Abstract
本发明涉及中药组合物领域,具体涉及一种适用于妊娠期高血压的活血降压组合物。所述组合物的制备原料包括以下组分,虎杖、炆黄精、荷叶、大黄、人参、薤白、冬虫夏草、芦荟、竹茹、知母、甘草、蒲公英、白薇、吴茱萸、桔梗、维生素A、维生素E、维生素D、维生素K和叶酸。本发明采用合理地组分和比例搭配,得到一种适合妊娠期高血压的活血降压组合物,可以直接磨粉得到成品,也可以通过合适提取方法得到更好的治疗效果。

技术领域
本发明涉及中药组合物领域,具体涉及一种适用于妊娠期高血压的活血降压组合物。
背景技术
妊娠期高血压疾病包括妊娠期高血压、子痫前期、子痫,以及慢性高血压并发子痫前期和妊娠期合并慢性高血压。如子痫前期,孕妇血压升高,同时可伴随血小板减少、肝肾功能损害、肺水肿及子痫等严重临床表现,并易导致子宫-胎盘的结构发生异常,影响腹中胎儿的生长,在分娩过程中严重威胁母婴的生命安全。
临床治疗妊娠期高血压疾病以降压、解痉、有指征的扩容与利尿、适时终止妊娠为主,但效果并不理想。对于妊娠高血压,孕妇与未孕妇有着不同的处理原则。虽然一些降压药物可降低血压,但与此同时也减少了其它脏器的血流量。尤其是子宫胎盘的血流量,这对胎儿有一定影响,所以说孕产妇应慎用降压药物:除非血压增高的较快,一般不宜服用降压药物;当平均动脉压≥140mm Hg,或舒张压≥110mm Hg时,可适当选用适宜的降压药物,避免恶化为先兆子痫及子痫;当高血压比较严重时,采用联合用药,以免影响子宫胎盘灌注量、肾血流量及心输出量。硫酸镁是目前产科临床上最常用的药物之一,镁离子可直接舒张外周血管平滑肌并且阻止交感神经神经元的传递,起到扩张血管,降低血压,且降压作用快而强。除此之外的降压药物还包括有β受体阻滞剂,钙拮抗剂等。
中医对妊娠高血压的治疗为益气、活血、化瘀,滋补肝肾阴虚的综合治疗,根据病因病机,辩证施治。不仅符合祖目医学理论,而且与现代医学的解痉、降压,改善微循环,增加组织血液灌流的理论是一致的。因此,中医药治疗妊娠高血压在临床医学的实践中不可或缺。
中国专利申请CN103301340A公开了一种治疗妊娠期高血压疾病的药物。它由下述重量的原料制成,灵芝20克、炒杜仲20克、桑寄生20克、续断15克、佛手15克、山药15克、水飞蓟10克、大枣10克、葛根10克、罗布麻叶10克、荷叶10克、石决明10克、山楂10克、玉米须10克、金钱草5克、芡实5克、鹿衔草5克、黄芩10克、党参5克和甘草5克。采用该中药治疗妊娠期高血压疾病,总有效率为100%,且不影响胎儿的正常发育。
中国专利申请CN103893548A公开了一种治疗妊娠期高血压病的中药制剂,它由以下原料按重量百分比配制而成:川芎10-20%,葛根10-20%,桑寄生6-12%,白术8-15%,黄精6-12%,山楂10-20%,夏枯草6-12%,黄瓜藤6-12%,茯苓8-16%。本发明针对妊娠生理因素血瘀证形成的实质,一是以活血祛风治法为主,提出治血须治风理念。应用桑寄生、葛根等祛风药物疏达升散,活跃气血运行,疏通经络瘀滞,风去血自然循经而行,与活血相辅相成,达到了单纯活血化瘀药难以达到的祛瘀效果,以弥补妊娠忌用祛瘀破血的不足、并通过治血达到气血充足,内风不能生,预防出现“子痫”。二是根据妊娠多郁特点,配合行血行水,从治气、治血、治水角度,益气固本,活血不忘安胎,达到扶正祛邪、安胎祛病之目的。.

虎杖5~15份、炆黄精10~30份、荷叶5~10份、大黄5~7份、人参10~20份、薤白5~10份、冬虫夏草5~10份、芦荟10~20份、竹茹5~10份、知母5~10份、甘草5~10份、蒲公英5~10份、白薇5~10份、吴茱萸5~10份、桔梗、5~10份、维生素A0.0002~0.0006份、维生素E0.02~0.06份、维生素D0.0002~0.0006份、维生素K0.0002~0.0006份和叶酸0.01~0.1份。
进一步地,所述组合物,按重量份数计,原料包括以下组分:

本发明的组合物也可以将所述重量份的原料通过合适的提取方法得到。
一种所述组合物的制备方法,包括以下步骤:
S1:将虎杖、炆黄精、荷叶、大黄、人参、薤白、冬虫夏草、芦荟和竹茹混合磨粉后,经过水提、过滤和干燥得提取物A;
S2:将知母、甘草、蒲公英、白薇、吴茱萸和桔梗混合磨粉后,经过乙醇-乙酸乙酯提取、过滤和干燥的提取物B;
S3:将提取物A、提取物B、维生素A、维生素E、维生素D、维生素K和叶酸混合,磨粉即得所述组合物。
进一步地,S1中,所述磨粉是将混合物磨成80-100目粉。
进一步地,S1中,所述水提是水用量为所述S1原料总重量份的20-30倍,温度为60-80℃提取8-10小时。
进一步地,S2中,所述磨粉是将混合物磨成80-100目粉。
进一步地,所述乙醇-乙酸乙酯提取是在乙醇和乙酸乙酯体积比为1:(5-10),乙醇-乙酸乙酯总用量为所述S2原料总重量份的10-15倍,温度为40-60℃提取4-6小时。
进一步地,所述S3中,所述磨粉是将混合物磨成80-100目粉。
本发明的优势在于:
1.采用合理地组分和比例搭配,得到一种适合妊娠期高血压的活血降压组合物,其中竹茹、知母、甘草、芦荟和桔梗的合理比例对降压效果影响较大。
2.本发明的组合物在实现活血降压的基础上,增加合理的维生素用量,补充妊娠其的维生素需求,巩固使用者的身体状况。
3.本发明的组合物可以直接磨粉得到成品,也可以通过合适提取方法得到更好的治疗效果,猜测可能是由于中药成分复杂,不同药物组合之后,通过合适的方法提取可以祛除一些抑制药效的物质。

虎杖15份、炆黄精10份、荷叶5份、大黄6份、人参20份、薤白5份、冬虫夏草5份、芦荟20份、竹茹5份、知母10份、甘草10份、蒲公英10份、白薇10份、吴茱萸5份、桔梗5份、维生素A0.0006份、维生素E0.02份、维生素D0.0006份、维生素K0.0002份和叶酸0.01份。
将各组分混合,一起磨至80目粉末,即得所述组合物。

所述组合物通过以下步骤制备得到:
S1:将虎杖、炆黄精、荷叶、大黄、人参、薤白、冬虫夏草、芦荟和竹茹混合磨至80目粉,加入此步骤原料总份数20倍重量份的水,升温至80℃,提取8小时,趁热过滤,回收滤液,浓缩,干燥得提取物A;
S2:将知母、甘草、蒲公英、白薇、吴茱萸和桔梗混合磨至80目粉,加入此步骤原料总份数15倍重量份的乙醇-乙酸乙酯混合液,其中乙醇和乙酸乙酯体积比为1:5,升温至40℃,提取4小时,趁热过滤,回收滤液,浓缩,干燥得提取物B;
S3:将提取物A、提取物B、维生素A、维生素E、维生素D、维生素K和叶酸混合,磨粉至80目,即得所述组合物,总量为36份。
实施例5
一种适用于妊娠期高血压的活血降压组合物,按重量份数计,原料包括以下组分:
虎杖10份、炆黄精20份、荷叶8份、大黄6份、人参15份、薤白7份、冬虫夏草8份、芦荟15份、竹茹7份、知母7份、甘草8份、蒲公英8份、白薇7份、吴茱萸7份、桔梗8份、维生素A0.0004份、维生素E0.04份、维生素D0.0004份、维生素K0.0004份和叶酸0.05份。
所述组合物通过以下步骤制备得到:
S1:将虎杖、炆黄精、荷叶、大黄、人参、薤白、冬虫夏草、芦荟和竹茹混合磨至80目粉,加入此步骤原料总份数30倍重量份的水,升温至60℃,提取10小时,趁热过滤,回收滤液,浓缩,干燥得提取物A;
S2:将知母、甘草、蒲公英、白薇、吴茱萸和桔梗混合磨至80目粉,加入此步骤原料总份数10倍重量份的乙醇-乙酸乙酯混合液,其中乙醇和乙酸乙酯体积比为1:10,升温至40℃,提取6小时,趁热过滤,回收滤液,浓缩,干燥得提取物B;
S3:将提取物A、提取物B、维生素A、维生素E、维生素D、维生素K和叶酸混合,磨粉至80目,即得所述组合物,总量为42份。

和实施例4相比,全部使用乙醇-乙酸乙酯提取
一种适用于妊娠期高血压的活血降压组合物,按重量份数计,原料包括以下组分:
虎杖10份、炆黄精20份、荷叶8份、大黄6份、人参15份、薤白7份、冬虫夏草8份、芦荟15份、竹茹7份、知母7份、甘草8份、蒲公英8份、白薇7份、吴茱萸7份、桔梗8份、维生素A0.0004份、维生素E0.04份、维生素D0.0004份、维生素K0.0004份和叶酸0.05份。
所述组合物通过以下步骤制备得到:
S1:将出维生素和叶酸外的原料混合,干燥,磨至80目粉,加入此步骤原料总份数15倍重量份的乙醇-乙酸乙酯混合液,其中乙醇和乙酸乙酯体积比为1:5,升温至40℃,提取4小时,趁热过滤,回收滤液,浓缩,干燥得提取物A;
S3:将提取物A、维生素A、维生素E、维生素D、维生素K和叶酸混合,磨粉至80目,即得所述组合物,总量为45份。
效果例-对妊娠期大鼠的降血压效果测试
妊娠大鼠获取:取成年SD大鼠,按雌性2:1合笼饲养,每天早上取雌鼠阴道分泌物进行涂片,显微镜下观察发现精子计为妊娠第0天,即得受孕大鼠。
受孕大鼠分组:空白对照组、妊娠高血压组、实施例3、实施例4、对比例1、对比例2和对比例3,每组5只。
造模和给药:除空白对照组外,其他组别大鼠自妊娠第12天起,连续7天灌胃亚硝基左旋精氨酸甲酯(L.NAME,100mg/kg)。
给药时间、方式和剂量:
空白对照组、妊娠高血压组,灌胃蒸馏水;
实施例3,实施例4、对比例1-3五个实验组,按生药量计算剂量给药,安生药量计,给药剂量为300mg/kg.BW;
所有组别每天同一时间灌胃给药一次。
检测指标:妊娠期高血压表现为血压升高和尿蛋白增加,因此主要检测各组合物对大鼠这两项指标的影响。
在妊娠期第10天(妊娠高血压造模前)、14天和21天分别测定各组大鼠的收缩压和24h尿蛋白。
实验结果
表1不同时间各实验组大鼠的血压和尿蛋白数据
和空白组相比:**P<0.01,*P<0.05。
可以看出,本申请的组合物,采用合适的组成和特定的份数,可以有效控制大鼠妊娠高血压的进程,采用合适的提取方法进行制备后,可以得到更好的效果。

1.一种适用于妊娠期高血压的活血降压组合物,按重量份数计,原料包括以下组分:
虎杖5~15份、炆黄精10~30份、荷叶5~10份、大黄5~7份、人参10~20份、薤白5~10份、冬虫夏草5~10份、芦荟10~20份、竹茹5~10份、知母5~10份、甘草5~10份、蒲公英5~10份、白薇5~10份、吴茱萸5~10份、桔梗、5~10份、维生素A0.0002~0.0006份、维生素E0.02~0.06份、维生素D0.0002~0.0006份、维生素K0.0002~0.0006份和叶酸0.01~0.1份。
2.根据权利要求1所述的组合物,其特征在于,按重量份数计,原料包括以下组分:
虎杖10份、炆黄精20份、荷叶8份、大黄6份、人参15份、薤白7份、冬虫夏草8份、芦荟15份、竹茹7份、知母7份、甘草8份、蒲公英8份、白薇7份、吴茱萸7份、桔梗8份、维生素A0.0004份、维生素E0.04份、维生素D0.0004份、维生素K0.0004份和叶酸0.05份。
3.一种如权利要求1或2任一项所述组合物的制备方法,将所述重量份数的原料混合,研磨成粉即得。
4.根据权利要求3所述的制备方法,其特征在于,所述研磨是研磨至60-80目的粉料。
5.一种如权利要求1或2任一项所述组合物的制备方法,包括以下步骤:
S1:将虎杖、炆黄精、荷叶、大黄、人参、薤白、冬虫夏草、芦荟和竹茹混合磨粉后,经过水提、过滤和干燥得提取物A;
S2:将知母、甘草、蒲公英、白薇、吴茱萸和桔梗混合磨粉后,经过乙醇-乙酸乙酯提取、过滤和干燥的提取物B;
S3:将提取物A、提取物B、维生素A、维生素E、维生素D、维生素K和叶酸混合,磨粉即得所述组合物。
6.根据权利要求5所述的制备方法,其特征在于,S1中,所述磨粉是将混合物磨成80-100目粉。
7.根据权利要求5所述的制备方法,其特征在于,S1中,所述水提是水用量为所述S1原料总重量份的20-30倍,温度为60-80℃提取8-10小时。
8.根据权利要求5所述的制备方法,其特征在于,S2中,所述磨粉是将混合物磨成80-100目粉。
9.根据权利要求5所述的制备方法,其特征在于,所述乙醇-乙酸乙酯提取是在乙醇和乙酸乙酯体积比为1:(5-10),乙醇-乙酸乙酯总用量为所述S2原料总重量份的10-15倍,温度为40-60℃提取4-6小时。
10.一种如权利要求1-2任一项所述的组合物或权利要求3-9任一项所述的制备方法制备得到的组合物的应用,所述应用为在制备妊娠期高血压相关产品中的应用。
