consistent, and the immune disinfection procedure is carried out according to the conventional method of a pig farm; the test is carried out by pre-feeding for 7 days before the test, and the formal test time is 30 days.
2 blood sample Collection and detection
Ear vein blood collection was performed on each test pig group on day 0, 14 and 28 of the main test period, and the blood was divided into two portions, one portion was anticoagulated with EDTA at a final concentration of 1.5mg/mL, and the other portion was not anticoagulated. Wherein the anticoagulant is added into the sample, and the blood component indexes including the content of Red Blood Cells (RBC), Hemoglobin (HGB) and average red blood cell hemoglobin (MCH) are measured by a red blood cell analyzer; blood without anticoagulant was centrifuged and plasma was collected and the plasma was assayed for cortisol and IgG content using a cortisol and immunoglobulin G (IgG) kit.
3 data analysis and processing
Data analysis statistical tests were performed on all data using SAS9.0 software and results are expressed as mean ± standard deviation and as (P <0.05) for significant differences.
3. Test result and evaluation of biochemical indexes of test pig blood
Table 1 shows the results of the powder prepared by the formula of the invention and the Sijunzi powder on the biochemical indexes of the blood of the test pigs. From the results in Table 1, it can be seen that the hemoglobin content of the blank group was increased by 0% and 2.8% on days 14 and 28 of the test, respectively, as compared with the case of day 0; the hemoglobin content of the test group is respectively improved by 18.3 percent and 19.2 percent; the hemoglobin content of the control group is respectively improved by 7.5 percent and 9.3 percent; compared with the blank group, the test group and the control group have the contents of red blood cells, hemoglobin and average hemoglobin which are obviously higher than those of the blank group, and have obvious difference (P <0.05), which shows that the oxygen carrying capacity of the red blood cells can be improved by adding the natural plant composition as a feed additive, and the contents of the red blood cells, the hemoglobin and the average hemoglobin of the test group are also obviously higher than those of the control group, so that the formula powder can enhance the oxygen carrying capacity of the red blood cells and obviously improve the health level of test animals compared with Sijunzi powder.
The results in table 1 show that the content of IgG in the test group and the control group is gradually reduced along with the increase of the feeding time, and the reduction range of the content of IgG in the test group is more obvious than that in the control group, and has a significant difference (P <0.05), which indicates that the powder of the invention can reduce the stress response of the body to the outside compared with Sijunzi san.
As can be seen from the results in table 1, the cortisol content of the vermilion blank was reduced by 1.2% and 2.1% on days 14 and 28 of the test, respectively, as compared with the case of day 0; the cortisol content of the test group is reduced by 8.5 percent and 25.7 percent respectively; the cortisol content of the control group was reduced by 6.2% and 4.2%, respectively; compared with the blank group, the cortisol content of the test group and the control group is obviously lower than that of the blank group, the difference is obvious (P is less than 0.05), the fact that the stress response of the animal can be reduced by adding the natural plant composition as the feed additive is proved, the cortisol content of the test group is obviously lower than that of the control group, and the fact that the powder prepared by the formula of the invention can reduce the stress response of the test animal and enhance the adaptability of the test animal to the external environment compared with the Sijunzi san is proved.
