Abstract
本发明提供一种医用人工皮肤制备方法,涉及组织工程技术领域。该医用人工皮肤制备方法,包括表皮层、真皮层和复合生物支架,所述复合生物支架嵌入表皮层的一侧,所述真皮层使用3D打印技术附着在复合生物支架处。通过医用人工皮肤制备方法,使用复合生物支架作为表皮层与真皮层的支撑架构,在附于烧伤皮肤处后,使用微电流刺激,可以使皮肤细胞趋电增殖,加快皮肤的愈合,表皮层具有自愈性,使表皮层在受到外界微小创伤后可以自我修复,确保皮肤愈合期间可以正常使用,真皮层各组分的组成成分,可以为烧伤皮肤愈合提供营养,并且在愈合后不易留疤,随着有效成分的吸收利用,可以为皮肤生长留出空间,同时不易感染,排异现象很小。

技术领域
本发明涉及组织工程技术领域,具体为一种医用人工皮肤制备方法。
背景技术
医用人工皮肤是利用工程学和细胞生物学的原理和方法,在体外使用的皮肤替代、修复组织,在治疗烧伤、烫伤方面应用较多。现有的医用人工皮肤在附着在损伤皮肤后,主要使用药物或人体的自愈能力使细胞生长、恢复,这种方式不能使皮肤细胞按照预定的方案生长,在医用人工皮肤损伤后,会在损伤处留下难看的疤痕影响美观,同时与人体的亲和性不好,需要使用抗排异反应的药物才可。
发明内容
(一)解决的技术问题
针对现有技术的不足,本发明提供了一种医用人工皮肤制备方法,解决了现有医用人工皮肤对皮肤细胞愈合不能调控、在损伤后会在损伤处留下难看的疤痕影响美观,与人体的亲和性不好的问题。
(二)技术方案
为实现以上目的,本发明通过以下技术方案予以实现:一种医用人工皮肤制备方法,包括表皮层、真皮层和复合生物支架,所述复合生物支架嵌入表皮层的一侧,所述真皮层使用3D打印技术附着在复合生物支架处;
所述表皮层由导电水凝胶制成;
所述复合生物支架由丝素蛋白、壳聚糖和金属钛组成,所述丝素蛋白、壳聚糖和金属钛经过混合后使用3D打印技术打印成树状;
所述真皮层由以下重量份成分组成:脂肪源干细胞10~15份、胶原蛋白15~25份、牛尾Ⅰ型胶原1~5份、鼠李糖脂2~3份、层粘连蛋白1~2份、纤连蛋白1~2份、透明质酸钠8~12份、大麻素1份、抗生素0.5份,取上述脂肪源干细胞、胶原蛋白、牛尾Ⅰ型胶原、鼠李糖脂、层粘连蛋白、纤连蛋白、透明质酸钠、大麻素、抗生素加入磷酸缓冲液,之后混合均匀制得打印原材。
优选的,所述丝素蛋白、壳聚糖和金属钛的占量比例为3:3:1。
优选的,所述导电水凝胶以磺化腐植酸钠、甲基丙烯酰氧乙基三甲基氯化铵和丙烯酰胺为原料制备。
优选的,所述磷酸缓冲液为磷酸一氢钠、磷酸二氢钠中的一种与去离子水调配制得,所述打印原材经磷酸缓冲液调节后的PH为7.3~7.4。
优选的,所述真皮层在3D打印附着在复合生物支架时,复合生物支架与表皮层预喷涂0.5份的抗菌素。
优选的,所述打印原材和医用人工皮肤宜保存在0~5℃的环境中,所述医用人工皮肤背离表皮层的一侧在真皮层打印附着完成后在其表面需喷涂0.5份大麻素。
(三)有益效果
本发明提供了一种医用人工皮肤制备方法。具备以下有益效果:
1、本发明,使用复合生物支架作为表皮层与真皮层的支撑架构,在附于烧伤皮肤处后,使用微电流刺激,可以使皮肤细胞趋电增殖,加快皮肤的愈合,表皮层具有自愈性,使表皮层在受到外界微小创伤后可以自我修复,确保皮肤愈合期间可以正常使用。
2、本发明,真皮层各组分的组成成分,可以为烧伤皮肤愈合提供营养,并且在愈合后不易留疤,随着有效成分的吸收利用,可以为皮肤生长留出空间,同时不易感染,排异现象很小。

实施例:
如图1所示,本发明实施例提供一种医用人工皮肤制备方法,其特征在于:包括表皮层1、真皮层2和复合生物支架3,复合生物支架3嵌入表皮层1的一侧,真皮层2使用3D打印技术附着在复合生物支架3处;
表皮层1由导电水凝胶制成,在导电水凝胶处可以使用微电流,使微电流传导至复合生物支架3,进而利用细胞的趋电性促进细胞增殖;
复合生物支架3由丝素蛋白、壳聚糖和金属钛组成,丝素蛋白、壳聚糖和金属钛经过混合后使用3D打印技术打印成树状,复合生物支架3与人体具有好的亲和性,同时可以减缓人体的炎症;
真皮层2由以下重量份成分组成:脂肪源干细胞10~15份、胶原蛋白15~25份、牛尾Ⅰ型胶原1~5份、鼠李糖脂2~3份、层粘连蛋白1~2份、纤连蛋白1~2份、透明质酸钠8~12份、大麻素1份、抗生素0.5份,取上述脂肪源干细胞、胶原蛋白、牛尾Ⅰ型胶原、鼠李糖脂、层粘连蛋白、纤连蛋白、透明质酸钠、大麻素、抗生素加入磷酸缓冲液,之后混合均匀制得打印原材。
丝素蛋白、壳聚糖和金属钛的占量比例为3:3:1。
导电水凝胶以磺化腐植酸钠、甲基丙烯酰氧乙基三甲基氯化铵和丙烯酰胺为原料制备,具备导电性、自愈能力和自黏附。
磷酸缓冲液为磷酸一氢钠、磷酸二氢钠中的一种与去离子水调配制得,打印原材经磷酸缓冲液调节后的PH为7.3~7.4,与人体皮肤的PH基本一致。
真皮层2在3D打印附着在复合生物支架3时,复合生物支架3与表皮层1预喷涂0.5份的抗菌素,防止在真皮层2打印期间感染有害菌。
打印原材和医用人工皮肤宜保存在0~5℃的环境中,医用人工皮肤背离表皮层1的一侧在真皮层2打印附着完成后在其表面需喷涂0.5份大麻素,便于烧伤皮肤的愈合。
尽管已经示出和描述了本发明的实施例,对于本领域的普通技术人员而言,可以理解在不脱离本发明的原理和精神的情况下可以对这些实施例进行多种变化、修改、替换和变型,本发明的范围由所附权利要求及其等同物限定。

1.一种医用人工皮肤制备方法,其特征在于:包括表皮层(1)、真皮层(2)和复合生物支架(3),所述复合生物支架(3)嵌入表皮层(1)的一侧,所述真皮层(2)使用3D打印技术附着在复合生物支架(3)处;
所述表皮层(1)由导电水凝胶制成;
所述复合生物支架(3)由丝素蛋白、壳聚糖和金属钛组成,所述丝素蛋白、壳聚糖和金属钛经过混合后使用3D打印技术打印成树状;
所述真皮层(2)由以下重量份成分组成:脂肪源干细胞10~15份、胶原蛋白15~25份、牛尾Ⅰ型胶原1~5份、鼠李糖脂2~3份、层粘连蛋白1~2份、纤连蛋白1~2份、透明质酸钠8~12份、大麻素1份、抗生素0.5份,取上述脂肪源干细胞、胶原蛋白、牛尾Ⅰ型胶原、鼠李糖脂、层粘连蛋白、纤连蛋白、透明质酸钠、大麻素、抗生素加入磷酸缓冲液,之后混合均匀制得打印原材。
2.根据权利要求1所述的一种医用人工皮肤制备方法,其特征在于:所述丝素蛋白、壳聚糖和金属钛的占量比例为3:3:1。
3.根据权利要求1所述的一种医用人工皮肤制备方法,其特征在于:所述导电水凝胶以磺化腐植酸钠、甲基丙烯酰氧乙基三甲基氯化铵和丙烯酰胺为原料制备。
4.根据权利要求1所述的一种医用人工皮肤制备方法,其特征在于:所述磷酸缓冲液为磷酸一氢钠、磷酸二氢钠中的一种与去离子水调配制得,所述打印原材经磷酸缓冲液调节后的PH为7.3~7.4。
5.根据权利要求1所述的一种医用人工皮肤制备方法,其特征在于:所述真皮层(2)在3D打印附着在复合生物支架(3)时,复合生物支架(3)与表皮层(1)预喷涂0.5份的抗菌素。
6.根据权利要求1所述的一种医用人工皮肤制备方法,其特征在于:所述打印原材和医用人工皮肤宜保存在0~5℃的环境中,所述医用人工皮肤背离表皮层(1)的一侧在真皮层(2)打印附着完成后在其表面需喷涂0.5份大麻素。
