Abstract
The invention discloses a microecological extract with an arteriosclerosis intervention function, which is prepared from the following components in parts by weight: 2-4 parts of garlic, 6-10 parts of black fungus, 5-7 parts of cinnamon, 6-10 parts of Chinese yam, 18-24 parts of grape, 8-12 parts of blueberry, 4-6 parts of hawthorn, 4-6 parts of pomegranate, 4-6 parts of pineapple, 3-5 parts of purple cabbage, 0.8-1.2 parts of peach kernel, 2.5-3.5 parts of ginkgo, 0.08-0.12 part of medlar powder, 0.25-0.35 part of spirulina platensis, 12-15 parts of fructo-oligosaccharide, 1.2-1.8 parts of oat wheat beta-glucan, 0.005-0.015 part of sucralose, 0.8-1.2 parts of kluyveromyces marxianus and 0.0008-0.0012 part of lycopene. The invention also provides a preparation method of the microecological extract. The microecological extract has the effects of arteriosclerosis intervention and the like.
Description
Technical Field
The invention relates to the field of health-care food, in particular to a microecological extract with an arteriosclerosis intervention function, and also relates to a preparation method of the microecological extract.
Background
With the improvement of living standard of people, various substances such as fat, sugar and the like are more and more ingested, but due to the reasons of high working pressure, little exercise, irregular work and rest time and the like at present, great harm is brought to the cardiovascular health of people, wherein the artery sclerosis becomes a common cardiovascular disease which has great harm to human bodies, and serious people can cause hypertension and even corresponding tissue and organ ischemia necrosis. Therefore, people can treat the diseases through chemical medicines, biological medicines, Chinese herbal medicines and other medicines, but the diseases can be treated after the diseases occur, and firstly, the treatment effect is difficult to ensure; secondly, medicines such as chemical medicines can also cause certain influences on the health of human bodies, such as sequelae, adverse reactions and the like, and although the traditional Chinese herbal medicines have small side effects, the traditional Chinese herbal medicines have more impurities, are inconvenient to take and have low extraction efficiency, so that the curative effect and the application are influenced.
For this reason, microbial extraction of Chinese herbal medicines, edible animal and plant foods, has been started to improve the active ingredients and effects:
for example, the Chinese patent publication No. CN102319278B discloses a red yeast rice compound preparation capable of regulating blood pressure and fat and improving arteriosclerosis, coronary heart disease and cerebral embolism; fermenting ginseng and ginkgo leaf liquid to obtain ginseng and ginkgo fermentation liquor, liquid culturing seeds of monascus 9901 or 9906 strain, adding water into ginseng and ginkgo leaf fermentation liquor and grain to prepare solid culture medium, inoculating, solid fermenting to obtain monascus mould, sterilizing malt juice or potato juice, sugar and agar to prepare flat plate, inoculating, culturing to obtain pure monascus mould, preparing strain expanding culture solution, inoculating, strain protecting and culturing, shaking table culturing, sterilizing, inoculating and culturing ginseng and ginkgo leaf extract and solid fermentation nutrient solution, fermenting to obtain solid culture, sterilizing, drying, crushing, mixing to obtain fine powder of monascus, and preparing the preparation according to corresponding preparation process.
However, people find that the existing microecological extraction has some defects through research:
1) the extraction effect and efficiency are not ideal, and the existing microorganism extraction is limited by the differences of fermentation ways and metabolites of different strains, so that the selection and proportion of microorganism strains can greatly influence the extraction efficiency;
2) the absorption efficiency of the extracted product for human body is not high, and the absorption degree of the produced extract for the gastrointestinal environment of human body still has certain deficiency due to the influence of the whole fermentation environment of the existing microbial extract;
3) harmful substances exist, and the existing Chinese herbal medicines and foods inevitably contain some harmful chemical substances such as pesticide ingredients and the like, which bring certain damage to human bodies.
Therefore, there is a need for a microecological extract which has a higher extraction efficiency, a low toxic component content, a high human absorption, and is capable of arteriosclerosis intervention.
Disclosure of Invention
The invention aims to overcome the defects in the prior art and provide a microecological extract with an arteriosclerosis intervention function, and also provides a preparation method of the microecological extract.
In order to achieve the purpose, the invention adopts the following technical scheme:
a microecological extract with an arteriosclerosis intervention function is prepared from the following components in parts by weight: 2-4 parts of garlic, 6-10 parts of black fungus, 5-7 parts of cinnamon, 6-10 parts of Chinese yam, 18-24 parts of grape, 8-12 parts of blueberry, 4-6 parts of hawthorn, 4-6 parts of pomegranate, 4-6 parts of pineapple, 3-5 parts of purple cabbage, 0.8-1.2 parts of peach kernel, 2.5-3.5 parts of ginkgo, 0.08-0.12 part of medlar powder, 0.25-0.35 part of spirulina platensis, 12-15 parts of fructo-oligosaccharide, 1.2-1.8 parts of oat beta-glucan, 0.005-0.015 part of sucralose, 0.8-1.2 parts of kluyveromyces marxianus and 0.0008-0.0012 part of lycopene.
In the invention, a further preferable scheme is that the microecological extract is prepared from the following components in parts by weight: 3 parts of garlic, 8 parts of black fungus, 6 parts of cinnamon, 8 parts of Chinese yam, 21.5 parts of grape, 10 parts of blueberry, 5 parts of hawthorn, 5 parts of pomegranate, 5 parts of pineapple, 4 parts of purple cabbage, 1 part of peach kernel, 3 parts of ginkgo, 0.1 part of medlar powder, 0.299 parts of spirulina platensis, 13.59 parts of fructo-oligosaccharide, 1.5 parts of oat beta-glucan, 0.01 part of sucralose, 1 part of kluyveromyces marxianus and 0.001 part of lycopene.
The invention also provides a preparation method of the microecological extract, which comprises the following steps:
step 1: taking the components according to the formula, mixing, sterilizing, and then breaking the wall by microwave to obtain a crushed raw material mixture;
step 2: carrying out microbial ecological extraction treatment on the raw material mixture obtained in the step 1 by using microbes, then filtering the mixture obtained after the microbial ecological extraction, removing filter residues, and collecting a microbial ecological extracting solution;
and step 3: and (3) sterilizing the microecological extracting solution obtained in the step (2), and then curing to obtain the microecological extract.
In the present invention, it is further preferable that the method further comprises, after the step 3, a step 4: adding food acceptable auxiliary materials into the microecological extract obtained in the step 3.
In the present invention, it is further preferable that step 4 is followed by step 5: heating the microecological extract treated in the step 4 until the auxiliary materials are dissolved, and then sterilizing; the adjuvant is soluble in water.
In the present invention, it is further preferable that the sterilization is performed by ozone.
In the invention, a further preferable scheme is that the extraction temperature in the step 2 is 35-39 ℃, and the extraction time is 2-4 years.
In the present invention, it is further preferable that the aging treatment in the step 3 specifically comprises: and (3) placing the microecological extracting solution into a sterilized curing barrel for sealed curing storage, wherein the temperature for sealed curing storage is room temperature.
In the present invention, it is further preferred that the microorganism in step 2 comprises the following strains in percentage by weight of the total amount of each component (the percentage by weight herein refers to the ratio of the amount of the strains to the total weight of each component of the microecological extract x 100%): 0.0133% of lactobacillus acidophilus, 0.0133% of lactobacillus casei, 0.0133% of lactobacillus rhamnosus, 0.01% of lactobacillus reuteri, 0.01% of streptococcus thermophilus and 0.02% of bifidobacterium animalis.
In the present invention, it is further preferable that the microorganism in step 2 further includes a microorganism for degrading an organophosphorus pesticide.
Compared with the prior art, the invention has the beneficial effects that the microecological extract has the following effects by screening and proportioning optimization of fermentation microbial strains:
1) the probiotic composition has high content of active ingredients, and can secrete protease, cellulase, hemicellulase, pectinase, amylase and other extracellular enzymes in the process of growing, reproducing and metabolizing by using each component as a nutrient substance, so that cells of the traditional Chinese medicine are cracked, the active ingredients of each component can be released from the cells, and the active ingredients in each component can be extracted and utilized to the maximum extent; has arteriosclerosis intervention function;
2) the beneficial component content is high, and the probiotic composition can generate substances such as polysaccharide beneficial to a human body in the secondary metabolism process, and finally enter the extracting solution to improve the content of the beneficial substances in the extracting solution;
3) the probiotic composition disclosed by the invention has high human body absorptivity, macromolecular substances which cannot be absorbed by a human body are degraded into active micromolecules which can be absorbed by the human body, and the micromolecular substances can be combined with cell proteins of an organism through a blood brain barrier, so that the absorption and utilization of the organism to effective components are improved;
4) the probiotics extracting method has the advantages that the variety of active ingredients is various, in the process of extracting the probiotics through combination, part of the ingredients in each component have inhibition/promotion effects on the growth and metabolism of microorganisms and the generation of the active ingredients, and the microorganisms can change own metabolic pathways in the special environment of each component, so that richer primary or secondary metabolites are generated, and more active ingredients are formed;

Through the screening and the proportioning optimization of the fermentation microbial strains, the microecological extract has the following effects:

The preparation method of the microecological extract comprises the following steps:
step 1: taking the components according to the formula, mixing, sterilizing, and then breaking the wall by microwave to obtain a crushed raw material mixture;
step 2: carrying out microbial ecological extraction treatment on the raw material mixture obtained in the step 1 by using microbes, then filtering the mixture obtained after the microbial ecological extraction, removing filter residues, and collecting a microbial ecological extracting solution;
and step 3: and (3) sterilizing the microecological extracting solution obtained in the step (2), and then curing to obtain the microecological extract.
In order to be more convenient for consumers to eat, auxiliary materials acceptable for food can be added into the microecological extract obtained in the step 3 to prepare different product dosage forms so as to enrich the mouthfeel of the product and facilitate storage and transportation, and specifically, a sweetening agent and the like can be added according to the needs or the preference of consumers, or the microecological extract is further prepared into oral liquid, tablets and the like; for example, step 4 is followed by step 5: heating the microecological extract treated in the step 4 until the auxiliary materials are dissolved, and then sterilizing; the adjuvants are soluble in water, and can be sweetener (such as granulated sugar, sucralose, mogroside), etc.
In order to improve the quality of the microecological extract, the fermentation temperature in the step 2 is 35-39 ℃, and the fermentation time is 2-4 years.
The sterilization treatment is not particularly limited, and for the sake of facilitating industrial operation, ozone sterilization treatment may be employed for 4 to 8 min.
For filtering the microorganism extract, a 100-mesh sieve can be selected for filtering; for the sterilization treatment of the microorganism extract in step 3, pasteurization can be carried out, such as sterilization at 85 + -5 deg.C for 30-35 min; after the microbial extracting solution is treated in the steps 4 and 5, the treatment such as sterilization, filling, packaging and the like can be continuously carried out for facilitating the storage and transportation of the product; in order to inspect and monitor the quality of the product, the physical and chemical indexes of the product, such as total acid, crude polysaccharide, free amino acid, short-chain fatty acid, microorganism and the like, can be detected.
In order to enhance the flavor of the microbial extract of the present invention, a curing treatment may be performed, and the curing treatment in step 3 is specifically: placing the micro-ecological extracting solution into a sterilized curing barrel for sealed curing storage, wherein the temperature for sealed curing storage is room temperature; by aging, fructo-oligosaccharide in the microorganism extractive solution can be aged to generate aromatic hydrocarbon, which can improve flavor and aromatic hydrocarbon effect.
In order to improve the active ingredient content and the human body availability of the microbial extract and reduce the content of harmful substances, the microbes in the step 2 comprise the following strains in percentage by weight according to the total amount of the components: 0.0133% of lactobacillus acidophilus, 0.0133% of lactobacillus casei, 0.0133% of lactobacillus rhamnosus, 0.01% of lactobacillus reuteri, 0.01% of streptococcus thermophilus and 0.02% of bifidobacterium animalis.
In order to further reduce the content of harmful substances in the microecological extract of the present invention, the microorganisms in step 2 further include microorganisms for degrading organophosphorus pesticides; as the microorganism for degrading the organophosphorus pesticide, bacteria, fungi and algae can be selected, and correspondingly such as:

step 2: carrying out microbial ecological extraction treatment on the raw material mixture obtained in the step 1 for 150 days by using microbes, then filtering (sieving by a 100-mesh sieve) the mixture obtained after the microbial ecological extraction, removing filter residues, and collecting a microbial ecological extracting solution; comprises the following strains in percentage by weight of the total amount of the components: lactobacillus acidophilus 0.0133%, lactobacillus casei 0.0133%, lactobacillus rhamnosus 0.0133%, lactobacillus reuteri 0.01%, streptococcus thermophilus 0.01%, and animal bifidobacterium 0.02%;
and step 3: sterilizing the microecological extracting solution obtained in the step 2, and then curing to obtain a microecological extract; the curing treatment in the step 3 specifically comprises the following steps: placing the micro-ecological extracting solution into a sterilized curing barrel for sealed curing storage, wherein the temperature for sealed curing storage is room temperature; obtaining the microecological extract.
Example 2
A microecological extract with an arteriosclerosis intervention function is prepared from the following components in parts by weight: 4 parts of garlic, 10 parts of black fungus, 7 parts of cinnamon, 6 parts of Chinese yam, 24 parts of grape, 8 parts of blueberry, 6 parts of hawthorn, 4 parts of pomegranate, 6 parts of pineapple, 5 parts of purple cabbage, 0.8 part of peach kernel, 2.5 parts of ginkgo, 0.08 part of medlar powder, 0.35 part of spirulina platensis, 12 parts of fructo-oligosaccharide, 1.2 parts of oat beta-glucan, 0.005 part of sucralose, 0.8 part of Kluyveromyces marxianus and 0.0008 part of lycopene; the microbial strains and the proportion in the step 2 are the same as those in the example 1; the preparation method is the same as that of example 1.
Example 3
A microecological extract with an arteriosclerosis intervention function is prepared from the following components in parts by weight: 2 parts of garlic, 6 parts of black fungus, 5 parts of cinnamon, 10 parts of Chinese yam, 18 parts of grape, 12 parts of blueberry, 4 parts of hawthorn, 6 parts of pomegranate, 4-6 parts of pineapple, 3 parts of purple cabbage, 0.8 part of peach kernel, 3.5 parts of ginkgo, 0.12 part of medlar powder, 0.25 part of spirulina platensis, 15 parts of fructo-oligosaccharide, 1.8 parts of oat beta-glucan, 0.015 part of sucralose, 1.2 parts of Kluyveromyces marxianus and 0.0012 part of lycopene; the microbial strains and the proportion in the step 2 are the same as those in the example 1; the preparation method is the same as that of example 1.
Experimental example 1
Taking the microecological extract with the arteriosclerosis intervention function in example 1, and then detecting the contents of polyphenol, flavone, saponin, amino acid and harmful substances in the microecological extract, wherein the specific detection results are shown in the following tables 1-3:

As can be seen from the data in table 1 above: the microecological extract with the arteriosclerosis intervention function has high contents of polyphenol, flavone, polysaccharide, saponin and the like, has high superoxide dismutase (SOD) activity, and can well intervene arteriosclerosis.

As can be seen from the data in table 2 above: the microecological extract with the arteriosclerosis intervention function has complete types and high content of essential amino acids for human bodies.

As can be seen from the data in table 3 above: the microecological extract with the arteriosclerosis intervention function has no other harmful substances except lactic acid, and has high safety.

From the data in table 4 above, it can be seen that the microecological extract of the present invention in example 1 was not detected for each heavy metal and pesticide, and was highly safe.

From the detection data in table 5 above, it can be seen that each harmful bacterium and saccharin sodium in the microecological extract of the present invention all meet the national standards, and the microecological extract of the present invention has high safety.
Comparative example 1
Taking the components according to the microecological components and the proportion thereof in the embodiment 1, mixing, sterilizing, and then carrying out superfine grinding to obtain a ground raw material mixture; and adding water, decocting for 3 times, decocting for 30min each time, collecting the filtrate obtained by 3 times of decoction through filtration, and combining the filtrates to obtain the filtrate of comparative example 1 (namely the extract of comparative example 1).
Respectively taking 2 parts of the same weight of the microecological formulation according to the example 1, then respectively adopting the methods of the example 1 and the comparative example 1 to obtain the microecological extract of the example 1 and the extract of the comparative example 1, concentrating the 2 parts of the extract to the same weight (concentrating the extract of the comparative example 1 to the same weight based on the microecological extract of the example 1), then respectively testing the content of each nutrient component and the total amount of hydrolyzed amino acid in the extract, wherein the test result data are shown in the following table:

As can be seen from the data in Table 6, compared with the conventional decoction process of comparative example 1, the microecological extraction of the present invention enables better release of active nutritional ingredients, so that the microecological extract of the present invention has higher nutritional active ingredients and better intervention effect on the anti-atherosclerosis ability of the human body.
Finally, it should be noted that: the above embodiments are only preferred embodiments of the present invention, and the protection scope of the present invention should not be limited thereby, and any insubstantial changes and substitutions made by those skilled in the art based on the present invention are within the protection scope of the present invention.
Claims (10)
1. A microecological extract with an arteriosclerosis intervention function is characterized by being prepared from the following components in parts by weight: 2-4 parts of garlic, 6-10 parts of black fungus, 5-7 parts of cinnamon, 6-10 parts of Chinese yam, 18-24 parts of grape, 8-12 parts of blueberry, 4-6 parts of hawthorn, 4-6 parts of pomegranate, 4-6 parts of pineapple, 3-5 parts of purple cabbage, 0.8-1.2 parts of peach kernel, 2.5-3.5 parts of ginkgo, 0.08-0.12 part of medlar powder, 0.25-0.35 part of spirulina platensis, 12-15 parts of fructo-oligosaccharide, 1.2-1.8 parts of oat beta-glucan, 0.005-0.015 part of sucralose, 0.8-1.2 parts of kluyveromyces marxianus and 0.0008-0.0012 part of lycopene.
2. The microecological extract according to claim 1, which is prepared from the following components in parts by weight: 3 parts of garlic, 8 parts of black fungus, 6 parts of cinnamon, 8 parts of Chinese yam, 21.5 parts of grape, 10 parts of blueberry, 5 parts of hawthorn, 5 parts of pomegranate, 5 parts of pineapple, 4 parts of purple cabbage, 1 part of peach kernel, 3 parts of ginkgo, 0.1 part of medlar powder, 0.299 parts of spirulina platensis, 13.59 parts of fructo-oligosaccharide, 1.5 parts of oat beta-glucan, 0.01 part of sucralose, 1 part of kluyveromyces marxianus and 0.001 part of lycopene.
3. A method for preparing the microecological extract according to claim 1, characterized in that it comprises the steps of:
step 1: taking the components according to the formula, mixing, sterilizing, and then breaking the wall by microwave to obtain a crushed raw material mixture;
step 2: carrying out microbial ecological extraction treatment on the raw material mixture obtained in the step 1 by using microbes, then filtering the mixture obtained after the microbial ecological extraction, removing filter residues, and collecting a microbial ecological extracting solution;
and step 3: and (3) sterilizing the microecological extracting solution obtained in the step (2), and then curing to obtain the microecological extract.
4. The method for preparing a microecological extract according to claim 3, further comprising the step 4 after the step 3: adding food acceptable auxiliary materials into the microecological extract obtained in the step 3.
5. The method for preparing a microecological extract according to claim 4, further comprising the step 5 after the step 4: heating the microecological extract treated in the step 4 until the auxiliary materials are dissolved, and then sterilizing; the adjuvant is soluble in water.
6. The method for preparing a microecological extract according to claim 3, wherein the sterilization is by ozone.
7. The method for preparing a microecological extract according to claim 3, wherein: the extraction temperature in the step 2 is 35-39 ℃, and the extraction time is 2-4 years.
8. The method for preparing a microecological extract according to claim 3, wherein the aging treatment in the step 3 is specifically: and (3) placing the microecological extracting solution into a sterilized curing barrel for sealed curing storage, wherein the temperature for sealed curing storage is room temperature.
9. The method for preparing a microecological extract according to claim 3, wherein the microorganisms of step 2 comprise the following species in percentage by weight based on the total amount of each component: 0.0133% of lactobacillus acidophilus, 0.0133% of lactobacillus casei, 0.0133% of lactobacillus rhamnosus, 0.01% of lactobacillus reuteri, 0.01% of streptococcus thermophilus and 0.02% of bifidobacterium animalis.
10. The method for preparing a microecological extract according to claim 3, wherein the microorganisms of step 2 further comprise microorganisms for degrading organophosphorus pesticides.
