Abstract
The invention provides a traditional Chinese medicine composition for preventing and treating liver tumor in an auxiliary way, a preparation method and application thereof, belonging to the technical field of traditional Chinese medicines. The traditional Chinese medicine composition comprises astragalus membranaceus, angelica sinensis, radix bupleuri, cornus officinalis, salvia miltiorrhiza, bighead atractylodes rhizome, rhizoma anemarrhenae, pericarpium citri reticulatae, radix rehmanniae preparata, medlar and cassia twig, and the astragalus membranaceus is used as a monarch drug, so that the curative effects of strengthening body resistance and eliminating evil, storing vital qi and keeping the evil dry can be achieved, the traditional Chinese medicine composition has good prevention and adjuvant therapy effects on liver tumors, the recurrence of postoperative liver tumors is effectively prevented, the survival time of patients is prolonged, and the life quality of the patients is improved. The traditional Chinese medicine composition has broad-spectrum applicability, easily obtained raw materials, safety, no toxic or side effect, simple preparation method and low preparation cost.

Technical Field
The invention belongs to the technical field of traditional Chinese medicines, and particularly relates to a traditional Chinese medicine composition for preventing and assisting in treating liver tumors, and a preparation method and application thereof.
Background
The primary liver tumor is one of the common clinical tumors, wherein primary hepatocellular carcinoma (HCC) accounts for more than 90% of primary liver cancer (hereinafter referred to as liver cancer). The newly discovered liver cancer patients and the patients who die of the liver cancer in China are about 11 thousands of people respectively, account for 45 percent of the liver cancer patients all over the world, and are one of three major life cancers. a traditional Chinese medicine composition for preventing and adjuvant therapy of liver tumor comprises the following raw materials in parts by weight: 15-25 parts of astragalus membranaceus, 5-15 parts of angelica sinensis, 2-10 parts of radix bupleuri, 10-20 parts of cornus officinalis, 5-15 parts of salvia miltiorrhiza, 7-15 parts of bighead atractylodes rhizome, 10-20 parts of rhizoma anemarrhenae, 5-15 parts of pericarpium citri reticulatae, 10-20 parts of radix rehmanniae preparata, 7-15 parts of wolfberry and 7-15 parts of cassia twig.

The invention also provides a preparation method of the traditional Chinese medicine composition, which comprises the following steps: mixing the astragalus, the angelica, the radix bupleuri, the cornus officinalis, the salvia miltiorrhiza, the bighead atractylodes rhizome, the rhizoma anemarrhenae, the dried orange peel, the prepared rhizome of rehmannia, the medlar and the cassia twig to obtain a traditional Chinese medicine mixture, mixing the traditional Chinese medicine mixture with water, soaking, decocting for 1-3 times, filtering, and combining filtrates to obtain the traditional Chinese medicine composition.

The invention provides a traditional Chinese medicine composition for preventing and treating liver tumor in an auxiliary way, which comprises the following raw materials in parts by weight: 15-25 parts of astragalus membranaceus, 5-15 parts of angelica sinensis, 2-10 parts of radix bupleuri, 10-20 parts of cornus officinalis, 5-15 parts of salvia miltiorrhiza, 7-15 parts of bighead atractylodes rhizome, 10-20 parts of rhizoma anemarrhenae, 5-15 parts of pericarpium citri reticulatae, 10-20 parts of radix rehmanniae preparata, 7-15 parts of wolfberry and 7-15 parts of cassia twig.
The astragalus root of the present invention is preferably 17 to 20 parts by weight, more preferably 18 parts by weight, and is astragalus root, i.e., astragalus mongholicus, which is a plant of the genus astragalus of the family leguminosae, which is perennial. Bitter in property and slightly cold in nature, entering liver and gallbladder meridians. Has the functions of harmonizing exterior and interior, soothing liver and invigorating yang. Can be used for treating common cold, fever, malaria, stagnation of qi due to depression of the liver, distending pain of chest and hypochondrium, rectocele, uterine prolapse, and menoxenia.

Warm in nature, pungent and bitter in flavor. It enters spleen, stomach and lung meridians. The main functions are regulating qi, stimulating appetite, eliminating dampness and phlegm, and treating spleen and stomach diseases. Belongs to qi regulating medicine, and also has the effects of resisting gastric ulcer, protecting liver, promoting bile flow, boosting pressure, tonifying heart, expanding coronary artery and resisting tumor.
The prepared rhizome of rehmannia of the invention is preferably 13 to 17 parts by weight, more preferably 15 parts by weight, and the prepared rhizome of rehmannia is rehmannia which is a plant of Scrophulariaceae. Sweet in nature and taste, slightly warm. It enters liver and kidney meridians. The main functions are nourishing yin and enriching blood. It is indicated for deficiency of yin and blood, atrophy and weakness of waist and knees, fatigue, cough, steaming bone, seminal emission, metrorrhagia, menoxenia, diabetes, scanty urine, deafness and blurred vision.
The medlar of the invention is preferably 8 to 12 parts by weight, more preferably 10 parts by weight, and the medlar is the fruit of medlar which belongs to the family of solanaceae and the genus of medlar. Sweet in nature and taste and mild in nature. It enters liver and kidney meridians. The main functions are nourishing kidney, moistening lung, tonifying liver and improving vision. It is indicated for liver and kidney yin deficiency, soreness and weakness of waist and knees, dizziness, blurred vision, hyperdacryosis, cough due to asthenia, diabetes, and spermatorrhea.
The cassia twig of the invention is preferably 8 to 12 parts by weight, more preferably 10 parts by weight, and belongs to cinnamoles, lauraceae and medium arbors; pungent, sweet and warm in nature. It enters heart, lung and bladder meridians. Has effects of inducing sweat, expelling pathogenic factors from muscles, warming and dredging channels, supporting yang, regulating qi, and lowering qi. It is commonly used for wind-cold type common cold, abdominal cold pain, amenorrhea due to blood cold, arthralgia, phlegm and fluid retention, edema and palpitation.
The theoretical formulation of the invention is based on the traditional Chinese medicine theory of 'yang moving, dispersing, solidifying and gasifying, yin static, solidifying and forming' of Zhangjing Yue of Ming dynasty physicians. In the invention, the mass ratio of the traditional Chinese medicine mixture to water is preferably 1 (7-13), more preferably 1:10, the soaking time is preferably 10-30min, more preferably 20min, the time of each decoction is preferably 40-120min, more preferably 60min, and the number of times of decoction is preferably 2; the decocting step comprises the steps of decocting for the first time, filtering, adding water into the decoction dregs for decocting for the second time, and filtering to obtain a second decoction filtrate, wherein the mass ratio of the decoction dregs to the water is preferably 1 (5-8), and more preferably 1: 6.
The traditional Chinese medicine composition prepared by the invention is taken 1 time each day in the morning and evening, 200 ml each time, and uncooked, cold, sticky, slippery, five-pungent and smelly foods are avoided during the taking period.
The invention also provides application of the traditional Chinese medicine composition or the traditional Chinese medicine composition obtained by the preparation method in preparation of a medicine for preventing and/or treating liver tumor.
The invention also provides a medicament for preventing and/or treating liver tumor, which is prepared into tablets, capsules, granules, pills or oral liquid by adding pharmaceutically acceptable auxiliary materials into the traditional Chinese medicine composition.
The technical solutions provided by the present invention are described in detail below with reference to examples, but they should not be construed as limiting the scope of the present invention.
Example 1
Accurately weighing 18g of astragalus membranaceus, 9g of angelica sinensis, 5g of radix bupleuri, 15g of cornus officinalis, 9g of salvia miltiorrhiza, 10g of bighead atractylodes rhizome, 12g of rhizoma anemarrhenae, 10g of pericarpium citri reticulatae, 15g of radix rehmanniae preparata, 10g of medlar and 10g of cassia twig, mixing the materials, mixing the mixture with water according to the weight ratio of 1:10, soaking the mixture in the water for 20min, decocting the mixture in the water for 2h, and filtering the mixture to obtain filtrate, thus obtaining the traditional Chinese medicine composition.
Example 2
Accurately weighing 15g of astragalus, 5g of angelica, 2g of radix bupleuri, 10g of cornus officinalis, 5g of salvia miltiorrhiza, 7g of bighead atractylodes rhizome, 10g of rhizoma anemarrhenae, 5g of dried orange peel, 10g of prepared rehmannia root, 7g of medlar and 7g of cassia twig, mixing the materials and water according to a weight ratio of 1: 7, soaking in water for 30min, decocting for 40min, filtering to obtain a first filtrate and a filter residue, mixing the filter residue with water according to the weight ratio of 1:6, decocting for 40min, filtering to obtain a second filtrate, and mixing with the first filtrate to obtain the traditional Chinese medicine composition.
Example 3
Accurately weighing 25g of astragalus membranaceus, 15g of angelica sinensis, 10g of radix bupleuri, 20g of cornus officinalis, 15g of salvia miltiorrhiza, 15g of bighead atractylodes rhizome, 20g of rhizoma anemarrhenae, 15g of pericarpium citri reticulatae, 20g of radix rehmanniae preparata, 15g of medlar and 15g of cassia twig, mixing the mixture with water according to the weight ratio of 1:13, soaking the mixture in water for 10min, decocting the mixture for 120min, filtering the mixture to obtain first filtrate and filter residue, mixing the filter residue with the water according to the weight ratio of 1:5, decocting the mixture for 40min, filtering the mixture to obtain second filtrate, and mixing the second filtrate with the first filtrate to obtain the traditional Chinese medicine composition.
Example 4
Accurately mixing 20g of astragalus membranaceus, 12g of angelica sinensis, 5g of radix bupleuri, 15g of cornus officinalis, 12g of salvia miltiorrhiza, 15g of bighead atractylodes rhizome, 15g of rhizoma anemarrhenae, 12g of pericarpium citri reticulatae, 20g of radix rehmanniae preparata, 12g of medlar and 12g of cassia twig, mixing the mixture with water according to the weight ratio of 1:10, soaking the mixture in water for 20min, decocting the mixture for 60min, filtering the mixture to obtain first filtrate and filter residue, mixing the filter residue with the water according to the weight ratio of 1:8, decocting the mixture for 40min, filtering the mixture to obtain second filtrate, and mixing the second filtrate with the first filtrate to obtain the traditional Chinese medicine composition.
Experimental example 1
The traditional Chinese medicine composition has the effect on liver cancer of mice
The experiment is mainly completed by the experimental animal center of university of Dalian medical science.
Experimental animals: the H-ras12V transgenic liver cancer model mouse is selected as an experimental object. The liver cancer model mouse directly induces the generation and development of spontaneous liver cancer by the Ras oncogene specifically expressed in the hepatocyte, the pathological generation and development process of the liver cancer model mouse is similar to that of human liver cancer, and the liver cancer model mouse has the advantages of single genetic background, high morbidity (up to 90%), timely morbidity (6-8 months old tumor), good repeatability, regular presentation of the whole process of the generation and development of the liver cancer and the like, and provides an excellent disease animal model for verifying the curative effect of the medicament on the generation and development of the liver cancer.
The experimental method comprises the following steps: (1) 28 male H-Ras12V transgenic liver cancer model mice are selected. Groups were randomized into 2, control and treatment groups of 14 individuals each. The treatment group started to drink the traditional Chinese medicine composition of the invention in example 1 freely at 3 months of age, and replaced fresh traditional Chinese medicines every day. The control group started drinking water normally at 3 months of age. The other feeding conditions of the control group and the treatment group were not changed. During the test period, the animals were observed daily for diet, drinking water, behavioral activity, and the animal body weights were measured and recorded every 5 days. (2) By the age of 6 months, the mice were sacrificed, the livers were dissected out, weighed, photographed, and the size and number of tumors were measured.
The diet, behavior and mental state of the treated mice were normal during the experiment, and no significant change was observed. The control group mice had normal diet, behavior and mental status in the early period of the experiment (3-5 months old), and had no significant change, but in the later period of the experiment (5-6 months old), the liver tumor grew to cause symptoms of enlargement of upper abdomen, emaciation, disorder of hair, listlessness, etc. in different degrees.
As can be seen from fig. 1 and fig. 2, the occurrence and development of the tumor in the treatment group of the present invention are significantly inhibited through the comparative experiment with the control group, which proves that the traditional Chinese medicine composition of the present invention has a significant inhibiting effect on the occurrence and development of the in situ spontaneous liver tumor.
Experimental example 2
The traditional Chinese medicine composition prepared by the embodiment is applied to clinical treatment
Zhangqi, male, 68 years old, Dalian city, Liaoning province, Zhenju: liver cancer. After the decoction is taken for half a month, the liver area is full and relieved, the body pain is relieved, the appetite is increased, the targeted medicine is stopped taking, the decoction is continuously taken, the aftereffect of two months disappears, the mental state is good, the medicine is stopped taking, and the normal state of the life is recovered at present.

A traditional Chinese medicine composition for preventing and adjuvant therapy of liver tumor is characterized by comprising the following raw materials in parts by weight: 15-25 parts of astragalus membranaceus, 5-15 parts of angelica sinensis, 2-10 parts of radix bupleuri, 10-20 parts of cornus officinalis, 5-15 parts of salvia miltiorrhiza, 7-15 parts of bighead atractylodes rhizome, 10-20 parts of rhizoma anemarrhenae, 5-15 parts of pericarpium citri reticulatae, 10-20 parts of radix rehmanniae preparata, 7-15 parts of wolfberry and 7-15 parts of cassia twig.

The traditional Chinese medicine composition according to claim 1, which is characterized by comprising the following raw materials in parts by weight: 17-20 parts of astragalus membranaceus, 7-10 parts of angelica sinensis, 3-8 parts of radix bupleuri, 13-17 parts of cornus officinalis, 7-10 parts of salvia miltiorrhiza, 8-12 parts of bighead atractylodes rhizome, 11-15 parts of rhizoma anemarrhenae, 7-10 parts of pericarpium citri reticulatae, 13-17 parts of radix rehmanniae preparata, 8-12 parts of wolfberry and 8-12 parts of cassia twig.

A method for preparing the traditional Chinese medicine composition of claim 1 or 2, which is characterized by comprising the following steps: mixing the astragalus, the angelica, the radix bupleuri, the cornus officinalis, the salvia miltiorrhiza, the bighead atractylodes rhizome, the rhizoma anemarrhenae, the dried orange peel, the prepared rhizome of rehmannia, the medlar and the cassia twig to obtain a traditional Chinese medicine mixture, mixing the traditional Chinese medicine mixture with water, soaking, decocting for 1-3 times, filtering, and combining filtrates to obtain the traditional Chinese medicine composition.

10. A medicament for preventing and/or treating liver tumor, which is characterized in that pharmaceutically acceptable auxiliary materials are added into the traditional Chinese medicine composition of claim 1 or 2 to prepare tablets, capsules, granules, pills or oral liquid.
