Abstract

本发明公开了一种外泌体负载电活性水凝胶敷料的制备方法,所述水凝胶敷料是通过下述方法制得:步骤一,制备水凝胶,步骤二,制备导电水凝胶,步骤三,骨髓间充质干细胞培养,步骤四,水凝胶敷料制备,将外泌体滴加于导电性水凝胶表面使其自由渗入水凝胶中,即得外泌体负载电活性水凝胶敷料。本发明所制备的敷料具有优良力学性能、电活性及促进神经修复性能,能够实现局部神经调节,便于治疗糖尿病患者的周围神经损伤。

一种外泌体负载电活性水凝胶敷料的制备方法
技术领域
本发明属于生物材料的技术领域,特别涉及一种外泌体负载电活性水凝胶敷料的制备方法。
背景技术
现有的导电电活性材料,如聚吡咯(PPy)、聚苯胺(PAni)、聚噻吩(PTh)等已得到广泛应用,并被用于人工神经导管的设计。
如专利申请202010181686.4公开了聚多巴胺和石墨烯或其衍生物复合聚吡咯导电神经支架的制备方法:通过电化学沉积方法,在静电纺丝聚乳酸丝膜上包覆复合有石墨烯或其衍生物的聚吡咯导电层;然后在弱碱性多巴胺溶液中浸泡1~8h,洗涤干燥后得到导电神经支架。
专利申请201910119503.3开了一种可自发电刺激的嵌段结构导电神经导管及其制备方法,该导管在导电基底上同时集成了阳极和阴极;导电基底是由基体材料复合导电成分构成;阳极是由导电基底复合葡萄糖氧化催化剂构成;阴极是由导电基底复合氧还原催化剂构成。本发明中的神经导管能够利用人体内存在的葡萄糖和氧气自发产生电刺激,促进神经生长,而不需要在人体内插入金属电极,减少了病人的痛苦和不便。此外该导管还可将电刺激集中的缺损神经处,提高电刺激的精准度和效率。
然而,这些传统的导管具有刚性结构,并且与神经组织不相容,而且具有粘性和非弹性。传统导管的这些特性通常需要进一步修剪残余神经纤维,将导管与远端缝合。手术干预包括缝线或插入导管以桥接间隙,对糖尿病神经损伤无效。这是因为糖尿病患者由于复杂的神经生理缺陷,损伤神经的再生极其缓慢(约1mm /天),并伴有严重的脱髓鞘。此外,侵入性干预和植入不匹配的植入物可能导致额外的创伤和并发症,如瘢痕、炎症增加。因此,用于治疗糖尿病神经挤压损伤的生物材料应该是柔软的、粘连的、生物相容性的,并且能够通过非侵入性程序插入。
因此,传统的聚吡咯(PPy)、聚苯胺(PAni)、聚噻吩(PTh)等人工神经导管材料具有刚性结构,并且与神经组织不相容,而且具有粘性和非弹性。传统导管这些特性通常需要进一步修剪残余神经纤维,将导管与远端缝合。此外,侵入性干预和植入不匹配的植入物可能导致额外的创伤和并发症,如瘢痕、炎症增加和进行性WD。
因此亟需一种具有优良力学性能、电活性及促进神经修复的外泌体负载生物相容导电水凝胶(ECH)敷料,以实现局部神经调节,治疗糖尿病患者的周围神经损伤。
发明内容
鉴于以上现有技术的缺点,本发明提供一种外泌体负载电活性水凝胶敷料的制备方法,该方法制备的敷料具有优良力学性能、电活性及促进神经修复性能,能够实现局部神经调节,便于治疗糖尿病患者的周围神经损伤。
本发明的另一个目的在于提供一种外泌体负载电活性水凝胶敷料的制备方法,该材料可通过NF-κ b途径调节巨噬细胞M2极化,以达到调控免疫反应效果促进成骨,并提供了更有利的骨免疫微环境,有利于骨髓间充质细胞进一步成骨分化。
基于此,本发明是这样实现的:
一种外泌体负载电活性水凝胶敷料的制备方法,其特征在于所述水凝胶敷料是通过下述方法制得:
步骤一,制备水凝胶,
将单宁酸溶解在去离子水中得到TA溶液。
将4mgTA溶于0.6ml去离子水中得到TA溶液。
步骤二,制备导电水凝胶,
在上述0.6 ml TA溶液中加入0.5 mmol吡咯,然后搅拌,得到溶液I。
0.316 g六水氯化铁粉末溶解在0.6 ml水中(4℃)制备溶液II。
溶液I和II被轻轻地混合在4℃促进凝胶,凝胶生成后,在蒸馏水(DI)中培养过夜在4℃去除任何剩余的反应试剂,得到生物相容性导电水凝胶(ECH)。
步骤三,骨髓间充质干细胞培养,
取自1-2周大雌性SD大鼠双侧肱骨和股骨冲洗骨髓腔获得骨髓间充质干细胞,获得的细胞在Dulbecco的改良的鹰培养基Dulbecco含有10%胎牛血清的培养基中培养,在37℃与5% CO2
RAW264.7细胞购自ATCC细胞库,在1640培养基中37℃和5% CO2 2.2培养。
首先,在4℃条件下,以120000 g超离心法(Optima-90 K, Beckman Coulter)离心14 h,制备出体外无外周胎牛血清,用于进一步的BMSCs培养。培养48 h后收集BMSCs培养基,然后进行多步离心,2000 g离心20 min, 10000 g离心30 min,最后100000 g离心90min分离外泌体。
步骤四,水凝胶敷料制备,将外泌体滴加于导电性水凝胶表面使其自由渗入水凝胶中,即得外泌体负载电活性水凝胶敷料。
本发明外泌体负载的ECH敷料由单宁酸(TA)、聚吡啶和外泌体组成,具有生物相容性,质地柔软,富含水分,粘连性强,具有自愈性。作为一种高度导电的薄膜,它附着在糖尿病模型损伤神经上,并自动包裹在尺寸匹配的管状结构中,与电原神经组织形成稳定而亲密的电桥,促进轴突再生和再髓鞘化。因此,外泌体负载生物相容导电水凝胶(ECH)敷料具有优良力学性能和电活性,以实现局部神经调节。
此外,外泌体可促进损伤的周围神经修复及调控免疫反应。因此,外泌体负载生物相容性导电水凝胶可多方面共同作用于损伤周围神经的修复,便于治疗糖尿病患者的周围神经损伤。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施仅用以解释本发明,并不用于限定本发明。
图1所示,本发明所实现的外泌体负载电活性水凝胶敷料的制备方法,所述水凝胶敷料是通过下述方法制得:
步骤一,制备水凝胶,
将单宁酸(TA,阿拉丁,中国上海)溶解在去离子水中得到TA溶液。
将4mgTA溶于0.6ml去离子水中得到TA溶液。
步骤二,制备导电水凝胶,
在上述0.6 ml TA溶液中加入0.5 mmol吡咯(py, 99%, Aldrich, St. Louis,USA),然后搅拌,得到溶液I。
0.316 g六水氯化铁粉末(FeCl3 6H2O, 98%,阿拉丁,上海,中国)溶解在0.6 ml水中(4℃)制备溶液II。
溶液I和II被轻轻地混合在4℃促进凝胶,凝胶生成后,在蒸馏水(DI)中培养过夜在4℃去除任何剩余的反应试剂,得到生物相容性导电水凝胶(ECH)。
步骤三,骨髓间充质干细胞培养,
取自1-2周大雌性SD大鼠双侧肱骨和股骨冲洗骨髓腔获得骨髓间充质干细胞,获得的细胞在Dulbecco的改良的鹰培养基Dulbecco (DMEM, GIBCO)含有10%胎牛血清(FBS,GIBCO)的培养基中培养,在37℃与5% CO2。
RAW264.7细胞购自ATCC细胞库,在1640培养基(GIBCO)中37℃和5% CO2 2.2培养。其中,髓间充质干细胞与材料共培养用于证明该材料有成骨效应,RAW264.7细胞同于与材料共培养用于证明该材料有调控免疫反应从而促进成骨作用。
首先,在4℃条件下,以120000 g超离心法(Optima-90 K, Beckman Coulter)离心14 h,制备出体外无外周胎牛血清,用于进一步的BMSCs培养。培养48 h后收集BMSCs培养基,然后进行多步离心,2000 g离心20 min, 10000 g离心30 min,最后100000 g离心90min分离外泌体。
步骤四,水凝胶敷料制备,将外泌体滴加于导电性水凝胶表面使其自由渗入水凝胶中,即得外泌体负载电活性水凝胶敷料。
总之,本发明针对治疗糖尿病引起的周围神经损伤的生物材料,通过大量研究,优选等原料并确定外泌体、单宁酸、吡咯、六水氯化铁(最优的配比所得外泌体负载电活性水凝胶敷料配方合理,所用原料与动物神经结合好,粘接效果佳,可以满足治疗糖尿病引起的周围神经损伤的人工材料的高弹性及高生物相容性,质地柔软,富含水分,粘连性强,可调控周围神经修复,具有自愈性要求。

Claims (4)

1.一种外泌体负载电活性水凝胶敷料的制备方法,其特征在于所述水凝胶敷料是通过下述方法制得:
步骤一,制备水凝胶,将单宁酸溶解在去离子水中得到TA溶液;
步骤二,制备导电水凝胶,
在TA溶液(中加入0.5 mmol吡咯,然后搅拌,得到溶液I;
0.316 g六水氯化铁粉末溶解在0.6 ml水中制备溶液II;
溶液I和II被轻轻地混合在4℃促进凝胶,得到生物相容性导电水凝胶(ECH);
步骤三,骨髓间充质干细胞培养,
取自1-2周大雌性SD大鼠双侧肱骨和股骨冲洗骨髓腔获得骨髓间充质干细胞,获得的细胞在Dulbecco的改良的鹰培养基Dulbecco (DMEM, GIBCO)含有10%胎牛血清(FBS,GIBCO)的培养基中培养,在37℃与5% CO2

4.如权利要求2所述的外泌体负载电活性水凝胶敷料的制备方法,其特征在于步骤二中,溶液I和II被轻轻地混合在4℃促进凝胶;凝胶生成后,在蒸馏水(DI)中培养过夜在4℃去除剩余的反应试剂。
