本发明涉及生物医药技术领域,具体而言,涉及一种PDGFR-β基因表达抑制剂及其用途。
背景技术
血小板衍生生长因子(Platelet-derived growth factor,PDGF)是普遍存在的一种由sis原癌基因编码的生长因子,主要由巨核细胞合成,在组织损伤修复,血管生成和细胞分化过程中起到非常重要的作用。PDGF分子是一个糖蛋白二聚体,可分为-AA,-BB和-AB三种类型。PDGF及其受体(Platelet-derived growth factor receptor,PDGFR)在多种人体肿瘤中都有比较高的表达,如胰腺癌,小细胞肺癌,胃癌和乳腺癌等。
PDGFR是一种位于细胞表面的受体酪氨酸激酶(Receptor tyrosine kinase,RTK),可分为两个亚型:PDGFR-α和PDGFR-β。其中,PDGFR-β可以特异性的与PDGF-BB或-AB结合而被激活,被激活后,两个PDGFR之间可以形成二聚体,并通过自身磷酸化激活下游的信号转到通路,以实现其调控细胞分化生长的功能。
PDGFR-β基因的异常表达可以引起多种疾病的产生,包括血管疾病,纤维紊乱以及癌症等。研究发现,通过药物靶向PDGFR-β启动子中NHE区域的G-四链体,可以抑制PDGFR-β基因转录,进而有望成为PDGFR-β异常表达引起的疾病的治疗手段。
G-四链体是一种由富含鸟嘌呤(G-Rich)核酸形成的四链结构。G-四链体在基因组DNA中广泛分布,并富集于基因调节区,例如基因启动子,参与转录的调节,DNA复制和重组。经典G-四链体通常具有两个或更多个的完整G-Quartet平面和四段连续G-tract。除了经典G-四链体之外,还存在包含不完整的G-Quartet平面或部分折叠的结构,例如携带的鸟嘌呤空缺的缺口G-四链体和G-三链体等。其中,缺口G-四链体(GVBQ)是一种具有不完整的末端G-Quartet平面的G-四链体,末端G-Quartet平面存在一个鸟嘌呤空缺,该空缺位置可以被鸟嘌呤及其衍生物填充。
PDGFR-β启动子的天然富含鸟嘌呤的DNA含有七段连续的G-tract,分析表明通过组合其中的四段G-tracts可以形成不同类型的G-四链体结构,研究发现,PDGFR-β基因启动子的天然富含鸟嘌呤的DNA可以形成缺口G-四链体(简称PDGFR-β缺口G-四链体,PDGFR-βGVBQ),它可以和dGMP,GMP,cGMP等鸟嘌呤类代谢物及含鸟嘌呤的药物分子结合并稳定。然而,目前还缺乏细胞内PDGFR-β缺口G-四链体生物学效应研究,也缺乏有效的靶向PDGFR-β基因启动子中的缺口G-四链体的药物分子。
研究表明,来自RHAU蛋白的G-四链体结构结合结构域的23个氨基酸(RHAU23)与鸟嘌呤碱基偶联的化合物分子(GRPC),具备高特异性和高亲和力的识别和稳定缺口G-四链体的能力。但GRPC分子无法单独应用于细胞内研究,需要进一步修饰以解决细胞内递送的问题,并改善其在细胞中的抗降解能力。
因此,如何实现药物分子在细胞内的递送是开发靶向细胞内PDGFR-β缺口G-四链体药物的难点。
发明内容
为了解决上述问题,本发明的第一目的在于提供一种PDGFR-β基因表达抑制剂,抑制剂含有mPG分子和修饰有细胞递送载体的mPG分子中的至少一种,其中,mPG分子能够实现在细胞内的递送,特异性识别和稳定PDGFR-β缺口G-四链体,从而能够在细胞内调控PDGFR-β基因的表达,抑制PDGFR-β基因的表达水平。
本发明的第二目的在于提供上述PDGFR-β基因表达抑制剂在调控PDGFR-β基因表达中的用途。
本发明的第三目的在于提供上述PDGFR-β基因表达抑制剂在制备用于治疗PDGFR-β基因表达异常相关的疾病的药物中的用途。
本发明公开的PDGFR-β基因表达抑制剂,含有mPG分子和修饰有细胞穿透肽的mPG分子中的至少一种,实现了mPG分子在细胞内的递送,能够通过mPG分子在细胞内特异性识别和稳定PDGFR-β基因的缺口G-四链体,从而实现了在细胞内调控PDGFR-β基因的表达,抑制了PDGFR-β基因的表达水平。

PDGFR-β是细胞表面受体酪氨酸激酶,是被广泛研究的疾病治疗靶标。研究表明,靶向PDGFR-β启动子中NHE区域的G-四链体,可以PDGFR-β抑制基因转录。此外,研究还发现PDGFR-β基因启动子的天然富含鸟嘌呤的DNA可以形成缺口G-四链体,它可以和dGMP,GMP,cGMP等鸟嘌呤类代谢物及含鸟嘌呤的药物分子结合并稳定。然而,目前还缺乏细胞内PDGFR-β缺口G-四链体生物学效应研究,也缺乏有效的靶向PDGFR-β基因启动子中的缺口G-四链体的药物分子。
为了至少部分解决上述技术问题的至少一个,本发明的第一方面提供了一种PDGFR-β基因表达抑制剂,该PDGFR-β基因表达抑制剂含有mPG分子,其中,mPG分子结构为:
mPG分子是一种卟啉偶联鸟嘌呤化合物,能够单独实现在细胞内的递送,特异性靶向细胞内的PDGFR-β缺口G-四链体,从而起到抑制PDGFR-β基因转录、降低PDGFR-β基因表达水平的作用。
可以理解的是,与经典G-四链体结构相比,靶向缺口G-四链体有以下优点:首先,由于结构缺陷,缺口G-四链体通常稳定性比经典G-四链体弱,结合缺口G-四链体的化合物可以更大程度上提高其稳定性,导致DNA结构状态和其结合蛋白质的显著变化,有利于调控基因的表达水平;其次,缺口G-四链体可以通过鸟嘌呤衍生物填充和稳定。因此,靶向缺口G-四链体的鸟嘌呤衍生物可以实现特异性靶向缺口G-四链体而不作用于其他类型的经典G-四链体结构。
而本发明提供的PDGFR-β基因表达抑制剂含有mPG分子,mPG分子不仅可以特异性识别和稳定PDGFR-β缺口G-四链体,还能单独实现在细胞内的递送,特异性靶向细胞内的PDGFR-β缺口G-四链体,从而抑制PDGFR-β基因的转录,降低基因的表达水平。
一些实施方案中,mPG分子可以通过在其C端修饰细胞运送载体而负载于细胞递送载体上,增强mPG分子的细胞穿透能力,提高mPG分子在细胞内的递送效率,进一步提高mPG分子与靶标的结合率,提高mPG分子对基因表达的抑制效果。
一些具体实施方案中,上述细胞递送载体包括细胞穿透肽、脂质体、纳米颗粒和聚合物载体中的至少一种,以增强mPG分子在细胞内的递送效率。
具体地,细胞穿透肽可以为TAT,TAT的氨基酸序列为GRKKRRQRRR。
一些实施方案中,上述PDGFR-β基因表达抑制剂用于靶向细胞内PDGFR-β基因启动子区形成的缺口G-四链体,从而实现抑制PDGFR-β基因的转录,降低PDGFR-β基因的表达水平。
一些具体实施方案中,PDGFR-β基因表达抑制剂用于靶向细胞内PDGFR-β基因启动子区M-2-3-4序列形成的缺口G-四链体,M-2-3-4序列也即PDGFR-β基因启动子区第2、3和4段G-tract的序列,具体地,M-2-3-4序列为:5′-GGGGGGGCGGCGGGG-3′,从而实现抑制PDGFR-β基因的转录,降低PDGFR-β基因的表达水平。
本发明的第二方面提供了上述PDGFR-β基因表达抑制剂在调控PDGFR-β基因表达中的用途,PDGFR-β基因表达抑制剂能够通过识别并稳定细胞内PDGFR-β基因形成的缺口G-四链体,特异性抑制PDGFR-β基因的转录,以降低PDGFR-β基因的表达水平。
一些实施方案中,将本发明提供的PDGFR-β基因表达抑制剂处理细胞,能够特异性靶向细胞内PDGFR-β基因启动子区M-2-3-4序列形成的缺口G-四链体,使得PDGFR-β基因的RNA和蛋白质水平显著的下调,从而实现利用缺口G-四链体结构在PDGFR-β基因启动子中的调节作用,特异性抑制PDGFR-β基因的转录,调控PDGFR-β基因表达水平。
本发明的第三方面提供了一种调控基因表达的方法,采用上述PDGFR-β基因表达抑制剂靶向细胞内的PDGFR-β缺口G-四链体,以实现对PDGFR-β基因表达水平的调控。
一些具体实施方案中,将本发明提供的PDGFR-β基因表达抑制剂mPG分子处理U2OS细胞后,PDGFR-β基因的RNA和蛋白质水平显著的下调,与之相比,启动子中的具有经典G-四链体结构的八个基因的表达水平则没有显著变化。由此说明,本发明提供的PDGFR-β基因表达抑制剂mPG分子能够特异性靶向PDGFR-β缺口G-四链体,特异性的抑制PDGFR-β表达。
本发明的第四方面提供了上述PDGFR-β基因表达抑制剂在制备用于治疗PDGFR-β基因表达异常相关的疾病的药物中的用途,以用于治疗PDGFR-β基因表达水平异常引起的相关疾病。
一些实施方案中,PDGFR-β基因的过度表达以及随之增加的PDGFR-β信号传导是各种疾病的致病因素,因而本发明还提供了上述PDGFR-β基因表达抑制剂在制备用于治疗PDGFR-β基因过度表达相关的疾病的药物中的用途,以用于治疗基因表达水平异常引起的相关疾病。
一些实施方案中,上述PDGFR-β基因表达异常相关的疾病包括血管疾病、纤维紊乱或者癌症,通过上述PDGFR-β基因表达抑制剂识别和稳定PDGFR-β基因启动子形成的缺口G-四链体,进而抑制PDGFR-β基因的表达水平,从而用于治疗PDGFR-β基因过度表达引起的相关疾病。
下面将结合实施例对本发明的实施方案进行详细描述。
本实施例使用的材料涉及U2OS细胞,MYC、Hif1a、VEGFA、BCL2、KRAS、BRCA1、MYB、ERBB2等基因荧光定量PCR引物,MYC、CSTB、MYOG、ABTB2、LRRC42、PDGFR-β启动子区的M2-3-4序列、M2-4-5序列和M2-4-5-6序列,以及发夹DNA(HP1和HP2),单链DNA(ssDNA),富含胞嘧啶的单链DNA(TEL-C和BCL-C)等寡核苷酸序列,具体如表1和表2所示,还涉及配体分子包括mPG、mPG-TAT、OGkk、MPIX、GRPC、TMPyP4等,分子合成中涉及的化学试剂HBTU,HATU,TFA等,具体如表3所示。

其中,U2OS细胞来源于中国科学院细胞库(上海),表1和表2所示的所有寡核苷酸序列均由Sangon Biotechnology(上海,中国)合成;鸟氨酸单磷酸盐(GMP),中卟啉IX二盐酸盐(MPIX)和MESO-5,10,15,20-四甲基-4-吡啶基卟啉(TMPyP4)从Sigma购买;GRPC购自北京赛百盛基因技术有限公司;MPIX-(PEG)2-G(PNA)-D-Lys-D-Lys(简称为mPG),mPG-(L-GRKKRRQRRR)(简称为mPG-TAT),(PEG)2-G(PNA)-D-Lys-D-Lys(简称为OGkk)均由TanzhenBiotechnologies(南昌,中国)公司合成。
实施例1 mPG分子和mPG-TAT分子制备
1、mPG分子的制备
本实施例将卟啉基团偶联鸟嘌呤并构建了卟啉-鸟嘌呤偶联化合物,卟啉MPIX是典型的G-四链体特异性结合化合物,主要与G-四链体的末端平面发生相互作用,其结构如图1所示,是NMM分子的一种衍生物。使用(PEG)2接头将如图2所示含两个D型赖氨酸的肽核酸鸟嘌呤OGkk与MPIX连接,得到如图3所示的卟啉-鸟嘌呤偶联化合物(MPIX-(PEG)2-G(PNA)-D-Lys-D-Lys),简称mPG。其中,含两个D型赖氨酸的肽核酸鸟嘌呤OGkk由如图4所示的鸟嘌呤以及鸟嘌呤N末端的两个D型-赖氨酸构成,含两个D型赖氨酸的肽核酸鸟嘌呤OGkk的ESI质谱鉴定图如图5所示,高效液相色谱图如6所示,mPG的ESI质谱鉴定图如图7所示,鉴定结果与分子式相一致,高效液相色谱图如8所示。
具体地,mPG化合物按照图9所示的合成路线制备而成:
步骤一:以Rink Amide MBHA树脂为起始原料,树脂用DMF浸泡半小时让树脂溶胀,再加入Fmoc-DLys(Boc)-OH(3eq),HBTU(3eq)和DIPEA(6eq),室温下反应1.5小时,抽去DMF,然后树脂用DMF洗涤3遍,用茚三酮检测树脂透明后加入20%哌啶/DMF溶液,室温下反应半小时,再用DMF洗涤树脂5遍,茚三酮检测树脂蓝色,得到化合物1,即Cmpd1;
步骤二:以Fmoc保护的D型氨基酸赖氨酸Fmoc-Dlys(Boc)-OH为单体继续添加D型氨基酸赖氨酸,在化合物1中加入Fmoc-DLys(Boc)-OH(3eq),HBTU(3eq)和DIPEA(6eq),室温下反应1.5小时,抽去DMF,然后树脂用DMF洗涤3遍,用茚三酮检测树脂透明后加入20%哌啶/DMF溶液,室温下反应半小时,再用DMF洗涤树脂5遍,茚三酮检测树脂蓝色,得到化合物2,即Cmpd2;
步骤三:Fmoc保护的肽核酸鸟嘌呤Fmoc-PNA-G(Bhoc)-OH为单体,在化合物3基础上在树脂中加入Fmoc-PNA-G(Bhoc)-OH(3eq),HBTU(3eq)和DIPEA(6eq),室温下反应1.5小时,抽去DMF,然后树脂用DMF洗涤3遍,用茚三酮检测树脂透明后加入20%哌啶/DMF溶液,室温下反应半小时,再用DMF洗涤树脂5遍,茚三酮检测树脂蓝色,得到化合物3,即Cmpd3。
步骤四:Fmoc保护的PEG2连接头Fmoc-PEG2-OH为单体,在化合物3基础上树脂中加入Fmoc-PEG2-OH(3eq),HBTU(3eq)和DIPEA(6eq),室温下反应1.5小时,抽去DMF,然后树脂用DMF洗涤3遍,用茚三酮检测树脂透明后加入20%哌啶/DMF溶液,室温下反应半小时,再用DMF洗涤树脂5遍,茚三酮检测树脂蓝色,得到化合物4,即Cmpd4。
步骤五:在化合物4基础上,将MPIX(2eq)和HATU(2eq)加入树脂中,再加入DIPEA(6eq),DMF作溶剂反应1.5小时,然后用DMF洗涤树脂3遍,二氯甲烷洗涤2遍,甲醇洗涤2遍,抽干树脂,将TFA(每100mg树脂加入5mL TFA)加入树脂中,室温反应2小时,过滤,滤液加入到10倍滤液体积的冰乙醚中,大量固体析出,离心得到粗品mPG化合物,产物经过HPLC分离纯化,冻干后得到黑褐色粉末产物,即mPG分子。经HPLC检测分子纯度,质谱检测分析mPG化学结构。
2、mPG-TAT分子的制备
mPG-TAT分子通过在mPG分子的肽核酸C末端连接一个细胞穿透肽TAT(氨基酸序列为GRKKRRQRRR)得到,以改善分子细胞递送能力,mPG-TAT分子合成由TanzhenBiotechnologies(南昌,中国)公司合成。mPG-TAT分子ESI质谱鉴定图和分子结构式如图10所示,高效液相色谱图如图11所示。
实施例2 mPG抑制PDGFR-β基因的表达水平
本实施例使用mPG、mPG-TAT以及经典G-四链体结构的配体分子TMPyP4分别处理U2OS细胞,并通过qRT-PCR和Western免疫印迹实验检测PDGFR-β基因的表达水平。
其中,U2OS细胞培养的步骤包括:
细胞在补充有10%胎牛血清和100U/mL青霉素和链霉素的DMEM培养基中,在37℃,5%CO2条件下培育,将U2OS细胞接种在6孔板(2×105个细胞/孔)上并培养12小时,再添加终浓度为0,2,10μM的mPG或mPG-TAT孵育后48小时,从每个培养板孔中收获细胞;
qRT-PCR定量检测步骤包括:
使用RNA提取套试剂盒(天根,中国)根据制造商的说明书步骤从细胞中提取总RNA,1.0μg总RNA使用第一链cDNA合成试剂盒(全式金,中国)进行cDNA反转录,使用2×荧光定量PCR MIX(GenStar,中国)进行实时荧光定量PCR,GAPDH基因被用做内参基因,qPCR引物信息见表2,使用ABI提供的QuantStudioTM实时荧光定量PCR软件计算阈值循环数(Ct)值;
Western免疫印迹实验步骤包括:
细胞在2×SDS上样缓冲液中直接裂解,然后煮沸10分钟,通过SDS-PAGE电泳分离蛋白质并转移至PVDF膜,在室温下将膜在5%脱脂牛奶(TBST缓冲液中配制(20mM Tris-HCl,pH 7.6,150mM NaCl,0.05%(v/v)Tween 20))封闭1小时,再用TBST洗涤三次,每次5分钟,然后将膜分别与指定的抗体在室温下孵育1小时,其中,抗PDGFR-β抗体,1:2000稀释,(AB32570,Abcam),alpha tubulin抗体,1:5000稀释,(66031-1-ig,proteintech),用TBST洗涤三次,将膜分别与1:5000稀释的HRP标记抗兔IgG(H+L)抗体(5220-0336,KPL)或HRP标记抗小鼠IgG(H+L)抗体(5220-0341,KPL)在室温下孵育1小时,在三次洗涤后,使用ECL发光液(K-12045,Advansta)根据制造商的指示方法来检测蛋白质显影。
如图12A和图12B所示,根据实验结果可知,在2μM和10μM mPG或mPG-TAT存在下,PDGFR-βRNA和蛋白质水平均显著的降低,表明mPG可以较好地渗透到细胞中并有效地调节PDGFR-β基因的表达。在与mPG分子相同浓度的条件下,MPIX和TMPyP4对PDGFR-β的RNA表达几乎没有影响,相关实验结果如图12C所示。
本实施例还测试了其他基因的RNA水平,例如,MYC、Hif1a、VEGFA、BCL2、KRAS、BRCA1、MYB和ERBB2等基因,上述基因受经典G-四链体结合配体分子的调控,根据图12D的实验结果可知,mPG分子对这些基因的转录水平影响很小,也即mPG对PDGFR-β基因的影响是特异性的,对于受经典G-四链体结合配体分子调控的基因的表达水平影响较小。
实施例3 mPG分子对U2OS细胞生长的影响
本实施例还检测了mPG和mPG-TAT对U2OS细胞生长的影响。
其中,U2OS细胞培养的步骤包括:
细胞在补充有10%胎牛血清和100U/mL青霉素和链霉素的DMEM培养基中,在37℃,5%CO2条件下培育,将U2OS细胞接种在96孔板(5×103个细胞/孔)上并培养12小时,再添加终浓度为10μM的mPG或mPG-TAT孵育后24小时或48小时,同时设置添加DMSO对照实验组,再进行细胞显微镜拍照或进行MTT细胞活力检测。
MTT细胞活力检测实验步骤包括:细胞添加10μM的mPG或mPG-TAT分子处理24小时或48小时后,每孔加10μL MTT溶液,使每个孔中的MTT的终浓度为0.5mg/ml,轻轻混匀后5%CO2,37℃培养箱中孵育4小时,然后小心吸取每个孔中的培养基以防止细胞单层破裂,每孔加100μL DMSO溶解甲臜,将96孔板放在振荡器上轻轻震荡10分钟,直至在普通光学显微镜下观察发现甲臜全部溶解,在酶标免疫检测仪570nm测定吸光度,计算相对细胞活力值。
实验结果如图13A和图13B所示,图13A表示分别在10μM mPG或mPG-TAT存在下培养细胞时,拍摄细胞24小时和48小时的图像,可以观察到添加了mPG或mPG-TAT分子后细胞生长状态与对照DMSO组相比较更差。图13B表示MTT方法测定的在具有10μM mPG或mPG-TAT存在下的相对细胞活力,根据实验结果可知mPG和mPG-TAT均有抑制细胞生长效应,且mPG-TAT引起的抑制作用更为明显。
实施例4 DMS足迹实验验证mPG识别并结合PDGFR-β启动子区序列形成的缺口G-四链体
本实施例通过DMS足迹实验证明mPG分子可以与PDGFR-β缺口G-四链体(M2-3-4-T)相互作用,DMS足迹实验步骤如下:将50mM LiCl或KCl溶液中的0.1μM寡核苷酸链,即PDGFR-β启动子区的序列M2-3-4-T,在95℃下加热5分钟,以0.1℃/s的速率冷却至室温,将样品分别在37℃下与特定浓度的GMP,GRPC和mPG一起孵育30分钟,将混合物在冰上用5%DMS处理4分钟。
实验结果如图14A和图14B所示,图14B是图14A中的凝胶的数字化统计结果,实验结果表明,当添加mPG时,PDGFR-β缺口G-四链体(M2-3-4-T)的两条过切割条带(图14A中1,2箭头所示)大部分消失(1,2箭头所示的两个切割带代表了两种类型的缺口G-四链体),与添加GMP,GRPC的效果类似,而GMP和GRPC分子均为能够填补PDGFR-β缺口G-四链体的鸟嘌呤空缺,稳定PDGFR-β缺口G-四链体(M2-3-4-T)的分子,表明mPG中的鸟嘌呤碱基填补了PDGFR-β缺口G-四链体(M2-3-4-T)的鸟嘌呤空缺,能够起到稳定PDGFR-β缺口G-四链体(M2-3-4-T)的作用。
实施例5 mPG结合PDGFR-β缺口G-四链体的能力检测
本实施例通过荧光滴定实验检测标记在DNA 5'末端的FAM基团的荧光信号变化来比较mPG与不同DNA结构之间的结合能力,以及MPIX、OGkk、mPG三种不同配体与PDGFR-β缺口G-四链体的结合能力,其中,mPG与PDGFR-β缺口G-四链体的结合后,FAM基团的荧光信号会被相邻的mPG淬灭。
其中,荧光滴定实验具体步骤如下:
将5'FAM标记的DNA(0.1μM)在含有50mM KCl的TE缓冲液(pH7.4)中稀释,在95℃下加热变性5分钟,并以0.1℃/s的速率冷却至25℃,将Tween-20和BSA分别加入最终0.05%(w/v)和0.2mg/mL,将不同浓度的配体加入到样品中并在室温下孵育2小时,在玻璃毛细血管中制备样品,并在NT.115仪器中扫描以测量荧光。
在MO.Affinity Analysis V2.3软件上选择初始荧光分析模式分析,配体与特定DNA的结合由平衡解离常数(KD)描述,使用单位点结合模型和配体耗尽的模型计算数据,在平衡情况下,荧光计算的方程可以写为:
其中,F代表原始荧光信号,Fmax代表F的最大值,Fmin代表F的最小值,CA,CB(nM)分别表示DNA和配体的初始浓度。
图15为检测mPG与不同DNA结构之间的结合能力的实验结果图,结合能力通过使用单位点结合模型拟合数据来获得图中括号中的解离常数(KD)来分析;其中,图15A表示PDGFR-β缺口G-四链体(M2-3-4-T)与MPIX,OGkk,mPG的结合能力比较图;图15B表示mPG与KRAS,MYC,CSTB,M2-4-5和M2-4-5-6形成的经典G-四链体的结合能力比较图;图15C表示mPG与MYOG,ABTB2和LRRC42等序列形成的缺口G-四链体的结合能力比较图;图15D表示检测mPG和非G-四链体的DNA的结合能力比较图,其中,非G-四链体的DNA是指无法形成G-四链体的DNA,包括发夹DNA,如HP1、HP2,单链DNA,如ssDNA,富含胞嘧啶的单链DNA,如TEL-C、BCL-C等。
如图15A和图15B所示,mPG与PDGFR-β缺口G-四链体结合的解离常数为10.2nM,远低于KRAS,MYC,CSTB,M2-4-5,M2-4-5-6等序列形成的非缺口G-四链体类型的经典G-四链体,其中,KRAS,MYC,CSTB,M2-4-5,M2-4-5-6对应的序列信息如表1所示。如图15C所示,mPG与MYOG、ABTB2、LRRC42序列形成的缺口G-四链体的结合能力都较低,其中,MYOG、ABTB2、LRRC42的序列信息如表1所示;根据图15D结果可知,低于1μM浓度的mPG分子无法识非G-四链体结构DNA,低于1μΜ浓度的mPG无法识别发夹DNA(HP1,HP2)、单链DNA(ssDNA)和富含胞嘧啶的单链DNA(TEL-C,BCL-C);根据图15的结果分析可知,MPIX和鸟嘌呤基团的组合赋予mPG以高特异性和高亲和力识别PDGFR-β缺口G-四链体的能力,是单独的MPIX或(PEG)2-G(PNA)-D-Lys-D-Lys(OGkk)分别无法实现的。
实施例6 mPG稳定PDGFR-β缺口G-四链体能力检测
本实施例使用PDGFR-β缺口G-四链体的核心序列进行了基于荧光共振能量转移(FRET)的融链分析,该DNA序列在3'端标记为FAM作为供体,5'端标记为TAMRA作为受体,以确定与不同配体复合是否可以增强PDGFR-β缺口G-四链体的稳定性。mPG的两个功能单元MPIX和OGkk用作对照配体分子,PDGFR-β缺口G-四链体与不同浓度的mPG、MPIX、OGkk孵育,荧光达到最小和最大荧光之间的中间值所需的温度,定义为T1/2,即解链温度,用于判断缺口G-四链体与配体复合物结合的稳定性。
其中,荧光能量共振转移-熔解实验(FRET-Melting)具体步骤如下:
荧光供体FAM和受体TAMRA标记的寡核苷酸在0.1μM浓度在含有50mM KCl和10mM胂酸锂缓冲液(pH 7.4)中溶解,在95℃下变性5分钟,0.1℃/s速率冷却至25℃,加入指定的浓度条件的配体分子,并在37℃下孵育1小时,在25℃下初始平衡10分钟后,在QuantStudio7Flex仪器(Thermo Scientific)通过监测上的FAM的荧光信号来进行荧光能量共振转移-熔解实验。
实验结果如图16所示,图16A表示通过mPG稳定PDGFR-β缺口G-四链体(M2-3-4-F)的荧光能量共振转移-熔解实验结果图,图16B表示MPIX对PDGFR-β缺口G-四链体(M2-3-4-F)的荧光能量共振转移-熔解实验影响结果图,图16C表示OGkk对PDGFR-β缺口G-四链体(M2-3-4-F)的荧光能量共振转移-熔解实验影响结果图,图16D表示不同配体对PDGFR-β缺口G-四链体T1/2的影响实验结果图,图16E表示mPG对MYOG缺口G-四链体的荧光能量共振转移-熔解实验影响结果图,图16F表示mPG对LRRC42缺口G-四链体的荧光能量共振转移-熔解实验影响结果图,图16G表示mPG对PDGFR-β启动子的M2-4-5序列形成G-四链体的荧光能量共振转移-熔解实验影响结果图,图16G表示mPG对PDGFR-β启动子的M2-4-5-6序列形成G-四链体的荧光能量共振转移-熔解实验影响结果图。
根据图16A、图16B和图16C可知,mPG提高了PDGFR-β缺口G-四链体的解链温度,并且mPG浓度越高,PDGFR-β缺口G-四链体的解链温度越高,而MPIX的加入对PDGFR-β缺口G-四链体的解链温度没有显著影响,OGkk对提高PDGFR-β缺口G-四链体的解链温度也有一定的影响,但与mPG分子相比弱很多。具体地,根据图16D可知,在10μM mPG存在下PDGFR-β缺口G-四链体的T1/2增加了26.1℃,远大于分别在MPIX和OGkk存在下PDGFR-β缺口G-四链体的T1/2增加的程度。
根据图16E和图16F可知,mPG稳定MYOG和LRRC42形成的缺口G-四链体的能力比PDGFR-β缺口G-四链体弱很多。根据图16G和图16H可知,mPG仅在1μM浓度以上才能稳定由PDGFR-β启动子序列M2-4-5和M2-4-5-6形成的经典G-四链体。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。

7.根据权利要求6所述的用途,其特征在于,采用如权利要求1-4任一项所述的PDGFR-β基因表达抑制剂靶向细胞内的PDGFR-β基因启动子区形成的缺口G-四链体。
8.根据权利要求1-5任一项所述的PDGFR-β基因表达抑制剂在制备用于治疗PDGFR-β基因表达异常相关疾病的药物中的用途。
9.根据权利要求8所述的用途,其特征在于,所述PDGFR-β基因表达异常是指PDGFR-β基因过度表达。
10.根据权利要求9所述的用途,其特征在于,所述疾病包括血管疾病、纤维紊乱以及癌症中的至少一种。
