Example 3 efficacy testing of antidepressant essential oils

The invention adopts an animal model for promoting blood circulation and suppressing depression, and researches the influence of the novel antidepressant compound on the behavior of a depression-like mouse by a mode of intraperitoneal administration.
1. Experimental animals: the male BALB/C mouse has the weight average of 20-22 g, is provided by the medical experimental animal center of Guangdong province, receives 12h of light/12 h of darkness every day, has the light cycle of 7: 00-19: 00, the temperature of a laboratory of 25 +/-2 ℃ and the humidity of 60 percent, can automatically take standard feed and clean water, and complies with the ethical requirements of international experimental animals in animal experiment operation. Two weeks after acclimation in the laboratory, mice were randomly divided into 16 groups of 8 mice per group by body weight.

a solution of reserpine was prepared by dissolving a quantity of reserpine in a dimethyl sulfoxide/normal saline mixture (0.1: 10 ml). The intraperitoneal injection of reserpine (6mg/kg) was given once at an amount of 0.1mL/10g of the body weight of the mice, and this dose resulted in depression symptoms lasting for 72 hours after the injection. c. Effect of antidepressant essential oil in depressed mouse tail suspension experiment
The tip of the experimental mouse tail is fixed at a height of 60cm above the ground and hung, the video is recorded for 3min, and the time of moving is counted.
And (4) analyzing results: compared with the normal saline group, the immobility time of mice of the essential oil groups 1-4 and the fluoxetine group is obviously reduced, and the immobility time of the mice of the essential oil groups 5-13 is longer than that of the essential oil groups 1-4. The results show that the antidepressant essential oils 1-4 can effectively relieve the depression state of the acute stress mice (see table 5).

d. Clinical experiments
100 testers are extracted to try out the antidepressant essential oil 1 and used by fragrance diffusing through a fragrance machine. The improvement in depression was examined once daily, 3 hours each, after 1 month of continuous use by questionnaire form. Depression symptoms were significantly improved in 66% of the subjects, somewhat improved in 21% of the subjects, and not improved in 13% of the subjects.

The antidepressant essential oil according to claim 4, which is prepared from the following components in parts by weight: 20-30 parts of lavender oil, 5-10 parts of vetiver root oil, 10-15 parts of jasmine oil, 10-30 parts of bergamot oil, 5-10 parts of sandalwood oil, 10-20 parts of geranium oil and 50-100 parts of absolute ethyl alcohol.

The antidepressant essential oil according to claim 4, which is characterized by consisting of the following components in parts by weight: 30 parts of lavender oil, 5 parts of vetiver root oil, 8 parts of jasmine oil, 28 parts of bergamot oil, 7 parts of sandalwood oil, 16 parts of geranium oil and 100 parts of absolute ethyl alcohol.

s1, extracting essential oil of lavender, vetiver roots, jasmine flowers, bergamot, sandalwood and pelargonium graveolens respectively by using a steam distillation method.
S2, uniformly mixing the essential oil extracted in the step S1 and absolute ethyl alcohol under the stirring condition to obtain the essential oil.

9. The method according to claim 7, wherein the stirring condition of step S2 is at a rotation speed of 900 to 1000 r/min.
