Abstract
The invention relates to an anti-depression traditional Chinese medicine formula and anti-depression essential oil, wherein the anti-depression traditional Chinese medicine formula comprises lavender oil, vetiver oil, jasmine oil, bergamot oil, sandalwood oil and geranium oil, is a composition of natural plant extracted essential oil, has wide raw material sources and simple preparation process, and has the effects of improving appetite decrease, locomotion reduction and cognitive disorder. And further develops an anti-depression essential oil which can specifically relieve depression mood, and can effectively improve symptoms of low mood, physically stiff, inappetence and slow reaction.
Description
Technical Field
The invention belongs to the technical field of traditional Chinese medicine compositions, and particularly relates to an anti-depression traditional Chinese medicine composition and anti-depression essential oil.
Background
With the development of society and economy, the rhythm is accelerated, the competition is fierce, and the incidence of various psychosomatic diseases in people of different ages is continuously increased under the influence of psychological factors, social factors, bad behaviors of human beings and life styles. A survey promulgated by the world health organization indicates that depression due to emotional stress is in the region of 2 to 4 million people worldwide each year, with 65% of U.S. professionals being mindful of negative emotions. Depression in the present invention refers to Major depressive disorder (Major depressive disorder): the three-low symptom is clinically called as the primary core symptom by the emotional depression, slow thinking and hypokinesia. The patients have extremely bad self-feelings, low emotions, slow response to the outside, slow association, and reduced speech movements, and have self-inferior, self-responsibility and self-guilt concepts. According to the current diagnostic classification, depression and reactive depression are commonly seen, and also trauma and toxic mental disorder are seen.
The action of aromatic plants is closely related to their action pathways. The most widely used and studied methods are currently the aromatherapy inhalation and the aromatherapy transdermal absorption. The aromatic transdermal absorption means that aromatic substances enter blood circulation (cell tissues such as nerve, bone, muscle and the like can be influenced) through skin in a short time by a massage method so as to achieve the purpose of preventing and treating diseases. The aromatic inhalation therapy is a therapy which directly acts on the central nervous system through olfactory pathway, and simultaneously enters blood circulation through gas exchange through the lung to treat diseases. Compared with the drug therapy, the aromatic inhalation therapy and the aromatic transdermal absorption therapy have the advantages of low irritation, small side effect and no influence on the immune system of a human body. Chinese patent with publication number CN106853172A discloses an antidepressant compound essential oil and application thereof, and the improvement effect is not obvious. Therefore, the invention of an anti-depression product which can effectively improve depression symptoms is very important.
Disclosure of Invention
The invention aims to provide an anti-depression traditional Chinese medicine formula which is a composition of natural plant extracted essential oil, has wide raw material sources and simple preparation process, and has the effects of improving appetite reduction, mobility reduction and cognitive impairment. And further develops an antidepressant essential oil which can effectively regulate low-falling mood and improve symptoms of body stiffness, inappetence and slow reaction.
The invention aims to provide an anti-depression traditional Chinese medicine formula.
Another object of the invention is to provide an antidepressant essential oil.
The above object of the present invention is achieved by the following technical solutions:
an antidepressant Chinese medicinal composition comprises oleum Lavandula Angustifolia, vetiver root oil, flos Jasmini sambac oil, Olium Citri Sarcodactylis, oleum Santali albi and Pelargonium graveolens oil.
In the formula, the effects of the components are as follows:
lavender oil: the lavender is called 'the king of all grass', has fresh and elegant fragrance and mild property, and is a plant which is acknowledged to have the effects of calming, soothing and hypnotizing. Relieving tension, calming mind, calming the nerves, healing wound, removing scar, controlling oil, regenerating, diminishing inflammation, and repairing. Lavender oil is a complex mixture of many different types of aromatic compounds, with over 30 components, the main components linalool, linalyl acetate, eucalyptol, B-rhomenene (including cis and trans) pairs, lavandula acetate, lavandulol, terpene-4-ol, and camphor, among others.
Vetiver oil: the appellation of "sedative essential oils" is known and clearly its appellation is indeed superior. It has effects in cleaning skin, astringing, and resisting infection; conditioning greasy and dirty skin; anti-inflammatory and antibacterial effects, and can be used for treating acne and skin ulcer; treating beriberi and various skin inflammations; awakening cells and improving damaged skin; repelling mosquitoes and flies, relieving itching and resisting bacteria.
Jasmine oil: pungent, sweet and warm. Regulating qi, resolving stagnation, removing filth, and regulating the middle warmer. It can be used for treating abdominal pain due to dysentery, conjunctival congestion, red swelling, and sore. The jasmine flower can be used for extracting jasmine flower oil, and the oil mainly contains benzyl alcohol and its esters, jasminoidin, linalool, and linalyl benzoate. The root contains alkaloid and sterol. Animal experiments show that the jasmine root alcohol immersion liquid can obviously reduce spontaneous activity of mice; the sleep time of the mice caused by the hexabarbital can be prolonged; can reduce the passive activity of the white mouse. Therefore, it is considered that jasmine root has an inhibitory effect on the central nervous system. Flos Jasmini sambac, leaf and root can be used for medicine, and generally root is dug after autumn, sliced and dried for use; collected in summer and autumn and dried in the sun for later use.
Bergamot oil: pungent, bitter, sweet, warm and nontoxic; it has the functions of regulating qi, eliminating phlegm, stopping vomiting, relieving flatulence, soothing liver, strengthening spleen, regulating stomach and other medicinal functions. It has obvious relieving effect on tracheitis and asthma of the old; has more obvious curative effect on dyspepsia and chest and abdomen swelling and stuffiness of general people.
Sandalwood oil: has effects in relaxing, tranquilizing, and resisting anxiety, depression and nervous tension. Experiments show that the sandalwood oil has the functions of diminishing inflammation and sterilizing, and can be clinically used for treating skin diseases.
Geranium oil: most terpene compounds such as citronellal and the like are used, and the compounds have the effects of resisting bacteria, diminishing inflammation, eliminating phlegm and relieving cough, and researches show that the geranium oil has the effects of balancing and enhancing skin compliance, moisturizing and repairing skin, improving rough and dry skin, diminishing inflammation, calming, stopping bleeding, promoting wound healing and the like.
Preferably, the antidepressant traditional Chinese medicine formula comprises the following components in parts by weight: 20-30 parts of lavender oil, 5-10 parts of vetiver root oil, 10-15 parts of jasmine oil, 10-30 parts of bergamot oil, 5-10 parts of sandalwood oil and 10-20 parts of geranium oil.
More preferably, the antidepressant traditional Chinese medicine formula comprises the following components in parts by weight: 30 parts of lavender oil, 5 parts of vetiver root oil, 8 parts of jasmine oil, 28 parts of bergamot oil, 7 parts of sandalwood oil and 16 parts of geranium oil.
The components in the anti-depression traditional Chinese medicine formula can be obtained commercially or prepared automatically.
As a preferred possible embodiment, the lavender oil, vetiver root oil, jasmine oil, bergamot oil, sandalwood oil, and geranium oil may be extracted by steam distillation.
The invention also provides an anti-depression essential oil containing the anti-depression traditional Chinese medicine formula, and the anti-depression essential oil consists of the anti-depression traditional Chinese medicine formula and absolute ethyl alcohol.
Preferably, the antidepressant essential oil comprises the following components in parts by weight: 20-30 parts of lavender oil, 5-10 parts of vetiver root oil, 8-15 parts of jasmine oil, 10-30 parts of bergamot oil, 5-10 parts of sandalwood oil, 10-20 parts of geranium oil and 50-100 parts of absolute ethyl alcohol.
More preferably, the antidepressant essential oil comprises the following components in parts by weight: 30 parts of lavender oil, 5 parts of vetiver root oil, 8 parts of jasmine oil, 28 parts of bergamot oil, 7 parts of sandalwood oil and 16 parts of geranium oil.
The invention also provides a preparation method of any one of the anti-depression essential oils, which comprises the following steps:
s1, extracting essential oil of lavender, vetiver roots, jasmine flowers, bergamot, sandalwood and pelargonium graveolens respectively by using a steam distillation method.
S2, uniformly mixing the essential oil extracted in the step S1 and absolute ethyl alcohol under the stirring condition to obtain the essential oil.
Preferably, the distillation time of the steam distillation method in the step S1 is 2-5 h.
Preferably, the rotation speed of the stirring condition in the step S2 is 900-1000 r/min.
Preferably, the stirring condition of step S2 is for 3-6 min.
Compared with the prior art, the invention has the following beneficial effects:
(1) the invention provides an anti-depression traditional Chinese medicine formula which is prepared by extracting essential oil from natural plants, has wide and easily obtained raw material sources, is safe and has no toxic or side effect, and has the effects of improving appetite reduction, mobility reduction and cognitive impairment.
(2) The invention also provides an anti-depression essential oil, wherein the essential oil of each component in the anti-depression essential oil plays a synergistic role, and further develops the anti-depression essential oil which can effectively aim at the expression symptoms of depression and has the functions of regulating low-grade mood, improving body stiffness, inappetence and slow reaction.
Detailed Description
The present invention is further illustrated by the following specific examples, which are not intended to limit the invention in any way. Reagents, methods and apparatus used in the present invention are conventional in the art unless otherwise indicated.
Unless otherwise indicated, reagents and materials used in the following examples are commercially available.
Example 1 antidepressant Chinese medicinal composition and preparation thereof
1. Preparation of anti-depression Chinese medicine composition
The lavender oil, vetiver oil, jasmine oil, bergamot oil, sandalwood oil and geranium oil are extracted by a steam distillation method, and the specific extraction steps are as follows:
(1) 200g of the pulverized sample powder was weighed and placed in a 5000mL round-bottom flask.
(2) 3500mL of deionized water is added for soaking for 12 h.
(3) Distilling with water vapor for 4 hr to obtain oleum Lavandula Angustifolia, vetiver root oil, flos Jasmini sambac oil, Olium Citri Sarcodactylis, lignum Santali albi oil, and Pelargonium graveolens oil.
(4) And stirring and mixing the obtained essential oil uniformly according to different proportions, wherein the rotating speed of the stirring condition is 900r/min, and the stirring time is 3 min.

EXAMPLE 2 antidepressant essential oils and their preparation

2. Preparation method of antidepressant essential oil

(4) Mixing the obtained oleum Lavandula Angustifolia, vetiver root oil, flos Jasmini sambac oil, Olium Citri Sarcodactylis, lignum Santali albi oil, Pelargonium graveolens oil and anhydrous alcohol at different ratios, stirring at 1000r/min for 5 min.
Example 3 efficacy testing of antidepressant essential oils
The positive effect of the present invention in improving the symptoms of depression is further illustrated by animal experimental data below.
The invention adopts an animal model for promoting blood circulation and suppressing depression, and researches the influence of the novel antidepressant compound on the behavior of a depression-like mouse by a mode of intraperitoneal administration.
1. Experimental animals: the male BALB/C mouse has the weight average of 20-22 g, is provided by the medical experimental animal center of Guangdong province, receives 12h of light/12 h of darkness every day, has the light cycle of 7: 00-19: 00, the temperature of a laboratory of 25 +/-2 ℃ and the humidity of 60 percent, can automatically take standard feed and clean water, and complies with the ethical requirements of international experimental animals in animal experiment operation. Two weeks after acclimation in the laboratory, mice were randomly divided into 16 groups of 8 mice per group by body weight.
2. Drug management: 1-13 parts of sleep-improving essential oil prepared according to each formula in table 2.
3. Building a blood-benefiting depression-suppressing animal model:
mice in a depression group need to be injected with reserpine in the abdominal cavity once, and the reserpine is used before injection. The specific experimental procedure is as follows:
a solution of reserpine was prepared by dissolving a quantity of reserpine in a dimethyl sulfoxide/normal saline mixture (0.1: 10 ml). The intraperitoneal injection of reserpine (6mg/kg) was given once at an amount of 0.1mL/10g of the body weight of the mice, and this dose resulted in depression symptoms lasting for 72 hours after the injection. The normal control group was administered with a DMSO/physiological saline mixture (0.1: 10mL) once per the same day in an amount of 0.1mL/10g of the body weight of the mouse.
4. The experimental method comprises the following steps: the 16 groups of mice were set as: normal control A control group of 0.9% physiological saline, a group of depression plus fluoxetine (200mg/kg), and a group of depression plus essential oil of antidepressant (200mg/kg) were given as normal controls. One day after the completion of intraperitoneal injection of reserpine, mice were administered by intragastric administration according to the above-mentioned groups, with an administration volume of 10ml/kg, i.e. 10ml for 1kg of mice. 1 hour after dosing, the mice were recorded and analyzed for sugar water preference and immobility time in forced swim and tail suspension experiments.
a. Experiment on influence of antidepressant essential oil on sugar water preference of depressed mice
Before the experiment, animals are trained to adapt to drinking sugar water in a noise-isolated and quiet room, 2 water bottles are placed in each cage, 1% of cane sugar water is filled in the two bottles in the first 24 hours, and 1% of cane sugar water and purified water are filled in the bottles in the subsequent 24 hours. Animals were subjected to sugar/water consumption experiments after 24 hours of fasting with water deprivation. Mice were given two bottles of pre-dosed 1% sucrose solution and one bottle of purified water. After 2 hours, two bottles were removed and weighed, and the mice were calculated for sugar water consumption, pure water consumption, sugar water preference (sugar water preference ═ sugar water consumption/total liquid consumption x 100%).
And (4) analyzing results: compared with the normal saline group, the sleep incidence rates of the mice in the essential oil 1-4 groups are significantly different (P <0.05), wherein the sugar water consumption of the essential oil 1 group reaches the highest value and approaches to the control group, which shows that the essential oil 1 has significant antidepressant effect, while the sugar water preference degree of the mice in the normal saline group is significantly lower than that of the control group, and the antidepressant effect of the essential oil 5-13 is not significant (see Table 3).

b. Function of antidepressant essential oil in forced swimming test of depressed mouse
The mice were placed in a glass cup with a water depth of 12cm, a water temperature of 22. + -. 1 ℃ and a water temperature of 21X 12 cm. The forced swim video analysis system recorded 4 minutes, the duration of the mice resting 3 minutes after recording.
And (4) analyzing results: compared with the normal saline group, the essential oil 1-4 group and the fluoxetine group obviously reduce the immobility time in the forced swimming experiment of the mice, namely the depression-like behavior of the mice is improved. The 1-4 groups of the antidepressant essential oil have a remarkable antidepressant effect in the depression model, the 5-13 groups of the essential oil have an unobvious antidepressant effect, and the 1-4 phases of the essential oil have a remarkable antidepressant effect compared with the 5-11 groups of the essential oil which lack one or more components, which shows that different components in the 1-4 formula of the essential oil have a remarkable synergistic effect (see table 4).

c. Effect of antidepressant essential oil in depressed mouse tail suspension experiment
The tip of the experimental mouse tail is fixed at a height of 60cm above the ground and hung, the video is recorded for 3min, and the time of moving is counted.
And (4) analyzing results: compared with the normal saline group, the immobility time of mice of the essential oil groups 1-4 and the fluoxetine group is obviously reduced, and the immobility time of the mice of the essential oil groups 5-13 is longer than that of the essential oil groups 1-4. The results show that the antidepressant essential oils 1-4 can effectively relieve the depression state of the acute stress mice (see table 5).

d. Clinical experiments
100 testers are extracted to try out the antidepressant essential oil 1 and used by fragrance diffusing through a fragrance machine. The improvement in depression was examined once daily, 3 hours each, after 1 month of continuous use by questionnaire form. Depression symptoms were significantly improved in 66% of the subjects, somewhat improved in 21% of the subjects, and not improved in 13% of the subjects.

1. An anti-depression traditional Chinese medicine formula is characterized by comprising the following components: lavender oil, vetiver oil, jasmine oil, bergamot oil, sandalwood oil and geranium oil.
2. The antidepressant traditional Chinese medicine formula according to claim 1, characterized by comprising the following components in parts by weight: 20-30 parts of lavender oil, 5-10 parts of vetiver root oil, 8-15 parts of jasmine oil, 10-30 parts of bergamot oil, 5-10 parts of sandalwood oil and 10-20 parts of geranium oil.

The antidepressant traditional Chinese medicine formula according to claim 2, characterized by comprising the following components in parts by weight: 30 parts of lavender oil, 5 parts of vetiver root oil, 8 parts of jasmine oil, 28 parts of bergamot oil, 7 parts of sandalwood oil and 16 parts of geranium oil.
4. An antidepressant essential oil, characterized by comprising the antidepressant traditional Chinese medicine formula of any one of claims 1 to 3.

The antidepressant essential oil according to claim 4, which is prepared from the following components in parts by weight: 20-30 parts of lavender oil, 5-10 parts of vetiver root oil, 10-15 parts of jasmine oil, 10-30 parts of bergamot oil, 5-10 parts of sandalwood oil, 10-20 parts of geranium oil and 50-100 parts of absolute ethyl alcohol.
6. The antidepressant essential oil according to claim 4, which is characterized by consisting of the following components in parts by weight: 30 parts of lavender oil, 5 parts of vetiver root oil, 8 parts of jasmine oil, 28 parts of bergamot oil, 7 parts of sandalwood oil, 16 parts of geranium oil and 100 parts of absolute ethyl alcohol.
7. The preparation method of the antidepressant essential oil as claimed in any one of claims 4 to 6, characterized by comprising the following steps:
s1, extracting essential oil of lavender, vetiver roots, jasmine flowers, bergamot, sandalwood and pelargonium graveolens respectively by using a steam distillation method.
S2, uniformly mixing the essential oil extracted in the step S1 and absolute ethyl alcohol under the stirring condition to obtain the essential oil.
8. The method according to claim 7, wherein the steam distillation in step S1 is carried out for 2-5 h.
9. The method according to claim 7, wherein the stirring condition of step S2 is at a rotation speed of 900 to 1000 r/min.
10. The method according to claim 7, wherein the stirring in step S2 is carried out for 3-6 min.
